Autism spectrum disorders (ASD) encompass neurodevelopment disorders characterized by atypical patterns of development that impact multiple areas of functioning beginning in early childhood. The etiology of ASD is unknown and there are currently no preventative treatment options. Gastrointestinal symptoms are commonly associated comorbidities. The microbiota-gut-brain axis is a multidirectional communication chain that connects the central and enteric nervous system that relates brain function to peripheral intestinal functions. Changes within this axis have been postulated in ASD. The ability of small extracellular vesicles (sEVs) to reprogram cancer cells is well established. However, the specific sEV components able to mediate aberrant effects in cancer cells have not been characterized. Integrins are major players in mediating sEV functions. We have previously reported that the αVβ3 integrin is detected in sEVs of prostate cancer (PαVβ3rCa) cells and transferred into recipient cells. Here, we investigate whether sEVs from -expressing cells affect tumour growth differently than sEVs from control cells that do not express αVβ3. One of the leading regulators of neuronal cell differentiation in the CNS is the family of basic helix–loop–helix (bHLH) transcription factors. One of these proteins, Nato3, is associated with the formation of dopaminergic neurons. Transcription factors can be regulated by kinase activity, and in order to detect the associated change of phosphorylation of Nato3, we have generated Nato3 with specific epitope tags that allow for the detection and isolation of the Nato3 protein. Idiopathic pulmonary fibrosis (IPF) is a chronic lung disease which affects about 5 to 8 million individuals in the world. Despite the high prevalence, there is currently no cure for IPF, and the cause of this disease is still unclear. Our laboratory and collaborators have shown that nudix hydrolase 21 (NUDT21, which is also known as cleavage factor 25, CFIm25) is a key regulator of alternative polyadenylation (APA). NUDT21 depletion causes 3’UTR shortening via APA leading to enhanced mRNA stability and protein translation. I will be exploring the impact of the large protein Titin on heart failure with a preserved ejection fraction (HFpEF). It was hypothesized that myocardial Titin plays a significant role in the progression of HFpEF through isoform, N2BA and N2B, changes. It was hypothesized that an increase in the quantity of N2B Titin, less compliant, and a decrease in N2BA Titin, more compliant, will be observed as HF progresses, contributing to an increase in passive stiffness involved in heart contraction. I will cover a detailed report on the role of Titin in HFpEF and propose research methods to explore further.

A method and system is provided for mass spectrometry for identification of a specific elemental formula for an unknown compound which includes but is not limited to a metabolite. The method includes calculating a natural abundance probability (NAP) of a given isotopologue for isotopes of non-labelling elements of an unknown compound. Intracapsular and well‑defined adenocarcinomas of the prostate are often surrounded by tissue areas that harbor molecular aberrations, including those of genetic, epigenetic and biochemical nature. This is known as field cancerization, or a field effect and denotes a state of pre‑malignancy. The G1/S cell cycle checkpoint is frequently dysregulated in cancer, leaving cancer cells reliant on a functional G2/M checkpoint to prevent excessive DNA damage. Wee1 regulates the G2/M checkpoint by phosphorylating CDK1 at Tyr15 to prevent mitotic entry. Previous drug development efforts targeting Wee1 resulted in the clinical-grade inhibitor, AZD1775. However, AZD1775 is burdened by dose-limiting adverse events, and has off-target PLK1 activity. In an attempt to overcome these limitations, we developed Wee1 degraders by conjugating AZD1775 to the cereblon (CRBN)-binding ligand, pomalidomide. The resulting lead compound, ZNL-02-096, degrades Wee1 while sparing PLK1, induces G2/M accumulation at 10-fold lower doses …

Kaposi’s sarcoma-associated herpesvirus (KSHV), also known as human herpesvirus 8 (HHV-8), is the etiological agent of Kaposi’s sarcoma (KS), and is also associated with two B cell malignancies, primary effusion lymphoma and multicentric Castleman's disease. The distribution of KSHV varies globally with high prevalence in some areas of sub-Saharan Africa (SSA), where seroprevalence can be as high as 80%. It is estimated that nearly 44,000 new cases of KS emerge annually globally, with the highest incidents occurring in Africa, where KSHV is endemic. Currently, there is no prophylactic vaccine against KSHV, and efforts to develop prophylactic vaccines have been limited. …

Lung cancer can be classified into two main categories: small cell lung cancer (SCLC) and non-small cell lung cancer (NSCLC), which are different in treatment strategy and survival probability. The lung CT images of SCLC and NSCLC are similar such that their subtle differences are hardly visually discernible by the human eye through conventional imaging evaluation. We hypothesize that SCLC/NSCLC differentiation could be achieved via computerized image feature analysis and classification in feature space, as termed a radiomic model. Physical inactivity, advancing age, limb immobilization, degenerative diseases and various systemic diseases (many cancers, sepsis, HIV, COPD, kidney disease) all lead to skeletal muscle wasting. The loss of muscle mass is of major clinical importance because it leads to an increased risk for morbidity, disability, and the loss of independence; collectively contributing to a substantive increase in healthcare utilization and cost. The prevalence of cachexia (disease-induced muscle wasting) can reach as high as 80% in certain patient populations and the average cost per hospital stay is $4,641 more than in non-cachectic patients. Dengue is an emerging arboviral disease endemic to many tropical countries including Malaysia. Due to its unspecific symptoms, dengue diagnosis can only be confirmed through diagnostic tests. Existing laboratory-based dengue diagnostics like enzyme linked immunosorbent assay (ELISA) is accurate in detecting dengue non-structural protein-1 (NS1), and anti-dengue immunoglobulin M (IgM) and G (IgG); but is also more complicated and time-consuming. Commercially available dengue rapid immunochromatographic test (RIT) is efficient but less sensitive. Pneumoviruses including human metapneumovirus (HMPV) and respiratory syncytial virus (RSV) are significant causes of respiratory tract infections globally. Children, elderly, and immunocompromised patients are at the greatest risk for developing severe infections, which can have devastating outcomes. Although these viruses are ubiquitous with significant impacts on human health, there are no antivirals or vaccines available. Acute inflammation begins with leukocyte P-selectin glycoprotein ligand-1 (PSGL-1) binding to P-selectin on inflamed endothelium and platelets. In pathologic conditions, this process may contribute to secondary organ damage, like sepsis-induced liver injury. Therefore, developing novel therapies to attenuate inflammation may be beneficial. We previously reported that recombinant human vimentin (rhVim) binds P-selectin to block leukocyte adhesion to endothelium and platelets. In this study, we used SPOT-peptide arrays to identify the rod domain as the active region within rhVim that interacts with P-selectin. Mucus serves as the chief protective barrier against pathogenic and mechanical insults in respiratory, gastrointestinal, and urogenital tracts. Altered mucin expression, the major component of mucus, in conjunction with differential glycosylation has been strongly associated with both benign and malignant pathologies of colon. BACKGROUND: Differential expression of mucins has been associated with several cancers including colorectal cancer (CRC). In normal physiological conditions, secretory mucin MUC5AC is not expressed in the colonic mucosa, whereas its aberrant expression is observed during development of colon cancer and its precursor lesions. To date, the molecular mechanism of MUC5AC in CRC progression and drug resistance remains obscure.

Kidney stone disease is a common urologic problem worldwide, especially in the tropics such as Thailand. It is known as a multifactorial condition, and aging increases the risk of stone development. The major type of stones is calcium oxalate (CaOx), and its formation is driven by increased urinary oxalate excretion and calcium oxalate monohydrate (COM) crystallization. มะเร็งสมองชนิดกลัยโอบลาสโตมา (Glioblastoma หรือ GBM) เป็นมะเร็งสมองชนิดที่พบมากที่สุด มีความรุนแรงและมีอัตราการเสียชีวิตสูง ทำให้คนไข้โดยเฉลี่ยเสียชีวิตภายในหนึ่งปี นอกจากนี้เซลล์ GBM ยังมีความหลากหลายทางสัณฐาน ดังนั้นผู้วิจัยจึงเล็งเห็นความสำคัญของเซลล์ต้นกำเนิดมะเร็งที่เป็นประชากรที่มีความสำคัญในแง่ของการตอบสนองต่อการรักษาและการกลับมาเป็นซ้ำของโรคมะเร็ง การที่จะสามารถพัฒนาการรักษาที่มีความจำเพาะในการกำจัดเซลล์ต้นกำเนิดมะเร็งนี้ จะต้องมีกระบวนการในการระบุเซลล์ต้นกำเนิดมะเร็ง (identification) ที่มีความจำเพาะและแม่นยำ ผู้วิจัยจึงต้องการค้นคว้าในเชิงลึกเพื่อหาการแสดงออกของยีนและโปรตีนที่เกี่ยวข้องกับความเป็นเซลล์ต้นกำเนิดมะเร็ง โดยทำการแยกประชากรเซลล์ U87MG ออกเป็น 3 กลุ่ม ได้แก่ ประชากรเซลล์รวม ประชากรเซลล์ใหญ่ และประชากรเซลล์เล็กออกจากกัน โดยใช้ความแตกต่างของขนาดเซลล์ ด้วยเทคนิค Fluorescence-activated cell sorting (FACS) จากนั้นนำประชากรดังกล่าวมาเลี้ยงแยกกัน โดยทำการศึกษาประชากรทั้งหมด ประชากรขนาดใหญ่และประชากรขนาดเล็ก แล้วศึกษาความเป็นเซลล์ต้นกำเนิดในระดับอาร์เอ็นเอด้วยเทคนิค Real-Time PCR โดยมุ่งเน้นไปที่การศึกษา (1) pluripotency markers/ transcription factor ได้แก่ OCT4, SOX2, NANOG และ c-MYC (2) cancer stem cell signalling ได้แก่ mTOR pathway โดยเน้นไปที่ mTORC2 และ LIN28/let-7 pathway และ (3) cellular phenotypes ได้แก่ วัฏจักรของเซลล์ (cell cycle) การแบ่งตัวของเซลล์ (cell proliferation) และการเคลื่อนที่ของเซลล์ (cell migration) งานวิจัยนี้เป็นรายงานแรกที่ศึกษาความเป็นเซลล์ต้นกำเนิดมะเร็ง GBM ใน U87MG โดยการแยกเซลล์ออกเป็นประชากรเซลล์ใหญ่และประชากรเซลล์เล็กด้วย FACS โดยพบว่าเซลล์ U87MG มีการแสดงออกของยีนที่บ่งชี้ความเป็นเซลล์ต้นกำเนิด ได้แก่ OCT4, SOX2, LIN28B,c-MYC, ZWINT, TYMS และ RCF4 ยีนที่เกี่ยวข้องกับ GBM ที่ทำการศึกษาการแสดงออก ได้แก่ SPARCL1, GDF15, LAMA1, …

Heart failure is a lifelong disability that for over half of those affected leads to mortality within five years after initial diagnosis. Left ventricular hypertrophy (LVH) is one of the most reliable independent predictors of heart failure. Pathological LVH is irreversible, but early diagnosis is often missed due to its asymptomatic nature. Obese Zucker rats (OZR), which develop obesity due to dysfunctional leptin signaling, naturally exhibit a LVH that mimics the obese human condition. Endothelial nitric oxide (NO) synthase (eNOS) plays a critical role in the maintenance of blood vessel homeostasis. Recent findings suggest that cytoskeletal dynamics play an essential role in regulating eNOS expression and activation. Here, we sought to test whether modulation of cytoskeletal dynamics through pharmacological regulation of histone deacetylase 6 (HDAC6)-mediated tubulin deacetylation affects eNOS expression and endothelial function in vitro and in vivo.Wefound that tubulin acetylation inducer (tubacin), a compound that appears to selectively inhibit HDAC6 activity, dramatically increased eNOS expression in several different endothelial cell lines, as determined by both immunoblotting and NO production assays. Apoptosis is a form of programmed cell death (PCD) that plays critical physiological roles in removing superfluous or dangerous cell populations that are unneeded or threatening to the health of the host organism. Although the molecular pathways leading to activation of the apoptotic program have been extensively studied and characterized starting in the 1970s, new evidence suggests that members of the gasdermin superfamily are novel pore-forming proteins that augment apoptosis by permeabilizing the mitochondria and participate in the final stages of the apoptotic program by inducing secondary necrosis/pyroptosis.
