Research documents that breast cancer is the leading cause of death in Latina females. The exact numbers are unknown, but studies reveal that Latinas with breast cancer underuse recommended follow-up chemotherapy, decreasing their rates of survival. Although several factors may be responsible, cultural influences are a possible barrier. However, there is a gap in the literature about how culture affects decisions about breast cancer treatment. This focused ethnographic study examined the role of cultural beliefs and perceptions in the decision-making process for Latina women about whether or not to receive chemotherapy following a breast cancer diagnosis. BACKGROUND: The polyglutamine expansion in huntingtin (Htt) protein is a cause of Huntington's disease (HD). Htt is an essential gene as deletion of the mouse Htt gene homolog (Hdh) is embryonic lethal in mice. Therefore, in addition to elucidating the mechanisms responsible for polyQ-mediated pathology, it is also important to understand the normal function of Htt protein for both basic biology and for HD. The purpose of this study is to explore whether storybooks designed to elicit talk about letters and sounds, termed "alphabet-storybooks" will generate more print referencing behaviors from Head Start teachers than traditional storybooks, and if there is a concomitant positive impact on the learning of the children in these classrooms. In addition, the meaning reference behaviors of adults and impact on children also will be measured to determine if meaning is sacrificed at the expense of print referencing.

Fasciolid flukes are among the largest and best known digenetic trematodes and have considerable historical and veterinary significance. Fasciola hepatica is commonly implicated in causing disease in humans. The origins, patterns of diversification, and biogeography of fasciolids are all poorly known. We have undertaken a molecular phylogenetic study using 28S, internal transcribed spacer 1 and 2 (ITS-1 and ITS-2) of nuclear ribosomal DNA, and mitochondrial nicotinamide dehydrogenase subunit 1 (nad1) that included seven of the nine recognized species in the family. The fasciolids examined comprise a monophyletic group with the most basal species recovered from African elephants. We hypothesize fasciolids …

Equine bone marrow-derived mesenchymal stromal cells (MSCs) and adipose tissue-derived stromal cells (ASCs) were compared for frequency within their respective tissues, cell doubling characteristics and differentiation multipotential in culture based on histochemical staining and compositional analysis. Equine MSCs and ASCs from young adult horses were harvested and isolated from sternal bone marrow and supragluteal subcutaneous adipose tissue, respectively, and grown up to passage 10 (P10) to determine cell doubling characteristics. The study purpose was to examine the effects of a perioperative music intervention on changes in mean arterial pressure (MAP), heart rate (HR), anxiety, and pain in women with a diagnosis of breast cancer undergoing mastectomy surgery. The convenience sample included 30 women between the ages of 42 and 70 who were undergoing mastectomy in a large urban hospital in the southern United States. Exclusion criteria included male gender, psychiatric disorder, use of psychotropic medications, cognitive mental disability, and American Society of Anesthesiologists (ASA) status 4 or greater. Increased levels of protein O-linked-N-acetylglucosamine (O-GlcNAc) have been correlated with increased cell survival following stress. Therefore the goal of this study was to determine whether enhanced flux through the hexosamine biosynthesis pathway (HBP), which leads to elevated levels of O-GlcNAc, improved the tolerance of isolated neonatal rat ventricular myocytes (NRVMs) to ischemia/reperfusion (I/R) injury. NRVMs were exposed to hypoxic medium for 4 hours, followed by oxygenated complete medium for 16 hours. Acute treatment with 5 mM glucosamine showed an increase in viability and decreased both necrosis and apoptosis following I/R. Glucosamine increased O-GlcNAc levels maximally at 2 hours and went …

This study was conducted in conjunction with the Birmingham Water Works Board to evaluate Kinetico ceramic filter media against GAC (Granular Activated Carbon) and Sand/Anthracite filters. The Birmingham Water Works Board Water Treatment Pilot Plant was located and operated on the Shades Mountain Filter Plant (SMFP) premises. The evaluation was conducted in three phases: plant matching, plant optimization, and pre-filter testing. A cross-sectional study was conducted to investigate the relationship of child, family, and medical home status to quality of life for mothers of children with and without special health care needs. Mothers of children with (n=100) and without (n=80) special health care needs, completed a questionnaire at a children’s hospital in the southeastern United States focusing on the child, family, and medical home and maternal quality of life. Standardized instruments were used to measure quality of life (SF-36) and social support (Social Support Questionnaire). The closely related bacterial species Xenorhabdus nematophila, Yersinia pestis and Y. pseudotuberculosis make biofilms capable of adhering to the head of the model nematode Caenorhabditis elegans. Y. pestis uses biofilms to block the digestive tract of its vector, the flea, in order to enhance its efficiency of transmission. I investigated the role of biofilms in X. nematophila and found that, like its Yersinia sp. counterparts, production of biofilm requires a four-gene operon: hmsHFRS. X. nematophila is an insect pathogen and an obligate symbiont of the nematode Steinernema carpocapsae. However, an X. nematophila hmsH mutant that failed to make biofilms on …

The regulation of signaling and migration is critical to B lymphocyte development, activation, and proliferation. This dissertation reports a functional role for Fc receptor-like molecule 5 (FCRL5), a B cell specific member of the Fc receptor-like family of molecules, and the discovery of a novel family of conserved transmembrane proteins with fibronectin, immunoglobulin, and leucine-rich repeat domains (FIGLER) expressed in bone marrow and B lineage cells. HCMV consists of a dsDNA genome enclosed by, an icosahedral capsid surrounded by a layer of tegument proteins; the virion structure is enclosed in a lipid envelope rich in virus expressed glycoproteins. The HCMV envelope contains an incompletely defined number of glycoproteins. The glycoprotein M (gM) and glycoprotein N (gN) form a gM/gN protein complex which is the most abundant protein component of the HCMV envelope. Interleukin-8 (IL-8) is a potent chemoattractant of numerous cells, particularly neutrophils, in the innate immune response. In addition to immune functions, IL-8 is known to contribute to the pathogenesis of a number of diseases, including cancer. Interferon-β (IFN-β), a Type I interferon, inhibits the expression of IL-8, but the details of this effect are not known. In this study we investigate transcriptional control of the IL- 8 gene and the mechanism by which IFN-β exerts inhibitory effects on IL-8. PURPOSE: This study compares vendor reagents used for cytometric bead-based assay (CBA) of cytokines in human tears and validates a protocol for use in evaluating punctal occlusion, a treatment for keratoconjunctivitis sicca. It has been proposed that punctal occlusion may alter tear cytokine concentrations based on the facts that: (1) tear clearance, a measure of the balance between tear production and elimination, declines following occlusion and (2) reduced tear clearance and increased levels of pro-inflammatory tear cytokines are correlated in dry eye patients. Phase III trials often have both efficacy and safety outcomes. Sometimes the safety outcomes are closely associated with the efficacy outcomes. We sought to find a statistical method to treat both efficacy and safety outcomes as bivariate primary outcomes and set up stopping rules at interim stages based on the following criteria: (1) greater safety; (2) greater efficacy; (3) futility. Regulated mRNA decay is a highly important process for the tight control of gene expression. Inherently unstable mRNAs contain AU-rich elements (AREs) in the 3’ untranslated regions that direct rapid mRNA decay by interaction with decay-promoting ARE-binding proteins (ARE-BPs). The decay of ARE-containing mRNAs is regulated by signaling pathways, which are believed to directly target ARE-BPs. Here, we show that BRF1 involved in ARE-mediated mRNA decay (AMD) is phosphorylated by MAPK-activated protein kinase 2, MK2. Exposure to ambient outdoor air pollution exposure is one of the many extraneous factors associated with preterm births. Many epidemiological studies involving preterm births and ambient air pollution have repeatedly indicated positive associations. However, for many such studies, results may be statistically significant but weakly associated or inconclusive. Overexpresssion of transforming growth factor (TGF)-β has been implicated in promoting immune suppression, tumor angiogenesis, tumor cell migration, and invasion in many cancers including carcinoma of the breast. Thus, targeted downregulation of TGF-β1 expression in breast cancer in situ and determination of its implications potentially could provide new treatment approaches for disease management. siRNA constructs targeting TGF-β1 were validated and used to develop clonal derivatives of the MDA-MB-435 metastatic breast cancer cell line. In the nervous system, PI 3-kinase has been implicated in neuronal differentiation. My studies have focused on a candidate neuronal PI 3-kinase target centaurin alpha-1, which binds PtdIns(3,4,5)P3, and is an Arf6 GAP. Centaurin alpha-1 is localized in dendrites, dendritic spines and synapses, and is required for neuronal differentiation and spine morphogenesis. In dissociated neuronal cultures, expression of centaurin alpha-1 enhances dendritic branching, and increases dendritic filopodia, lamellipodia and spine-like protrusions. Expression of centaurin alpha-1 GAP inactive mutant or knocking down centaurin alpha-1 levels using siRNA leads to inhibition of dendritic outgrowth and branching. Since an antibody to LTA has been shown to protect from Gr+ bacterial infections or colonization, and the mutant Gr+ bacteria which produce less amount of LTA have less pathogenic than WT, LTA may be an important role in Gr+ bacterial infection and inflammation. Synaptic transmission mediates the processing of incoming information, and recent studies have demonstrated that long-term changes in synaptic strength at CA1 synapses are important for learning and memory. Pancreatic adenocarcinoma (PAC) is the fourth leading cause of cancer mortality in the United States. Despite the use of highly aggressive treatment regimens (surgery, chemotherapy and radiation), almost all patients succumb to metastatic disease within 6-10 months of diagnosis. The hedgehog (HH) signaling pathway was originally discov-ered to play a critical role in mammalian embryological development. Interestingly, re-cent studies have suggested that aberrant expression of this pathway is involved in the initiation and continued growth of PAC. Childhood immunization coverage in Pakistan remains sub-optimal despite universal and free of cost availability of vaccines. Structural impediments to progress may include the immunization card that is hard to read and interpret. A randomized controlled trial was conducted to assess the effect of redesigned immunization card and center-based education to mothers on immunization completion at rural immunization centers in Karachi, Pakistan. The study purpose was to examine the effects of a 15-min live-music therapy intervention on heart rate, oxygen saturation, level of motor activity, behavioral distress, and behavioral state levels in premature infants in the neonatal intensive care unit. The convenience sample included 20 infants born at 26 to 29 weeks’ gestational age who were hospitalized in a large teaching hospital in the southern United States.
