The Er Retention Protein Rer1 Promotes Alpha-Synuclein Degradation Via The Proteasome, Hyo-Jin Park, Daniel Ryu, Mayur Parmar, Benoit I. Giasson, Nikolaus R. Mcfarland

HPD Articles

Abnormal accumulation of α-synuclein (αSyn) has been linked to endoplasmic-reticulum (ER) stress, defective intracellular protein/vesicle trafficking, and cytotoxicity. Targeting factors involved in ER-related protein processing and trafficking may, therefore, be a key to modulating αSyn levels and associated toxicity. Recently retention in endoplasmic reticulum 1 (RER1) has been identified as an important ER retrieval/retention factor for Alzheimer's disease proteins and negatively regulates amyloid-β peptide levels. Here, we hypothesized that RER1 might also play an important role in retention/retrieval of αSyn and mediate levels. We expressed RER1 and a C-terminal mutant RER1Δ25, which lacks the ER retention/retrieval function, in HEK293 and …

Although membrane proteins represent most therapeutically relevant drug targets, the availability of atomic resolution structures for this class of proteins has been limited. Structural characterization has been hampered by the biophysical nature of these polytopic transporters, receptors, and channels, and recent innovations to in vitro techniques aim to mitigate these challenges. One such class of membrane proteins, the ATP-binding cassette (ABC) superfamily, are broadly expressed throughout the human body, required for normal physiology and disease-causing when mutated, yet lacks sufficient structural representation in the Protein Data Bank. However, recent improvements to biophysical techniques (e.g., cryo-electron microscopy) have allowed for previously …

The Effects Of Fetal Alcohol Exposure On Gene Expression And Associated Behavioral Markers In Sprague Dawley Rats, Sarah Willey

Biology Honors Papers

Alcohol is a dangerous recreational substance because of its availability and the economic, social, and biological problems that it can cause. Fetal alcohol exposure, although preventable, is particularly harmful to a developing embryo. This exposure often causes fetal alcohol spectrum disorder (FASD), the symptoms of which include learning disabilities and psychological hardships with comorbidities such as anxiety, depression, and attention deficit disorder. One of the main teratogenic effects of alcohol is the effect on epigenetic modifications and altered gene expression during development. These modifications involve changes in the methylation and condensation of DNA that then alters the expression of specific …
