Biochemistry and Microbiology
Other than genetic regulation of salt sensitivity of blood pressure, many factors have been shown to regulate renal sodium handling which contributes to long-term blood pressure regulation and have been extensively reviewed. Here we present our progress on the Na/K-ATPase signaling mediated sodium reabsorption in renal proximal tubules, from cardiotonic steroids-mediated to reactive oxygen species (ROS)-mediated Na/K-ATPase signaling that contributes to experimental salt sensitivity.

Non-specific quantitation of [14C]sucrose in blood and brain has been routinely used as a quantitative measure of the in vivo blood-brain barrier (BBB) integrity. However, the reported apparent brain uptake clearance (Kin) of the marker varies widely (∼100 fold). We investigated the accuracy of the use of the marker in comparison with a stable isotope of sucrose ([13C]sucrose) measured by a specific LC-MS/MS method. Rats received single doses of each marker, and the Kin values were determined. Surprisingly, the Kin value of [13C]sucrose was 6-7 fold lower than that …

The present invention provides, among other things, oligonucleotide modulators of human 5'-HT2C receptor (HTR2C) and improved methods and composition for treating HTR2C-related diseases, disorders or conditions based on such modulators. In particular, oligonucleotides modulators according to the invention target specific regions in the Exon V/Intron V junction of the human HTR2C pre-mRNA and drive expression of HTR2C Vb splice isoform, leading to increased generation of non-edited strong HTR2C receptor and enhanced serotonin receptor activity.

Tailed bacteriophages and herpesviruses assemble infectious particles via an empty precursor capsid (or 'procapsid') built by multiple copies of coat and scaffolding protein and by one dodecameric portal protein. Genome packaging triggers rearrangement of the coat protein and release of scaffolding protein, resulting in dramatic procapsid lattice expansion. Here, we provide structural evidence that the portal protein of the bacteriophage P22 exists in two distinct dodecameric conformations: an asymmetric assembly in the procapsid (PC-portal) that is competent for high affinity binding to the large terminase packaging protein, and a symmetric ring in the mature virion (MV-portal) that has negligible affinity …

Apoptosis is a genetically regulated cell suicide programme mediated by activation of the effector caspases 3, 6 and 7. If apoptotic cells are not scavenged, they progress to a lytic and inflammatory phase called secondary necrosis. The mechanism by which this occurs is unknown. Here we show that caspase-3 cleaves the GSDMD-related protein DFNA5 after Asp270 to generate a necrotic DFNA5-N fragment that targets the plasma membrane to induce secondary necrosis/pyroptosis. Cells that express DFNA5 progress to secondary necrosis, when stimulated with apoptotic triggers such as etoposide or vesicular stomatitis virus infection, but disassemble into small apoptotic bodies when DFNA5 …

Oxidative stress, as a consequence of the elevated reactive oxygen species (ROS), has been implicated in various chronic diseases including cancers by, for examples, causing oxidative DNA damage and altering epigenetic regulation. Long interspersed element-1 (LINE-1) is an only active retrotransposable elements in human DNA, comprising up to 17% of the whole genome. LINE-1 hypomethylation is reported in bladder cancer and evidently it is a consequence of oxidative stress. In this study, we investigated the expression of 5-methylcytosine (5-mC), 8-hydroxydeoxyguanosine (8-OHdG), LINE-1-endoced protein (ORF1p), histone H3K9me3 and HP1 alpha in bladder cancer tissues. Hormone replacement therapy (HRT), used by many women to alleviate menopausal symptoms such as hot flashes and mood swings, is often a combination of horomones such as estrogens, progesterone, and conjugated equine estrogens (CEE), extracted from the urine of pregnant mares. Previous studies have found positive correlations between estradiol and cellular protection, but recent research has concluded CEE provide less protective mechanisms as compared to endogenous hormones. Our previous study indicated that overexpression of nicotinamide phosphoribosyltransferase (NAMPT) aggravated acute lung injury, while knockdown of NAMPT expression attenuated ventilator-induced lung injury. Recently, we found that meta-carborane-butyl-3-(3-pyridinyl)-2E-propenamide (MC-PPEA, MC4), in which the benzoylpiperidine moiety of FK866 has been replaced by a carborane, displayed a 100-fold increase in NAMPT inhibition over FK866. Here, we determined the effects of MC4 and FK866 on cecal ligation and puncture (CLP) surgery-induced sepsis in C57BL/6J mice. Preeclampsia is a hypertensive disease of pregnant women that is known to cause detrimental physiological complications to both the mother and fetus. The hypertension hypothesized to result from endothelial dysfunction may be improved therapeutically by increasing Vascular Endothelial Growth Factor ligand concentration and it’s Vascular Endothelial Growth Factor Receptor 2 (VEGF and VEGFR2). Together, this receptor pathway may help overall vasodilation of key blood vessels linking the mother and fetal placental unit, allowing for increased clinical pathologies of both. Microabubiminuria is a condition in which small amounts of albumin are excreted in the urine without overt nephropathy. Based on the
American Diabetic Association classification, microalbuminuria is defined as urinary excretion of albumin of 30–300 mg per 24 hours or 20–200 mcg/min or 30–300 mcg/mg creatinine. Various conditions such as diabetes, hypertension, and impaired renal tubular reabsorption can lead to microalbuminuria. Microalbuminuria is an indicator and predictor of the development of overt nephropathy. If identified and treated early, overt nephropathy can be prevented and averted. In December 2016, a panel of experts in microbiology, nutrition and clinical research was convened by the International Scientific Association for Probiotics and Prebiotics to review the definition and scope of prebiotics. Consistent with the original embodiment of prebiotics, but aware of the latest scientific and clinical developments, the panel updated the definition of a prebiotic: a substrate that is selectively utilized by host microorganisms conferring a health benefit. This definition expands the concept of prebiotics to possibly include non-carbohydrate substances, applications to body sites other than the gastrointestinal tract, and diverse categories other than food. Neuropilins (NRPs) are non-tyrosine kinase cell surface glycoproteins expressed in all vertebrates and widely conserved across species. The two isoforms, such as neuropilin-1 (NRP1) and neuropilin-2 (NRP2), mainly act as coreceptors for class III Semaphorins and for members of the vascular endothelial growth factor family of molecules and are widely known for their role in a wide array of physiological processes, such as cardiovascular, neuronal development and patterning, angiogenesis, lymphangiogenesis, as well as various clinical disorders. Poly (ADP-Ribose) Polymerase I (PARP-1) is a first responder to DNA damage and participates in the regulation of gene expression. The interaction of PARP-1 with chromatin and DNA is complex and involves at least two different modes of interaction. In its enzymatically inactive state, PARP-1 binds native chromatin with similar affinity as it binds free DNA ends. Automodification of PARP-1 affects interaction with chromatin and DNA to different extents. Here we describe a series of biochemical and biophysical techniques to quantify and dissect the different binding modes of PARP-1 with its various substrates. Metalloproteins requiring one or more metal ions for normal function make up 30% of all known proteins, and many critical biological pathways contain at least one metallo-enzyme. Di-nuclear metallo-proteins constitute a large class of these proteins yet we currently lack effective methods of inhibiting these enzymes for the development of new medical therapies, particularly for the discovery of new antibiotics. Our work has focused on developing novel functionalities that selectively interact with di-nuclear catalytic centers, and we are targeting three separate di-zinc-metallo-enzymes that are unique to bacteria and play key roles in their growth and development. โรคมะเร็งตับ (HCC) เป็นโรคที่มีอุบัติการณ์สูงเมื่อเทียบกับโรคมะเร็งชนิดอื่น ในประเทศแถบเอเชียมักพบผู้ป่วยโรคมะเร็งตับซึ่งมีสาเหตุมาจากการติดเชื้อไวรัสตับอักเสบบี จากการศึกษาก่อนหน้านี้พบว่าเอนไซม์ Sulfatase-2 (SULF2) มีการแสดงออกเพิ่มสูงขึ้นในบริเวณเนื้อเยื่อมะเร็งตับ เอนไซม์ชนิดนี้สามารถกระตุ้นการเจริญของเซลล์มะเร็งตับได้ผ่านกระบวนการ desulfation ภายในโมเลกุล Glypican-3 (GPC3) ซึ่งทำหน้าที่เป็นแหล่งกักเก็บของลิแกนด์ชนิด Wnt และมีการรายงานว่า GPC3 มีการแสดงออกเพิ่มสูงขึ้นในบริเวณเนื้อเยื่อมะเร็งตับเช่นเดียวกับ SULF2 การศึกษานี้จึงมีวัตถุประสงค์เพื่อประเมินบทบาทในการวินิจฉัยโรคมะเร็งตับของ SULF2 และ GPC3 ในผู้ป่วยโรคมะเร็งตับจากการติดเชื้อไวรัสตับอักเสบบี ผลการวิเคราะห์การแสดงออกที่เปลี่ยนไปในระดับ mRNA ของ SULF2 และ GPC3 ด้วยเทคนิค Real-time PCR (RT-PCR) พบว่าในตัวอย่างเลือดของผู้ป่วยโรคมะเร็งตับ (N=93) มีระดับ mRNA ของทั้งสองยีนสูงที่สุดเมื่อเทียบกับกลุ่มผู้ป่วยโรคติดเชื้อไวรัสตับอักเสบบี (N=98) และกลุ่มผู้มีสุขภาพดี (N=50) (SULF2: 25.26± 38.10 และ 8.74±17.30 และ 5.33±12.78, P<0.001; GPC3: 32.47±35.38 และ 16.27±28.30 และ 3.67 ±4.45, P<0.001) ผลการวิเคราะห์ระดับโปรตีนในซีรั่มด้วยเทคนิค Enzyme-linked immunosorbent assay (ELISA) พบปริมาณ SULF2 สูงสุดในผู้ป่วยโรคมะเร็งตับ (N=146) เมื่อเทียบกับกลุ่มผู้ป่วยโรคติดเชื้อไวรัสตับอักเสบบี (N=119) และกลุ่มผู้มีสุขภาพดี (N = 50) เช่นกัน (27.51±10.17 และ 18.56±5.21 และ 15.81±4.30 ng/ml, P<0.001) ขณะที่สามารถตรวจพบ GPC3 ได้มากที่สุดในกลุ่มผู้ป่วยโรคมะเร็งตับ เมื่อเปรียบเทียบความสามารถในการเป็นตัวบ่งชี้ทางชีวภาพพบว่าระดับ SULF2 และ GPC3 ในซีรั่มเพียงชนิดเดียวมีประสิทธิภาพด้อยกว่า AFP แต่เมื่อนำ AFP ร่วมกับ SULF2 มาพิจารณาร่วมด้วยพบว่า AFP ร่วมกับ SULF2 มีประสิทธิภาพในการจำแนกผู้ป่วยมะเร็งตับจากผู้ไม่ได้เป็นมะเร็งตับได้ดีที่สุด นอกจากนี้ระดับของ SULF2 และ GPC3 ภายในซีรั่มสัมพันธ์กับระยะเวลาการรอดชีวิตของผู้ป่วยโรคมะเร็งตับอีกด้วย (P < 0.001). การศึกษานี้จึงสรุปได้ว่า SULF2 และ GPC3 ภายในซีรั่มของผู้ป่วยสามารถนำไปใช้เป็นตัวบ่งชี้ทางชีวภาพและตัวพยากรณ์โรคมะเร็งตับที่มีสาเหตุมาจากการติดเชื้อไวรัสตับอักเสบบีได้

โรคมะเร็งตับเป็นหนึ่งในโรคมะเร็งที่สามารถพบได้ทั่วโลกและมีอัตราการตายเป็นอันดับต้นๆของโลก สาเหตุหลักที่พบเกิดจากการติดเชื้อไวรัสตับอักเสบบี ผู้ป่วยโรคมะเร็งตับส่วนใหญ่จะได้รับการวินิจฉัยในระยะท้ายของโรค ส่งผลให้มีอัตราการรอดชีวิตต่ำ ในปัจจุบันไมโครอาร์เอ็นเอมีบทบาทสำคัญในกระบวนการต่างๆภายในร่างกาย รวมทั้งการเกิดมะเร็ง เพราะฉะนั้นจึงสนใจศึกษาระดับการแสดงออกของไมโครอาร์เอ็นเอในซีรั่มที่มีประสิทธิภาพในการวินิจฉัยโรคมะเร็งตับระยะแรกที่เกิดจากการติดเชื้อไวรัสตับอักเสบบี โดยทำการวิเคราะห์รูปแบบการแสดงออกของไมโครอาร์เอ็นเอด้วยเทคนิค NanoString จากนั้นทำการวัดระดับการแสดงออกของตัวแทนไมโครอาร์เอ็นเอ ได้แก่ miR-21-5p และ miR-125b-5p ในชิ้นเนื้อจำนวน 22 ตัวอย่าง และในซีรั่มของกลุ่มควบคุมที่มีสุขภาพดี 50 ราย กลุ่มผู้ป่วยที่ติดเชื้อไวรัสตับอักเสบบี 92 ราย และกลุ่มผู้ป่วยโรคมะเร็งตับที่เกิดจากการติดเชื้อไวรัสตับอักเสบบี 98 ราย ด้วยเทคนิคเรียลไทม์พีซีอาร์ ผลการศึกษาพบว่า miR-21-5p มีแนวโน้มการแสดงออกสูงขึ้นในชิ้นเนื้อบริเวณส่วนที่เป็นมะเร็ง เมื่อเปรียบเทียบกับบริเวณข้างเคียง (P=0.068) นอกจากนั้น miR-21-5p ในซีรั่มของกลุ่มผู้ป่วยที่ติดเชื้อไวรัสตับอักเสบบีมีการแสดงออกสูงกว่าในกลุ่มควบคุมที่มีสุขภาพดี และกลุ่มผู้ป่วยโรคมะเร็งตับที่เกิดจากการติดเชื้อไวรัสตับอักเสบบี (P=0.0019 และ P<0.001 ตามลำดับ) ในขณะที่การแสดงออกของ miR-21-5p ในซีรั่มของกลุ่มผู้ป่วยโรคมะเร็งตับที่เกิดจากการติดเชื้อไวรัสตับอักเสบบีมีแนวโน้มสูงขึ้นเมื่อเปรียบเทียบกับกลุ่มควบคุมที่มีสุขภาพดี (P=0.2332) ในทางตรงกันข้าม miR-125b-5p มีการแสดงออกที่ลดลงในชิ้นเนื้อบริเวณส่วนที่เป็นมะเร็ง เมื่อเปรียบเทียบกับบริเวณข้างเคียง (P=0.0391) เช่นเดียวกับการแสดงออกของ miR-125b-5p ในซีรั่มของกลุ่มผู้ป่วยโรคมะเร็งตับที่เกิดจากการติดเชื้อไวรัสตับอักเสบบี พบว่ามีระดับลดลงเมื่อเปรียบเทียบกับกลุ่มผู้ป่วยที่ติดเชื้อไวรัสตับอักเสบบี และกลุ่มควบคุมที่มีสุขภาพดี (P=0.0234 และ P<0.001 ตามลำดับ) จากการวิเคราะห์ Receiver operating characteristic (ROC) พบว่าพื้นที่ใต้กราฟของการใช้ miR-125-5p ร่วมกับ alpha fetoprotein (AFP) มีค่าเท่ากับ 0.94 ซึ่งสูงกว่าการใช้ AFP หรือ miR-125b-5p เพียงอย่างเดียว ดังนั้น miR-125b-5p และ AFP น่าจะมีประสิทธิภาพในการวินิจฉัยโรคมะเร็งตับระยะแรกที่เกิดจากการติดเชื้อไวรัสตับอักเสบบี

A subgroup of breast cancers has several metabolic compartments. The mechanisms by which metabolic compartmentalization develop in tumors are poorly characterized. TP53 inducible glycolysis and apoptosis regulator (TIGAR) is a bisphosphatase that reduces glycolysis and is highly expressed in carcinoma cells in the majority of human breast cancers. Hence we set out to determine the effects of TIGAR expression on breast carcinoma and fibroblast glycolytic phenotype and tumor growth. The overexpression of this bisphosphatase in carcinoma cells induces expression of enzymes and transporters involved in the catabolism of lactate and glutamine. The relevance of mitochondrial phosphate carrier (PiC), encoded by SLC25A3, in bioenergetics is well accepted. However, little is known about the mechanisms mediating the cellular impairments induced by pathological SLC25A3 variants. To this end, we investigated the pathogenicity of a novel compound heterozygous mutation in SLC25A3 First, each variant was modeled in yeast, revealing that substituting GSSAS for QIP within the fifth matrix loop is incompatible with survival on non-fermentable substrate, whereas the L200W variant is functionally neutral. Type III secretion systems (T3SS) are central virulence factors for many pathogenic Gram-negative bacteria, and secreted T3SS effectors can block key aspects of host cell signaling. To counter this, innate immune responses can also sense some T3SS components to initiate anti-bacterial mechanisms. The Yersinia pestis T3SS is particularly effective and sophisticated in manipulating the production of pro-inflammatory cytokines IL-1β and IL-18, which are typically processed into their mature forms by active caspase-1 following inflammasome formation. The gene expression profile of a eukaryotic cell is responsive to a variety of extracellular stimuli, including nutrient availability, which allows cells to toggle between anabolism and catabolism based on the favorability of their environment. Much of this information is relayed through signaling complexes, such as the target of rapamycin complex 1 (TORC1), to downstream chromatin modifying enzymes. These enzymes impact the gene regulatory process through altered histone post-translation modifications, changes in chromatin structure, and docking of chromatin regulatory complexes. Protein Kinase D1 (PKD1) is a serine threonine kinase which is downregulated in Prostate, Breast and Colon Cancer. It functions as a tumor suppressor in different cancer cells. Downregulation of PKD1 is known to be associated with aggressiveness of the cancer. Desmosomes are prominent adhesive junctions present between many epithelial cells as well as cardiomyocytes. The mechanisms controlling desmosome assembly and remodeling in epithelial and cardiac tissue are poorly understood. We recently identified protein palmitoylation as a mechanism regulating desmosome dynamics. In this study, we have focused on the palmitoylation of the desmosomal cadherin desmoglein-2 (Dsg2) and characterized the role that palmitoylation of Dsg2 plays in its localization and stability in cultured cells. We identified two cysteine residues in the juxtamembrane (intracellular anchor) domain of Dsg2 that, when mutated, eliminate its palmitoylation. Epoxyeicosatrienoic acid (EET) contributes to browning of white adipose stem cells to ameliorate obesity/diabetes and insulin resistance. In the current study, we show that EET altered preadipocyte function, enhanced peroxisome proliferation-activated receptor γ coactivator α (PGC-1α) expression, and increased mitochondrial function in the 3T3-L1 preadipocyte subjected to adipogenesis. Cells treated with EET resulted in an increase, P < 0.05, in PGC-1α and a decrease in mitochondria-derived ROS (MitoSox), P < 0.05. The EET increase in heme oxygenase-1 (HO-1) levels is dependent on activation of PGC-1α as cells deficient in PGC-1α (PGC-1α knockout adipocyte cell) have an impaired ability to express HO-1, P < 0.02. Additionally, adipocytes treated with EET exhibited an increase in mitochondrial superoxide dismutase (SOD) in a PGC-1α-dependent manner, P < 0.05. The increase in PGC-1α was associated with an increase in β-catenin, P < 0.05, adiponectin expression, P < 0.05, and lipid accumulation, P < 0.02. EET decreased heme levels and mitochondria-derived ROS (MitoSox), P < 0.05, compared to adipocytes that were untreated. EET also decreased mesoderm-specific transcript (MEST) mRNA and protein levels (P < 0.05). Adipocyte secretion of EET act in an autocrine/paracrine manner to increase PGC-1α is required for activation of HO-1 expression. This is the first study to dissect the mechanism by which the antiadipogenic and anti-inflammatory lipid, EET, induces the PGC-1α signaling cascade and reprograms the adipocyte phenotype by regulating mitochondrial function and HO-1 expression, leading to an increase in healthy, that is, small, adipocytes and a decrease in adipocyte enlargement and terminal differentiation. This is manifested by an increase in mitochondrial function and an increase in the canonical Wnt signaling cascade during adipocyte proliferation and terminal differentiation.

The recent paradigm shift in infectious disease diagnosis from culture-based to molecular-based approaches is exemplified in the findings of a national study assessing the detection of verotoxigenic Escherichia coli infections in Ireland. Peroxiredoxins (Prxs) are ubiquitous cysteine-based peroxidases that guard cells against oxidative damage, are virulence factors for pathogens, and are involved in eukaryotic redox regulatory pathways. We have analyzed catalytically active crystals to capture atomic resolution snapshots of a PrxQ-subfamily enzyme (from Xanthomonas campestris) proceeding through thiolate, sulfenate, and sulfinate species. These analyses provide structures of unprecedented accuracy for seeding theoretical studies, and show novel conformational intermediates giving insight into the reaction pathway. Based on a highly non-standard geometry seen for the sulfenate intermediate, we infer that the sulfenate formation itself can strongly promote local unfolding of the active site to …

The master regulator of the macrophage development, differentiation, proliferation, survival, phagocytosis, cytokine secretion, motility, adhesion, migration, and spreading is the receptor tyrosine kinase (RTK) colony stimulating factor-1 receptor (CSF-1R). Aberrant CSF-1R signaling is present amongst a variety of highly prevalent and devastating human diseases in the United States such as atherosclerosis, cancer, inflammatory bowel disease, arthritis, and neuro-demyelination/neuro-degeneration. A better understanding of basic mechanisms that govern macrophage development and function is of vital importance in treating patients afflicted with these conditions/diseases. Over the past two decades, advances in RNA structural biology have improved our understanding of the structures and folding properties of naturally occurring RNAs. RNA sequences and structures participate in many specific biological functions, such as those performed by messenger RNA (mRNA), ribosomal RNA (rRNA), transfer RNA (tRNA), micro RNA (miRNA), short-interfering RNA (siRNA), small nuclear RNA (snRNA) and many others.
