An intact B-box 2 domain is essential for the antiretroviral activity of TRIM5α. We modeled the structure of the B-box 2 domain of TRIM5α based on the existing three-dimensional structure of the B-box 2 domain of human TRIM29. Using this model, we altered the residues predicted to be exposed on the surface of this globular structure. Most of the alanine substitutions in these residues exerted little effect on the antiretroviral activity of human TRIM5αhu or rhesus monkey TRIM5α_rh. However, alteration of arginine 119 of TRIM5α_hu or the corresponding arginine 121 of TRIM5α_rh diminished the abilities of …

The aim of this study was to determine which symptom domains in ulcerative colitis (UC) are important in the evaluation of disease activity. An important symptom is that which 1) occurs during flares, 2) improves during effective therapy, and 3) resolves during remission. Twenty eight symptom domains were evaluated. Sixty subjects were surveyed, rating each symptom on three criteria with a 100 mm Visual Analog Scale (VAS). Important symptoms were defined a priori as those whose median VAS rating for all 3 criteria was significantly greater than 50. Thirteen of the 28 symptom domains proved to be both frequent in …

Late long-term potentiation (L-LTP) denotes long-lasting strengthening of synapses between neurons. L-LTP appears essential for the formation of long-term memory, with memories at least partly encoded by patterns of strengthened synapses. How memories are preserved for months or years, despite molecular turnover, is not well understood. Ongoing recurrent neuronal activity, during memory recall or during sleep, has been hypothesized to preferentially potentiate strong synapses, preserving memories. This hypothesis has not been evaluated in the context of a mathematical model representing ongoing activity and biochemical pathways important for L-LTP. Perceptions of good parent–adolescent relationships were explored among 19 Mexican American high school students aged 14–17 who participated in focus group interviews on what it means for Mexican American teenagers to have good relationships with parents. Using a grounded theory approach, five general themes emerged in the responses, corresponding to open communication, instrumental and emotional support, indirect expressions of caring, parental control, and valued relationship qualities. Both genders described distinct relationships with mothers and fathers. Relationships with mothers were closer and more open than relationships with fathers, and mothers were seen as being more affectionate, lenient, and emotionally supportive, whereas …

Project ACT is a randomized controlled trial designed to test the effectiveness of a non-pharmacological home-based intervention to reduce behavioral and psychological symptoms of dementia (BPSD) and caregiver distress. The study targets 272 stressed racially diverse family caregivers providing in-home care to persons with moderate stage dementia with one or more behavioral disturbances. All participants are interviewed at baseline, 4-months (main trial endpoint), and 6-months (maintenance). The four-month intervention involves up to 13 visits from an occupational therapist who works with families to problem-solve potential triggers (communication style, environmental clutter) contributing to behaviors, and instruct in strategies to reduce caregiver …

The objective of this paper is to explore the perceptions and experiences of providers with regard to the process of informed consent in the provision of female sterilization services in Chennai, south India. It describes the quality of information provided to clients of sterilization, providers’ attitudes toward clients and the quality of decisionmaking, as well as the quality of the consent-taking process in this setting. In addition, providers’ perceptions of the importance of informed consent are discussed. Although common in the educational and developmental areas, multilevel models are not often utilized in the analysis of data from experimental designs. This article illustrates how multilevel models can be useful with two examples from experimental designs with repeated measurements not involving time. One example demonstrates how to properly examine independent variables for experimental stimuli or individuals that are categorical, continuous, or semicontinuous in the presence of missing data. In this study we present results from a high-fidelity simulator study of Air Traffic Controller (ATCo) workload, in which routine and non-routine scenario events are examined. Specifically, we test the ability of a task load metric (Airservices Australia’s Workload Assessment Tool: WAT) to predict controller workload in these situations. In the study, sector was a between-subjects variable and task load a within-subjects variable. Twenty-one professional ATCos worked three 30-minute segments in Airservices Australia’s TAAATS simulator, and rated their workload on a scale from 1 to 10 every two minutes. Background

Routine, initial, empiric vancomycin dosing by clinicians in postoperative coronary artery bypass grafting (CABG) patients was identified as a potential patient safety issue in the Cardiovascular Intensive Care Unit (CVICU) because the rate of postoperative acute renal insufficiency (ARI) and average patient Body Mass Index (BMI) > 35 kg/m2 were significantly higher in our institution than those of the Society of Thoracic Surgeons (STS) database. A vancomycin dosing nomogram was derived from the local patient population in the attempt to improve patient safety by convincing clinicians to use an evidence-based approach to vancomycin prescription.

Cytoplasmic polyadenylation regulates mRNA stability and translation and is required for early development and synaptic plasticity. The GLD-2 poly(A) polymerase catalyzes cytoplasmic polyadenylation in the germline of metazoa. Among vertebrates, the enzyme is encoded by two isoforms of mRNA that differ only in the length of their 3′-UTRs. Here we focus on regulation of vertebrateGLD-2 mRNA. We show that the 3′-UTR of GLD-2 mRNA elicits its own polyadenylation and translational activation during frog oocyte maturation. We identify the sequence elements responsible for repression and activation, and demonstrate that CPEB and PUF proteins likely mediate repression in the resting oocyte. …

Initially, FAA regulators dismissed this technology as gaming software and hardware, but the successful collegiate use of PCATD technology for training pilots convinced the FAA in 1997 (AC 61-126) to approve the use of PCATDs as an equivalency to actual flight time, but for only a portion of that equivalency allowed Flight Training Devices (FTDs). Motor vehicle manufacturers are installing Head-Up Displays (HUDs) in motor vehicles to provide drivers with a variety of vehicle information such as vehicle speed. Despite the decade-long focus on pilot understanding of automated systems, knowledge gaps continue to suggest that current training programs are incomplete. There are several different types of knowledge that contribute to an effective understanding of automated systems, including more complex knowledge structures. Synthetic Vision Systems (SVS) create a synthetic clear-day view of the terrain in front of ownship to prevent controlled flight into terrain. To investigate how spatial biases manifest themselves in SVS displays, an experiment was conducted. Eighteen pilots made spatial judgments (relative angle, distance, height, and abeam time) regarding the location of terrain points displayed in 112 5-second videos of a SVS head down display. Judgment errors revealed expected and unexpected spatial biases. Knowledge of these biases will allow SVS engineers to compensate for them and to improve spatial awareness in future SVS designs.

ABSTRACT: BACKGROUND: Shedding of the Alzheimer amyloid precursor protein (APP) ectodomain can be accelerated by phorbol esters, compounds that act via protein kinase C (PKC) or through unconventional phorbol-binding proteins such as Munc13-1. We have previously demonstrated that application of phorbol esters or purified PKC potentiates budding of APP-bearing secretory vesicles at the trans-Golgi network (TGN) and toward the plasma membrane where APP becomes a substrate for enzymes responsible for shedding, known collectively as alpha-secretase(s). The purpose of this study was to determine whether branched-chain amino acid (BCAA) supplementation attenuates indirect indicators of muscle damage during endurance exercise as compared with an isocaloric, carbohydrate (CHO) beverage or a noncaloric placebo (PLAC) beverage. Nine untrained men performed three 90-min cycling bouts at 55% VO_2peak. Subjects, blinded to beverage selection, ingested a total of 200 kcal of energy via the CHO or BCAA beverage before and at 60 min of exercise, or they drank the PLAC beverage. Yet researchers have long worried about negative effects of Teflon and the chemical compounds used to make it, perfluorooctanoic acid (PFOA): is PFOA a carcinogen? Yellow grubs give the fish's flesh a wonny, unappetizing appearance and are a problem for commercial fish fanners in that heavily infected fish are not suitable for marketing. The parasite is common in smallmouth bass (Micropterus dolomieu) living in upland streams of Arkansas where the bass may serve as a wild reservoir for contamination of commercial fish ponds. Introduction: This report analyzes how well the residents of Washington Heights/Inwood (WH/IN) have fared on selected health indicators set forth by the NYC Department of Health and Mental Hygene between 2000 and 2005.

Methods: Data on Latinos and other racial/ethnic groups were obtained from the U.S. Census Bureau American Community Survey, reorganized for public use by the Minnesota Population Center, University of Minnesota, IPUMSusa. Cases in the dataset were weighted and analyzed to produce population estimates.

Research looks for the sources of state anxiety for individual athletes prior to competition, which can be debilitating (Gould, 1993; Gould, Horn & Spreeman, 1983). Non-elite athletes who are high trait anxious (possibly fear of failure) have higher state anxiety than elite athletes (Conroy, 2002; Gould, Horn & Spreeman, 1983). Fencers may be prone to elevated state anxiety. No research links these variables together in fencing.
