Crizotinib is a small molecule tyrosine kinase inhibitor that is currently being marketed by Pfizer under the trade name Xalkori®. After its development in 2005, an appropriate target was discovered in 2007. By 2011 the FDA had approved crizotinib for use in patients with ALK+ non small cell lung cancer (NSCLC). ALK+ NSCLC is caused by a genetic mutation that truncates and flips a section of the EML4 gene and attaches it to a section of the ALK gene. The result is the EML4-ALK oncogene. The resulting protein from this mutation is capable of undergoing ligand independent dimerization, …

Tempat pembuangan akhir (TPA) sampah domestik dengan sistem penampungan terbuka sangat berisiko mencemari air tanah milik warga yang bermukim di sekitarnya melalui proses perlindian. Untuk mengetahui kandungan logam berat dalam air tanah di sekitar TPA, sebanyak 68 sampel air sumur gali (45 sumur Dusun I dan 23 sumur Dusun IV) dari Desa Namobintang Kecamatan Pancurbatu Kabupaten Deli Serdang, Sumatera Utara, telah dianalisis dengan inductively couple plasma atomic emission spectroscopy. Hubungan jarak sumur dengan konsentrasi kromium, merkuri, dan timbal diuji dengan Mann-Whitney, Spearman’s Correlation dan analisis regresi linier sederhana. Pada tahun 2009, dilakukan penelitian deskriptif di Kecamatan Jatibarang dan Kecamatan Kedokan Bunder untuk mengetahui faktor-faktor sanitasi lingkungan, dan perilaku ibu-ibu dan kejadian penyakit infeksi pada bayi dan anak. Data dikumpulkan menggunakan kuesioner dengan responden ibu rumah tangga yang mempunyai bayi/ anak balita berjumlah 401 orang. Caveolae are cell membrane invaginations that are highly abundant in adipose tissue, endothelial cells and the lung. The formation of caveolae is dependent on the expression of various structural proteins that serve as scaffolding for these membrane invaginations. Cavin1 is a newly identified structural protein whose deficiency in mice leads to loss of caveolae formation and to development of a lipodystrophic phenotype. In this study, we sought to investigate the functional role of Cavin1 in the lung. Cavin1 deficient mice possessed dramatically altered distal lung morphology and exhibited significant physiological alterations, notably, increased lung elastance. The changes in distal lung …

Cerebral palsy, the most common cause of physical childhood disability, is a non-progressive disorder that results from an anoxic brain injury to the fetal or newborn brain. Kabupaten Kulonprogo merupakan salah satu daerah endemis malaria di Daerah Istimewa Yogyakarta dan Jawa Tengah. Lonjakan jumlah kasus malaria terjadi hampir dua kali lipat sejak tahun 2010 hingga bulan Agustus 2011. Penelitian ini bertujuan untuk mengetahui jenis Plasmodium pada penyakit malaria. Penelitian eksplorasi ini menggunakan desain studi cross sectional. Diagnosis kasus malaria diperiksa menggunakan pemeriksaan darah tebal secara mikroskopis dan untuk mengetahui faktor sosiodemografi dilakukan wawancara menggunakan kuesioner. Computationally driven drug design/discovery efforts generally rely on accurate assessment of the forces that guide the molecular recognition process. HINT (Hydropathic INTeraction) is a natural force field, derived from experimentally determined partition coefficients that quantifies all non-bonded interactions in the biological environment, including hydrogen bonding, electrostatic and hydrophobic interactions, and the energy of desolvation. The overall goal of this work is to apply the HINT-based atomic level description of molecular systems to biologically important proteins, to better understand their biochemistry – a key step in exploiting them for therapeutic purposes. Background and purpose: Strength and range of motion of the foot and ankle have been shown to be related to measures of balance and fall risk in older adults. The primary purpose of this pilot investigation was to evaluate the feasibility of a 6-week home-based exercise program focusing on the foot and ankle and any associated changes in balance, muscle performance and range of motion in older adults. Methods: This single-group repeated measures study involved a convenience sample of 21 healthy communitydwelling older adults age 60-90. Objectives

A community-based intervention is described that targets oral health self-care practices among Hispanic children in the United States and is being tested in an ongoing trial. Descriptive results of baseline oral health variables are presented.

Methods

As of January 2013, 284 Hispanic children of ages 5–7 enrolled in the Healthy Families Study in Nashville, TN, USA. Families are randomized to one of two culturally appropriate interventions.

Results

At baseline, 69.6 % of children brushed at least twice daily, and 40.6 % brushed before bed daily. One-third of parents did not know if their children’s toothpaste contained fluoride.

Previous research has shown that embedded questions hurt the reading comprehension of high comprehenders. All participants had high reading comprehension skills scoring at or above 66 percent on the ACT/SAT. Participants either read a packet with questions embedded about every paragraph or a packet where questions were all located at the end of the reading. Participants answered the questions as they came to them. After a week of delay, the participants came back and took a test with questions that were either: target, non-target, or related. The purpose of this project was to assess first semester freshman college students’ opinions about smoking, secondhand smoke exposure, and smoking behaviors on a smoke-free campus. This research used an online cross-sectional survey. For two years, surveys were emailed via Zoomerang to all first semester freshmen students at Valparaiso University. Survey questions contained 60 forced-choice or open-ended options. Data were analyzed using descriptive statistics. A total of 630 students responded. Subjects were primarily female (58.4%) and Caucasian (84.9%). Twenty-five percent of the respondents reported being exposed to secondhand smoke in their environments. Introduction: According to the Institute of Medicine (IOM), over 90 million people in the U.S. have difficulty understanding and using health information including medication labels. Purpose: To determine how individuals interpret medication labels and the relationship between medication label interpretation and level of health literacy. Oxycodone (OXY) is one of the most commonly abused opiates during pregnancy. Perinatal opiate exposure (POE) is associated with neurobehavioral and hormone changes. Little is known about the effects of perinatal OXY on the cardiovascular (CV) responses to stress.

Objectives: to determine the effects of POE on: (1) CV responses to acute stress and ability to discriminate using a classical conditioning paradigm; (2) changes in CV response to the paradigm and retention of the ability to discriminate from postnatal day (PD) 40 to young adulthood.

To improve relatively poor outcomes for locally-advanced lung cancer patients, many current efforts are dedicated to minimizing uncertainties in radiotherapy. This enables the isotoxic delivery of escalated tumor doses, leading to better local tumor control. The current dissertation specifically addresses inter-fractional uncertainties resulting from patient setup variability. An automatic block-matching registration (BMR) algorithm is implemented and evaluated for the purpose of directly localizing advanced-stage lung tumors during image-guided radiation therapy. In this algorithm, small image sub-volumes, termed “blocks”, are automatically identified on the tumor surface in an initial planning computed tomography (CT) image. Drug safety and quality is an essential assumption of clinical medicine, but there is growing concern that this assumption is not always correct. Poor manufacturing and deliberate fraud occasionally compromises the drug supply in the United States, and the problem is far more common and serious in low- and middle-income countries with weak drug regulatory systems. An Institute of Medicine consensus committee report identified the causes and possible solutions to the problem of falsified and substandard drugs around the world.

Introduction:

Microwave tomography recovers images of tissue dielectric properties, which appear to be specific for breast cancer, with low-cost technology that does not present an exposure risk, suggesting the modality may be a good candidate for monitoring neoadjuvant chemotherapy.

Methods:

Eight patients undergoing neoadjuvant chemotherapy for locally advanced breast cancer were imaged longitudinally five to eight times during the course of treatment. At the start of therapy, regions of interest (ROIs) were identified from contrast-enhanced magnetic resonance imaging studies. OBJECTIVE: Randomized controlled trials have shown that growth hormone (GH) therapy has effects on growth, metabolism, and body composition. GH therapy is prescribed for children with growth failure and adults with GH deficiency. Carefully conducted observational study of GH treatment affords the opportunity to assess long-term treatment outcomes and the clinical factors and variables affecting those outcomes, in patients receiving GH therapy in routine clinical practice.

DESIGN: The NordiNet® International Outcome Study (IOS) and the American Norditropin® Studies: Web Enabled Research (ANSWER Program®) are two complementary, non-interventional, observational studies that adhere to current guidelines for pharmacoepidemiological data.

Perceptions of children restrained and secluded (R&S) were explored using Yin's methodology and critical theory. The literature presented reviewed proponent's and opponent's views of the R&S intervention used on inpatient psychiatric units for the containment of violent patients. Despite reports of physical and emotional injury to staff and patient during restraining and secluding children, the intervention is still in use. Using Machover's framework, a semi-structured interview and drawings were used to illicit information from a 12 year old boy about his perceptions of R&S. Timely detection and follow-up of abnormal cellular changes can aid in early diagnosis of breast cancer, thus leading to better treatment outcomes. However, despite substantial breast cancer screening initiatives, the proportion of female breast cancer cases diagnosed at late stages remains high. Distance to screening clinics may affect access to care, particularly for women living in impoverished areas with limited means of reliable transportation. Journal of Health Disparities Research and Practice

This study examines the relationship between racial/ethnic discrimination in medical settings, distrust in conventional medicine, and attitudes toward complementary and alternative medicine (CAM) among a racially/ethnically diverse sample. We also investigate how this relationship differs by nativity. Data are from a 2008 statewide stratified sample of publicly insured adults in Minnesota (N=2,194). Discrimination was measured as self-reported unfair treatment in medical settings due to race, ethnicity, and/or nationality. Outcomes are trust in conventional providers/medicine and attitudes toward CAM modalities. The purpose of this pilot study was 1) to modify the Diabetes Prevention Program protocol collaboratively with home care nurses, health professionals, and residents of public housing: 2) to evaluate the feasibility (reach and implementation) and the preliminary efficacy of the modified diabetes prevention program (mDPP) provided by home care nurses to adults at-risk for T2D on clinical, behavioral, and psychosocial outcomes compared to an enhanced standard care control group.
