Biomedical Sciences Faculty Research and Publications

Ghrelin receptor, also known as growth hormone secretagogue receptor (GHS-R1a), is coexpressed with its truncated isoform GHS-R1b, which does not bind ghrelin or signal, but oligomerizes with GHS-R1a, exerting a complex modulatory role that depends on its relative expression. D1 dopamine receptor (D1R) and D5R constitute the two D1-like receptor subtypes. Previous studies showed that GHS-R1b also facilitates oligomerization of GHS-R1a with D1R, conferring GHS-R1a distinctive pharmacological properties. Those include a switch in the preferred coupling of GHS-R1a from Gq to Gs and the ability of D1R/D5R agonists and antagonists to counteract GHS-R1a signaling. Activation of ghrelin …

The intestinal microbiota of Sprague-Dawley (SD) rats were investigated following consumption of diets containing refined high-amylose cornstarch (RS), waxy whole-grain flour from maize (WG), or high-amylose whole-grain flour (WG + RS) together with moderate (MF, 30% of energy) or high (HF, 42% of energy) levels of fat. Compared with control (CON) diets, RS reduced bacterial alpha-diversity and enriched Bacteroidetes and WG increased Lactobacillus proportions by three-fold. WG + RS conferred a combined effect, but the changes were attenuated and greater inter-individual variation was observed. Proportions of fiber-degrading bacterial taxa were inversely correlated with cecal pH. Notably, there was little influence …

Manuscripts, Articles, Book Chapters and Other Papers

Hyperfiltration is an important underlying cause of glomerular dysfunction associated with several systemic and intrinsic glomerular conditions leading to chronic kidney disease (CKD). These include obesity, diabetes, hypertension, focal segmental glomerulosclerosis (FSGS), congenital abnormalities and reduced renal mass (low nephron number). Hyperfiltration-associated biomechanical forces directly impact the cell membrane, generating tensile and fluid flow shear stresses in multiple segments of the nephron. Ongoing research suggests these biomechanical forces as the initial mediators of hyperfiltration-induced deterioration of podocyte structure and function leading to their detachment and irreplaceable loss from the glomerular filtration barrier. Membrane lipid-derived polyunsaturated fatty acids (PUFA) and their …

Hormone replacement therapies have become important for treating diseases such as premature ovarian failure or menopausal complications. The clinical use of bioidentical hormones might significantly reduce some of the potential risks reportedly associated with the use of synthetic hormones. In the present study, we demonstrate the utility and advantage of a microfluidic chip culture system to enhance the development of personalized, on-demand, treatment modules using embryoid bodies (EBs). Functional EBs cultured on microfluidic chips represent a platform for personalized, patient-specific treatment cassettes that can be cryopreserved until required for treatment. We assessed the viability, differentiation, and functionality of EBs cultured …

The most common host entry point of human adapted coronaviruses (CoV) including SARS-CoV-2 is through the initial colonization in the nostril and mouth region which is responsible for spread of the infection. Most recent studies suggest that the commercially available oral and nasal rinse products are effective in inhibiting the viral replication. However, the anti-viral mechanism of the active ingredients present in the oral rinses have not been studied. In the present study, we have assessed in vitro enzymatic inhibitory activity of active ingredients in the oral mouth rinse products: aloin A and B, chlorhexidine, eucalyptol, hexetidine, menthol, triclosan, methyl …

Health & Biomedical Sciences Faculty Publications

Breast cancer is one of the leading causes of death in the female population worldwide. Standard treatments such as chemotherapy show noticeable results. However, along with killing cancer cells, it causes systemic toxicity and apoptosis of the nearby healthy cells, therefore patients must endure side effects during the treatment process. Implantable drug delivery devices that enhance therapeutic efficacy by allowing localized therapy with programmed or controlled drug release can overcome the shortcomings of conventional treatments. An implantable device can be composed of biopolymer materials, nanocomposite materials, or a combination of both. This review summarizes the recent research and current state-of-the …

Polarized epithelial cells form an essential barrier against infection at mucosal surfaces. Many pathogens breach this barrier to cause disease, often by co-opting cellular endocytosis mechanisms to enter the cell through the lumenal (apical) cell surface. We recently discovered that the loss of the cell polarity gene PARD6B selectively diminishes apical endosome function. Here, we find that in response to the entry of certain viruses and bacterial toxins into the epithelial cells via the apical membrane, PARD6B and aPKC, two components of the PARD6B-aPKC-Cdc42 apical polarity complex, undergo rapid proteasome-dependent degradation. The perturbation of apical membrane glycosphingolipids by toxin- or …

Generalized Estimating Equations (Gee) Approach For Clustered Binary Data With Application To Covid-19 Treatment., Sadixa Sanjel Feb 2022

Generalized Estimating Equations (Gee) Approach For Clustered Binary Data With Application To Covid-19 Treatment., Sadixa Sanjel

SDSU Data Science Symposium

Clustered binary data frequently occur in epidemiology and other applied fields such as clinical trial studies, where observations within the respective samples are correlated. In such situations, the standard logistic regression method is not valid as logistic regression requires the observations to be independent of one other. This situation arises when treating COVD-19 patients. Patients from certain clusters, such as geographic areas or the same family, are highly correlated, and we need to fit the model using the GEE approach. In this paper, Standard Logistic Regression (LS), Generalized Linear Models (GENMOD), and GEE procedures have been utilized for comparison purposes. …

General Adversarial Networks In Tumor-Related Research: A Review And Agenda For Moving Forward, Andrew James Behrens, Cherie Noteboom

SDSU Data Science Symposium

Recent advances in Generative Adversarial Networks (GANs) have led to many new variants and uses of GANs. The latest advancements have allowed researchers and practitioners to apply this technique to tumor-related problems with limited data. One of the trends in this problem domain is to develop different variants of GANs suited explicitly to particular problems. The variants of GANs are numerous but share a common characteristic of expanding the dataset by creating synthetic data from the original dataset. This paper aims to develop a research agenda through a systematic literature review that investigates practitioners' and researchers' emerging issues and current …

Selective Elimination Of Pluripotent Stem Cells By Pikfyve Specific Inhibitors., Arup R Chakraborty, Alex Vassilev, Sushil K Jaiswal, Constandina E O'Connell, John F Ahrens, Barbara S Mallon, Martin Pera, Melvin L Depamphilis

Faculty Research 2022

Inhibition of PIKfyve phosphoinositide kinase selectively kills autophagy-dependent cancer cells by disrupting lysosome homeostasis. Here, we show that PIKfyve inhibitors can also selectively eliminate pluripotent embryonal carcinoma cells (ECCs), embryonic stem cells, and induced pluripotent stem cells under conditions where differentiated cells remain viable. PIKfyve inhibitors prevented lysosome fission, induced autophagosome accumulation, and reduced cell proliferation in both pluripotent and differentiated cells, but they induced death only in pluripotent cells. The ability of PIKfyve inhibitors to distinguish between pluripotent and differentiated cells was confirmed with xenografts derived from ECCs. Pretreatment of ECCs with the PIKfyve specific inhibitor WX8 suppressed their …

Life During The Covid-19 Pandemic Was, And Still Is, Dispiriting, Anonymous

Community Reflections

For me personally, I did not enjoy quarantining at all. I did enjoy seeing my family every day, however, I was very lonely. I couldn’t see any of my friends, which was very disappointing. Thankfully with social media I could facetime them and text them, but face-to-face contact with other people is very necessary; it is something you wouldn’t normally think is important, but when you are unable to physically be with other people, you realize how lonely you feel.
