Odors can be powerful stimulants. It is well-established that odors provide strong cues for recall of locations, people and events. The effects of specific scents on other cognitive functions are less well-established. We hypothesized that scents with different odor qualities will have a different effect on attention. To assess attention, we used Inter-Subject Correlation of the EEG because this metric is strongly modulated by attentional engagement with natural audiovisual stimuli. We predicted that scents known to be “energizing” would increase Inter-Subject Correlation during watching of videos as compared to “calming” scents. In a first experiment, we confirmed this for eucalyptol and …

The heterogeneity of glioblastomas, the most common primary malignant brain tumor, remains a significant challenge for the treatment of these devastating tumors. Therefore, novel combination treatments are warranted. Here, we showed that the combined inhibition of TRAP1 by gamitrinib and histone deacetylases (HDAC1/HDAC2) through romidepsin or panobinostat caused synergistic growth reduction of established and patient-derived xenograft (PDX) glioblastoma cells. This was accompanied by enhanced cell death with features of apoptosis and activation of caspases. The combination treatment modulated the levels of pro- and anti-apoptotic Bcl-2 family members, including BIM and Noxa, Mcl-1, Bcl-2 and Bcl-xL. Silencing of Noxa, BAK and …

Fast scan cyclic voltammetry (FSCV) allows for real -time analysis of phasic neurotransmitter levels. Tryptophan (TRP) is an aromatic amino acid responsible for facilitating electron transfer kinetics in oxidoreductase enzymes. Previous work with TRP-modified electrodes showed increased sensitivity for cyclic voltammetry detection of dopamine (DA) when used with slower scan rates (0.05 V/s). Here, we outline an in vitro proof of concept for TRP-modified electrodes in FSCV detection of DA, and decreased sensitivity for ascorbic acid (AA). TRP-modified electrodes had a limit of detection (LOD) for DA of 2.480 ± 0.343 nM compared to 8.348 ± 0.405 nM for an …

The aim of this study was to derive an equation that validly and reliably estimates the VO2 reached in the IPTL, a maximum stress test for lifeguards. From the results obtained in the first part of the research, a multiple linear regression model was identified. A "stepwise" procedure was established as a variable selection method, where the VO2IPTL was selected as the dependent variable and the maximum time reached in the IPTL (s), the percentage of muscle mass (%) and weight of the participant (kg), as independent variables. This procedure generated a regression model where a high correlation …

A Study Of Covid-19 Mortality Under Varying Patient Frailty, Alison N. Sifuentes
Theses and Dissertations
Modeled the spread and mortality of COVID-19 throughout the city of Chicago. By incorporating group frailty into a classic SEIR infectious disease model, we were able to differentiate the population of Chicago by their response to COVID-19. Three age groups with different COVID-19-induced death rates were examined, and the model sought to showcase the multiplicative deviation of each age group death rate from the average disease-induced death rate. This adjustment for different death rates among age groups accounted for heterogeneity within the population, and sought to introduce a more accurate manner for modeling the spread of infectious diseases.

Background: Oral antipsychotic (AP) medications are frequently prescribed to people with bipolar I disorder (BD-I). A cross-sectional online survey examined the experiences of people living with BD-I with a history of recent AP use. Methods: Adults with self-reported physician-diagnosed BD-I (N = 200) who received oral APs during the prior year completed a survey on AP-related experiences, including side effects and their perceived burden on social functioning, adherence, and work. Items also assessed preferences for trade-offs (balancing symptom management and side effects) when considering a hypothetical new AP. The perceived impact of specific, prevalent side effects on adherence, work, and …
A Study On Ecological Distribution Of And Selected Gene Expression In Amblyomma Americanum: An Insight Into The Alpha-Galactose Allergy Caused By Tick Bites, Leah Cuthill
Electronic Theses & Dissertations
Ticks transmit a wide variety of pathogens including viruses, bacteria, protozoa, and helminthes to vertebrates. Their life cycle depends on blood meals from various hosts as well as on environmental conditions such as the temperature and habitat type. A newly recognized allergic disease has been identified and is characterized by a delayed reaction following the consumption of some mammalian meats. The allergy often results in life threatening reactions such as anaphylaxis, hives, and breathing problems 3-6 hours after the consumption of mammalian meat. Not everyone develops the allergy when they are bitten by a lone star tick, and the cause …

Immune checkpoint inhibitors (ICI) have shown positive results in patients with hepatocellular carcinoma (HCC). As liver function contributes to prognosis, its precise assessment is necessary for the safe prescribing and clinical development of ICI in HCC. We tested the accuracy of the albumin-bilirubin (ALBI) grade as an alternative prognostic biomarker to the Child-Turcotte-Pugh (CTP). In a prospectively maintained multi-centre dataset of HCC patients, we assessed safety and efficacy of ICI across varying levels of liver dysfunction described by CTP (A to C) and ALBI grade and evaluated uni- and multi-variable predictors of overall (OS) and post-immunotherapy survival (PIOS). We studied …

"In his introduction to Epidemics and Ideas, Paul Slack calls to revive the study of social history of epidemics, wanting to show how societies cope with, react to, and interpret crises of disease. He reviews historian Richard Evans’ notion of the “common dramaturgy” to all epidemics, which states that human society responds to mass infection through an inherent response mechanism. Disease presents common dilemmas - including decisions on how the disease is transmitted, whom it infects, who is to blame, and incites common responses. Furthermore, Slack suggests that …

"By 1992, the AIDS epidemic in the United States had reached seemingly catastrophic proportions. Over ten years after the first published report of AIDS-related lung infection, the number of AIDS cases in the United States far exceeded 100,000. It would be four years until the FDA approval of the first protease inhibitor. Over ten thousand women had been diagnosed with the disease, and experts expected over ninety thousand more were already infected. The disease, lacking effective treatment, increasingly struck women and people of color in the early 1990s; …

"As the world has become steadily more interdependent over the last century, disease has been able to spread with increasing ease. As a consequence of this new danger, the international community has changed the way that it understands contagion, and public health has taken on new roles and new meanings around the world. I argue that the globalization of public health and the relatively new perception of diseases as “global” threats have dramatically altered practices of health screening, both at the border and beyond. Above all, these changes …

Genes Adapt To Outsmart Gene-Targeting Strategies In Mutant Mouse Strains By Skipping Exons To Reinitiate Transcription And Translation., Vishnu Hosur, Benjamin E. Low, Daniel Li, Grace Stafford, Vivek Kohar, Leonard D. Shultz, Michael V. Wiles
Faculty Research 2020
BACKGROUND: Gene disruption in mouse embryonic stem cells or zygotes is a conventional genetics approach to identify gene function in vivo. However, because different gene disruption strategies use different mechanisms to disrupt genes, the strategies can result in diverse phenotypes in the resulting mouse model. To determine whether different gene disruption strategies affect the phenotype of resulting mutant mice, we characterized Rhbdf1 mouse mutant strains generated by three commonly used strategies-definitive-null, targeted knockout (KO)-first, and CRISPR/Cas9.
RESULTS: We find that Rhbdf1 responds differently to distinct KO strategies, for example, by skipping exons and reinitiating translation to potentially yield gain-of-function alleles …
