The pharmacokinetics of a humanized anti-RNLS monoclonal antibody used for the treatment of melanoma in a congenic mouse species

Authors: Oriyomi Alimi1, Gary Desir1

1Department of Internal Medicine, Yale School of Medicine, New Haven, CT

Cancer cells can overcome signaling that restrains their growth and promotes senescence and cell death. Renalase (RNLS) is a secreted flavoprotein that functions as a survival factor after ischemic and toxic injury, signaling through the plasma calcium channel PMCA4b to activate the PI3K/AKT and MAPK pathways. In addition, recent studies, indicate that dysregulated RNLS signaling promotes survival of melanoma cells due to its capacity to …

Effect Of Silver Diamine Fluoride On Candida Albicans Associated With Early Childhood Caries, Sarah Al Kukash Jan 2020

Effect Of Silver Diamine Fluoride On Candida Albicans Associated With Early Childhood Caries, Sarah Al Kukash

Student Theses, Dissertations and Capstones

BACKGROUND: Early childhood caries (ECC) is one of the most prevalent infectious diseases in childhood worldwide. It afflicts 23% of the preschoolers in the U.S. It most frequently targets children from poor socioeconomic families and racial/ethnic minority backgrounds and becomes a serious public health problem. The major pathogen of ECC is Streptococcus mutans (S. mutans). However, the occurrence of ECC is positively associated with the presence of Candida albicans (C. albicans). It is well established that Silver Diamine Fluoride (SDF), a recent Food and Drug Administration (FDA)-off labeled approved agent for arresting dental caries, exhibits a robust antibacterial effect on …

Teaming For Transition: Interprofessional Collaboration, Megan Sturdivant, Emily Thompson, Sam Skelton, Rebecca Mallory

2020 Honors Symposium

The transition from youth to young adult for individuals with severe disabilities is difficult, provides few opportunities for employment, and offers less of a chance to connect with friends and engage with people. This research presents ways that speech-language pathologists, occupational therapists, school counselors, and special educators can collaborate to create a successful transition process for individuals with severe disabilities.

Development Of Early Social Interactions In Infants Exposed To Artificial Intelligence From Birth, Anna Vest Jan 2020

Development Of Early Social Interactions In Infants Exposed To Artificial Intelligence From Birth, Anna Vest

2020 Honors Symposium

Emersion in a ‘smart’ world of technology starts the day an infant enters the door of a home shared with Amazon Alexa, Siri, Nest, and perhaps a smart baby bassinet. This exposure raises a question about how artificial intelligence might yield a different experience in social and interactive life for infants that have been exposed to it from their earliest days.

Ethical imperatives critical to effective disease control in the coronavirus pandemic: Recognition of global health interdependence as a driver of health and social equity

George A. Gellert MD, MPH, MPA

ABSTRACT

Decades into the era of emerging infectious diseases, the 2019-2020 coronavirus pandemic has caught the world, and the United States in particular, poorly prepared to engage effective public health disease prevention and control measures. In part, this reflects poor public health planning, response, logistical preparation and pandemic readiness, and complacency by governments and disease control agencies. In terms of future responses to emerging infection pandemics, these deficiencies can be …

Journal Articles: Physical Therapy

INTRODUCTION: Men who are overweight or obese in the rural Midwestern USA are an unrepresented, at-risk group exhibiting rising rates of cardiovascular disease, poor access to preventive care and poor lifestyle behaviours that contribute to sedentary lifestyle and unhealthy diet. Self-monitoring of eating and activity has demonstrated efficacy for weight loss. Use of mobile technologies for self-monitoring eating and activity may address rural men's access disparities to preventive health resources and support weight loss. Our pilot trial will assess the feasibility and acceptability of two mobile applications for weight loss in rural men to inform a future, full-scale trial.

The Potential Effects Of Hormonal Therapy And Stress On The Oral Health Of The Transitioning Population, Elena G. Cantwell, Devon Mcclure

Dental Hygiene Student Scholarship

Problem: In terms of healthcare, the transgender population is underserved. Unfortunately, these individuals often experience stress related to seeking preventative care and fear discrimination. These factors preventing them from seeking care, in addition to hormone therapy taken during the transition process, could have severe impacts on their dental health. The purpose of this study is to establish a link between the hormone therapy used during gender transitioning and the effect on oral health. While there is a correlation between hormone replacement therapy and clinical evidence that sex hormones can impact on periodontal tissues, few studies have linked this knowledge to …

© 2019 The Author(s). Published by Oxford University Press on behalf of the European Society of Cardiology. Pacemakers, implantable cardiac defibrillators, and cardiac resynchronization therapy devices are potentially life-saving treatments for a number of cardiac conditions, but are not without risk. Most concerning is the risk of a cardiac implantable electronic device (CIED) infection, which is associated with significant morbidity, increased hospitalizations, reduced survival, and increased healthcare costs. Recommended preventive strategies such as administration of intravenous antibiotics before implantation are well recognized. Uncertainties have remained about the role of various preventive, diagnostic, and treatment measures such as skin antiseptics, pocket …

© 2020, The Author(s). Purpose: To study the association between early initiation of intra-arrest therapeutic hypothermia and neurologic outcome in out-of-hospital cardiac arrest. Methods: A prespecified sub-analysis of the PRINCESS trial (NCT01400373) that randomized 677 bystander-witnessed cardiac arrests to transnasal evaporative intra-arrest cooling initiated by emergency medical services or cooling started after hospital arrival. Early cooling (intervention) was defined as intra-arrest cooling initiated < 20 min from collapse (i.e., ≤ median time to cooling in PRINCESS). Propensity score matching established comparable control patients. Primary outcome was favorable neurologic outcome, Cerebral Performance Category (CPC) 1–2 at 90 days. Complete recovery (CPC 1) was among secondary outcomes. Results: In total, 300 patients were analyzed and the proportion with CPC 1–2 at 90 days was 35/150 (23.3%) in the intervention group versus 24/150 (16%) in the control group, odds ratio (OR) 1.92, 95% confidence interval (CI) 0.95–3.85, p =.07. In patients with shockable rhythm, CPC 1–2 was 29/57 (50.9%) versus 17/57 (29.8%), OR 3.25, 95%, CI 1.06–9.97, p =.04. The proportion with CPC 1 at 90 days was 31/150 (20.7%) in the intervention group and 17/150 (11.3%) in controls, OR 2.27, 95% CI 1.12–4.62, p =.02. In patients with shockable rhythms, the proportion with CPC 1 was 27/57 (47.4%) versus 12/57 (21.1%), OR 5.33, 95% CI 1.55–18.3, p =.008. Conclusions: In the whole study population, intra-arrest cooling initiated < 20 min from collapse compared to cooling initiated at hospital was not associated with improved favorable neurologic outcome. In the subgroup with shockable rhythms, early cooling was associated with improved favorable outcome and complete recovery.

PURPOSE: Patients with acute myeloid leukemia with high-risk cytogenetics in first complete remission (CR1) achieve better outcomes if they undergo allogeneic hematopoietic cell transplantation (HCT) compared with consolidation chemotherapy alone. However, only approximately 40% of such patients typically proceed to HCT. METHODS: We used a prospective organized approach to rapidly identify donors to improve the allogeneic HCT rate in adults with high-risk acute myeloid leukemia in CR1. Newly diagnosed patients had cytogenetics obtained at enrollment, and those with high-risk cytogenetics underwent expedited HLA typing and were encouraged to be referred for consultation with a transplantation team with the goal of …

The COVID-19 pandemic seemingly is peaking now in New York City and has triggered significant changes to the standard management of gastrointestinal diseases. Priorities such as minimizing viral transmission, preserving personal protective equipment, and freeing hospital beds have driven unconventional approaches to managing gastroenterology (GI) patients. Conversion of endoscopy units to COVID units and redeployment of GI fellows and faculty has profoundly changed the profile of most GI services. Meanwhile, consult and procedural volumes have been reduced drastically. In this review, we share our collective experiences regarding how we have changed our practice of medicine in response to the COVID …

© 2020 Ferrata Storti Foundation. Leukemia stem cells (LSC) are more resistant to standard chemotherapy and their persistence during remission can cause relapse, which is still one of the major clinical challenges in the treatment of acute myeloid leukemia (AML). A better understanding of the mutational patterns and the prognostic impact of molecular markers associated with stemness could lead to better clinical management and improve patients’ outcomes. We applied a previously described 17-gene expression score comprising genes differently expressed between LSC and leukemic bulk blasts, for 934 adult patients with de novo AML, and studied associations of the 17-gene LSC …
