Breast cancer is the most commonly diagnosed malignancy in women, with more than two million new cases diagnosed world-wide each year. Improvements in screening technology and diagnostics have increased survival rates, but cancer-associated sleep disruptions persist for the majority of breast cancer survivors well beyond the treatment period. In addition to reducing quality of life, reduced sleep quality is associated with greater incidences of affective dysfunction, reduced treatment efficiency, aggressive cancer progression, increased symptom severity, and increased mortality by all causes. Substance use disorders impose a staggering toll on the United States (U.S.), straining healthcare systems, destabilizing communities, and profoundly impacting both individuals and families. Approximately one in six Americans aged 12 and older had a past-year substance use disorder in 2022 (Key Substance Use, 2022), and nearly one-third of adults experience a substance use disorder in their lifetime (Wu, 2010), contributing to an economic burden exceeding $400 billion annually (National Drug Threat Assessment, 2011). The placebo effect is well-studied on pain perception and is often observed to have a pain-reducing effect in patients. A few studies suggest that the placebo effect can influence physiological changes in people. To date, not many studies have examined the placebo effect on stress; thus, the current study explored the placebo effect on participants’ cortisol stress response and self-reported stress while undergoing a modified acute stress task, the Sing-a-Song Stress Test (SSST). Descending neurons (DNs) play a critical role in sensorimotor integration by relaying motor information to the ventral nerve cord (VNC). Within the VNC, local interneurons, other DNs, and ascending neurons (ANs) make circuits with sensory and motor neurons to modify flies’ behavior. One important AN pair in Drosophila Melanogaster is mesothoracic ascending histaminergic neurons (MsAHNs), which are known to convey predictive wing motor signals to the brain. Sleep plays a crucial role in memory consolidation, neuronal plasticity, and cognitive function. Individuals with autism spectrum disorder (ASD) often experience sleep disturbances, which are positively correlated with symptom severity. However, on the surface, sleep architecture is remarkably normal under baseline conditions in many rodent models of ASD. The goals if this dissertation were to investigate whether regulation of sleep, biochemical signaling during sleep, and downstream consequences of sleep on behavior are altered in two mouse models of ASD. Sex differences in the morphology of the brain are thought to underlie the sex differences we see in anxiety and depression, where women are twice as likely to be diagnosed when compared to men. Stress, or a disruption to homeostasis, can exacerbate these mood disorders. Corticotropin releasing factor (CRF), and its receptors, play an important role in regulating the stress response. Insulin has widespread actions within the brain, including energy homeostasis, modulation of feeding, and as a key component of memory processes. Further, brain insulin dysfunction correlates with Alzheimer's disease and other neurodegenerative disorders. To this point, measurement of hippocampal insulin has been limited. Determining in vivo hippocampal insulin levels both at a healthy baseline and after, e.g., induction of a disease state or in response to cognitive challenge, is essential to advance our understanding of both insulin's procognitive actions and to guide potential therapeutic interventions.

One mechanism for generating diversity at the single cell level is selective expression of a unique set of transcription factors (TFs) and RNA binding proteins (RBPs). These regulatory factors contribute to functions of cells by controlling cell fate, turning genes on and off, and processing of transcripts either co- or post-transcriptionally. Generating and drafting ideas are creative processes linked to internal neural mechanisms, such as directed attentional states, reflected in alpha band synchronization patterns. Previous research has demonstrated that these mechanisms vary based on levels of expertise during creative tasks. Many animals use visual signals to evade predators, forage for food, and communicate with conspecifics. Some animals that shift into and out of breeding stages, such as fish, can modulate their visual sensitivity to better adapt to changing internal and external conditions. Previous work in our lab demonstrated that females of the cichlid fish Astatotilapia burtoni have improved visual sensitivity when they enter reproductive readiness. However, little is known about how fish and vertebrates more broadly modulate visual sensitivity across their reproductive cycle. Significance. Memory disturbances for emotional content are central to PTSD, yet most research focuses on trauma memories. Recent evidence suggests that enhancing access to positive memories could improve mood, affect regulation, self-esteem, and integration of trauma memories. However, the neurocognitive mechanisms underlying positive memory encoding in PTSD are poorly understood. No study has examined memory for positive social items in PTSD, which is important because PTSD entails social withdrawal and reduced accessibility to positive memory. Introduction: Major Depression Disorder (MDD) is a prevalent, recurrent, and chronic disorder that represents a leading cause of disability worldwide. Millions are affected with symptoms that go from constant sadness to even suicidal thoughts. In the U.S., major depression and fatal suicide have increased by 57.4% among adolescents and young adults. Myelin and inflammation-related genes have been found to play a role in individuals with MDD.

Introduction: The median survival time in ALS is approximately 3 years, but survival times range from less than a year to more than 10 years and much variance in disease course remains to be explained. As is true for physical outcomes, there is considerable variance in QOL, which is influenced by psychological health, coping, and social support, among other psychosocial factors. The Seattle ALS Patient Project Database (SALSPPD) provides a unique opportunity for researchers to address established and novel hypotheses about disease progression and QOL in ALS.

This dissertation investigates the mechanisms underlying variations in credit assignment to reward cues, with a focus on the lateral orbitofrontal cortex (lOFC). The lOFC has long been implicated in signaling reward expectancies, with controversy as to whether it signals the economic value of future outcomes, their sensory features to guide behavior or both. However, whether the lOFC signals the relative (competition-dependent) or absolute (contiguity-dependent) credit of cues is unknown. The capacity to initiate actions endogenously is critical for goal-directed behavior. Spontaneous voluntary actions are typically preceded by slow-ramping activity in medial frontal cortex that begins around two seconds before movement, which may reflect spontaneous fluctuations that influence action timing. However, the mechanisms by which these slow ramping signals emerge from single-neuron and network dynamics remain poorly understood. Here, we developed a spiking neural-network model that produces spontaneous slow ramping activity in single neurons and population activity with onsets ~2 s before threshold crossings. External cues can become powerful motivators of behavior when paired with rewarding stimuli, such as natural rewards or drugs of abuse, as is the case in individuals with substance use disorder (SUD). The dorsomedial prefrontal cortex (dmPFC) has been implicated as a neural substrate which encodes cue-reward associations and has been shown to exert top-down control over both natural and drug reward seeking. We and others have shown that activity within the dmPFC is heterogeneous during reward seeking, with some neurons displaying excitatory responses to reward-predictive cues, and others showing inhibitory responses to the same cues. Investigating The Role Of Insulin-Like Signaling In Paclitaxel-Induced Nociceptive Hypersensitivity Using Drosophila Melanogaster, Sreepradha Sridharan

Paclitaxel (PTX), a chemotherapeutic that alters microtubule dynamics, is known to induce nociceptive hypersensitivity and sensory neuron damage in human patients and in Drosophila larvae. We used a modified feeding protocol to establish a genetically tractable Drosophila model of PTX-induced hypersensitivity. Larvae fed PTX exhibited hypersensitivity to thermal and mechanical stimuli. Thermal hypersensitivity was observed at low concentrations of PTX (even below 1 μM per ml of food), begins within 8 hours of PTX feeding, and did not completely resolve at the larval stage. Live imaging of peripheral thermal nociceptors showed that this hypersensitivity precedes observable neuronal damage. At low …

Neutral sphingomyelinase-2 (nSMase2), gene name sphingomyelin phosphodiesterase-3 (Smpd3), is a key regulatory enzyme responsible for generating the sphingolipid cer- amide. The function of nSMase2 in the brain is still controversial. To better under- stand the functional roles of nSMase2 in the aging mouse brain, we applied RNA-seq analysis, which identified a total of 1462 differentially abundant mRNAs between +/fro and fro/fro, of which 891 were increased and 571 were decreased in nSMase2-deficient mouse brains. Cranial radiation therapy plays an integral role in the treatment of brain tumors but can lead to progressive cognitive deficits in survivors by mechanisms that are poorly understood. To develop preventive or mitigative strategies, it is crucial to better understand the underlying pathogenesis of radiation-induced cognitive impairments. The study investigated single-cell transcriptomics and DNA methylation changes as potential drivers of persistent cellular dysfunction after radiation exposure, specifically concentrating on the CA1-3 regions of the hippocampus and the prefrontal cortex due to their role in cognitive functions. This dissertation investigates host-microbial interactions within the teleost brain, previously considered sterile. Reviewing current literature highlights significant gaps in understanding teleost brain immunity, despite prevalent neurotropic diseases. We mapped the immune profiles of rainbow trout brains and meninges, revealing the olfactory bulbs as the immune effector part of the brain and the optic tectum as a nod2 expression hub. High il6 and il10 levels were noted in the telencephalon. A persistent bacterial community within healthy salmonid brains shares diversity with gut and blood microbiota. Our understanding of human brain function can be greatly aided by studying analogous brain structures in other organisms. One brain structure with neurochemical and anatomical homology throughout vertebrate species is the locus coeruleus (LC), a small collection of norepinephrine (NE)-containing neurons in the brainstem that project throughout the central nervous system. Addiction is facilitated by social, environmental, genetic, and neurobiological mechanisms. Nicotine use disorders (NUDs) pose a persistent global health threat, contributing to significant morbidity and mortality. It is a complex issue arising from a combination of different factors; thus, it is crucial to approach it from different perspectives. In this study, we aimed to determine whether the inbred High Drinking in the Dark (iHDID) mice replicate lines (iHDID-1 and iHDID-2) represent a good model for nicotine use disorders (NUDs). We measured nicotine (50µg/mL) intake in a Drinking in the Dark (DID) assay (n=12/sex/line). At the end of the DID assay, we measured serum cotinine levels using a cotinine ELISA kit. …

This thesis looks at the potential benefits and cohesive experience of sensory exploration for children with multiple diagnoses through a trauma informed lens. The researcher was inspired by Prendiville’s (2021) idea of how interactive sensory exploration and art making can help foster and develop reciprocal communication and socio-affective relationships influenced the researcher to design a method that would be used to collect data about potential similar findings. In the method, the interaction of the sensory-specific art mediums were reflected through the expressive therapies continuum (ETC) framework. Visual operators (e.g. edge detectors) are classically modelled using small circuits involving canonical computations, such as template-matching and gain control. Circuit models explain many aspects of the empirical descriptors that are used to characterize local visual operators, from sensitivity to classification images. Notwithstanding their utility, these models fail to provide a unified framework encompassing the variety of effects observed experimentally, such as the impact of contrast, SNR, and attention on the above descriptors. Δ9-tetrahydrocannabinol (Δ9-THC) is used during pregnancy to mitigate symptoms such as nausea, pain, and insomnia. It is established that Δ9-THC exposure early in life causes adverse effects in children’s development including behavioral disorders. Our goal was to determine the latent and generational impacts on growth, behavior, and mitochondrial bioenergetics following exposure to Δ9-THC.
