Plant waxes provide a marker profile of individual plants that, when sufficiently distinct, can be used to estimate the diet composition of grazing cattle. They also may provide a tool for reliably predicting feed intake. The traditional method (nonnegative least squares) to use these markers to predict diet composition has limitations. A newer statistical approach (Bayesian linear unmixing) deemed more efficient was tested with simulation. Samples from 8 forage species in Nebraska were analyzed for their plant- wax marker contents. A 2- year study evaluated the effect of corn residue baling or grazing on subsequent yields, as well as, nutrient removal by baling at five locations in Nebraska. Three treatments were applied to each field: baled, grazed, and control (no baling and no grazing). Grain and stover yields were measured by hand harvest at maturity. No differences were observed among treatments for corn yield with baled having yields of 234 bu grain / acre, grazed with 239 bu grain / acre and control with 223 bu grain / acre. Th e objective of this study was to compare a modified estrus detection and fixed time AI vs. no estrus detection and fixed time AI on subsequent pregnancy rates. Yearling heifers were estrus synchronized and AI at 72 ± 2 h after prostaglandin injection. Th is study evaluated effects of a single stocker implant (Revalor G) on growth and reproductive performance of yearling beef heifers in the Nebraska Sandhills. Crossbred heifers, grazing native Sandhills range, were randomly assigned to either be implanted 82 ± 2 days prior to estrus synchronization, or not implanted, to determine the effects of growth implants on heifer performance. Heifers were bred via artificial insemination followed with clean- up bulls. Implanted heifers gained more and were heavier at the end of the trial, but had a reduced pregnancy rate than non- implanted heifers. Implanted heifers also had a lower pregnancy …

Replacement heifers from March and May calving herds were offered ad libitum meadow hay and 4 lb/d supplement or grazed meadow and offered 1 lb/d supplement from mid- January to mid- April. Heifers fed hay gained more during the treatment; however, heifers grazing meadow experienced compensatory gain, resulting in similar body weight at pregnancy diagnosis in both calving herds. Pregnancy rates were similar between treatment groups in March and May heifers. A reduced input winter management system is a viable option to maintain pregnancy rates in early and late summer breeding seasons.

Increased probability for fecal shedding and colonization by Escherichia coli O157:H7 has been observed in previous studies when steers are fed increased inclusions of distillers grains. A feeding study was conducted to determine if distillers grains or fiber from distillers grains in the finishing ration of feedlot steers affected fecal shedding prevalence of seven serogroups of enterohemorrhagic Escherichia coli (EHEC- 7). An experiment was conducted to evaluate the effect of long- term (16 years) corn residue grazing with cattle on soil compaction, soil structure, soil organic matter, and nutrients. Three treatments: 1) fall grazing (November through January; 1.8 to 2.5 AUM/ac), 2) spring grazing (February to middle April; 2.3 to 3.1 AUM/ac), and 3) control (no grazing) under a no- till irrigated corn- soybean system in eastern Nebraska were studied. A growing study evaluated the effects of increasing supplemental rumen undegradable protein (RUP) on performance of calves fed an 85% corn silage diet. Five levels of supplementation were evaluated with 12 individually fed steers per level of supplement. Supplement levels consisted of 0, 3.25, 6.5, 9.75 and 13% supplemental RUP (% of diet DM) fed as a blend of 60% Soypass and 40% Empyreal. Increasing supplemental RUP in the diet increased ending body weight and average daily gain linearly, and decreased F:G linearly while DMI remained constant among treatments. Plant density can be changed by altering row width and/or number of plants within a row. The use of narrower rows at seeding may increase yield and reduce plant-toplant competition. Corn seeds were planted at 2 row widths (15 and 30- inch rows) and 3 plant populations (25,200, 35,000 and 54,000 seeds / acre). Contrary to popular belief, pet overpopulation is caused more by owners failing to keep their dog than with accidental or intentional breeding (Moulton et al., 1991). Factors related to relinquishment have been researched, and concluded that behavioral problems are the most reoccurring reasons observed (Bailey, 1992; Miller at al., 1996; Patronek et al., 1996; Serpell, 1966; Salman et al., 1998, 2000; Scarlett et al, 1999; New et al., 2000; Shore et al., 2003, 2005; Mondelli 2004, Blackwell et al., 2008). BVDV is an important pathogen of cattle that affects both the dairy and beef industry causing severe economic losses. The main problem of BVDV is the immune suppression that is associated with lymphoid depletion shortly post infection. The mechanism of this immune suppression is not well understood. Porcine reproductive and respiratory syndrome virus (PRRSV) is an economically devastating virus that causes a persistent infection and spreads quickly. The virus causes abortions in sows and respiratory diseases in young piglets. Current PRRSV vaccines appear to provide limited cross-protection. A virus-like particles (VLPs) vaccine is made up of key PRRSV structural proteins, which could be cross protective among the different strains. Accurate and rapid diagnostic tests are essential for the management and control of emerging infectious diseases. Therefore, the overall goals of studies presented in this dissertation were to provide improved diagnostic and surveillance capabilities for several emerging viral diseases impacting the livestock industry and to provide insight into appropriate control strategies. Recent, severe outbreaks of porcine epidemic diarrhea virus (PEDV) in Asia and North America highlighted the need for well-validated diagnostic tests for identification of PEDV infected animals and evaluation of their immune status. The purpose of this project was to define monoclonal antibodies against viral proteins from Epizootic Hemorrhagic Disease Virus (EHDV) to be used in quality control of vaccines produced by an industrial partner, and development of tools to identify EHDV. EHDV is an Orbivirus, which also includes Bluetongue Virus and African Horse Sickness, which are all transmitted through arthropod vectors. EHDV causes significant morbidity and mortality in white-tailed deer, but has recently been found to infect cattle. Senecavirus A (SVA) is a positive sense single-stranded virus that belongs to the genus Senecavirus within the family Picornaviridae. Senecavirus has been present in US swine population since 1988. However, there have been only a few reports of SVA associated with disease in last two decades. In late 2014 early 2015, a sudden increase in the number of cases of Senecavirus A associated with vesicular disease was observed in Brazil, the US and China. The purpose of the study was to use decision trees to predict increased levels of somatic cells in cow's milk. The material for the study comprised data collected in 2012-2014 from five farms in Poland equipped with an automatic milking system. Data on 803 Polish Holstein-Friesian cows were collected. In order to predict somatic cell count data mining techniques were used to build a graphical model of a decision tree. Thirty Holstein calves presented with infectious diarrhea during the second week following birth were utilized to evaluate the effects of a standardized product containing a combination of probiotics, ß-glucans, and glyconutrients (GLY) in support of antibiotic therapy during the early rearing phase (day 7 to day 45 following birth). Treatments were 1) antibiotic therapy (trimethoprim and sulfas; ANTIB), and 2) ANTIB plus 5 g/calf per day GLY. Treatments did not affect diarrhea duration, averaging 16.7 days. However, GLY support decreased (P < 0.05) the mortality rate and increased (P < 0.05) live weight gain. Increased weight gain was attributable to enhanced (P < 0.05) consumption of calf starter. Supplemental GLY increased (P < 0.05) serum glucose and lowered (P < 0.05) serum urea nitrogen, nonesterified fatty acids, and cortisol concentration. We conclude that daily provision of 5 g of a standardized product containing probiotics, ß-glucans, and glyconutrients to diarrheic calves under antibiotic therapy will promote health and enhance early growth performance.

Mitochondria play a fundamental role in the regulation of cell death during accumulation of oxidants. High concentrations of atmospheric oxygen (hyperoxia), used clinically to treat tissue hypoxia in premature newborns, is known to elicit oxidative stress and mitochondrial injury to pulmonary epithelial cells. A consequence of oxidative stress in mitochondria is the accumulation of peroxides which are detoxified by the dedicated mitochondrial thioredoxin system. This system is comprised of the oxidoreductase activities of peroxiredoxin-3 (Prx3), thioredoxin-2 (Trx2), and thioredoxin reductase-2 (TrxR2). Intrauterine growth restriction (IUGR) is a leading cause of perinatal morbidity and mortality. Low birth weight resulting from preterm birth and/or IUGR is an underlying factor in 60–80% of perinatal death worldwide, and is particularly common in developing countries (UNICEF, 2008). Furthermore, studies have linked IUGR and the associated fetal malnutrition to increased incidence of metabolic syndrome in adult life (Barker et al., 1993; Godfrey and Barker, 2000). The “thrifty phenotype hypothesis” developed by David Barker (Hales et al., 1991) states that IUGR-associated fetal malnutrition forces the fetus to spare nutrients by altering tissue-specific metabolism in order to survive. Poxviruses have evolved multiple strategies to subvert signaling by Nuclear Factor κB (NF-κB), a crucial regulator of host innate immune responses. Here, we describe an orf virus (ORFV) virion-associated protein, ORFV119, which inhibits NF-κB signaling very early in infection (≤30 min post infection). ORFV119 NF-κB inhibitory activity was found unimpaired upon translation inhibition, suggesting that virion ORFV119 alone is responsible for early interference in signaling. A C-terminal LxCxE motif in ORFV119 enabled the protein to interact with the retinoblastoma protein (pRb) a multifunctional protein best known for its tumor suppressor activity. Equine foot pain can be a result of many conditions and disease processes, all of which lead to a loss of function in the horse. When the horse loads the foot, pressure applied to the frog translates to the digital cushion and lateral cartilages, compressing the blood vessels, perfusing the area, and circulating blood back up the leg; this constant flow of blood stimulates growth of the hoof wall. Estrous synchronization and artificial insemination (ESAI) are reproductive technologies that cattlemen can use to improve the reproductive performance of their herds. Controlling the gender ratio of the calf-crop can also improve the opportunity for increased revenue and profit. Articular cartilage in mammals has a limited intrinsic capacity to repair structural injuries and defects, a fact that contributes to the chronic and progressive nature of osteoarthritis. Current treatment modalities do not enable articular cartilage to achieve a complete and permanent restoration of normal structure and function with large or partial thickness lesions. Progesterone in the mare declines to almost undetectable concentrations in late gestation. It’s metabolized into several pregnanes, some circulating at very high concentrations. Although the function of many pregnanes remains unclear, 5α-dihydroprogesterone and allopregnanolone are bioactive. Measurements of pregnanes in late gestation are typically by immunoassay, although results are confounded by cross-reactivity with related pregnanes. Conversely, liquid chromatography tandem mass spectrometry (LC-MS/MS) allows differentiation of individual pregnanes. The mare experiences a transient innate immune response to breeding, the resolution of which is crucial for optimal fertility. The majority of mares are able to modulate this inflammation in a timely fashion, but a subpopulation exists which fail to do so and are considered susceptible to persistent breeding-induced endometritis (PBIE). Seminal plasma has been shown to modulate aspects of this inflammation. Recently, two seminal plasma proteins have garnered interest for their immune modulating properties: cysteine-rich secretory protein-3 (CRISP-3) and lactoferrin. Equine protozoal myeloencephalitis (EPM), caused by the protozoan parasite Sarcocystis neurona, is one of the most important neurological diseases of horses in the Americas. While seroprevalence of S. neurona in horses is high, clinical manifestation of EPM occurs in less than 1% of infected horses. Factors governing the occurrence and severity of EPM are largely unknown, although horse immunity might play an important role in clinical outcome.
