The Thai Journal of Veterinary Medicine

Trifluralin, a herbicide, was observed for it's side effects when used, extralabel, as an ectoparasiticide in shrimp farming. Black tiger shrimp were exposed to trifluralin at a therapeutic level, commonly employed in shrimp farming, 0.1 or 0.2 ppm, for up to 96 hrs. Shrimp were examined 96 hrs after the primary dosage of trifluralin. The total haemocyte count in the exposed shrimp was not different from the control, unexposed shrimp. A significant reduction of muscle acetylcholinesterase was found in all exposed shrimp (p<0.05). Some of the exposed shrimp also demonstrated clinical signs of cholinergic overload characterized as abnormal movement. This study suggests sublethal effects when trifluralin is used for the treatment of shrimp ectoparasitic disease.

Tuberculosis is a term that encompasses various diseases caused by bacteria of the Mycobacterium tuberculosis complex, including M tuberculosis, M bovis, M africanum, and other mycobacterial species. Whereas M tuberculosis infection is largely spread from human to human, M bovis infection has been identified as a zoonotic disease with most cases of human infection attributable to animal sources. The mycobacteria other than tuberculosis complex (MOTT), which includes M avium subsp avium and M avium subsp intracellulare isolated from animals, has been isolated from immune-compromised humans (ie, those with human immunodeficiency virus [HIV] infection), but seldom from immunocompetent humans. Cattle are a major reservoir of Escherichia coli 0157:H7, an important zoonotic pathogen that causes hemorrhagic colitis and hemolytic uremic syndrome (HUS). Colonization of cattle occurs predominantly in the large intestine, and may especially target follicle-associated epithelium (FAE) in the terminal rectum. Bacterial colonization involves induction of attaching- effacing (A/E) lesions, mediated by type III secreted proteins and an outer membrane protein called intimin. ToxB, encoded on plasmid p0157, contributes to adherence of E. coli 0157:H7 through promotion of the production and/or secretion of type III secreted proteins. The latency-related (LR) gene of bovine herpesvirus 1 (BHV-1) is abundantly expressed during latency. A mutant BHV-1 strain that contains three stop codons at the 5′ terminus of the LR gene (LR mutant) does not reactivate from latency. This study demonstrates that the LR mutant does not express open reading frame 2 or an adjacent reading frame that lacks an initiating ATG (reading frame C). Since the LR mutant and wild-type BHV-1 express similar levels of LR RNA, we conclude that LR protein expression plays an important role in regulating the latency reactivation cycle in cattle.

Semi-quantitative analysis of the interleukin 10 (IL-10) in bronchoalveolar lavage cells (BALC), from pigs challenged with porcine reproductive and respiratory syndrome virus (PRRSV), was undertaken in this study. Twenty-one, three weeks old, crossbred pigs from a PRRSV-free commercial farm were separated into three groups, a control, non-infected group (3 pigs/group), and 2 groups (9 pigs/group), the first inoculated with a Thai-isolated European (O2SB3) strain and the second a Thai-isolated, North American (O1NP1) PRRSV strain. Cattle are an important reservoir of Escherichia coli O157:H7 leading to contamination of food and water, and subsequent human disease. This pathogen colonizes its hosts by producing several proteins such as Tir and EspA that are secreted by a type III secretion system. These proteins play a role in colonization of the intestine, suggesting that they might be useful targets for the development of a vaccine to reduce levels of this organism in cattle. Apparent prevalence, although useful as a consistent index, may underestimate the true prevalence of disease. In Michigan, the ability to estimate the true prevalence of bovine tuberculosis (TB; caused by Mycobacterium bovis) in free-ranging white-tailed deer (Odocoileus virginianus) will become increasingly important to accurately assess progress towards eradication. Our objectives were threefold: to estimate the true prevalence of M. bovis in free-ranging deer in Michigan, to evaluate the effectiveness of existing TB surveillance methods, and to indirectly assess whether TB epidemiologic data from captive cervid herds can be meaningfully extrapolated to free-ranging populations. Pathogenesis and transmission of the prion disorders (transmissible spongiform encephalopathies, TSEs) are mediated by a modified isoform of the prion protein (PrP). Prion protein gene (PRNP) alleles associated with relative susceptibility to TSE have been identified in sheep, humans and possibly elk. Comparable data have not been derived for mule deer, a species susceptible to the TSE chronic wasting disease (CWD). Initial analysis of the open reading frame (ORF) in exon 3 of the mule deer PRNP gene revealed polymorphisms in all 145 samples analyzed, with 10 potential polymorphic sites. Scrapie is a prion disease in sheep that, similar to Kuru or variant Creutzfeldt-Jakob disease (vCJD), requires both prion exposure and a genetic susceptibility. Susceptibility is defined by amino acid polymorphisms in the single copy prion protein (PrP) gene, Prnp. Specifically, in sheep homozygous for A at position 136 and R at position 154, susceptibility is defined by R or Q at position 171. The genotype ARQ/ARQ confers susceptibility, whereas (apart from a few reported exceptions) genotypes ARR/ARQ and ARR/ARR confer resistance. Similarly, a M/V polymorphism at Prnp codon 129 determines susceptibility to vCJD and Kuru,4 with partial resistance conferred …

Chronic wasting disease (CWD) status and PrP genotypes were determined for a group of 133 wild white-tailed deer in a 780 acre enclosure in western Nebraska, USA. Approximately half of the deer tested showed evidence of PrPd in the brainstem or lymphoid tissues. Four PRNP alleles encoding amino acid substitutions were identified, with substitutions at residues 95 (Q→H), 96 (G→S) or 116 (A→G), each with serine (S) at residue 138. In addition, a processed pseudogene with two alleles encoding five or six copies of the octapeptide repeat was identified in 26% of the deer. Both alleles encoded asparagine (N) …

To determine the transmissibility of transmissible mink encephalopathy (TME) agent to raccoons and to provide information about clinical course, lesions, and suitability of currently used diagnostic procedures for detection of transmissible spongiform encephalopathies (TSEs) in raccoons, 4 raccoon kits were inoculated intracerebrally with a brain suspension from mink experimentally infected with TME. One uninoculated raccoon kit served as a control. All 4 animals in the TME-inoculated group showed clinical signs of neurologic disorder and were euthanized between 21 and 23 weeks postinoculation (PI). Sections of medulla oblongata. taken at the level of the obex, palatine tonsil and medial retropharyngeal lymph node from 10,269 captive Rocky Mountain elk (Cervus elaphus nelsoni), were examined by immunohistochemical staining with monoclonal antibody for the prion protein associated with the transmissible spongiform encephalopathy of cervids, chronic wasting disease (PrPCWD). The protein was detected in 226 of them. Sheep scrapie is a prion disease that requires interaction of exogenous prions with host prion protein (PrP) supporting prion formation. Disease is associated with deposition of a host-generated conformational variant of PrP, PrPsc, in a variety of tissues, including brain, resulting in fatal spongiform encephalopathy. Efficiency of PrPsc formation is determined by polymorphisms in the PrP-coding sequence. This article adds to previous data of natural sheep scrapie, concentrating on the effect of host genotype and age on PrPsc accumulation patterns during preclinical and clinical disease. Bovine tuberculosis (BTB) was introduced into Swedish farmed deer herds in 1987. Epidemiological investigations showed that 10 deer herds had become infected (July 1994) and a common source of infection, a consignment of 168 imported farmed fallow deer, was identified (I).

As trace-back of all imported and in-contact deer was not possible, a control program, based on tuberculin testing, was implemented in July 1994. As Sweden has been free from BTB since 1958, few practicing veterinarians had experience in tuberculin testing. In this test, result relies on the skill, experience and conscientiousness of the testing veterinarian. Deficiencies in performing the …

Livestock face complex foraging options associated with optimizing nutrient intake while being able to avoid areas posing risk of parasites or disease. Areas of tall nutrient-rich swards around fecal deposits may be attractive for grazing, but might incur fitness costs from parasites. We use the example of dairy cattle and the risks of tuberculosis transmission posed to them by pastures contaminated with badger excreta to examine this trade-off. A risk may be posed either by aerosolized inhalation through investigation or by ingestion via grazing contaminated swards. We have investigated the detection, confirmation, and metabolism of the beta-adrenergic agonist ractopamine administered as Paylean to the horse. A Testing Components Corporation enzyme-linked imunosorbent assay (ELISA) kit for ractopamine displayed linear response between 1.0 and 100 ng/ml, with an 1-50 of 10 ng/ml, and an effective screening limit of detection of 50 ng/mL. The kit was readily able to detect ractopamine equivalents in unhydrolyzed urine up to 24 h following a 300-mg oral dose. Gas chromatography-mass spectrometry (GC-MS) confirmation comprised glucuronidase treatment, solid-phase extraction, and trimethylsilyl derivatization, with selected-ion monitoring of ractopamine-tris(trimethylsilane) (TMS) m/z 267, 250, 179, and 502 …

Amitraz (N′-(2,4-dimethylphenyl)-N-[[(2,4-dimethylphenyl)imino] methyl]-N-methyl-methanimidamide) is an alpha-2 adrenergic agonist used in veterinary medicine primarily as a scabicide- or acaricide-type insecticide. As an alpha-2 adrenergic agonist, it also has sedative/tranquilizing properties and is, therefore, listed as an Association of Racing Commissioners International Class 3 Foreign Substance, indicating its potential to influence the outcome of horse races. We identified the principal equine metabolite of amitraz as N-2,4-dimethylphenyl-N′-methylformamidine by electrospray ionization(+)-mass spectrometry and developed a gas chromatographic-mass spectrometric (GC-MS) method for its detection, quantitation, and confirmation in performance horse regulation. The phosphoprotein (P) of vesicular stomatitis virus (VSV) is an essential subunit of the viral RNAdependent RNA polymerase (RdRp) complex. It is phosphorylated at two different domains. Using defective interfering (DI) RNA or minigenomic RNA templates, we previously demonstrated that phosphorylation within the amino-terminal domain I is essential for transcription, whereas phosphorylation within the carboxyterminal domain II is necessary for replication. For the present study, we examined the role of the phosphorylation of residues in these domains in the life cycle of VSV. This study occurred in the Rural Academic Unit-Tiahuanaco installations, of the Bolivian Catholic University-La Paz, Bolviia, with the objective of evaluating macro- and microscopic characteristics of sperm ejaculation from bulbourethrectomized llamas. Six q'ara-variety male llamas of 3, 4, and 5 years of age were used over 8 weeks during which they were fed with natural and cultivated pastures. Ejaculate was collected with an artificial vagina with stimulation (libido) of male llamas by female llamas, for macro- and microscopic evaluation (volume, pH, color, appearance, motility, concentration, and sperm vitality). Isoxsuprine is used to treat navicular disease and other lower-limb problems in the horse. Isoxsuprine is regulated as a class 4 compound by the Association of Racing Commissioners, International (ARCI) and, thus, requires regulatory monitoring. A gas chromatography-mass spectrometry method utilizing electron impact ionization was developed and validated for the quantitation of isoxsuprine in equine plasma or equine urine. The method utilized robotic solid-phase extraction and tri-methyl silyl ether products of derivatization. Products were bis-trimethylsilyl (TMS) isoxsuprine and tris-TMS ritodrine, which released intense quantifier ions m/z 178 for isoxsuprine and m/z 236 for ritodrine that were products of C-C cleavage. …

Reviewing the mare reproductive loss syndrome (MRLS), it is proposed that the fundamental mechanism of this syndrome, which includes early fetal loss, late fetal loss, uveitis, pericarditis, and encephalitis, is tissue penetration by septic barbed setal fragments (septic penetrating setae) from Eastern tent caterpillars (Malacosoma americanum). Once ingested, these barbed setal fragments migrate through moving tissues, followed by rapid hematogenous spread of bacteria, bacterial emboli, and/or septic fragments of setae (septic penetrating setal emboli), collectively referred to as septic materials. The sex composition, age, length and body mass relationship, Von Bertalanffy growth equation and condition factor of 342 individuals were investigated to determine some population parameters of goldfish (Carassius auratus L., 1758) between June 1998 and May 1999 in Lake Eğirdir. Sex composition was 32.75 % females and 67.25 % males and the fish were 0-IV years old. The length-body mass relationship was W = 0.0210 L^{3,0598}, the Von Bertalanffy growth equation was Lt = 122.7 [1-e^{-0.033(t+3.74)}] and the average condition factor was 2.520 ± 0.012. The aim of this study was to evaluate the effects of the addition of 0.5% urea or 0.5% urea plus 4% molasses into some sorghum varieties (Gözde, P-988, Grazer and Grass-II) harvested at the dough stage on silage quality, in vitro dry matter (DM) digestibility and digestible DM yields. Triplicate silage samples from each variety and treatment group, a total of 36 silage samples, were prepared in 1 l mino-silos and incubated for 90 days. The spermatological characteristics of merino rams' semen collected with an artificial vagina (AV) and electro-ejeculator (EE) were compared. The semen was evaluated for volume (ml), mass activity (0-5, no activity to rapid wave), motility (%), sperm concentration (x 10^9/ml) and abnormal spermatozoa percentage. In this study, 212 Acanthobrama marmid Heckel, 1843 individuals were caught at monthly periods from Keban Dam Lake between January 1997 and December 1998 and transported live to the laboratory. After haematological analysis was carried out on the blood samples taken from their hearts, total lengths and weights were measured. The sex of the fish was identified and their ages were determined from the scales. The purpose of this study was to determine the presence of lice species on wild geese in the Elazığ district. Twenty wild geese were inspected for lice species between January and February in 1998 and 2001. Nine of these geese had been exposed to lice infestations and Anaticola anseris (Linnaeus, 1758) composed all the collected 92 lice. The morphological description of A. anseris was based on microscopy and microphotography pictures were taken.

In this study Cu, Zn, Pb and Cd contents were analysed in 3 molluscs (Octopus vulgaris, Sepia officinalis, Loligo vulgaris) and in one crustacean sample (Parapenaeus longirostris) caught in the Gulf of Antalya in January-March 2000. In molluscs, Cu content was determined to be between 1.82 and 6.22 mg/kg, and in crustacea, it was determined to be between 4.24 and 7.40 mg/kg. In the mollusc and crustacea specimens Zn content was 10.95-21.52 mg/kg and 11.73-14.27 mg/kg, respectively. Pb and Cd are the 2 most important heavy metals for human health.
