All eukaryotic cells require that transcribed mRNAs undergo export form the nucleus to the cytoplasm where they can be translated into proteins. This process requires a host of proteins which are conserved between the unicellular budding yeast, S. cerevisiae, and humans. During this process, Mex67 and other associated proteins facilitate the mRNA to travel across the nuclear pore complex (NPC), doorways embedded in the nuclear envelope. Upon the exit of mRNA, Mex67 is released and recycled back into the nucleus to provide the export of more mRNA. This release occurs through the action of Dbp5, whose activity is regulated …

The Lake Wales Ridge is important scrub habitat that has been increasingly altered since the post-Columbian settlement in Florida. This loss of habitat has caused extreme anthropogenic fragmentation within the Lake Wales Ridge resulting in isolation among extant scrub patches. To expand the geographic scope of previous studies and answer questions concerning population connectivity, we characterized genetic diversity and differentiation using cytochrome-b and microsatellite genetic markers for two endemic skink species: the Florida Sand Skink (Plestiodon reynoldsi) and Blue-tailed Mole Skink (Plestiodon egregius lividus). Psychopathology is a risk factor for accelerated biological aging and early mortality. We examined associations between broad underlying dimensions of psychopathology (reflecting internalizing and externalizing psychiatric symptoms), PTSD, and age-adjusted GrimAge (“GrimAge residuals”), a DNA methylation biomarker of mortality risk relative to age. We also examined neurobiological correlates of GrimAge residuals, including neurocognitive functioning, blood-based biomarkers (of inflammation, neuropathology, metabolic disease), and cortical thickness. Data from two independent trauma-exposed military cohorts (n = 647 [62.9% male, Mage = 52], n = 434 [90% male, Mage = 32]) were evaluated using linear regression models to test associations between …

In this study I attempted to further resolve the population genetic structure in the Prothonotary Warbler (Protonotaria citrea), and conducted an outlier SNP analysis and exploratory gene ontology analysis to investigate potential ongoing natural selection in the species. This analysis of population structure confirms previous work by DeSaix et al. (2019), where weak population structure was observed between eastern sites along the Atlantic Coastal Plain, and western sites in the Mississippi Alluvial Valley, possibly due to a genetic discontinuity across the Appalachian Mountains. This Senior Thesis project aimed to be able to screen samples that came in from a PhD student in Georgia for the presence of two oyster diseases, MSX and Dermo. This study was done using PCR followed by visualizing on E-gels for initial presence of disease. These were then followed by qPCR and visualization on Criterion gels. These Criterion gels were able to be analyzed in order to calculate the pg/uL of MSX and Dermo in the original ocean samples. สุกรเป็นอีกหนึ่งแหล่งกักเก็บเชื้อแบคทีเรียที่มียีนดื้อยาสร้างเอนไซม์ Extended-Spectrum β-Lactamases (ESBLs) ซึ่งสามารถแพร่กระจายยีนดื้อยามาสู่มนุษย์ สัตว์ และสิ่งแวดล้อมได้ ในการศึกษานี้จึงได้พัฒนาการตรวจหายีนสร้างเอนไซม์ ESBLs จากมูลสุกรโดยตรง ด้วยเทคนิค Multiplex polymerase chain reaction (mPCR), mPCR-Lateral flow assay (mPCR-LFA) และเทคนิค Nested PCR (nPCR) เปรียบเทียบกับการตรวจจากเชื้อ ESBL-producing Escherichia coli ซึ่งเพาะแยกจากมูลสุกรตัวอย่างเดียวกัน ด้วยเทคนิค PCR-sequencing เชื้อ ESBL-producing E. coli จำนวน 149 isolates ที่เพาะแยกจากมูลสุกรทั้งหมด 139 ตัวอย่าง พบความชุกของยีนชนิด blaCTX-M-15 (34.2%) มากที่สุด รองลงมาคือ blaCTX-M-15+TEM-1 (17.5%) และ blaCTX-M-55+TEM-1 (13.4%) โดยเชื้อ ESBL-producing E. coli ส่วนใหญ่มีการดื้อยาหลายขนาน โดยดื้อต่อยา Ceftriaxone (100%), Cefotaxime (100%), Ciprofloxacin (31.5%) และ Trimethoprim/Sulfamethoxazole (30.9%) ความไว ความจำเพาะ และขีดจำกัดของการตรวจจากเชื้อ ESBL-producing E. coli ด้วยเทคนิค mPCR-LFA เท่ากับ 100% และ 0.1-1 ng ตามลำดับ เทคนิค mPCR ที่พัฒนาขึ้น สำหรับตรวจจากมูลสุกรโดยตรง พบว่าต้องใช้ปริมาณเชื้อมากถึง 107 CFU จึงจะตรวจพบ โดยมีความไวในการตรวจยีน blaCTX-M และ blaTEM เท่ากับ 30.43% และ 87.65% ตามลำดับ ขีดจำกัดและความไวของการตรวจพบยีน blaCTX-M โดยตรง …

Proper formation of the visual system requires the precise interaction of several embryonic cell lineages, including the neuroectoderm (forms the retina and retinal pigment epithelium), surface ectoderm (forms the lens), mesoderm and cranial neural crest cells (form the ocular blood vessels and anterior ocular structures). When this process is disrupted structural birth defects such as coloboma result, leading to pediatric visual deficits. Ocular developmental defects are often present in larger syndromic disorders. The root system of the plant plays a vital role in water and nutrient uptake. Native North American grapevines adapted to a broad range of climatic and soil conditions, which led to the evolution of diverse root system architecture (RSA) within the Vitis genus. Despite the importance of RSA in viticulture, little is known about the genetic basis of the RSA in grapevine. Flavobacterium psychrophilum is a known fish pathogen, causing bacterial cold-water disease (BCWD) and rainbow trout fry syndrome (RTFS), predominantly in salmonids. The disease causes tissue damage and tail rot in young and adult fish. The issue is prevalent in fisheries in the Pacific Northwest and the treatments often entail the use of antibiotics. Genetic manipulations in F. psychrophilum are scarce, due to most of the strains’ ability to destroy foreign DNA via restriction enzymes. Gene therapy has a wide range of applications for various types of pathologies. Viral methods of gene delivery provide high levels of gene expression but have various safety concerns. Non-viral methods are largely known to provide lower levels of expression. We aim to address this issue by using plasmid DNA with smaller backbones to increase gene expression levels when delivered using non-viral methods. After myocardial infarction, the massive death of cardiomyocytes leads to cardiac fibroblast proliferation and myofibroblast differentiation, which contributes to the extracellular matrix remodelling of the infarcted myocardium. We recently found that myofibroblasts further differentiate into matrifibrocytes, a newly identified cardiac fibroblast differentiation state. Cardiac fibroblasts of different states have distinct gene expression profiles closely related to their functions. Ixodes scapularis is a medically important tick that transmits several microbes to humans, including rickettsial pathogen Anaplasma phagocytophilum. In nature, these ticks encounter several abiotic factors including changes in temperature, humidity, and light. Many organisms use endogenously generated circadian pathways to encounter abiotic factors. In this study, we provide evidence for the first time to show that A. phagocytophilum modulates the arthropod circadian gene for its transmission to the vertebrate host. BACKGROUND: Recent high-throughput technologies have opened avenues for simultaneous analyses of thousands of genes. With the availability of a multitude of public databases, one can easily access multiple genomic study results where each study comprises of significance testing results of thousands of genes. Researchers currently tend to combine this genomic information from these multiple studies in the form of a meta-analysis. The outer epithelial layer covering an organism, commonly known as the epidermis, is crucial for maintaining homeostasis and for the wound healing processes after injury. The planarian epidermis allows flatworms to heal their wounds and virtually restore any missing tissues. Immediately after amputation, planarians contract their muscle and stretch their epidermis to heal the wound area. However, how the planarian epidermis coordinates with other tissues and mechanisms after the initial wound healing processes begins is not understood in detail. Twins have long been of interest for illuminating genetic and environmental influences on human traits and diseases. These explorations have been accelerated by the substantial amount of information collected from twins and their families participating in twin registers around the world. In this dissertation, I review what is known and what remains unknown about the biology and genetics of twins and the underlying twinning process. Small Cell Lung Cancer (SCLC) is a devastating disease characterized by a very low two-year survival rate and almost universal acquisition of chemoresistance. Nearly all patients have tumors driven by functional inactivation of the tumor suppressors Rb and p53, but despite the uniform origins of this tumor, not all patients are genetically or phenotypically identical. SCLC can be subtyped into four unique molecular subtypes, determined by the expression of ASCL1, NEUROD1, POU2F3, or YAP1. These subtypes are plastic, and subtype switching after chemotherapy has been documented. Without the understanding of how tumor heterogeneity arises, we cannot solve the challenge of …

Lake sturgeon were extirpated throughout much of their historic range in the early 1900s due to overharvest, habitat degradation, and reduced water quality. Interagency management plans have worked to identify historic locations and remnant populations suitable for stocking and habitat remediation to rebuild self-sustaining populations of lake sturgeon within their historic range. The St. Louis River population was extirpated in the early 1900s until lake sturgeon were stocked from 1983-2000 from two different source populations (Wolf River, Sturgeon River). Objective: Mexican Americans are disproportionally affected by non-alcoholic fatty liver disease (NAFLD), liver fibrosis and hepatocellular carcinoma. Noninvasive means to identify those in this population at high risk for these diseases are urgently needed. Approach: The Cameron County Hispanic Cohort (CCHC) is a population-based cohort with high rates of obesity (51%), type 2 diabetes (28%) and NAFLD (49%). In a subgroup of 564 CCHC subjects, we evaluated 339 genetic variants previously reported to be associated with liver injury markers aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in United Kingdom and Japanese cohorts. Evolutionary Developmental Biology seeks to answer fundamental questions about the mechanisms underlying the evolution and innovation towards increasing structural complexity within the body plan. The ambrosia symbiosis, a polyphyletic group of xylem-feeding beetles, provides a wealth of diversity in novel pouch-like structures called mycangia. This diversity could serve as a rich model for understanding how mutualisms may prompt structural innovation and diversification in a host organism. The ambrosia symbiosis has become a fast-growing research subject in entomology and forest pathology but has yet to experience the same attention under the lens of evolutionary developmental genetics.

Proteins are an important class of biologics, but there are several recurring challenges to address when designing protein-based therapeutics. These challenges include: the propensity of proteins to aggregate during formulation, relatively low loading in traditional hydrophobic delivery vehicles, and inefficient cellular uptake. This last criterion is particularly challenging for anionic proteins as they cannot cross the anionic plasma membrane. Here we investigated the complex coacervation of anionic proteins with a block copolymer of opposite charge to form polyelectrolyte complex (PEC) micelles for use as a protein delivery vehicle. Long Interspersed Nuclear Elements (LINEs) are retrotransposons. They function by making a cDNA copy, reverse-transcribing it into DNA and reinserting itself back into the genome; thereby, increasing its copy number. Sequencing of the human genome revealed that LINEs (L1) are underrepresented on the short arm of human chromosome 21 (HC21), but it is enriched in full-length copies. A number of previous studies have investigated the DNA methylation patterns of L1s in cancer cells, but these studies are typically done in euchromatic regions. Atlantic sturgeon are a long-lived anadromous fish that ranged from Labrador, Canada to Florida, US. Due to overharvest in the late 1800’s and 1900’s, Atlantic sturgeon populations across the coast experienced a dramatic population crash. Recovery of this species has faced challenges due to anthropogenic threats, such as vessel strikes, bycatch, and habitat degradation. In 2012, Atlantic sturgeon were added to the United States Endangered Species Act (ESA). Hybridization offers unique insight into the evolutionary process of speciation and the forces that maintain barriers to reproduction. In northeast Florida, a hybrid zone between two species of killifish, Fundulus heteroclitus and F. grandis, has been identified near Flagler Beach, FL, although the exact boundaries of the hybrid zone remain unknown (Gonzalez et al. 2009). This study aims to determine the fine-scale spatial genetic patterns of the hybrid zone and test the hypothesis that species ranges are influenced by changes in dominant vegetation. ไลโพโปรตีนชนิดความหนาแน่นสูง (High-density lipoprotein; HDL) มีบทบาทสำคัญในการป้องกันการเกิดโรคหัวใจขาดเลือด ต้านการอักเสบของหลอดเลือด และยับยั้งการเกิด LDL-oxidation แต่การทำงานของ HDL จะสูญเสียไปเมื่อ HDL ถูกทำลายด้วยอนุมูลอิสระ กลายเป็น oxidized HDL (oxHDL) การวัดระดับ HDL-cholesterol เป็นการประเมินด้านปริมาณแต่ไม่สามารถบอกการทำงานของ HDL ได้ หากสามารถวัดระดับ oxHDL ควบคู่ไปกับระดับ HDL-cholesterol ทำให้การพยากรณ์การเกิดโรคหัวใจขาดเลือดได้แม่นยำขึ้น แต่การวิเคราะห์ระดับ oxHDL ต้องใช้เครื่องมือจำเพาะและน้ำยามีราคาสูง งานวิจัยนี้มีวัตถุประสงค์เพื่อพัฒนาชุดตรวจวิเคราะห์ oxHDL ด้วยหลักการ Paper-based Immunogold silver-enhanced assay โดยออกแบบฐานกระดาษมีลักษณะเดียวกับ microtiter plate ตรึงแอนติบอดีต่อ 4-Hydroxynonenal (anti-HNE) ซึ่งเป็นผลผลิตจากกรดไขมันที่ถูกออกซิไดซ์ oxHDL ในตัวอย่างจับกับ anti-HNE บนกระดาษ ใช้ anti-APO AI ที่มีความจำเพาะต่อ HDL ที่ติดฉลากกับอนุภาคของทองคำและขยายสัญญาณด้วย silver ion อ่านผลด้วยเครื่องสแกนและวิเคราะห์ความเข้มสีด้วยโปรแกรม Photoshop CS64 ผลการวิเคราะห์ความเป็นเส้นตรงระหว่าง oxHDL กับ ความเข้มสี พบว่า มีความสัมพันธ์เป็นเส้นตรงที่ความเข้มข้นตั้งแต่ 0-300 ng/mL ( r2 = 0.946) ผลการทดสอบความถูกต้องของชุดทดสอบ ใช้ซีรัมจำนวน 32 ราย วิเคราะห์ปริมาณ oxHDL ด้วยวิธีที่พัฒนาขึ้นและวิธี Sandwich ELISA หาความสัมพันธ์ระหว่างปริมาณ oxHDL ที่วิเคราะห์ได้จากทั้งสองวิธีด้วย Pearson correlation coefficient พบว่า ค่า correlation coefficient (r) = 0.527 และมีค่า p-value =0.002 แสดงให้เห็นว่าทั้งสองวิธีมีความสัมพันธ์มีความสัมพันธ์กันในระดับกลางอย่างมีนัยสำคัญทางสถิติ อุปกรณ์ตรวจวิเคราะห์ออกซิไดซ์ไลโพโปรตีนชนิดความหนาแน่นสูงฐานกระดาษเป็นต้นแบบที่สามารถนำไปต่อยอดและพัฒนา เพื่อประยุกต์ใช้ในการตรวจปริมาณ oxHDL …

มะเร็งช่องปากชนิดสความัสเซลล์คาร์ซิโนมา (Oral squamous cell carcinoma, OSCC) จัดเป็นมะเร็งที่พบได้บ่อยที่สุดของ มะเร็งศรีษะและลำคอ เคอคิวมินเป็นสารออกฤทธิ์หลักซึ่งพบได้ในขมิ้นชันและถูกใช้เป็นสมุนไพรสำหรับการรักษาโรคต่าง ๆ หลายชนิด รวมทั้ง ได้รับการพิสูจน์ในห้องปฏิบัติการแล้วว่ามีคุณสมบัติในการยับยั้งเซลล์มะเร็งได้หลากหลายประเภท ในขณะที่โพรไบโอติกแบคทีเรีย Lactobacillus rhamnosus GG (LGG) ได้รับการพิสูจน์ในห้องปฏิบัติการถึงคุณสมบัติในการยับยั้งเซลล์มะเร็งได้หลากหลายชนิดเช่นกัน และ มีรายงานถึงการนำ LGG ไปใช้ร่วมกับสารผลิตภัณฑ์ธรรมชาติชนิดอื่น ๆ เพื่อเพิ่มประสิทธิภาพของสารธรรมชาตินั้นอีกด้วย ดังนั้นวัตถุประสงค์ ของการศึกษาในครั้งนี้คือ เพื่อศึกษาผลของการใช้เคอคิวมินร่วมกับน้ำเลี้ยงส่วนใสของแบคทีเรีย LGG (LGG cell-free supernatant, LGG CFS) ในการยับยั้งการมีชีวิตและกระตุ้นให้เซลล์มะเร็งช่องปากชนิด OSCC (SCC-9 cells) เกิดการตายแบบอะพอพโตซิส ผลการศึกษาพบว่า เคอคิวมินและ LGG CFS มีคุณสมบัติในการยับยั้งการมีชีวิตของเซลล์มะเร็งช่องปาก SCC-9 cells โดยขึ้นอยู่กับความเข้มข้นที่ใช้ การใช้เคอคิว มินและ LGG CFS ที่ความเข้มข้นสูง (Curcumin 40 µg/ml และ 25% (volume/volume) LGG CFS ที่ความเข้มข้น 108 CFU/ml หรือความ เข้มเข้นเริ่มต้น (undiluted condition)) ในรูปแบบการใช้สารเดี่ยวหรือสารผสมร่วมกัน ส่งผลยับยั้งการมีชีวิตของทั้งเซลล์มะเร็งช่องปาก SCC9 cells และเซลล์เนื้อเยื่อเหงือก (human gingival fibroblast, HGF cells) เมื่อเปรียบเทียบกับสภาวะควบคุม (p<0.0001 สำหรับการใช้สาร เดี่ยวในเซลล์มะเร็งช่องปาก SCC-9 และการใช้สารเดี ่ยวและสารผสมในเซลล์เนื ้อเยื ่อเหงือก HGF และ p=0.02 สำหรับการใช้สารผสมใน เซลล์มะเร็งช่องปาก SCC-9) ในขณะที ่การใช้เคอคิวมินและ LGG CFS ในรูปแบบของสารผสมร่วมกันที ่ระดับความเข้มข้นต่ำ (Curcumin 5 µg/ml และ 25% (v/v) LGG CFS ความเข้มข้น 1:100 เท่าจากระดับความเข้มข้นเริ ่มต้น) สามารถยับยั ้งการมีชีวิตของเซลล์มะเร็งช่องปาก SCC-9 ได้อย่างมีประสิทธิภาพเมื่อเปรียบเทียบกับสภาวะการใช้สารเดี่ยวของเคอคิวมินและ LGG CFS ที่ระดับความเข้มข้นต่ำ และสภาวะควบคุม (p<0.0001, p=0.002 และ p=0.02 ตามลำดับ) และไม่ส่งผลต่อการมีชีวิตของเซลล์เนื้อเยื่อเหงือก HGF เมื่อเทียบกับกลุ่มควบคุม (p=0.14) ผลการวิเคราะห์ Flow analysis พบว่าสภาวะการใช้เคอคิวมินและ LGG CFS ร่วมกันที่ระดับความเข้มข้นต่ำ สามารถเหนี่ยวนำให้ เซลล์มะเร็ง SCC-9 เกิดการตายแบบอะพอพโตซิสได้มากขึ้นอย่างมีนัยสำคัญทางสถิติเมื่อเปรียบเทียบกับการใช้สารเดี่ยวและสภาวะควบคุม (p<0.0001 ในทุกสภาวะ) นอกจากนั้นสภาวะการใช้เคอคิวมินและ LGG CFS ร่วมกันที่ระดับความเข้มข้นต่ำ สามารถเหนี่ยวนำให้เซลล์มะเร็ง SCC-9 มีระดับการแสดงออกของ Bax/Bcl-2 ratio ที่สูงขึ้น ซึ่งมีผลให้ให้เซลล์มะเร็งเกิดการตายแบบอะพอพโตซิสต่อไป อย่างไรก็ตามสภาวะ เคอคิวมินและ LGG CFS ร่วมกันที่ระดับความเข้มข้นต่ำ ส่งผลต่อระดับการแสดงออกของโปรตีน caspase-3 ที ่สูงขึ้นอย่างไม่มีนัยสำคัญ (p=0.12) แต่ไม่มีผลต่อระอัตราส่วนการแสดงออกของโปรตีน Bcl-xL/Bak และโปรตีน Mcl-1/Bak เมื่อเทียบกับกลุ่มควบคุม (p=0.53 และ p=0.34 ตามลำดับ) การวิเคราะห์ LGG CFS ที่ความเข้มข้น 1:100 เท่าจากระดับความเข้มข้นเริ่มต้น ด้วยเทคนิค LC-MS/MS พบสารประเภท โปรตีนทั้งหมด 30 ชนิด ได้แก่ metabolic enzyme and protein จำนวน 26 ชนิด และ surface layer protein จำนวน 4 ชนิด การใช้สาร เคอคิวมินและ LGG CFS ร่วมกันที่ระดับความเข้มข้นต่ำ น่าจะมีผลในการยับยั้งเซลล์มะเร็งช่องปากชนิด SCC-9 แต่ไม่มีผลกระทบต่อเซลล์ เหงือก HGF ผลการศึกษาในครั้งนี้อาจนำไปสู่การพัฒนาวิธีป้องกันหรือรักษามะเร็งช่องปากชนิด OSCC รูปแบบใหม่ได้ในอนาคต

The shedding propensity of a person can assist data interpretation in casework when assessing the possibility of passive transfer for DNA analysis. Past studies on shedding propensity evaluated palmar skin (washed and unwashed) deposits. This study compared different skin locations with respect to shedding propensity, and explored the potential of tape-lifts as a skin surface collection method. Cytokine production is a critical component of cell-extrinsic responses to DNA damage and cellular senescence. Here, we demonstrated that expression of the gene encoding interleukin-19 (IL-19) was enhanced by DNA damage through pathways mediated by c-Jun amino-terminal kinase (JNK) and cGAS-STING and that IL19 expression was required for the subsequent production of the cytokines IL-1, IL-6, and IL-8. IL19 expression was stimulated by diverse cellular stresses, including inhibition of the DNA replication checkpoint kinase ATR (ataxia telangiectasia and Rad3-related protein), oncogene expression, replicative exhaustion, oxidative stress, and DNA double-strand breaks.
