Chronic lymphocytic leukemia (CLL) is a heterogeneous disease characterized by the accumulation of mature CD5+ B-cells in the peripheral blood, bone marrow, and secondary lymphoid tissues (e.g., spleen and lymph nodes).Despite the efficacy of front-line therapies, CLL is still an incurable disease, highlighting the need for development of novel therapeutics and further study of resistance mechanisms. Within the lymph node CLL tumor microenvironment (TME), there is an upregulation of gene signatures associated with B-cell receptor (BCR) and downstream nuclear factor kappa B (NF-κB) signaling compared to CLL cells found within the blood or bone marrow niches. In this study, we investigated the color-changing capabilities of Trinidadian killifish, Anablepsoides hartii, through exposure to varied color conditions over defined periods of time. Our experiment aims to understand adaptive responses when killifish are subjected to different color backgrounds, exploring their mechanisms to change color. By subjecting the species to different environmental color variations, we sought to discern color change patterns, which could indicate adaptive responses related to camouflage, stress, behavior clues, and physiological adjustments. Few of the world's rivers remain free-flowing for over 1000 km, significantly impacting anadromous fish populations due to dam-induced habitat fragmentation. Three partial migration barriers (low-head dams) on North Carolina's Cape Fear River (USA)—lock and dam 1 (LD1), lock and dam 2 (LD2), and lock and dam 3 (LD3)—impact American shad Alosa sapidissima, Atlantic sturgeon Acipenser oxyrinchus oxyrinchus, and striped bass Morone saxatilis populations. Mitigation strategies to pass fish upstream have included conservation locking, nature-like fishway construction, and environmental flows (e-flows). Pancreatic Neuroendocrine Tumors (PanNETs) are the most common and lethal neuroendocrine malignancies where treatments used in advanced patients have limited efficacy, adverse side effects, and acquire resistance. Thus, there is a critical need to uncover novel precision therapeutics for PanNET patients. Additionally, pre-clinical models that more accurately represent disease are an urgent necessity for translational studies.
This dissertation directly addresses these challenges by identifying histone lysine methyltransferase (KMT) NSD3 as a critical oncogenic driver of PanNETs through di-methylation of histone H3K36 (H3K36me2).

Methamphetamine abuse amongst adolescents has been on the rise in the United States. In this study, we assessed the effects of manipulation of KORs on methamphetamine-induced behavioral sensitization in adolescent male and female rats. Methamphetamine-induced behavioral sensitization is an animal model to study methamphetamine addiction. Behavioral sensitization usually occurs in animals who have been treated with methamphetamine followed by a period of withdrawal. It is hypothesized that brain circuits are altered during the period of withdrawal after methamphetamine exposure resulting in sensitized (increased) responses to methamphetamine at the end of the withdrawal period. How traits at multiple levels of biological organization evolve in a correlated fashion in response to directional selection is poorly understood, but two popular models are the very general “behavior evolves first” (BEF) hypothesis and the more specific “morphology-performance-behavior-fitness” (MPBF) paradigm. Both acknowledge that selection often acts relatively directly on behavior and that when behavior evolves, other traits will as well but most with some lag. However, this proposition is exceedingly difficult to test in nature. Studying the involvement of nicotinic acetylcholine receptors (nAChRs), specifically α7-nAChRs, in neuropsychiatric brain disorders such as autism spectrum disorder (ASD) has gained a growing interest. The flavonoid apigenin (APG) has been confirmed in its pharmacological action as a positive allosteric modulator of α7-nAChRs. Effects of cadmium and obesity on accumulation of essential metals in mouse kidney were evaluated. Starting in utero and continuing until 24 weeks postweaning, male/female mice were exposed to cadmium (0, 0.5, or 5 ppm) and were fed on Normal Diet (ND)/High-Fat Diet (HFD) post weaning. Sudden unexpected death in epilepsy (SUDEP), the primary cause of mortality in epilepsy, remains poorly understood. Studies suggest seizures may trigger dangerous signals affecting the heart and lungs leading to collapse and death. The Kv1.1 deficiency mouse model mirrors clinical SUDEP cases, showing spontaneous seizures, cardiorespiratory issues, and premature death. However, this model lacks regional specificity in Kv1.1 deletion, hindering insights into SUDEP’s mechanisms and anatomical substrates.

For this research project, we studied three separate pastures containing Fort Hays State University’s Holstein cows. One pasture was our control group, with no horn fly protection applied. Another pasture had cattle that were fed with garlic in their mineral salts, an all-natural horn fly prevention method. Our final pasture contained cows sprayed with a larvicide. This was a single-blind study, meaning I did not know which pasture had what treatment, but my professor knew. Background
Biologic TNF-α inhibitors (bTNFIs) can block cerebral TNF-α in Alzheimer’s disease (AD) if these macromolecules can cross the blood–brain barrier (BBB). Thus, a model bTNFI, the extracellular domain of type II TNF-α receptor (TNFR), which can bind to and sequester TNF-α, was fused with a mouse transferrin receptor antibody (TfRMAb) to enable brain delivery via BBB TfR-mediated transcytosis. Previously, we found TfRMAb-TNFR to be protective in a mouse model of amyloidosis (APP/PS1) and tauopathy (PS19), and herein we investigated its effects in mice that combine both amyloidosis and tauopathy (3xTg-AD).

The human gastrointestinal system is a complex ecosystem crucial for well-being. During sepsis-induced gut injury, the integrity of the intestinal barrier can be compromised. Lipopolysaccharide (LPS), an endotoxin from Gram-negative bacteria, disrupts the intestinal barrier, contributing to inflammation and various dysfunctions. The current study explores the protective effects of limonene, a natural compound with diverse biological properties, against LPS-induced jejunal injury in mice. Oral administration of limonene at dosages of 100 and 200 mg/kg was used in the LPS mouse model. It is so important to explore the potential side effects of antidepressants especially if these side effects could lead to a future detriment to an individual’s financial and social well-being. A better understanding of these side effects in regards to human developmental issues is imperative considering issues related to mental health and depression affect a large percentage of the population. In this study, we tested if Caenorhabditis (C.) elegans could be a potential model organism to predict possible side effects associated with select antidepressants. Their developmental problems potentially encountered upon drug treatment may be encountered in humans.

Drone flies, Eristalis tenax (Diptera: Syrphidae), are important generalist pollinators and visit flowers globally that range widely in color. The flies’ photoreceptors allow them to sense light wavelengths between 300-600 nm and E. tenax exhibit a positive phototactic response. To understand the effects of light on E. tenax movement, we conducted two-choice behavioral tests to determine their phototactic response to different wavelengths of light across the spectrum (ultraviolet to red light, plus full spectrum white light). The use of unmanned aerial vehicles (UAVs) for agricultural and environmental research has increased markedly in recent years, and a key application has been to conduct damage assessments. Invasive wild pigs (Sus scrofa) cause both agricultural and environmental damage, and past studies have utilized UAVs to quantify this damage. Each methodology has advantages, but they also have drawbacks such as high rates of damage omission or reliance upon a near-infrared camera, which may be prohibitively expensive for some land managers and scientists. Non-small cell lung cancer (NSCLC) heterogeneity is a major challenge for determining effective treatment strategies. Adenocarcinomas (ADCs) and squamous cell carcinomas (SCCs) are histologically and epigenetically distinct subtypes of NSCLC. Patients with ADC tumors harboring mutations in both KRAS and LKB1 (aka STK11) have lower survival rates than those with KRAS-only tumors. KRAS/LKB1 tumors are not only aggressive, but also respond poorly to immunotherapy. The Wnt/β-catenin pathway’s significance in cancer initiation, progression, and stem cell biology underscores its therapeutic potential. However, the clinical application of Wnt inhibitors remains limited due to challenges posed by off-target effects and complex cross-talk of Wnt signaling with other pathways. In this study, we leveraged a zebrafish model to perform a robust and rapid drug screening of 773 FDA-approved compounds to identify Wnt/β-catenin inhibitors with minimal toxicity. Utilizing zebrafish expressing a Wnt reporter, we identified several drugs that suppressed Wnt signaling without compromising zebrafish development. The striatum is highly innervated by dopamine neurons and is involved in movement control (dorsal striatum) reward learning (ventral striatum). Fast-scan cyclic voltammetry (FSCV) is a technique that uses microelectrodes (diameter ~5 μm, length ~150 μm) to monitor dopamine signaling in the brain. Although urethane and isoflurane anesthetics are commonly used in in vivo studies, isoflurane’s potential influence on dopamine signaling has not been thoroughly investigated. BACKGROUND: Oxytocin (OXT) has been used as a post-treatment for post-traumatic stress disorder (PTSD). Previous research suggests that pre-treatment of intranasal OXT attenuates signs of fear in rats; however, the impact of intraperitoneal (i.p) OXT pre-treatment on signs of fear in rats is unknown. Obesity is a multifactorial disease with many comorbidities leading to multiorgan dysfunction. Current studies point towards dysregulation of platelet signaling as a driver for increased risk of cardiovascular disease and stroke. The exact mechanisms remain elusive due to the complex interplay of factors involved, warranting the need to further understand how platelets are affected. Our preliminary data evaluating changes in platelet and coagulation parameters from bariatric patients showed that obesity did not affect platelet count or clotting time as demonstrated in INR values. However, female patients had increased bleeding events before surgery that were decreased after surgery. Due to limitations …

Central to these advancements are many orthopedic implants, including anchors and joint replacements, which have been designed to restore mobility. Despite their functional efficacy, these products have limited lifespan and are associated with many complications, such as implant-associated infection. The success of implant design and function, both in the short and long term, is paramount for enhancing patient outcomes and overall well-being. Significant knowledge gaps exist in how land-cover impacts ground-hunting spider populations. To fill these gaps, this study investigates a common family of ground-hunting spiders, Lycosidae, to determine differences in their abundance and diversity in deciduous leaf litter and managed turfgrass (lawn). The study was conducted within a forested ecosystem at Virginia Commonwealth University's Rice Rivers Center in Charles City County, Virginia. I placed 10 belt transects (1m x 20m) on lawn substrate and 10 identical transects in deciduous forest leaf litter substrate. This project seeks to evaluate pharmacological dosing of melatonin on the cell proliferation of MDA-MB-231 breast cancer cell line in culture, using the two solubility carriers: DMSO/water and ethanol. This experiment is a component of ongoing research at the Eaton lab which will elucidate melatonin dose-response toxicology in zebrafish and a possible breast cancer xenograft model for novel treatment therapies.

Geckos move efficiently through a variety of substrates, from swaying branches to wooden fences to the sides of structures. Previous studies have studied locomotion and adhesion on manmade and rough substrates; however, the work is limited when discussing locomotion and adhesion as a relationship of compliance and roughness. Ease of movement was quantified by speed, under the assumption that the faster a gecko moves, the easier it is for the gecko to cross the substrate. The purpose of this paper was to use a tube feeding method on crayfish to expose them to substances through ingestion rather than the gill. This was done to mimic human ingestion of substance to aid in crayfish research involving substances that are normally ingestion. This was done by using a pipette to inject grains of sand into the mouth of the crayfish. To determine if the method worked and successfully went into the stomach the grains of sand were recovered and counted under a microscope after certain increments of time post injection.

The neural mechanisms that control the onset of puberty are not completely understood. In livestock, the onset of puberty relies on an increase in gonadotropin releasing hormone (GnRH) and luteinizing hormone (LH). Kisspeptin, neurokinin B, and dynorphin neurons (KNDy) and their receptors, Kiss1r, NK3R, and KOR, respectively, are important regulators of puberty. Previous data from our laboratory has shown that KNDy neurons may be responding to other inputs, including the melanocortin system, that will affect the timing of puberty. Given this, we examined kisspeptin receptor (Kiss1r) mRNA expression in GnRH neurons as well as examining melanocortin 3 and 4 receptor …
