DNA breaks are inevitable as they mainly occur due to cells’ own reactive oxygen species (ROS). While DNA breaks can be single-stranded or double-stranded, the double-stranded DNA (dsDNA) breaks are more dangerous. If such damage is not repaired, it can lead to genetic instability and serious health issues including cancers. One way dsDNA breaks can be repaired is via a process called homologous recombination (HR), which involves several DNA-binding proteins. The inhalation of toxic inhaled agents (TIAs) such as methyl isocyanate and cyanide can compromise an individual’s general health due to their extremely toxic nature. Nonetheless, these gases are used extensively in the electronics, chemical, welding, electroplating industries, etc., increasing the risk of exposure to both civilians and industrial workers. It is therefore very crucial to develop analytical methods for TIA metabolites and therapeutic agents. Several Bacillus amyloliquefaciens isolates obtained from wheat residue have been studied as biological agents to control wheat diseases. Notable traits of isolates 1BA and 1D3 include growth with high salt (10% NaCl), at temperatures up to 50°C and over a wide pH range. BIOLOG Gen III plates were used to study and further characterize Bacillus amyloliquefaciens 1BA and 1D3. Both isolates showed tolerance to high salt concentration supporting previous studies. They also grew in the presence of lithium chloride, potassium tellurite, and sodium bromate. Both isolates grew at pH 5, with almost identical carbon source utilization fingerprints. This thesis describes a chemical method for alternative oligonucleotide purification that is non-chromatographic and gel-free and allows to routinely synthesize and purify long functional RNA strands. The purification of long RNAs is based on the bio-orthogonal inverse electron demand Diels-Alder (IEDDA) chemistry between trans-cyclooctene (TCO) and tetrazine (Tz). Target oligonucleotide strands are selectively tagged with Tz and can be captured and purified from the failure sequences with immobilized TCO. RNA strands are synthesized on solid support through a photolabile linker to avoid the loss of Tz tag. Purity of the isolated oligonucleotides was evaluated using gel electrophoresis, HPLC and mass …

Cataract, or clouding of the ocular lens, among the most common types of eye diseases, is the leading cause of blindness worldwide. With the opacity or clouding of the lens, light incident on the lens is scattered rather than being transmitted and is thus prevented from focusing on the retina. การพัฒนาพาสต้าปราศจากกลูเตนที่มีเส้นใยอาหารสูงจากฟลาวร์มันสำปะหลัง, พลิตา อุดมสิรินิกร

งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาอิทธิพลของฟลาวร์ข้าวเจ้า สตาร์ชข้าวโพด เส้นใยมันสำปะหลัง และกัวกัม ต่อสมบัติทางกายภาพและสมบัติทางประสาทสัมผัสของพาสต้าปราศจากกลูเตนที่มีเส้นใยอาหารสูงจากฟลาวร์มันสำปะหลัง และเพื่อพัฒนาพาสต้าปราศจากกลูเตนที่มีเส้นใยอาหารสูงจากฟลาวร์มันสำปะหลังที่ขึ้นรูปด้วยกระบวนการอัดรีด จากการศึกษาสมบัติทางเคมีกายภาพของสตาร์ชและฟลาวร์ พบว่า สตาร์ชมันสำปะหลัง สตาร์ชข้าวเจ้า และสตาร์ชข้าวโพด มีปริมาณแอมิโลสร้อยละ 16.31, 21.35 และ 23.15 ตามลำดับ สตาร์ชมันสำปะหลังมีกำลังการพองตัวและการละลายสูงกว่าสตาร์ชข้าวโพดและสตาร์ชข้าวเจ้า จากการวิเคราะห์องค์ประกอบทางเคมีของฟลาวร์มันสำปะหลังและเส้นใยมันสำปะหลัง พบว่า มีปริมาณเส้นใยอาหารร้อยละ 7.42 และ 54.75 โดยน้ำหนักแห้ง ตามลำดับ จึงคำนวณสูตรและนำเส้นใยมันสำปะหลังมาเสริมแหล่งของเส้นใยอาหาร และนำฟลาวร์ข้าวเจ้าและสตาร์ชข้าวโพดมาทดแทนฟลาวร์มันสำปะหลังที่สัดส่วนต่าง ๆ จากนั้นศึกษาคุณภาพพาสต้าที่ขึ้นรูปด้วยวิธีการรีดและตัด พบว่า พาสต้าสูตร RF50 ที่มีส่วนผสมจากฟลาวร์มันสำปะหลัง ฟลาวร์ข้าวเจ้า สตาร์ชข้าวโพด เส้นใยมันสำปะหลัง ในปริมาณร้อยละ 35 : 50 : 0 : 15 มีคุณภาพหลังปรุงสุกและลักษณะเนื้อสัมผัสของพาสต้าดีกว่าพาสต้าสูตรอื่น (p≤0.05) จากนั้นนำกัวกัมมาเสริมในพาสต้าสูตร RF50 ที่ร้อยละ 1 และ 2 โดยน้ำหนักฟลาวร์ผสม พบว่า พาสต้าสูตร RF50GG2 ที่มีส่วนผสมจากฟลาวร์มันสำปะหลัง ฟลาวร์ข้าวเจ้า เส้นใยมันสำปะหลัง ในปริมาณร้อยละ 35 : 50 : 15 และเสริมกัวกัมร้อยละ 2 โดยน้ำหนัก ฟลาวร์ผสม มีคุณภาพหลังปรุงสุกและลักษณะเนื้อสัมผัสของพาสต้า ไม่แตกต่างจากพาสต้ามันสำปะหลังทางการค้า (p>0.05) และได้คะแนนการทดสอบทางประสาทสัมผัสทางด้านเนื้อสัมผัสและความชอบโดยรวม มากกว่าพาสต้าสูตรอื่น (p≤0.05) ในกระบวนการอัดรีดด้วยสกรูเดี่ยว กำหนดสภาวะการตั้งค่าพารามิเตอร์ในส่วนของอัตราการป้อนวัตถุดิบ 70 กิโลกรัมต่อชั่วโมง, อุณหภูมิน้ำ 70 องศาเซลเซียส, อุณหภูมิบาเรลส่วนนวดผสม 65 องศาเซลเซียส, ความเร็วรอบสกรูส่วนนวดผสม 70 rpm, อุณหภูมิบาเรลส่วนอัดรีด 45 องศาเซลเซียส, ความเร็วรอบสกรูส่วนอัดรีด 25 rpm, อุณหภูมิหัวได 55 องศาเซลเซียส และแปรปริมาณน้ำที่ …

ปัจจุบันการแพร่ระบาดของเชื้อก่อโรคมีความรุนแรงเพิ่มมากขึ้น ทำให้ผู้บริโภคให้ความสนใจเกี่ยวกับสุขลักษณะที่ดี นำมาสู่ความสนใจในเทคโนโลยีกำจัดจุลินทรีย์ที่อยู่ในสภาพแวดล้อมในอุปกรณ์เครื่องมือชีวิตประจำวัน ได้แก่ เครื่องปรับอากาศ และเครื่องฟอกอากาศ ซึ่งปัจจุบันบริษัท บิทไว้ส์ (ประเทศไทย) จำกัด ผู้ผลิตเครื่องปรับอากาศได้มีการพัฒนาเทคโนโลยี 2 ชนิด ได้แก่ negative ion generator (NIG) และพลาสมาชนิด dielectric barrier discharge (DBD plasma) เพื่อตอบโจทย์ผู้บริโภคดังกล่าว งานวิจัยนี้มีวัตถุประสงค์เพื่อหาภาวะที่เหมาะสมในการทดสอบประสิทธิภาพในการกำจัดจุลินทรีย์ ด้วยเทคโนโลยีพลาสมา และทดสอบประสิทธิภาพการกำจัดจุลินทรีย์สำคัญในกลุ่มแบคทีเรีย รา และไวรัส ด้วยเทคโนโลยีพลาสมา พร้อมทั้งทดสอบผลของเทคโนโลยีพลาสมาต่อเซลล์มนุษย์ โดยสร้างกล่องทดลองอะคริลิค ที่ติดตั้ง NIG กำลังไฟ 1 วัตต์ และสร้าง กล่องทดลองอะคริลิค ที่ติดตั้งพลาสมาชนิด DBD กำลังไฟ 15 วัตต์ เพื่อใช้ในการทดสอบ ซึ่งในการทดสอบแบคทีเรียนั้นใช้แบคทีเรียแกรมบวก แบคทีเรียแกรมลบ และแบคทีเรียที่ทนต่อสารปฏิชีวนะ ได้แก่ Escherichia coli ATCC25922, Staphylococcus aureus ATCC6538P และ Pseudomonas aeruginosa ATCC9027 ตามลำดับ โดยเลี้ยงและนำมากระจายเชื้อบนอาหารแข็งชนิด LB จากนั้นนำมาสัมผัส NIG และพลาสมาชนิด DBD เป็นเวลา 60 นาที พบว่าพลาสมาชนิด DBD สามารถกำจัดแบคทีเรียทั้ง 3 ชนิดได้ 100% แต่ NIG ไม่มีประสิทธิภาพในการกำจัดแบคทีเรียดังกล่าว ในการทดสอบผลต่อยีสต์โดยเลือก Saccharomyces cerevisiae ATCC4105 เป็นตัวแทนเนื่องจากโครงสร้างใกล้เคียงยีสต์ที่ก่อโรคในมนุษย์ โดยทดสอบนำยีสต์มากระจายเชื้อบนอาหารแข็งชนิด YPD และสัมผัส NIG และพลาสมาชนิด DBD เป็นเวลา 60 นาที พบว่าพลาสมาชนิด DBD มีประสิทธิภาพในการฆ่ายีสต์ 100% และ NIG ไม่มีประสิทธิภาพในการฆ่ายีสต์ …

Notch signaling is an evolutionarily conserved signaling pathway involved in cell fate decisions and cell renewal of multiple systems, including skeletal and muscle homeostasis. As the multifunctional nuclear complex of Notch1 is essential for its pathological function, identifying these interacting partners represents a potentially broad range of druggable vulnerabilities. We have completed a BioID proteomics study and found a large set of nuclear proteins associated with NOTCH1, including transcriptional regulators and epigenetic modulators, of which Histone Deacetylase 1 (HDAC1) is a promising interacting partner of Notch1. วัตถุประสงค์ของการวิจัยครั้งนี้ 1. ศึกษาลักษณะตลาดเครื่องปรับอากาศ แนวโน้มการตลาด และลักษณะของตลาดผลิตภัณฑ์ เครื่องควบแน่นแบบระเหยสำหรับระบบปรับอากาศ (Evaporative Condensing Unit) ที่บริษัทต้องการพัฒนา 2. ศึกษาแนวทางการพัฒนาตลาดสำหรับผลิตภัณฑ์ใหม่ในสองรูปแบบ คือ แบบติดตั้งเพิ่มเติม (Retrofitting) และแบบที่ติดตั้งสำเร็จ (Built-in) โดยเก็บข้อมูลจากการสัมภาษณ์กลุ่มตัวอย่างผู้จัดการดูแลงานโครงการภาคใต้ และผู้เชี่ยวชาญด้านการติดตั้งซ่อมบำรุงระบบเครื่องปรับอากาศ 10 คน, บุคคลทั่วไป 10 คน และเจ้าของสถานประกอบการ 4 คน ร่วมกับการเก็บข้อมูลโดยใช้แบบสอบถาม 2 ชุด ชุดแรกสำหรับผู้จัดการดูแลงานโครงการภาคใต้ และผู้เชี่ยวชาญด้านการติดตั้งซ่อมบำรุงระบบเครื่องปรับอากาศ 10 คน ชุดที่ 2 สำหรับกลุ่มตัวอย่างผู้ที่เคยซื้อ หรือกำลังตัดสินใจซื้อผลิตภัณฑ์ใหม่ 400 คน โดยใช้วิธีแจกแบบสอบถามออนไลน์ในการเก็บข้อมูล ซึ่งใช้สถิติพรรณนา และสถิติอนุมาน ในการวิเคราะห์รายงานผล ผลการวิจัยพบว่า ปัจจุบันมีการใช้เครื่องปรับอากาศที่มีเครื่องควบแน่นแบบระเหย เพียงร้อยละ 0.8 โดยผู้บริโภคส่วนใหญ่มีพฤติกรรมค้นหาข้อมูลของผลิตภัณฑ์ประเภทเดียวกันจากหลายยี่ห้อ จากผู้จัดจำหน่ายหลายแห่งก่อนการตัดสินใจซื้อ ซึ่งส่วนใหญ่สนใจจะซื้อผลิตภัณฑ์ใหม่แบบที่ติดตั้งสำเร็จ (Built-in) และแบบติดตั้งเพิ่มเติม (Retrofitting) ช่วง 6-10 ปี ข้างหน้าโดยผู้บริโภคจะยอมจ่ายเพื่อซื้อเครื่องปรับอากาศที่มีเครื่องควบแน่นแบบระเหยแบบที่ติดตั้งสำเร็จ (Built-in) ขนาด 25,000 btu/h ประเภทธรรมดา (Fix speed) ในช่วงราคา 30,252-31,421 บาท สำหรับเครื่องปรับอากาศที่มีเครื่องควบแน่นแบบระเหยแบบที่ติดตั้งสำเร็จ (Built-in) ขนาด 36,000 btu/h ประเภทธรรมดา (Fix speed) ในช่วงราคา 47,369-52,020 บาท สำหรับผู้บริโภคกลุ่มเจ้าของกิจการจะยอมจ่ายเพื่อซื้อเครื่องปรับอากาศที่มีเครื่องควบแน่นแบบระเหยแบบที่ติดตั้งสำเร็จ (Built-in) ขนาด 25,000 btu/h ประเภทธรรมดา (Fix speed) ไม่เกิน 37,828 บาท สำหรับเครื่องปรับอากาศที่มีเครื่องควบแน่นแบบระเหยแบบที่ติดตั้งสำเร็จ (Built-in) ขนาด 36,000 btu/h ประเภทธรรมดา (Fix speed) ไม่เกิน 66,587.81 …

หน่วยควบแน่นแบบระเหย (Evaporative condenser unit) คือการอาศัยหลักการระเหยของน้ำมาช่วยเพิ่มความสามารถในการระบายความร้อนของเครื่องปรับอากาศ งานวิจัยนี้มีจุดประสงค์เพื่อพัฒนาหน่วยควบแน่นแบบระเหยและศึกษาปัจจัยที่มีผลต่อการเพิ่มประสิทธิภาพของเครื่องปรับอากาศ ได้แก่ ความชื้นสัมพัทธ์โดยรอบ (Ambient relative humidity) และอัตราการไหลของน้ำ (Water flow rate) โดยออกแบบคอยล์เสริมเพื่อเป็นบริเวณที่ใช้ในการฉีดพ่นน้ำ พบว่าการเชื่อมต่อคอยล์เสริมบริเวณดิสชาร์จสูงกว่าบริเวณลิควิด 3.5% การทดลองเพื่อศึกษาเปลี่ยนแปลงค่าความชื้นสัมพัทธ์ควบคุมโดยปรับค่าความเร็วอากาศพบว่าเมื่อความเร็วอากาศลดลงทำให้ความชื้นสัมพัทธ์เพิ่มขึ้น และความสามารถในการระบายความร้อนมีค่าสูงสุดในช่วงความชื้นสัมพัทธ์ระหว่าง 73.1% ถึง 80.9% การทดลองศึกษาการเปลี่ยนแปลงอัตราการไหลของน้ำพบว่าพื้นที่สัมผัสบริเวณผิวคอยล์เสริมมากจะสามารถลดอุณหภูมิของสารทำความเย็นได้มากเช่นกัน นอกจากนี้ขนาดของหัวฉีดพ่นและความชื้นสัมพัทธ์ยังส่งผลต่อการลดลงของอุณหภูมิสารทำความเย็นอีกด้วย ยิ่งไปกว่านั้นในการทดลองเพื่อหาประสิทธิภาพการทำงานของต้นแบบหน่วยควบแน่นแบบระเหยพบว่าสามารถเพิ่มประสิทธิภาพจากเครื่องปรับอากาศที่ระบายความร้อนด้วยลมปกติได้สูงถึง 20.36%

Prostate apoptosis response-4 (Par-4) is a proapoptotic tumor suppressor protein that has been linked to a large number of cancers. This 38 kilodalton (kDa) protein has been shown to be predominantly intrinsically disordered in vitro. In vivo, Par-4 is cleaved by caspase-3 at Asp-131 to generate the 25 kDa functionally active cleaved Par-4 protein (cl-Par-4) that inhibits NF-κB-mediated cell survival pathways and causes selective apoptosis in tumor cells. Pyrogenic dissolved organic matter (pyDOM) is known to be an important biogeochemical constituent of aquatic ecosystems and the carbon cycle. While recent studies have examined how pyDOM production, composition, and photolability varies with parent pyrogenic solid material type, we lack an understanding of potential microbial mineralization and transformation of pyDOM in the biogeosphere. Thus, leachates of oak, charred at 400 °C and 650 °C, as well as their photodegraded counterparts were incubated with a soil‐extracted microbial consortium over 96 days. Studies dealing with plants’ mineral nutrient status under elevated atmospheric CO2concentration (eCO2) are usually conducted in closed hydroponic systems, in which nutrient solutions are entirely renewed every several days. Here, we investigated the contribution of the fluctuation of concentrations of N ([N]), P ([P]), and K ([K]) in nutrient solutions in this short period on their concentrations in cucumber plants exposed to different [CO2] and N levels.

The aggregation of the 37-amino acid polypeptide human islet amyloid polypeptide (hIAPP), as either insoluble amyloid or as small oligomers, appears to play a direct role in the death of human pancreatic β-islet cells in type 2 diabetes. hIAPP is considered to be one of the most amyloidogenic proteins known. The quick aggregation of hIAPP leads to the formation of toxic species, such as oligomers and fibers, that damage mammalian cells (both human and rat pancreatic cells). In recent years, many medically promising antibiotics have been discovered in nature, especially in insect-microbe symbioses. One of the better-studied examples of this kind of defensive relationship is that of fungus-growing ants and the antibiotic-producing Actinobacteria. These bacteria produce several defensive chemicals with myriad uses, including one antibiotic that inhibits the growth of several bacterial strains, including other Actinobacteria. According to the Centers of Disease Control and Prevention, antibiotics decrease in effectiveness as bacteria gain resistance for previously treatable illnesses. Currently, antibiotic susceptibility is typically carried out via the Kirby-Bauer method. Even with automation, this process requires two incubation periods so a less time-consuming technique is desirable. Single entity electrochemistry (SEE) detects changes in current when collisions of individual particles at an ultramicroelectrode (UME) are linked with an electrochemical event. Macrolide antibiotics are in high demand for clinical applications. Macrolides are biosynthesized via giant assembly line polyketide synthases (PKS) which are arranged in a modular fashion. Combinatorial biosynthetic methods have been used to produce diversified macrolides by reprograming these modules and modifying tailoring enzymes required for post synthetic modifications. However it is challenging due to the size and complexity of PKSs. Microwave-assisted deep eutectic solvent and gamma-valerolactone metallic chloride catalyzed conversion of xylan to furfural were investigated using a 2x3 factorial experimental design at two levels of percent microwave power, reaction time, and catalyst concentration. The levels of each factor studied were (20%, 60% microwave power, 2, 4 minutes, and 10, 20mg) respectively. The effect of three metallic chloride catalysts (LiCl, FeCl3.6H20, CuCl2) on the conversion of xylan to furfural was also investigated. The primary objective of my thesis was to develop and utilize a biocompatible multifunctional trityl spin probe for concurrent measurement of pO2, pHe, and [Pi] in vivo by electron paramagnetic resonance (EPR) spectroscopy (Chapter 2). My first goal was to synthesize the proposed probe we are terming HOPE71. Secondly, HOPE71 was characterized by X-band and L-band EPR spectroscopy. Next, the biocompatibility of HOPE71 was assessed through an albumin binding test, cytotoxicity assays, and in vivo intravenous tolerance. Huntington’s Disease (HD) is a genetic neurodegenerative disease caused by the expansion of polyglutamine (polyQ) domain within the first exon (exon1) of the huntingtin (htt) protein. Due to this mutation within the polyQ domain, htt aggregates into various toxic species such as oligomers, fibrils, and other amorphous aggregates. While the aggregation of htt strongly correlates with polyQ length, other factors, e.g. interaction with membranes or organelles and posttranslational modifications (PTMs), modulate aggregation. The first 17 N-terminal amino acids (Nt17) that precede the polyQ in htt-exon1 enhances aggregation and facilitated binding of htt to membranous organelles, promoting morphological changes and disfunction. …
