The coat proteins of the RNA phages MS2 and Qβ are structurally homologous, yet they specifically bind different RNA structures. In an effort to identify the basis of RNA binding specificity we sought to isolate mutants that convert MS2 coat protein to the RNA binding specificity of Qβ. A library of mutations was created which selectively substitutes amino acids within the RNA binding site. Genetic selection for the ability to repress translation from the Qβ translational operator led to the isolation of several MS2 mutants that acquired binding activity for Qβ RNA. Despite adequate hormonal stimulation, oocytes collected for the purpose of in vitro fertilization and embryo transfer display several levels of nuclear maturity. Preovulatory or mature oocytes, technically those that are Metaphase I or II, are inseminated shortly after aspiration and assessed for fertilization the following day. Prophase I oocytes, also called germinal vesicle-bearing or immature oocytes, require a 24-36 hour period in culture before being exposed to spermatozoa. During this time, the majority of Prophase I oocytes complete nuclear maturation in vitro, progressing from germinal vesicle breakdown through first polar body extrusion. Cancers of the oral cavity and pharynx have increased throughout the decades. Since oral cancer represents a small percentage of the total cancers in the United States, it is often overlooked. Though this is the case, the results of this cancer can be devastating. To address this issue, an Oral Cancer Screening was organized on August 14, 1996 at the West Michigan Cancer Center in Kalamazoo. It is considered that the implementation of this policy will protect the natural environment and native crayfish species. It will also allow for the development of a redclaw industry, and provide the first step towards self regulation of that industry.

The complex mechanisms of heredity are little appreciated by nonspecialists, in some measure, because of misunderstandings that are perpetuated when words used for technical terms have other, more widely understood, folk meanings. When a word has both technical and folk meanings, it is the responsibility of the specialist to avoid promoting confusion by either using extremely cautious and precise language when using the term or, in cases when confusion is inevitable, abandoning the term in favor of one without a widely understood folk meaning. Y-Chromosome Effects On Drosophila Geotaxis Interact With Genetic Or Cytoplasmic Background, Scott F. Stoltenberg, Jerry Hirsch

Previously, all of the major fruit fly, Drosophila melanogaster, chromosomes (I, II, and III) have been shown to be associated with geotaxis, but the Y chromosome has not. Using two methods (back-crossing and chromosome substitution), Y chromosomes from lines that have evolved stable, extreme expressions of geotaxis were placed into different genetic and cytoplasmic backgrounds to test the resulting males for geotaxis. The results of the back-crossing do not support the interpretation of Y-chromosome effects on geotaxis. These tests do not have sufficient statistical power, however, to detect small genetic effects. Identification Of Poinsettia Cultivars Using Rapd Markers, Jing-Tian Ling, Roger Sauve, Nick Gawel

Randomly amplified polymorphic DNA (RAPD) techniques were used to compare the DNA from leaf tissues of nine commercial poinsettia (Euphorbia pulcherrima Wild ex Klotzsch) cultivars. Amplification occurred in 57 out of 60 (95%) tested primers. Nine primers that revealed polymorphisms among cultivars were selected for further evaluation. Forty-eight RAPD bands were scored from these primers, and 33 (69%) were polymorphic. All tested cultivars could be discriminated with seven bands generated from primers OPB7 and OPC13. Results of a UPGMA cluster analysis and principal components analysis placed the nine cultivars into two groups: one group consisted of `Jingle Bells', `Supjibi', …

Wild-type fungal population 851-WT was selected for shortened latent period on cv. CI 13227 for five uredinial generations to study the adaptation of Puccinia recondita f. sp. tritici to partially resistant wheat cultivars. Differences among wild-type and selected populations for traits contributing to parasitic fitness (i.e., latent period, infection frequency, and uredinium area and growth rate) were assessed in monocyclic infection experiments on susceptible cv. Monon and partially resistant cvs. Suwon 85, Sw 72469-6, L-574-1, and CI 13227. Differences were greatest among fungal populations on cv. CI 13227. The study was conducted to determine the effects of lowered genetic variance in the Western mosquitofish (Gambusia affinis) on the physiological parameters of critical thermal maximum and minimum pH. Mosquitofish were obtained from two small central Illinois ponds, Lost Pond and Shadow Pond, by seines and dip nets. Once caught, fish were returned to the laboratory and acclimated for a minimum of five days at a temperature of 27°C and a pH of 8.25. At the beginning of each experimental run, fish were subjected to the physiological experiments for determination of critical thermal maximum and minimum pH, and then measured …

The purpose of the project was to assess the effects of inbreeding on the genetic constitution of two Pakistani bradaris (literally defined as brotherhoods). Both bradaris contain children born to consanguineous (first cousin) and nonconsanguineous marriages. DNA samples have been supplied by Dr Subaib Ahmed of the Armed Forces Institute of Pathology in Rawalpindi, for a total of 91 individuals. The specific allele frequencies, and levels of homozygosity of each bradari, were determined using twenty fluorescence-labelled microsatellite markers for chromosomes 13 and 15. The project has studied the inheritance of microsatellite alleles across four chromosomes within two multi-generation families from a highly endogamous community, the Sankethi of Karnataka, India. The two families each have members from four generations participating in the study. Finger prick blood samples and detailed pedigree information were collected from subjects in India. DNA was extracted from the blood spot samples using a phenol-chloroform technique, and analysed by spectrophotometry to determine DNA concentrations. Fluorescent markers from chromosomes 15, 16, 17 & 18 were amplified using polymerase chain reaction (PCR) techniques on a Perkin Elmer 96 well thermocycler. The PCR products …

Macroparasites including helminths constitute a potential threat to economically important resources in agriculture and conservation biology. The distamycin A inducible fragile site, FRA8E, Fra(8) (q24.11) has been previously mapped to 8q24.11, proximal to the MYC gene, by fluorescent in situ hybridization (Takahasi et al., 1991). This fragile site has been shown to be present in about 1 out of every 140 healthy Japanese individuals (Takahasi et al., 1988). To more precisely map FRA8E, we have used cosmids isolated from the Langer-Giedion chromosomal region (LGCR) for FISH analysis. An endogenous circadian biological clock controls the temporal aspects of life in most organisms, including rhythmic control of genes involved in clock output pathways. In the fungus Neurospora crassa, one pathway known to be under control of the clock is asexual spore (conidia) development. To understand more fully the processes that are regulated by the N. crassa circadian clock, systematic screens were carried out for genes that oscillate at the transcriptional level. A subregion of the mitochondrial large subunit (16s) rRNA gene was amplified by polymerase chain reaction (PCR) from nine species of blackflies (Diptera: Simuliidae) which serve as natural or experimental vectors of human or bovine Onchocerca parasites. PCR products from each species of blackfly were tested by directed heteroduplex analysis (DHDA), and their genotypes established according to diagnostic banding patterns of the heteroduplex products. Three alleles of mitochondrial 16s rRNA were found to exist in members of the Simulium (Edwardsellum) damnosum sensu lato complex from West Africa, and two alleles were found in the Neotropical Simulium (Psilopelmia …

The Offshore Constitutional Settlement OCS is a legal arrangement between the Commonwealth and State Governments, and defines control over the fisheries which operate off each State of Australia. For the Western Australian coast, the first set of arrangements took effect in 1987. The OCS of 1995 specifies five arrangements, under which the fisheries inside the 200 nautical mile approximately 370 km limit of the Australian Fishing Zone AFZ come under either State, Commonwealth, or joint control.

In June 1995, a self-completion survey of fishermen in the rock lobster fishery was sent out by The Marketing Centre to 589 licensees. A total of 206 valid responses were received. The questionnaire was fairly long and dealt with a number of complex issues. As such, this response rate of 36.0% compares favorably with the average for postal surveys of around 40.0%. The following represents the main findings from the survey.
