Neutrophils Are Mediators Of Metastatic Prostate Cancer Progression In Bone, Diane L. Costanzo-Garvey, Tyler Keeley, Adam J. Case, Gabrielle F. Watson, Massar Alsamraae, Yangsheng Yu, Kaihong Su, Cortney E. Heim, Tammy Kielian, Colm Morrissey, Jeremy S Frieling, Leah M. Cook

Journal Articles: Pathology and Microbiology

Bone metastatic prostate cancer (BM-PCa) significantly reduces overall patient survival and is currently incurable. Current standard immunotherapy showed promising results for PCa patients with metastatic, but less advanced, disease (i.e., fewer than 20 bone lesions) suggesting that PCa growth in bone contributes to response to immunotherapy. We found that: (1) PCa stimulates recruitment of neutrophils, the most abundant immune cell in bone, and (2) that neutrophils heavily infiltrate regions of prostate tumor in bone of BM-PCa patients. Based on these findings, we examined the impact of direct neutrophil-prostate cancer interactions on prostate cancer growth. Bone marrow neutrophils directly induced apoptosis …

Monocyte Metabolic Reprogramming Promotes Pro-Inflammatory Activity And Staphylococcus Aureus Biofilm Clearance, Kelsey J. Yamada, Cortney E. Heim, Xinyuan Xi, Kuldeep S. Attri, Dezhen Wang, Wenting Zhang, Pankaj K. Singh, Tatiana K. Bronich, Tammy Kielian

Journal Articles: Pathology and Microbiology

Biofilm-associated prosthetic joint infections (PJIs) cause significant morbidity due to their recalcitrance to immune-mediated clearance and antibiotics, with Staphylococcus aureus (S. aureus) among the most prevalent pathogens. We previously demonstrated that S. aureus biofilm-associated monocytes are polarized to an anti-inflammatory phenotype and the adoptive transfer of pro-inflammatory macrophages attenuated biofilm burden, highlighting the critical role of monocyte/macrophage inflammatory status in dictating biofilm persistence. The inflammatory properties of leukocytes are linked to their metabolic state, and here we demonstrate that biofilm-associated monocytes exhibit a metabolic bias favoring oxidative phosphorylation (OxPhos) and less aerobic glycolysis to facilitate their anti-inflammatory activity and biofilm …

Integrating Image Caption Information Into Biomedical Document Classification In Support Of Biocuration., Xiangying Jiang, Pengyuan Li, James A. Kadin, Judith A. Blake, Martin Ringwald, Hagit Shatkay

Faculty Research 2020

Gathering information from the scientific literature is essential for biomedical research, as much knowledge is conveyed through publications. However, the large and rapidly increasing publication rate makes it impractical for researchers to quickly identify all and only those documents related to their interest. As such, automated biomedical document classification attracts much interest. Such classification is critical in the curation of biological databases, because biocurators must scan through a vast number of articles to identify pertinent information within documents most relevant to the database. This is a slow, labor-intensive process that can benefit from effective automation.

We present a document classification …

Faculty Research 2020

Background: Oligodendroglioma is a rare primary central nervous system (CNS) tumor with highly variable outcome and for which therapy is usually not curative. At present, little is known regarding the pathways involved with progression of oligodendrogliomas or optimal biomarkers for stratifying risk. Developing new therapies for this rare cancer is especially challenging. To overcome these challenges, the neuro-oncology community must be particularly innovative, seeking multi-institutional and international collaborations, and establishing partnerships with patients and advocacy groups thereby ensuring that each patient enrolled in a study is as informative as possible.

Methods: The mission of the National Cancer Institute's NCI-CONNECT program …

Understanding Costs Of Healthcare: Patient Perspectives And Beyond, Edwin Yirong Chan Jan 2020

Understanding Costs Of Healthcare: Patient Perspectives And Beyond, Edwin Yirong Chan

Yale Medicine Thesis Digital Library

Introduction: Healthcare spending in the US is among the highest in the world and continues to grow year-over-year despite outcomes comparable to or worse than other nations. This spending has translated to extensive patient burden and high out-of-pocket costs for many individuals. Despite high costs to both systems and individuals, patients continue to pursue expensive care options such as emergency department care over other appropriate alternatives. Here, we assess patient understanding of emergency room costs and influences on their decisions to pursue emergency department care.

Methods: Adult patients presenting to the emergency department for conditions not meriting hospital admission or …

Human Amniotic Membrane Improves Healing In A Chronic, Massive Rotator Cuff Repair Model, Maarouf Saad Jan 2020

Human Amniotic Membrane Improves Healing In A Chronic, Massive Rotator Cuff Repair Model, Maarouf Saad

Yale Medicine Thesis Digital Library

Rotator cuff tendon heals by fibrovascular scar that is weaker than native tissue leading to repairs that are prone to failure. To investigate the utility of an amniotic-membrane-derived human allograft in improving tendon-bone healing and skeletal muscle architecture, a rotator cuff repair model in Sprague-Dawley rats was established, tendon biomechanical testing, single muscle fiber biomechanical testing, microcomputed tomography analysis, tendon and muscle histological analysis, and gene expression analysis were performed. Our findings demonstrated single muscle-fibers with larger cross-sectional area and greater maximum isometric and specific force in the augmented experimental repair (ER) group. Additionally, microcomputed tomography revealed a larger volume …

Coronary Revascularization Options For Females: A Meta-Analysis Of Randomized Controlled Trials, Tayyab Hussain Shah Jan 2020

Coronary Revascularization Options For Females: A Meta-Analysis Of Randomized Controlled Trials, Tayyab Hussain Shah

Yale Medicine Thesis Digital Library

The totality of evidence for revascularization decisions between coronary artery bypass grafting (CABG) and percutaneous coronary intervention (PCI) for the treatment of multivessel coronary artery disease comes from predominantly male patients. Although societal guidelines do not provide sex specific recommendations, growing evidence suggests that revascularization outcomes may not be consistent between sexes. This study evaluates outcomes of CABG versus PCI in female patients with multivessel disease (MVD) and/or left main disease (LMD). We conducted a systematic search to identify randomized controlled trials (RCTs) comparing the long-term outcomes after PCI versus CABG in MVD and/or LMD. We extracted female specific data …

Characterization Of Temperature Curves In The Intensive Care Unit, Patricia Lourdes Valda Toro Jan 2020

Characterization Of Temperature Curves In The Intensive Care Unit, Patricia Lourdes Valda Toro

Yale Medicine Thesis Digital Library

Fevers are extremely common in hospitalized patients, particularly in those admitted to the intensive care unit (ICU). Up to 50% of fevers in the ICU are non-infectious in origin. Yet, the development of a fever commonly prompts extensive, costly and invasive infectious workups. Patients with persistent fevers are usually treated with antimicrobials long after infectious workups prove unrevealing. While this is viewed as a ‘safe’ approach, inappropriate antibiotic use has shown to be harmful to patients – even effecting an increase in mortality – in addition to contributing to the spread of antibiotic resistance. The purpose of this study is …

Exploratory Study Of The Lived Experiences Of Adoptees In The Caribbean, Kelley T. Knowles Jan 2020

Exploratory Study Of The Lived Experiences Of Adoptees In The Caribbean, Kelley T. Knowles

Department of Family Therapy Dissertations and Applied Clinical Projects

This qualitative research study offers exploration into Caribbean adoptee experiences related to their adoptive parent-child relationships. Existing studies focus on adoptive parent perspectives. Few accounts focus on Caribbean adoptees' experiences with adoption (Caughman, 2007). The existing research serves a purpose by helping adopted parents discuss adoption with adoptees. However, this approach does not include thevoice of the adoptee and fails to encompass the totality of adoptee lived experiences. The research does not focus on the amplification of the adoptees' voice, which in turn, neglects the adoptees' role inunderstanding their family processes. Therefore, it is pertinent to include lived experiences of …

White Marriage And Family Therapy Students’ Experiences In Diversity Courses: An Interpretative Phenomenological Analysis, Porshia Cunningham Jan 2020

White Marriage And Family Therapy Students’ Experiences In Diversity Courses: An Interpretative Phenomenological Analysis, Porshia Cunningham

Department of Family Therapy Dissertations and Applied Clinical Projects

American minority racial groups are on the rise and the majority racial group (White) will decrease in size (Vespa, Medina, & Armstrong, 2018). The U.S. population is becoming progressively diverse and projections indicate that by 2050, the elderly population will be 42% minority for the first time in history (U.S. Census Bureau, 2002). Racial minority experiences and needs are uniquely influenced by race due to systematic structures historically constructed in the United States (Cole, 2019; Sue, 2019). For many marriage and family therapy (MFT) students, particularly White students, the culture and diversity course is the primary mechanism for exposure to …

Journal Articles: Pulmonary & Critical Care Med

COPD, chronic bronchitis (CB) and active smoking have all been associated with goblet cell hyperplasia (GCH) in small studies. Active smoking is strongly associated with CB, but there is a disconnect between CB clinical symptoms and pathology. Chronic cough and sputum production poorly correlate with the presence of GCH or COPD. We hypothesized that the primary determinant of GCH in ever smokers with or without airflow obstruction is active smoking. Goblet Cell Density (GCD) was measured in 71 current or former smokers [32 subjects without COPD and 39 COPD subjects]. Endobronchial mucosal biopsies were stained with Periodic Acid Schiff-Alcian Blue, …
