Chronic wasting disease (CWD) is a transmissible spongiform encephalopathy, or prion disease, that affects deer, elk, and moose. Human susceptibility to CWD remains unproven despite likely exposure to CWD-infected cervids. We used 2 nonhuman primate species, cynomolgus macaques and squirrel monkeys, as human models for CWD susceptibility. CWD was inoculated into these 2 species by intracerebral and oral routes. After intracerebral inoculation of squirrel monkeys, 7 of 8 CWD isolates induced a clinical wasting syndrome within 33–53 months. The monkeys’ brains showed spongiform encephalopathy and proteaseresistant prion protein (PrPres) diagnostic of prion disease. After oral exposure, 2 squirrel monkeys had …

Since the discovery of the Marburg and Ebola species of filovirus, seemingly random, sporadic fatal outbreaks of disease in humans and nonhuman primates have given impetus to identification of host tropisms and potential reservoirs. Domestic swine in the Philippines, experiencing unusually severe outbreaks of porcine reproductive and respiratory disease syndrome, have now been discovered to host Reston ebolavirus (REBOV). Although REBOV is the only member of Filoviridae that has not been associated with disease in humans, its emergence in the human food chain is of concern. REBOV isolates were found to be more divergent from each other than from the …

Light-Induced Fos Expression In Intrinsically Photosensitive Retinal Ganglion Cells In Melanopsin Knockout (Opn4-/-) Mice, Gary E. Pickard, Scott B. Baver, Malcolm D. Ogilvie, Patricia J. Sollars
School of Veterinary and Biomedical Sciences: Faculty Publications
Retinal ganglion cells that express the photopigment melanopsin are intrinsically photosensitive (ipRGCs) and exhibit robust synaptically driven ON-responses to light, yet they will continue to depolarize in response to light when all synaptic input from rod and cone photoreceptors is removed. The light-evoked increase in firing of classical ganglion cells is determined by synaptic input from ON-bipolar cells in the proximal sublamina of the inner plexiform layer. OFF-bipolar cells synapse with ganglion cell dendrites in the distal sublamina of the inner plexiform layer. Of the several types of ipRGC that have been described, M1 ipRGCs send dendrites exclusively into the …

Selective breeding of sheep for arginine (R) at prion gene (PRNP) codon 171 confers resistance to classical scrapie. However, other effects of 171R selection are uncertain. Ovine progressive pneumonia/Maedi-Visna virus (OPPV) may infect up to 66% of a flock thus any affect of 171R selection on OPPV susceptibility or disease progression could have major impact on the sheep industry. Hypotheses that the PRNP 171R allele is 1) associated with the presence of OPPV provirus and 2) associated with higher provirus levels were tested in an Idaho ewe flock. OPPV provirus was found in 226 of 358 …

The examination of rectoanal mucosa-associated lymphoid tissue (RAMALT) biopsy specimens for the diagnosis of transmissible spongiform encephalopathies has been described in sheep, elk, and small numbers of mule and white-tailed deer. Previous sample numbers have been too small to validate examination of this type of tissue as a viable antemortem diagnostic test. In this study, we examined RAMALT collected postmortem from 76 white-tailed deer removed from a farm in Wisconsin known to be affected by chronic wasting disease (CWD) and from 210 free-ranging white-tailed deer harvested from an area in Wisconsin where the overall prevalence of CWD among the deer …

Vaccination With Mycobacterium Bovis Bcg Strains Danish And Pasteur In White-Tailed Deer (Odocoileus Virginianus) Experimentally Challenged With Mycobacterium Bovis, M. V. Palmer, T. C. Thacker, W. R. Waters
Michigan Bovine Tuberculosis Bibliography and Database
Wildlife reservoirs of Mycobacterium bovis represent serious obstacles to the eradication of tuberculosis in domestic livestock and the cause for many faltering bovine tuberculosis eradication programs. One approach in dealing with wildlife reservoirs of disease is to interrupt inter-species and intraspecies transmission through vaccination of deer or cattle. To evaluate the efficacy of BCG vaccination in white-tailed deer, 35 deer were assigned to one of three groups; one s.c. dose of 107 CFU of M. bovis BCG Pasteur (n = 12); 1 s.c. dose of 107 CFU of M. bovis BCG Danish (n = 11); or unvaccinated deer …

The Effects Of Dietary Conjugated Linoleic Acid (Cla) On Fatty Acid Composition And Key Enzymes Of Fatty Acid Oxidation In Liver And Muscle Of Geese, Xuhui Zhang, Baowei Wang, Fangyu Long, Lei Wang, Zhigang Yang
Turkish Journal of Veterinary & Animal Sciences
The objective of this study was to determine the influence of conjugated linoleic acid (CLA) on fatty acid composition and key enzymes of fatty acid oxidation in liver and muscle of geese. A total of 192 1-day-old geese were randomly assigned to 1 of 4 dietary groups, and were fed for 56 days on diets containing 2.5% soybean oil (group A, control group) or 2.0% soybean oil with 0.5% CLA (group B), 1.0% soybean oil with 1.5% CLA (group C), and 2.5 % CLA (group D). Geese fed CLA had a decreased abdominal fat percentage (AFP) (P < 0.01) compared with the control. It was observed that dietary CLA levels had a significant effect on the body weight, weight gain, feed conversion rate (FCR) (P < 0.05), and feed intake (P < 0.01). Fatty acid composition showed a significant increase of the biologically active cis-9, trans-11 and trans-10, and cis-12 CLA isomers in both liver and muscle of geese fed CLA (P < 0.01). Dietary CLA led to an increase (P < 0.01) in saturated fatty acid (SFA) and a reduction in monounsaturated fatty acid (MUFA) (P < 0.05) concentrations in both liver and muscle. In liver, dietary CLA increased (P < 0.05) acyl CoA oxidase (ACO) activity, but did not affect carnitine palmitoyltransferase-I (CPT-I) activity compared with the control. CPT-I activity was significantly increased by 2.5% dietary CLA in muscle, where ACO activity was decreased at 1.5% CLA level. The results obtained suggested that geese can successfully incorporate CLA in both liver and muscle, which could be beneficial in the human diet, through provision of bioactive fatty acids with no detrimental effects on n-3 PUFA levels.

The Detection And Analysis Of Selected Pathogens In The Water Supply Of Dairy Cohort Herds., Niall Drummond
Theses
The issue of food safety is a growing concern to Irish agriculture and to the food industry. In the last twenty years, many novel pathogens, which can cause concern to the food industry, have been identified. Organisms such as Verocytotoxigenic Escherichia coli (VTEC) cause hemorrhagic colitis and Haemolytic Uremic Syndrome (HUS), which is the major cause of acute renal failure in children in many countries, and it’s presence in food is a cause of growing concern. Other pathogens causing concern are those implicated as causative agents of Acute Gastroenteritis (AGE) and these can be bacterial, viral and parasitic organisms.

Pyrilamine is an antihistamine used in human and veterinary medicine. As antihistamines produce central nervous system effects in horses, pyrilamine has the potential to affect the performance of racehorses. In the present study, O-desmethylpyrilamine (O-DMP) was observed to be the predominant equine urinary metabolite of pyrilamine. After intravenous (i.v.) administration of pyrilamine (300 mg/horse), serum pyrilamine concentrations declined from about 280 ng/mL at 5 min postdose to about 2.5 ng/mL at 8 h postdose. After oral administration of pyrilamine (300 mg/horse), serum concentrations peaked at about 33 ng/mL at 30 min, falling to <2 ng/mL at 8 h postdose. Pyrilamine was not detected in serum samples at 24 h postdosing by either route. After i.v. injection of pyrilamine (300 mg/horse) O-DMP was recovered at a level of about 20 ̄g/mL at 2 h postdose thereafter declining to about 2 ng/mL at 168 h postdose. After oral administration, the O-DMP recovery peaked at about 12 ̄g/mL at 8 h postdose and declined to <2 ng/mL at 168 h postdose. These results show that pyrilamine is poorly bioavailable orally (18%), and can be detected by sensitive enzyme-linked immunosorbent assay tests in urine for up to 1 week after a single administration. Care should be taken as the data suggest that the withdrawal time for pyrilamine after repeated oral administrations is likely to be at least 1 week or longer.

Industrial Copper Intoxication Of Iranian Fat-Tailed Sheep In Kerman Province, Iran, Ali Asghar Mozaffari, Amin Derakhshanfar, Jamileh Salar Amoli
Turkish Journal of Veterinary & Animal Sciences
An outbreak of industrial chronic copper poisoning in Iranian fat-tailed sheep in a region of southeast Iran is described. At least 10,000 out of 75,000 sheep (13.3%) originating from 50 flocks died over a period of 3 years in the Khatoon Abad region in Kerman province, Iran. The diagnosis was based on epidemiological, clinical, hematological, serum biochemical, urinalysis, necropsy and histopathological evaluations. Then findings were confirmed by toxicological analysis of liver and kidney samples. The details of epidemiological, clinical, laboratory and pathologic findings are described in the manuscript. These findings were indicative of chronic copper poisoning in affected sheep. Toxicological …

The expression of the calcium-binding protein S100 and its subunits \alpha and \beta in the intracellular protozoon Eimeria tenella was investigated. For this purpose, anti-S100 whole molecule polyclonal antibody and anti-S100 \alpha and anti-S100 \beta monoclonal antibodies were used on paraffin sections collected from the cecum, ileum, colon, rectum, and bursa of Fabricius of chicks experimentally infected with E. tenella. One hundred seventy-five 2- to 3-day-old specific pathogen-free chicks were used. The chicks were divided into experiment and control groups, to include 94 and 81 chicks, respectively. All the chicks in the experimental group received 10,000 spored oocytes in 0.5 …

A total of 181 Jersey cows were used to investigate how leptin gene polymorphisms affect milk production traits such as milk yield, fat and protein yield, and fat and protein content. Two single-nucleotide polymorphisms (SNPs) were genotyped, including the A59V in exon 3 and Sau3AI in intron 2. The genotype, allele, and haplotype frequencies were estimated. Statistical analysis revealed that the A59V polymorphism significantly affected the milk, protein, and fat yield (P Keywords: Leptin, SNP, Jersey cows, milk production traits

Turkish Journal of Veterinary & Animal Sciences
Fatal pulmonary aspergillosis and AA type amyloid accumulations in the liver and spleen are described in a female ostrich (Struthio camelus). The animal had had respiratory problems and long-term inappetence over an 8-week period. Necropsy revealed that several soft, grayish to white nodules ranging from 1 to 3 mm in diameter were scattered throughout the lungs and thoracic air sacs. Prominent enlargement of the liver and spleen was observed. No gross lesions were found in the other organs studied. The microscopic examination showed severe, necrotizing, granulomatous pneumonia, and air sacculitis. Aspergillus fumigatus was recovered from the lungs and air sacs …
