The neuro-oncological ventral antigen 2 (NOVA2) protein is a major factor regulating neuron-specific alternative splicing (AS), previously associated with an acquired neurologic condition, the paraneoplastic opsoclonus-myoclonus ataxia (POMA). We report here six individuals with de novo frameshift variants in NOVA2 affected with a severe neurodevelopmental disorder characterized by intellectual disability (ID), motor and speech delay, autistic features, hypotonia, feeding difficulties, spasticity or ataxic gait, and abnormal brain MRI. The six variants lead to the same reading frame, adding a common proline rich C-terminal part instead of the last KH RNA binding domain. We detected 41 genes differentially spliced after NOVA2 …

Phosphoinositides In Retinal Function And Disease, Theodore G Wensel Apr 2020

Phosphoinositides In Retinal Function And Disease, Theodore G Wensel

Faculty, Staff and Students Publications

Phosphatidylinositol and its phosphorylated derivatives, the phosphoinositides, play many important roles in all eukaryotic cells. These include modulation of physical properties of membranes, activation or inhibition of membrane-associated proteins, recruitment of peripheral membrane proteins that act as effectors, and control of membrane trafficking. They also serve as precursors for important second messengers, inositol (1,4,5) trisphosphate and diacylglycerol. Animal models and human diseases involving defects in phosphoinositide regulatory pathways have revealed their importance for function in the mammalian retina and retinal pigmented epithelium. New technologies for localizing, measuring and genetically manipulating them are revealing new information about their importance for the …

Crisis Communications: Facing Covid-19 Together Could Lead To Positive Psychological Growth, Lowri Dowthwaite

Social Space

Although news reports of hoarding, and panic-buying might make it hard to believe, research shows that natural disasters, like the COVID-19 pandemic, can actually bring out the best in people. Although times of significant threat or crisis can cause post-traumatic stress, research shows that so-called “adversarial growth” is just as common as a response. This is our capacity to not only overcome a crisis, but to actually grow stronger, wiser and more resilient.

In Patients With End-Stage Ankle Arthritis, How Does Total Ankle Arthroplasty Compared To Arthrodesis Affect Ankle Pain And Function?, Zachary Whipple

Physician Assistant Capstones

End-stage ankle arthritis is a debilitating degenerative disease commonly located at the tibiotalar joint. The prevalence of symptomatic arthritis is about nine times lower than the rates associated with those of the knee or hip.1 Though less common than knee and hip arthritis, the US estimates greater than 50,000 new cases are reported each year.2 The most common etiology of ankle arthritis is post-traumatic pathology. Other etiologies related to ankle degeneration include primary degenerative disease, rheumatoid disease, osteonecrosis, neuropathologies, infection, and gout.3 Injury to the joint damages the intraarticular cartilage leading to changes in joint mechanics. These alterations cause the …

Integrated Multimodal Genomic Analyses Reveal Novel Mechanisms Of Glucocorticoid Resistance In Acute Lymphoblastic Leukemia, Robert J. Autry

Theses and Dissertations (ETD)

Acute lymphoblastic leukemia (ALL) is the most common childhood cancer. Much has been discovered in recent decades regarding ALL biology, and the outcome of patients with ALL has vastly improved, especially in pediatric ALL patients. Despite very promising overall cure rates, patients who relapse have a greatly decreased prognosis with survival rates ranging from 30-60%. These numbers stand to improve even further with new targeted therapies that seek to improve or maintain cure rates while reducing treatment related toxicities which affect patients both acutely and chronically. Glucocorticoids (GCs) are essential components of modern chemotherapeutic intervention for ALL. Resistance to glucocorticoids …
