Soil and root microbial communities have been studied for decades, and the incorporation of high-throughput techniques and analysis has allowed the identification of endophytic/non-culturable organisms. This has helped characterize and establish the core microbiome of many model plant species which include underground and aboveground organs. Unfortunately, the information obtained from some of these model plants is not always transferable to other agronomic species. In this project, we decided to study the microbiome of the Nicotiana genus because of its importance in plant physiological and plant-microbe interactions studies. Biodiesel is an important alternative energy that can reduce CO2 emission, the main cause of Global warming. Algal biomass has been considered to be the third generation biofuels. One of the important strategies that enhance lipid accumulation; precursor of biodiesel production, is stress cultivation. High salt concentration is one of a potential approach to enhance lipid accumulation in microalgae. Here, we investigated the impacts of salt on lipid accumulation in green microalga Chlamydomonas reinhardtii (137c). There are only two natural ways by which molecular nitrogen can be available to support life, either by free-living or by symbiotic nitrogen fixing bacteria. The best studied diazotrophs form symbiotic associations with plants, primarily legumes and certain tree species, but little is known about how non-leguminous plants such as grasses obtain nitrogen in their environment. Natural prairies have few legumes, thus have less symbiotic nitrogen fixer interaction. This indicates presence of free-living nitrogen (FLN) fixation activity towards the balance of the N cycle. Systemic health effects from exposure to a complex natural dust containing heavy metals from the Nellis Dunes Recreation Area (NDRA) near Las Vegas, NV, were evaluated. Several toxicological parameters were examined following lung exposure to emissive dust from three geologic sediment types heavily used for recreational off-road activities: yellow sand very rich in arsenic (termed CBN 5); a shallow cover of loose dune sand overlying a gravelly subsoil bordering dune fields (termed CBN 6); and brown claystone and siltstone (termed CBN 7). Agroecosystems occupy a large portion of the United States and receive a disproportionally high amount of terrestrial nitrogen inputs. As application rates increase, nitrogen use efficiency declines leading to higher rates of nitrogen loss. To develop sustainable agricultural practices, the long-term effects of management practices on the soil microorganisms that perform central roles in the transformation of nitrogen need to be studied. Carotenoids are used as natural colorants and have been shown to have numerous health benefits, making these molecules highly desirable in the aquaculture, pharmaceutical, and nutraceutical markets. There is increasing consumer demand for the natural synthesis of carotenoids, but current methods are limited by the cost and difficulty of extraction from organisms. Bile acids (cholate and chenodeoxycholate) are synthesized from cholesterol in the human liver, and secreted in small intestine for food digestion. However, a portion of these primary bile acids is converted in the colon to toxic secondary bile acids, deoxycholate and lithocholate. Secondary bile acid formation is the result of 7α-dehydroxylating anaerobes, and one of the key players in this conversion is Clostridium scindens. Aspergillus flavus is an opportunistic fungal plant and human pathogen and producer of mycotoxins including aflatoxin B1 (AFB1). As part of our ongoing studies to elucidate biological functions of the A. flavus rtfA gene, we examined its role in pathogenicity of both plant and animal model systems. rtfA encodes a putative RNA-Pol II transcription elongation factor, previously characterized in Saccharomycese cerevisiae, Aspergillus nidulans, Aspergillus fumigatus, where it was shown to regulate several important cellular processes, including morphogenesis, secondary metabolism. Waterborne diseases continue to plague the poorest people in low-income countries and are estimated to cause 4,600,000 acute incidents of diarrhea resulting in over 2,000 deaths daily. A major challenge is performing microbiology tests to monitor drinking water quality. Friends of the Old (FOTO) implemented a novel strategy using evidence-based microbiology to educate communities about the relationship between contaminated water and disease. Two commercially available tests for E.coli, adapted for fieldwork, provided easily interpreted results of contamination that correlate with WHO's disease risk categories. Of particular interest to this research was the variation in microbial community structure at high, intermediate and poor water quality impacted areas. Cyanobacteria are a group of photo-oxygenic bacteria found in nearly every ecosystem, but much cyanobacterial diversity, in various habitats, has yet to be explored. Cyanobacteria are often conspicuous components of photosynthetic flora, providing significant carbon and nitrogen inputs to surrounding systems. As possible primary colonizers of stone substrates not native to this region, cyanobacteria isolated from headstones may provide biogeographically informative data. An exploratory study of lichen-dominated microbial consortia, growing on headstones, was conducted to isolate and identify novel microaerophytic cyanobacteria, and resulted in the establishment of four novel cyanobacterial taxa. TRIM5α is an anti-viral restriction factor that inhibits the lifecycle of retroviruses. TRIM5α binds to and forms a hexameric lattice around the retroviral capsid, thereby initiating its antiviral activities, which include: (1) inhibition of viral infection; (2) inhibition of viral reverse transcription; (3) disassembly of the capsid; and (4), activation of innate signaling pathways. The formation of this assembly also activates the E3 ubiquitin ligase function of TRIM5α. Ubiquitin modification is associated with directing substrates to particular cellular pathways. Vaccination is historically the most effective tool for preventing infectious disease but current vaccine strategies fail to generate robust immunity to major infectious diseases such as HIV and malaria. Therefore, newer vaccine approaches are needed. Vaccines generated from viral, adenovirus based, vectors (AdVs) have proven highly immunogenic in multiple disease models. However, the clinical use of many AdVs is limited by the presence of pre-existing antibodies in human populations, which prevent expression of antigenic genes during immunization with AdVs based on common adenovirus (Ad) serotypes, such as HAdV-5C. Nitric oxide (NO) is an important signal secreted by the squid, Euprymna scolopes, and is thought to limit symbiotic biofilm formation, the initial step in colonization by the bacterium, Vibrio fischeri. Previous work demonstrated that a mutant defective for NO-sensor HnoX (heme nitric oxide/oxygen binding) had a competitive advantage over wild-type V. fischeri, but the mechanism by which this occurred remains unknown. HnoX is encoded upstream of hahK, a recently identified positive regulator of biofilm formation. I thus hypothesized that HnoX inhibits colonization by controlling HahK-induced biofilm formation during the initiation of colonization.

Two coronaviruses (CoVs)—severe acute respiratory syndrome (SARS) virus and Middle East respiratory syndrome (MERS) virus—have emerged in the 21st century from animal reservoirs into the human population, each causing an epidemic associated with significant disease and mortality. CoV epidemics are currently only controllable by rigorous public health measures; no targeted therapeutics or vaccines exist to treat or prevent any human CoV infection. An important part of the HIV-1 infection cycle is the attachment of the intracellular viral core to the host microtubule network, facilitated by attachment of the viral capsid to cargo adaptor proteins. One such cargo adaptor is Bicaudal D Homolog Protein 2 (BICD2). BICD2 can attach to both the HIV-1 capsid and the dynein/dynactin complex and facilitate the trafficking of the viral core towards the host nucleus. Tuberculosis is a zoonotic disease that can be transmitted from infected animals to humans. Wild animals such as elephants and non-human primate have been reported to be infected by M. tuberculosis (MTB) complex, which can cause tuberculosis in animals as well. The common TB diagnostic approaches in human such as chest X-ray and tuberculin skin test are not practical in the elephants. Moreover, the gold standard of the bacteria culture from trunk wash has low sensitivity, is time-consuming and can only detect tuberculosis in the active stage. มันสำปะหลังเป็นหนึ่งในพืชเศรษฐกิจที่นิยมเพาะปลูกในเกือบทุกภาคของประเทศไทย ปกติหลังการเก็บเกี่ยวผลผลิต ใบและลำต้นจะถูกทิ้งไว้เพื่อรอการไถกลบสำหรับเตรียมแปลงปลูกในครั้งถัดไป งานวิจัยนี้มีวัตถุประสงค์เพื่อคัดแยกและศึกษาสมบัติของแบคทีเรียที่สามารถย่อยสลายเซลลูโลสซึ่งเป็นส่วนประกอบหลักของพืช จากดินบริเวณที่ปลูกมันสำปะหลัง 5 สายพันธุ์ ได้แก่ เกษตรศาสตร์ 50, พิรุณ 1, พิรุณ 2, ห้วยบง 60 และ ห้านาที ณ ศูนย์วิจัยพืชไร่ จังหวัดระยอง โดยคัดแยกบนอาหารแข็งที่มีสับสเตรตต่างกัน 3 ชนิด ได้แก่ คาร์บอกซีเมทิลเซลลูโลส, Avicel และเซลลูไบโอส จากนั้นคัดเลือกแบคทีเรียที่มีความสามารถในการย่อยเซลลูโลส 2 ใน 3 หรือ 3 ใน 3 ชนิดของสับสเตรต มาวิเคราะห์ลำดับนิวคลีโอไทด์ 16S rDNA และวิเคราะห์หาปริมาณน้ำตาลรีดิวซ์ คัดเลือกแบคทีเรีย 3 ไอโซเลทที่มีความสามารถในการย่อยสลายเซลลูโลสดีที่สุด ซึ่งได้แก่ Bacillus amyloliquefaciens, Bacillus pumilus และ Paenibacillus kribbensis แล้วนำแบคทีเรียทั้ง 3 ไอโซเลทมาทดสอบหาภาวะที่เหมาะสม พบว่าที่อุณหภูมิ 37 องศาเซลเซียส และความเป็นกรดด่าง (pH) 7 แบคทีเรียสามารถย่อยสลายเซลลูโลสได้ดีที่สุดจากสับสเตรตทั้ง 3 ชนิด จากนั้นนำมาทดสอบในการย่อยกระดาษกรอง ขนาด 1.5*1.5 เซนติเมตร ภาพจากกล้องจุลทรรศน์แบบส่องกราดแสดงให้เห็นว่า สภาพเส้นใยของกระดาษกรองเปลี่ยนแปลงไปมากที่สุด เมื่อเลี้ยงรวมกับ Paenibacillus kribbensis ในขณะที่ชุดการทดลองการย่อยใบมันสำปะหลังโดยตรง และชุดการทดลองการย่อยใบมันสำปะหลังในดิน เห็นผลการเปลี่ยนแปลงโครงสร้างของใบมันสำปะหลังมากที่สุดเมื่อใช้แบคทีเรียผสมกันทั้ง 3 ไอโซเลท หลังจากนำดินจากชุดการทดลองการย่อยใบมันสำปะหลังในดินไปวิเคราะห์คุณสมบัติ โดยศึกษาพารามิเตอร์ ความเป็นกรดด่าง, ปริมาณอินทรียวัตถุ, ฟอสฟอรัส, โพแทสเซียม, แคลเซียม, แมกนีเซียม, ไนโตรเจน และ ปริมาณอินทรีย์คาร์บอนรวมในดิน พบว่าไม่มีความแตกต่างอย่างมีนัยสำคัญในทุกการทดลอง เมื่อเปรียบเทียบกับชุดควบคุม อย่างไรก็ดีแบคทีเรียผสมจากการทดลองนี้ มีแนวโน้มที่สามารถนำไปประยุกต์ในสภาพแวดล้อมจริงได้ แต่ควรมีการยืนยันประสิทธิภาพที่เห็นผลชัดเจนมากกว่านี้ต่อไป.

Curvularia lunata เป็นราสายใยที่พบทั่วไปในดินซึ่งก่อให้เกิดโรคในพืชเศรษฐกิจหลายชนิด และส่งผลให้เกิดความเสียหายทางด้านเศรษฐกิจเป็นอย่างมาก เกษตรกรจึงจำเป็นต้องควบคุมและกำจัดรานี้ ซึ่งแนวทางที่น่าสนใจในการยับยั้งและกำจัดรา คือ การใช้วิธีการทางชีวภาพซึ่งไม่ก่อให้เกิดอันตรายต่อสิ่งมีชีวิตและไม่ส่งผลเสียต่อสิ่งแวดล้อม งานวิจัยก่อนหน้าพบว่าสารออกฤทธิ์ทางชีวภาพจาก Bacillus subtilis N3 สามารถยับยั้งการเจริญของ C. lunata ได้อย่างมีประสิทธิภาพ เมื่อวิเคราะห์สารออกฤทธิ์ทางชีวภาพดังกล่าว พบว่าเป็นโปรตีนที่มีความคล้ายกับแฟลกเจลลิน เอ ซึ่งเป็นยูนิตโปรตีนที่เป็นส่วนประกอบของแฟลกเจลลาในแบคทีเรีย งานวิจัยนี้จึงมุ่งศึกษายีนที่ประมวลรหัสให้โปรตีนแฟลกเจลลินจาก B. subtilis N3 โดยให้แสดงออกใน Escherichia coli BL21 ผลการวิจัยพบว่ายีนแฟกเจลลินที่สมบูรณ์จาก B. subtilis N3 มีขนาด 990 คู่เบส ที่แปลรหัสให้โปรตีนที่มีกรดอะมิโน 330 เรซิดิว โดยมีความใกล้เคียงกับโปรตีนแฟลกเจลลิน เอ ของแบคทีเรียในกลุ่ม Bacillus สูงถึง 100% เมื่อถ่ายโอนรีคอมบิแนนท์พลาสมิดที่มียีนแฟลกเจลลินดังกล่าวเข้าสู่ E. coli BL21 แล้วชักนำให้เกิดการแสดงออกของโปรตีนด้วย IPTG พบว่าส่วนของเซลล์และส่วนของน้ำเลี้ยงเชื้อที่ปราศจากเซลล์มีการแสดงออกโปรตีนขนาด 37 kDa และ 37-40 kDa ตามลำดับ ภาวะที่เหมาะสมในการชักนำให้เกิดการแสดงออกของโปรตีน คือ IPTG ความเข้มข้นสุดท้าย 0.1 มิลลิโมลาร์ เป็นเวลา 8 ชั่วโมงในส่วนของเซลล์ และ 16 ชั่วโมงในส่วนของน้ำเลี้ยงเชื้อ เมื่อนำมาทดสอบประสิทธิภาพในการยับยั้ง C. lunata พบว่าน้ำเลี้ยงเชื้อปราศจากเซลล์สามารถยับยั้ง C. lunata ได้ 21.3 % เมื่อทำบริสุทธิ์โปรตีนพบว่าโปรตีนบริสุทธิ์มีขนาดประมาณ 41 kDa และมีความสามารถในการยับยั้ง C. lunata ได้ โดยทำให้เกิดความผิดปกติของสายใยและยับยั้งการงอกของสปอร์ เมื่อโคลนและแสดงออกโดเมน N และโดเมน C ของแฟลกเจลลินพบว่าไม่เกิดการแสดงออกโปรตีนใน E. coli BL 21 เมื่อใช้วิธีการกลายแบบสุ่มบนยีนแฟลกเจลลินด้วยปฏิกิริยาลูกโซ่พอลิเมอเรสในภาวะที่มีความแม่นยําต่ำและเอื้อต่อความผิดพลาด พบว่าโคลนที่แสดงออกโปรตีนที่เกิดการกลายบริเวณโดเมน C โดยเกิดการเปลี่ยนแปลง A330T …

คัดกรองได้ยีสต์น้ำมัน Pseudozyma tsukubaensis YWT 7-2 จากยีสต์ทั้งหมด 28 สายพันธุ์ที่แยกมาจากป่าชายเลน จังหวัดจันทบุรี ซึ่งสะสมน้ำมันภายในเซลล์ 31.36% (น้ำหนัก/น้ำหนัก) ของน้ำหนักเซลล์แห้ง งานวิจัยนี้เป็นการรายงานครั้งแรกว่ายีสต์ P. tsukubaensis เป็นยีสต์น้ำมัน P. tsukubaensis YWT 7-2 สามารถผลิตน้ำมันเมื่อเจริญในอาหารที่มีอัตราส่วนของคาร์บอนต่อไนโตรเจนสูง (high C/N medium) และในสารละลายกลูโคส แสดงว่าการผลิตน้ำมันของยีสต์ P. tsukubaensis YWT 7-2 ในบางครั้งอาจไม่มีความเกี่ยวข้องกับการเจริญ แต่ให้ผลผลิตน้ำมันใน high C/N medium มากกว่าในสารละลายกลูโคส P. tsukubaensis YWT 7-2 ผลิตน้ำมันและเพิ่มจำนวนในไฮโดรไลเสตแป้งมันสำปะหลังที่ปรับด่างเกินด้วยแคลเซียมไฮดรอกไซด์ (DCSH) ได้มากกว่าในไฮโดรไลเสตแป้งมันสำปะหลัง (CSH) ปริมาณน้ำมันที่ผลิตได้เท่ากับ 1.49 กรัม/ลิตร (อัตราการผลิตน้ำมัน 0.75 กรัม/ลิตร/วัน) เมื่อเติม 0.25% (น้ำหนัก/ปริมาตร) แอมโมเนียมซัลเฟตลงใน DCSH พบว่าผลผลิตน้ำมันเพิ่มขึ้นเป็น 3.85 กรัม/ลิตร (อัตราการผลิตน้ำมัน 1.28 กรัม/ลิตร/วัน) พีเอชเริ่มต้นทีเหมาะสมที่สุดสำหรับการผลิตน้ำมันคือ 5.5 การเพิ่มจำนวนเซลล์ของ P. tsukubaensis YWT7-2 ใน DCSH ที่เติม 0.2% (น้ำหนัก/ปริมาตร) แอมโมเนียมซัลเฟต และ 0.1% (น้ำหนัก/ปริมาตร) โพแทสเซียมไดไฮโดรเจนฟอสเฟต แทนการเพิ่มจำนวนเซลล์ใน CSH ที่เติม 0.2% (น้ำหนัก/ปริมาตร) แอมโมเนียมซัลเฟต ทำให้ผลผลิตน้ำมันเพิ่มขึ้นเป็น 5.42 กรัม/ลิตร (อัตราการผลิตน้ำมัน 1.36 กรัม/ลิตร/วัน) น้ำมันที่ยีสต์ P. tsukubaensis YWT7-2 ผลิตเมื่อเจริญใน high C/N medium และใน DCSH …

วิถีสัญญาณ Notch ประกอบด้วย รีเซบเตอร์ 4 ชนิด (Notch 1-4) และลิแกนด์ 5 ชนิด (Delta-1, 3, 4 และ Jagged 1, 2) การเกิดอันตรกิริยาระหว่างลิแกนด์และรีเซบเตอร์ จะส่งผลให้เกิดการเปลี่ยนแปลงบริเวณ NRR ทำให้เกิดการเผยตำแหน่งที่มีการตัดโดยเอนไซม์ซึ่งนำไปสู่การตัดโดยแกมมาซีครีเทส ดังนั้น NRR จึงเป็นตำแหน่งสำคัญเพื่อรักษาเสถียรภาพของ NRR ไว้เพื่อป้องกันการตัดของ Notch รีเซบเตอร์ วิถีสัญญาณ Notch ทำหน้าที่ควบคุมการเพิ่มจำนวน การเปลี่ยนสภาพ ตลอดจนถึงการตายของเซลล์หลายประเภท ความผิดปกติของสัญญาณ Notch ส่งผลให้เกิดการพัฒนาเป็นมะเร็งชนิดต่าง ๆ ได้ มีการใช้ยากดการทำงานของเอนไซม์แกมมาซีครีเทส (GSI) เพื่อการยับยั้งวิถีสัญญาณ Notch แต่มีรายงานว่า GSI ทำให้เกิดผลข้างเคียงที่ไม่พึงประสงค์ เนื่องจากมีซับสเตรทหลายชนิด รีคอมบิแนนท์แอนติบอดีในรูป scFv-Fc ซึ่งเป็นโปรตีนลูกผสมประกอบด้วย scFv และ Fc ของอิมมูโนโกลบูลินจีของมนุษย์ ได้ถูกนำมาประยุกต์ใช้ทางการรักษาบำบัดเนื่องจากมีคุณสมบัติที่ดีกว่ามอโนคลอนอลแอนติบอดีทั้งโมเลกุลหลายด้าน ในงานวิจัยนี้มีวัตถุประสงค์ในการผลิตรีคอมบิแนนท์แอนติบอดีชนิด scFv-Fc ต่อบริเวณ NRR ของ Notch1 และ Notch2 โดยใช้เซลล์ไลน์ HEK-293T และศึกษาลักษณะสมบัติของรีคอมบิแนนท์แอนติบอดี scFv-Fc โดยทดสอบการจับกับ Notch1 และ Notch2 และผลกระทบต่อวิถีสัญญาณ Notch ในเซลล์มะเร็งเม็ดเลือดขาว Jurkat จากผลการทดลองพบว่าสามารถผลิตรีคอมบิแนนท์แอนติบอดี scFv-Fc ต่อ Notch1 และ 2 ได้ในเซลล์ไลน์ HEK-293T เมื่อทำการทรานส์เฟคชันพลาสมิดที่มีชิ้นยีนของ scFv-Fc เข้าสู่เซลล์ โดยนำโปรตีนที่ได้ในอาหารเลี้ยงเซลล์ ไปแยกบริสุทธิ์ด้วยคอลัมน์โครมาโทกราฟีสัมพรรคภาพ พบว่าบน SDS-PAGE ปรากฏโปรตีนเพียงแถบเดียว เมื่อผ่านการแยกบริสุทธิ์ด้วยสองขั้นตอนโดยใช้คอลัมน์ต่อ His-tag และ คอลัมน์โปรตีน A นำแอนติบอดีที่ได้ไปทดสอบรีคอมบิแนนท์แอนติบอดีต่อบริเวณ NRR โดย anti-Notch1 …
