The Humboldt marten (Martes caurina humboldtensis) is a species facing conservation concern in Oregon and California. Historical populations were subjected to unregulated fur trapping and timber harvest throughout the late 19th and early 20th centuries leading to a 90% decline in their distribution. As such, the Humboldt marten is listed as a state endangered species in California, and the analogous ‘coastal marten’ is listed as federally threatened under the Endangered Species Act and classified as a Species of Greatest Conservation Need in Oregon and California. To date, most genetic studies on the American alligator (Alligator mississippiensis) have focused on population genetics and multiple paternity. There have not been any studies looking at how relatedness affects alligator behavior and movement. This study focused on three main questions: 1) what is the genetic diversity among alligators in this location? 2) can parentage be accurately defined among parents and offspring? and 3) how does individual relatedness affect their spatial distribution? Turner syndrome (TS) is a genetic disorder seen in phenotypically female (pf) individuals who have either a complete or partial absence of the second sex (X) chromosome. TS includes different karyotypes, and it presents with a variety of phenotypic and genotypic features. In general, the neuropsychological profiles for individuals diagnosed suggest that TS can contribute to challenges in various aspects of daily life, including social and emotional functioning. การดื้อยา colistin มีแนวโน้มเพิ่มสูงขึ้นในเชื้อแบคทีเรียกลุ่ม Enterobacterales เนื่องมาจากการนำยา colistin กลับมาใช้ในการรักษาเชื้อดื้อยาหลายขนาน โดยเฉพาะอย่างยิ่งเชื้อแบคทีเรียแกรมลบที่ดื้อยา carbapenem ประกอบกับการถ่ายทอดยีน mobile colistin-resistant (mcr) ผ่านทาง plasmid ซึ่งส่งผลให้การแพร่ระบาดของเชื้อดื้อยา colistin เกิดขึ้นในวงกว้างอย่างรวดเร็ว สถานการณ์เชื้อกลุ่ม Enterobacterales ที่ดื้อยา colistin ในประเทศไทยกำลังเป็นที่สนใจ เนื่องมาจากเริ่มพบอัตราเชื้อดื้อยาที่สูงขึ้นอย่างต่อเนื่องทั้งในภาคการปศุสัตว์และในโรงพยาบาล การศึกษาครั้งนี้ได้ศึกษาลักษณะจีโนมของเชื้อ E. coli และ K. pneumoniae ที่ดื้อยา colistin ที่เพาะแยกได้จากผู้ป่วยของโรงพยาบาลเลิดสิน กรุงเทพมหานคร ในช่วงระหว่างเดือนตุลาคม พ.ศ. 2564 ถึงเดือนกันยายน พ.ศ. 2565 และศึกษาความสัมพันธ์และการกระจายตัวของเชื้อในแผนกต่างๆ ของโรงพยาบาล เชื้อที่เก็บได้ทั้งหมดจำนวน 54 ตัวอย่าง ประกอบด้วยเชื้อ E. coli และ K. pneumoniae จำนวน 14 และ 40 ตัวอย่าง ตามลำดับ ซึ่งให้ผลดื้อยา colistin เมื่อทดสอบด้วยเครื่องอัตโนมัติ Sensititre™ Gram Negative MIC Plate เชื้อส่วนใหญ่เพาะแยกจากปัสสาวะ (46.30%, p-value=0.38) และเสมหะ (29.63%, p-value=0.63) โดยเพาะแยกเชื้อได้จากผู้ป่วยใน (81.48%) มากกว่าผู้ป่วยนอก (18.52%) อย่างมีนัยสำคัญ (p-value=0.000) ภายหลังนำเชื้อทั้งหมดไปวิเคราะห์ลำดับเบสด้วยเทคนิค Illumina whole genome sequencing (WGS) สามารถจำแนกสายพันธุ์ของเชื้อได้อย่างมีประสิทธิภาพ โดยเชื้อ E. coli และ K. pneumoniae มีรูปแบบยีนก่อโรคที่แตกต่างกันอย่างเห็นได้ชัด และพบยีน blaCTX-M, blaTEM, blaOXA ,blaSHV ,ยีน aac, ยีน qnr …

โรคหลอดเลือดแดงโคโรนารี (Coronary Artery Disease, CAD) เป็นโรคไม่ติดต่อชนิดเรื้อรังที่เกิดจากหลอดเลือดแดงที่กล้ามเนื้อหัวใจตีบหรือตัน สาเหตุหลักเกิดจากไขมันและเนื้อเยื่อสะสม มีผลทำให้เยื่อบุผนังหลอดเลือดชั้นในหนาตัวขึ้นและเกิดการอุดตัน จากการศึกษาที่ผ่านมาพบว่า ox-LDL เป็นจุดเริ่มต้นของโรคดังกล่าว เนื่องจากมีคุณสมบัติต่างจาก normal LDL โดยไม่สามารถจับได้กับ LDL receptor ได้ แต่สามารถจับได้กับ scavenger receptor ที่ผิวเซลล์แมคโคฟาจจึงทำให้ ox-LDL สามารถเข้าเซลล์ได้อย่างไม่มีสิ้นสุด เกิดเป็นโฟมเซลล์ (foam cells) สะสมอยู่ในผนังหลอดเลือดเกิดเป็นรอยขีดไขมัน (fatty streak) ประกอบกับการกระตุ้นเกล็ดเลือดและเซลล์อิมมูน ทำให้เกิดเป็นลิ่มเลือดอุดตันทางเดินของเส้นเลือด แม้ว่า ox-LDL จะเป็นสาเหตุสำคัญของการเกิดโรคแต่การตรวจวัดระดับไขมัน ox-LDL ยังไม่แพร่หลายในห้องปฏิบัติการเทคนิคการแพทย์ เนื่องจากชุดตรวจมีราคาแพง ใช้เวลาในการวิเคราะห์นาน และยังต้องใช้เครื่องมือที่มีความจำเพาะ ดังนั้นการศึกษาในครั้งนี้มีวัตถุประสงค์เพื่อพัฒนาเทคนิคการตรวจวัดระดับ ox-LDL ด้วยเทคนิคการตรวจวัดบนฐานกระดาษร่วมกับ immunogold silver enhancement โดยออกแบบกระดาษกรองให้มีรูปแบบเป็น microtiter plate อาศัยเทคนิค wax printing ในการสร้างขอบเขต หาสภาวะที่เหมาะสมสำหรับออกซิไดซ์กระดาษ เพื่อให้มีคุณสมบัติเหมาะสมกับการตรึงแอนติบอดี โดยเปลี่ยนเซลลูโลสเป็นหมู่แอลดีไฮด์ งานวิจัยนี้ได้นำสารละลายลิเธียมคลอไรด์มาช่วยเพิ่มประสิทธิภาพการออกซิไดซ์ร่วมกับสารละลายโซเดียมเพอร์ริออเดท พบว่ากระดาษที่ออกซิไดซ์แล้วสามารถคงสภาพนาน 72 ชั่วโมง และแอนติบอดีที่ตรึงไว้คงสภาพนานที่สุด 4 ชั่วโมง ทดลองวิเคราะห์วัดปริมาณ std. ox-LDL และ ox-LDL ที่เตรียมเองในห้องปฏิบัติการด้วยสภาวะเทคนิคดังกล่าว พบว่าสีปฏิกิริยาที่เกิดขึ้นยังไม่ชัดเจน อันเนื่องมากจากข้อจำกัดของความจำเพาะของแอนติบอดี การทำให้เกิดสีด้วย silver enhancement ซึ่งยังคงต้องมีการพัฒนาต่อไปเพื่อให้ผลการวิเคราะห์มีความชัดเจน นอกจากนี้ได้พัฒนากล่องถ่ายภาพเพื่อใช้เก็บผลการวิเคราะห์บนฐานกระดาษ ที่สามารถถ่ายภาพด้วยแสงและระยะที่คงที่ สามารถเคลื่อนย้ายได้ง่ายในภาคสนาม อย่างไรก็ตามแม้การพัฒนาวิธีการวิเคราะห์ ox-LDL ยังไม่ประสบผลสำเร็จ แต่ยังพอได้แนวทางและต้นแบบในการศึกษาต่อไปในอนาคต

Aspergillosis เป็นโรคติดเชื้ออันตรายถึงแก่ชีวิต มีสาเหตุมาจากเชื้อราในจีนัส Aspergillus การวินิจฉัยโรคในปัจจุบัน การเพาะแยกเชื้อและจัดกลุ่มตามลักษณะทางสัณฐานวิทยายังคงเป็นวิธีมาตรฐาน การตรวจหา galactomannan มักใช้ตรวจวินิจฉัยโรคในผู้ป่วยกลุ่มเสี่ยงเท่านั้น ในขณะที่เทคนิคทางอณูชีววิทยาอาศัยข้อมูลสารพันธุกรรมของ “its region” ในการจำแนกกลุ่มเชื้อระดับสปีชีส์ แต่อย่างไรก็ตามข้อมูลดังกล่าวถูกพิสูจน์ว่าสามารถจำแนกกลุ่มเชื้อราก่อโรคในคนในระดับสปีชีส์ได้ต่ำ ในด้านการรักษาสปีชีส์ของเชื้อ Aspergillus มักไม่ถูกนำมาพิจารณาร่วมเพื่อการเลือกกลุ่มยาแม้ว่ายังคงมีผู้ป่วยบางรายที่ตอบสนองต่อการรักษาไม่ดีเนื่องจากเชื้อสาเหตุดื้อต่อยาต้านเชื้อรา ด้วยเหตุดังกล่าวเพื่อศึกษาความสัมพันธ์ระหว่างสปีชีส์และรูปแบบความไวรับต่อยา การค้นหายีนที่สามารถจำแนกเชื้อ Aspergillus ในระดับสปีชีส์ได้อย่างแม่นยำจึงเป็นสิ่งจำเป็นยีน “tef-1α” เป็นสารพันธุกรรมที่ได้รับการพิสูจน์แล้วว่ามีคุณสมบัติสามารถจำแนกเชื้อราเส้นสายในระดับสปีชีส์ได้สูง ในงานวิจัยนี้ ผู้วิจัยได้เปรียบเทียบผลการจำแนกกลุ่มเชื้อ Aspergillus ที่แยกได้จากผู้ป่วยจำนวน 227 ตัวอย่าง โดยใช้ข้อมูลสารพันธุกรรม “its region” และ“tef-1α” จากผลการศึกษาพบว่าสารพันธุกรรมจากยีน“tef-1α” (D=0.8461) มีความสามารถในการจำแนกเชื้อในระดับสปีส์สูงกว่าบริเวณ “its” (D=0.796) โดยเฉพาะอย่างยิ่งใน section Fumigati ข้อมูลสารพันธุกรรมจาก “its region” สามารถแบ่งเชื้อกลุ่มดังกล่าวออกได้เพียง 1 สปีชีส์ คือ A. fumigatus ในขณะที่ยีน“tef-1α” สามารถแบ่งออกเป็น A. fumigatus และอีก 2 สปีชีส์ คือ A. lentulus และ A. udagawa ซึ่งเป็นเชื้อที่ดื้อต่อยา amphotericin B โดยกำเนิด โดยสรุป ผลจากงานวิจัยนี้ชี้ให้เห็นว่าการจำแนกสปีชีส์ของเชื้อ Aspergillus โดยใช้ยีนทุติยภูมิ “tef-1α” สามารถบ่งชี้กลุ่มยาที่เหมาะสมต่อการรักษาได้

Honey bees (Apis mellifera) are the most economically important agricultural pollinator in North America, as well as being the most frequently studied bee species. Many agricultural systems, such as fruit tree orchards, benefit from having a diversity of bee species present. In this article, we present information about the types of bees that can be found in orchards and explore their mating behaviors, life cycles, genetic differences, flower preferences, and foraging activities. Recent genome wide association studies have identified 89 common genetic variants robustly associated with ischemic stroke and primarily located in non-coding regions. To evaluate the contribution of coding variants, which are mostly rare, we performed an exome array analysis on 106,101 SNPs for 9721 ischemic stroke cases from the SiGN Consortium, and 12,345 subjects with no history of stroke from the Health Retirement Study and SiGN consortium. We identified 15 coding variants significantly associated with all ischemic stroke at array-wide threshold (i.e., p < 4.7 × 10-7), including two common SNPs in ABO that have previously been associated with stroke. The nematode Caenorhabditis elegans is a powerful tool for studying nervous system genetics. Though relatively simple compared to mammals, C. elegans boasts a remarkably well-conserved neuronal genome and proteome, and its utility in the characterization of neuronal genes has been well-established. However, gene expression is often controlled by complex interactions between multiple genes, and teasing apart the functions of individual genes within such networks remains a challenge. Hsp100 chaperones, also known as Clp proteins, constitute a family of ring-forming ATPases that differ in 3D structure and cellular function from other stress-inducible molecular chaperones. While the vast majority of ATP-dependent molecular chaperones promote the folding of either the nascent chain or a newly imported polypeptide to reach its native conformation, Hsp100 chaperones harness metabolic energy to perform the reverse and facilitate the unfolding of a misfolded polypeptide or protein aggregate. Smut, caused by Sporisorium scitamineum and brown rust, caused by Puccinia melanocephala, are two of the most important diseases that affect sugarcane production in Louisiana and worldwide. Smut continues to cause losses in the breeding program in Louisiana. Therefore, a QTL analysis was conducted to identify molecular markers associated with resistance in F1 progeny (162) of a biparental cross between susceptible cultivar L 99-233 and resistant HoCP 96-540. Using 1,574 single-dose SNP markers, a total of 253 linkage groups (LG) were obtained with a genome coverage of 24,580.15 cM. Six QTLs were localized on five LGs. The highest …

Lipoprotein(a) (Lp(a)) represents a unique subclass of circulating lipoprotein particles and consists of an apolipoprotein(a) (apo(a)) molecule covalently bound to apolipoprotein B-100. The metabolism of Lp(a) particles is distinct from that of low-density lipoprotein (LDL) cholesterol, and currently approved lipid-lowering drugs do not provide substantial reductions in Lp(a), a causal risk factor for cardiovascular disease. Somatic genome editing has the potential to be a one-time therapy for individuals with extremely high Lp(a). Background: A critical unmet medical need in clinical management of colorectal cancer (CRC) pivots around lack of noninvasive and or minimally invasive techniques for early diagnosis and prognostic prediction of clinical outcomes. Because DNA methylation can capture the regulatory landscape of tumors and can be measured in body fluids, it provides unparalleled opportunities for the discovery of early diagnostic and prognostics markers predictive of clinical outcomes. During meiosis, recombination between homologous chromosomes (homologs) generates crossovers that promote proper segregation at the first meiotic division. Recombination is initiated by Spo11-catalyzed DNA double strand breaks (DSBs). 5' end resection of the DSBs creates 3' single strand tails that two recombinases, Rad51 and Dmc1, bind to form presynaptic filaments that search for homology, mediate strand invasion and generate displacement loops (D-loops). D-loop processing then forms crossover and noncrossover recombinants. Meiotic recombination occurs in two temporally distinct phases. During Phase 1, Rad51 is inhibited and Dmc1 mediates the interhomolog recombination that promotes homolog synapsis. In Phase 2, Rad51 becomes active …

Theta and gamma oscillations in the hippocampus have been hypothesized to play a role in the encoding and retrieval of memories. Recently, it was shown that an intrinsic fast gamma mechanism in medial entorhinal cortex can be recruited by optogenetic stimulation at theta frequencies, which can persist with fast excitatory synaptic transmission blocked, suggesting a contribution of interneuronal network gamma (ING). Camelina is an alternative low-input oil plant grown in marginal areas in Turkey. This study aimed to determine some quality characteristics of 39 camellia genotypes in ecological conditions in Bolu, Turkey. The field experiments were carried out in an augmented design with randomized blocks. In this work, some important properties such as thousand seed weight, oil content, fatty acid composition, and protein ratio were investigated. Results showed that mean data for thousand seed weight, oil ratio, and protein ratio were 0.63 – 1.5 g, 26.69 – 39.17%, and 21.81 – 29.81%, respectively. The effect of Spirulina platensis inclusion in feed of commercial broilers was evaluated over one experiment. The study aimed to investigate the effects of Spirulina platensis freshwater algae inclusion on live performance and processing characteristics of commercial broilers subjected to daily cyclic heat stress. Day old Ross 708 male broilers were placed into 8 environmentally controlled chambers. At d 21, four chambers remained on a control diet containing no algae (CON) while 4 chambers received a diet that included algae at an inclusion rate of 2.5% (ALG). Food production must increase by 70% to feed this future population. The Mediator complex (MED) is a multi-subunit protein complex integral to the eukaryotic transcription machinery. MED12 is a Cdk8- regulatory kinase module subunit directly implicated in human disease and is genetically altered in neurological disease and cancer. Numerous attempts at generating an in vivo system to study the role of Med12 failed due to embryonic lethality associated with germline or developmental disruption of Med12 gene. Background: Human subtelomeric DNA regulates the length and stability of adjacent telomeres that are critical for cellular function, and contains many gene/pseudogene families. Large evolutionarily recent segmental duplications and associated structural variation in human subtelomeres has made complete sequencing and assembly of these regions difficult to impossible for many loci, complicating or precluding a wide range of genetic analyses to investigate their function.
Results: We present a hybrid assembly method, NanoPore Guided REgional Assembly Tool (NPGREAT), which combines Linked-Read data with mapped ultralong nanopore reads spanning subtelomeric segmental duplications to potentially overcome these difficulties. One of the current major issues in the control of vector-borne disease is resistance to insecticides. Resistance to one or more insecticides has appeared in over 580 different arthropod species. Widespread resistance has led to the field of Insecticide Resistance Management (IRM). The goal of IRM is to develop insecticide-use strategies that prevent or slow down the evolution of resistance. Graphical models determine associations between variables through the notion of conditional independence. Gaussian graphical models are a widely used class of such models, where the relationships are formalized by non-null entries of the precision matrix. However, in high-dimensional cases, covariance estimates are typically unstable. Moreover, it is natural to expect only a few significant associations to be present in many realistic applications. This necessitates the injection of sparsity techniques into the estimation method. Classical frequentist methods, like GLASSO, use penalization techniques for this purpose. Quiescence is a distinct cell cycle phase, termed G0, in which growth, transcriptional and translational activity, and replication are halted. Importantly, quiescent cells are viable and can return to the cell cycle. Quiescence is a feature of all organisms and a key hallmark of stem cells; however, the regulation of quiescence is still poorly understood.

Pediatric central nervous system (CNS) tumors represent the most common cause of cancer-related death in children aged 0-14 years. They differ from their adult counterparts, showing extensive clinical and molecular heterogeneity as well as a challenging histopathological spectrum that often impairs accurate diagnosis. Here, we use DNA methylation-based CNS tumor classification in combination with copy number, RNA-seq, and ChIP-seq analysis to characterize a newly identified CNS tumor type. Over half of the global human population suffers from lactase nonpersistence, a condition marked by losing the ability to digest lactose after infancy. However, a minority of the global population, primarily located in Central and Northern Europe, has a genetic mutation that results in lactase persistence, which is the continued ability to process lactose after infancy. This interdisciplinary analysis blends archaeology, cultural anthropology, evolutionary biology, and archaeogenetics to explore the origin and rise of lactase persistence in Europe and its contribution to the end of hunter-gatherer societies in Scandinavia. Rapid and accurate detection of carriers of carbapenemase-producing organisms (CPO) in hospitalized patients is critical for infection control and prevention. This study aimed to evaluate a pooling strategy for the detection of carbapenem resistance genes (CRG) in multiple specimens using the Xpert Carba-R test. Two rectal swabs each were collected from 415 unique patients. One swab was tested by Carba-R on the five specimen-pooled strategy. The other swab was tested individually by culture followed by DNA sequence analysis for CRG as the reference. At the first 5:1 pooling testing, 22 of 83 pools were positive, which yielded 34 positives from …
