The alternansucrase (ALT, EC 2.4.1.140) catalyses transferring of glucose from sucrose to produce a polymer with α-1,6 and α- 1,3 linkages, so-called "alternan". The ALT-encoding gene from Leuconostoc citreum ABK-1 (WTalt) was successfully cloned and expressed in Escherichia coli BL21 (DE3). The two versions of truncated SH3-like domains, ∆3SHALT and ∆7SHALT, were successfully constructed. The WTALT, ∆3SHALT and ∆7SHALT possessed optimum temperature of 40 °C, while optimum pH of WTALT was at 5.0 but ∆3SHALT's and ∆7SHALT's were shifted to pH 4.0. Their activities could be enhanced by Mn²⁺. Kinetic studies of all ALTs were mainly responsible for transglycosylation activity …

Alpha-glucosidase [EC 3.2.1.20; α-D-glucoside glucohydrolase] is an exohydrolase which catalyzes non-reducing end of substrates to release D-glucose. In addition, alpha-glucosidase also displays a transferase activity, which bring about the formation of α-glucosylated compounds such as soluble starch and glycogen. This research aims to express and characterize α-glucosidase from Weissella confusa BBK-1 (WcAG). WcAG gene was expressed in modified pET28b vector. Fisetin is a flavonoid which was widely exerted in pharmacological industries. However, fisetin has low water solubility and unstable in aqueous solution. So the uses of fisetin have been limited. The purpose of this study was to increase water solubility of fisetin by transglycosylation reaction using cyclodextrin glycosyltransferase (CGTase) from Paenibacillus sp. RB01. The reaction had to perform in co-organic solvent system and DMSO at 40% (v/v) was proved to be the most suitable co-organic solvent since it provided the high enzyme stability and high production yield. Lung neuroendocrine tumors (NETs) incidences are increasing in recent years. Women with lung NETs have significantly better survival rates as compared to men suggesting the involvement of sex steroids and their receptors in the progression of lung NETs. ในปัจจุบัน ออทิซึมสเปกตรัม (Autism spectrum disorder, ASD) ยังคงเป็นโรคที่ยังคงไม่ทราบกลไกระดับโมเลกุล แน่ชัด ยีนและโปรตีนที่เคยมีการรายงานการแสดงออกผิดปกติไปในผู้ป่วยออทิซึมสเปกตรัมมีความสามารถในการทำซ้ำ (Reproducibility) ต่ำหรืออาจไม่มีเลย ซึ่งน่าจะมาจากความหลากหลายของอาการแสดงของผู้ป่วย จากการศึกษาล่าสุด แสดงให้เห็นว่าการจัดกลุ่มออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยตามความรุนแรงของลักษณะทางคลินิกสามารถช่วยในการระบุยีนและกลไกที่ผิดปกติไปในกลุ่มย่อยของผู้ป่วย อย่างไรก็ตาม การแบ่งกลุ่มผู้ป่วยเป็นกลุ่มย่อยตามความรุนแรงของลักษณะทางคลินิกใน ออทิซึมสเปกตรัมยังไม่มีการใช้กันอย่างแพร่หลายในการค้นหาโปรตีน Biomarker ที่เกี่ยวข้องกับออทิซึมสเปกตรัม ในการศึกษาครั้งนี้ ผู้วิจัยทำการวิเคราะห์โดยการแบ่งกลุ่มผู้ป่วยจำนวน 85 รายออกเป็นกลุ่มย่อยทางคลินิกตามคะแนน ADI-R และทำการวิเคราะห์รูปแบบการแสดงออกของยีนในออทิซึมสเปกตรัมในแต่ละกลุ่มย่อยเปรียบเทียบกับกลุ่มควบคุมคนปกติที่อายุใกล้เคียงกันและเพศเดียวกันกับผู้ป่วย (Sex/age-matched control) ซึ่งสามารถระบุยีนที่มีการแสดงออกผิดปกติ (Differentially expressed genes, DEGs) ในออทิซึมสเปกตรัม นอกจากนี้ ผู้วิจัยได้ทำการวิเคราะห์รูปแบบการแสดงออกของโปรตีโอมจากเซลล์ไลน์เม็ดเลือดขาว (Lymphoblastoid cell lines,LCLs) ด้วยเทคนิค Two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) ร่วมกับ Nano-liquid chromatography-tandem mass spectrometry (Nano-LC-MS/MS) รายชื่อของโปรตีนที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญที่ได้จากการวิเคราะห์ถูกนำมาเปรียบเทียบกับ DEGs และผลการวิเคราะห์ Meta-analysis ของโปรตีนและโปรตีโอมในการศึกษาก่อนหน้า หน้าที่ทางพยาธิสภาพที่มีความเกี่ยวข้องกับโปรตีนถูกวิเคราะห์โดยโปรแกรม Ingenuity Pathway Analysis (IPA) และได้ทำการคัดเลือกโปรตีนที่น่าสนใจมาทำการตรวจสอบโดยใช้ Western blot analysis จากการศึกษาครั้งนี้พบว่าผลการวิเคราะห์การแบ่งกลุ่มผู้ป่วยออกเป็นกลุ่มย่อยทางคลินิกตามคะแนน ADI-R สามารถแบ่งผู้ป่วยออกเป็น 4 กลุ่มย่อย ซึ่งพบว่าหนึ่งในกลุ่มนั้น เป็นกลุ่มออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษารุนแรง จากผลวิเคราะห์รูปแบบ ทรานสคริปโตมของเซลล์ไลน์เม็ดเลือดขาว พบว่ามีจุดโปรตีนจำนวน 82 จุดที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญในกลุ่มออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษารุนแรง และได้ทำการคัดเลือก 18 จุดโปรตีนจากทั้งหมดมาวิเคราะห์ชนิดของโปรตีนโดยเทคนิค Nano-LC-MS/MS ซึ่งพบว่ามีความเกี่ยวข้องกับการทำงานของระบบประสาทและกระบวนการอักเสบ จากการศึกษานี้พบว่าจากโปรตีนทั้งหมด การแสดงออกของ Isocitrate dehydrogenase (IDH2) และ Diazepam binding inhibitor (DBI) ลดลงในออทิซึมสเปกตรัม ซึ่งโปรตีนเหล่านี้ยังพบการแสดงออกผิดปกติในการวิเคราะห์ Meta-analysis ของทรานสคริปโตมและโปรตีโอมร่วมด้วย การแบ่งกลุ่มออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยทางคลินิกตามด้วยการวิเคราะห์ทรานสคริปโตมและโปรตีโอมแบบบูรณาการร่วมกัน ทำให้การศึกษานี้สามารถระบุโปรตีนที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญในกลุ่มย่อยของออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษาอย่างรุนแรง ซึ่งโปรตีนเหล่านี้อาจทำหน้าที่เป็นตัวแทนในการช่วยระบุกลไกทางโมเลกุลของออทิซึมสเปกตรัมในอนาคต ตลอดจน อาจพัฒนาไปเป็น …

ออทิซึมสเปกตรัมเป็นความผิดปกติที่เกี่ยวกับการพัฒนาของระบบประสาทและสมองที่มีความชุก 1 ใน 68 คนของเด็กในสหรัฐอเมริกา โดยจะมีระดับความรุนแรงและอาการแสดงทางคลินิกแตกต่างกันออกไปในแต่ละบุคคล ออทิซึมสเปกตรัมจัดเป็นโรคที่ไม่ทราบสาเหตุการเกิดที่แน่ชัด แต่เป็นที่เชื่อกันว่าสาเหตุของการเกิดโรคนั้นอาศัยปัจจัยร่วมหลายปัจจัย ได้แก่ ทั้งปัจจัยทางพันธุกรรมและปัจจัยทางสิ่งแวดล้อม การวินิจฉัยออทิซึมสเปกตรัมในปัจจุบัน ใช้วิธีอิงจากการสังเกตพฤติกรรมผู้ป่วย วินิจฉัยโรคและความรุนแรงตามลักษณะอาการทางคลินิกเท่านั้น ยังไม่มีสารบ่งชี้ทางห้องปฏิบัติการทางการแพทย์ที่สามารถใช้ในการวินิจฉัย หรือติดตามความรุนแรงของโรคอย่างจำเพาะ โดยผลการศึกษาก่อนหน้านี้ แสดงให้เห็นว่าผู้ป่วยโรคออทิซึมสเปกตรัมมีความผิดปกติในระดับอณูชีวโมเลกุลมากมายทั้งในสมองและเลือด ซึ่งมีหลายประการที่สอดคล้องกับพยาธิสภาพระดับชีวโมเลกุลที่เกิดขึ้นในสมอง อย่างไรก็ตาม ยังไม่มียีนหรือโปรตีนชนิดใดที่สามารถใช้ในการอธิบายโรคออทิซึมสเปกตรัมได้ทั้งหมดในปัจจุบัน แสดงให้เห็นว่าอาจมีกลไกอื่น ๆ ที่สามารถทำให้เกิดพยาธิสภาพได้ ยิ่งไปกว่านั้น แม้โรคออทิซึมสเปกตรัมจะเป็นโรคที่มีอัตราการถ่ายทอดทางพันธุกรรมสูง แต่มีฝาแฝดร่วมไข่จำนวนไม่น้อยที่คนหนึ่งเป็นโรคแต่อีกคนไม่เป็น หรือมีระดับความรุนแรงของโรคไม่เท่ากัน เนื่องจากฝาแฝดร่วมไข่มีลำดับเบสของดีเอ็นเอที่เหมือนกันทุกประการเมื่อแรกเกิด แสดงให้เห็นว่า กลไกเหนือพันธุกรรมหรือเอพิเจเนติกส์อาจเป็นสาเหตุหรือมีความเกี่ยวข้องกับการเกิดโรคออทิซึมสเปกตรัม รีโทรทรานสโพซอนไลน์วัน (retrotransposon LINE-1) เป็นส่วนของจีโนมที่สามารถเคลื่อนที่ได้ และมีอยู่ถึงประมาณร้อยละ 20 และในจีโนมมนุษย์ อย่างไรก็ตามบทบาทของ LINE-1 ในผู้ที่เป็นออทิซึมสเปกตรัมยังคงไม่ทราบแน่ชัด แต่มีรายงานว่าพบการควบคุมที่ผิดปกติของ LINE-1 ในสมองของหนูที่ขาด MECP2 และมีพฤติกรรมคล้ายออทิซึมสเปกตรัม ดังนั้นคณะผู้วิจัยจึงทำการวิเคราะห์ระดับเมธิเลชันของ LINE-1 เพื่อตรวจสอบการเปลี่ยนแปลงระดับเมธิเลชันของ LINE-1 ในเซลล์ของผู้ที่เป็นออทิซึมสเปกตรัม และผลต่อการแสดงออกของยีน รวมไปถึงบทบาทหน้าที่ทางชีวภาพของยีนนั้น ๆ โดยทำการศึกษาในเซลล์ไลน์เม็ดเลือดขาว (lymphoblastoid cell lines; LCLs) ของผู้ที่เป็นออทิซึมสเปกตรัมและคนปกติ นอกจากนี้ยังได้ทำการวิเคราะห์ทางชีวสารสนเทศศาสตร์ร่วมด้วย โดยได้ทำการวิเคราะห์ร่วมระหว่างรายชื่อยีนที่มี LINE-1 แทรกตัวอยู่และมีการแสดงออกที่ผิดปกติในผู้ที่เป็นโรคออทิซึมสเปกตรัม และได้คัดเลือกรายชื่อยีนมาทำการศึกษาบาทหน้าที่ทางชีวภาพ และความสัมพันธ์ของเครือข่ายการควบคุมยีนของยีนเหล่านั้นด้วยโปรแกรม Ingenuity Pathway Analysis (IPA) ซึ่งพบว่ายีนเหล่านั้นอาจจะมีความสัมพันธ์กับโรคทางระบบประสาทและสมอง ในส่วนของการวิเคราะห์ระดับเมธิเลชันของ LINE-1 ในดีเอ็นเอซึ่งสกัดมาจาก LCLs ของผู้ที่เป็นโรคออทิซึมสเปกตรัมและคนปกติ และดำเนินการโดยใช้เทคนิคการวิเคราะห์ Combined Bisulfite Restriction Analysis (COBRA) ซึ่งพบว่าใน LCLs ของกลุ่มตัวอย่างที่เป็นออทิซึมสเปกตรัมมีระดับเมธิเลชันของ LINE-1 ที่เปลี่ยนแปลงไปเมื่อเทียบกับคนปกติ และเมื่อทำการวิเคราะห์ระดับเมธิเลชันของ LINE-1 ในแต่ละกลุ่มของ LCLs ที่เป็นออทิซึมสเปกตรัม ซึ่งถูกจัดแบ่งกลุ่มตามหลักเกณฑ์ของ ADI-R เนื่องจากออทิซึมสเปกตรัมเป็นโรคที่มีลักษณะอาการและความรุนแรงที่หลากหลายมาก คณะผู้วิจัยพบว่าแต่ละกลุ่มมีรูปแบบการเปลี่ยนแปลงของระดับเมธิเลชันของ LINE-1 ที่จำเพาะ และในบางกลุ่มไม่พบการเปลี่ยนแปลงของระดับเมธิเลชันของ LINE-1 นอกจากนี้ยังพบว่าการเมธิเลชันของ …

กระบวนการสร้างนิวไรท์ เป็นกระบวนการที่สำคัญในการเจริญพัฒนา การซ่อมแซมส่วนที่เสียหาย รวมถึงการสื่อสารระหว่างเซลล์ของเซลล์ประสาท ซึ่งหากกระบวนการการสร้างนิวไรท์เกิดความผิดปกติสามารถก่อให้เกิดโรคความผิดปกติของระบบประสาท เช่น โรคอัลไซเมอร์ โรคพาร์กินสัน และโรคฮันติงตัน เป็นต้น ในการศึกษาวิจัยนี้ศึกษาฤทธิ์การต้านสารอนุมูลอิสระ และการกระตุ้นการสร้างนิวไรท์จากสารสกัดเมล็ดของหมามุ่ยไทย และหมามุ่ยอินเดียในเซลล์ประสาทเพาะเลี้ยงชนิด Neuro2a การศึกษาคุณสมบัติในการต้านสารอนุมูลอิสระโดยวิธี DPPH และ ABTS assays นอกจากนั้นทำการศึกษาอัตราการรอดชีวิตของเซลล์ประสาทโดยวิธี MTT พร้อมกับศึกษาการแสดงออกของยีนที่เกี่ยวข้องกับการกระตุ้นการงอกของนิวไรท์ ประกอบด้วย Ten-4 และ GAP-43 โดยวิธี Real-time PCR ซึ่งจากการทดสอบพบว่าสารสกัดจากเมล็ดหมามุ่ยไทย และเมล็ดหมามุ่ยอินเดียด้วยตัวทำละลายเอทานอลมีความสามารถในการต้านสารอนุมูลอิสระ และพบว่าสารสกัดจากเมล็ดหมามุ่ยไทย และเมล็ดหมามุ่ยอินเดียไม่เป็นพิษต่อเซลล์ประสาท จากนั้นทำการศึกษาฤทธิ์ และกลไกของสารสกัดจากสมุนไพรต่อการกระตุ้นการงอกของนิวไรท์ โดยพบว่าสารสกัดจากเมล็ดหมามุ่ยไทยด้วยตัวทำละลายเอทานอล สามารถกระตุ้นการสร้างนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน Ten-4 และ GAP-43 ในขณะที่สารสกัดจากเมล็ดของหมามุ่ยอินเดียด้วยตัวทำละลายเฮกเซนกระตุ้นการสร้างนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน Ten-4 อีกทั้งศึกษาผลการยับยั้งการแสดงออกของยีน Ten-4 ต่อการกระตุ้นการสร้างนิวไรท์ของสารสกัดสมุนไพร ซึ่งพบว่ามีการสร้างนิวไรท์ และความยาวของนิวไรท์ลดลง จากการศึกษาสามารถบ่งชี้ได้ว่าสารสกัดจากเมล็ดหมามุ่ยไทย และหมามุ่ยอินเดียสามารถปกป้องเซลล์ประสาทจากสารอนุมูลอิสระได้ และสามารถกระตุ้นการงอกของนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน GAP-43 และ Ten-4 ซึ่งเป็นอีกทางเลือกหนึ่งในการพัฒนายาการกระตุ้นการเจริญพัฒนาของเซลล์ประสาทสำหรับป้องกัน หรือรักษาโรคความผิดปกติของระบบประสาท

โรคออทิซึมสเปกตรัม (Autism Spectrum Disorder, ASD) คือ โรคทางพัฒนาการระบบประสาทที่พบในเพศชายมากกว่าเพศหญิงโดยไม่ทราบสาเหตุ สาเหตุการเกิดโรคออทิซึมสเปกตรัมยังไม่เป็นที่ทราบแน่ชัดแต่การได้รับบิสฟีนอลเอขณะตั้งท้องมีความสัมพันธ์ต่อการเกิดโรคออทิซึมสเปกตรัมในลูกหนู อย่างไรก็ตามยังไม่มีการศึกษาแน่ชัดว่าการได้รับบิสฟีนอลเอขณะตั้งท้องส่งผลต่อการแสดงออกของยีนที่เกี่ยวข้องกับโรคออทิซึมสเปกตรัมหรือไม่ โดยเฉพาะเมื่อวิเคราะห์แบบแยกเพศ ในการศึกษาครั้งนี้ ทางคณะผู้วิจัยจึงทำการศึกษาผลของการได้รับบิสฟีนอลเอในขณะตั้งท้องต่อการแสดงออกของยีนในสมองและความเกี่ยวข้องกับโรคออทิซึมสเปกตรัมรวมไปถึงหน้าที่ของเซลล์ระบบประสาท ในการทำนายว่าการได้รับบิสฟีนอลเอสามารถเปลี่ยนแปลงการแสดงออกของยีนและมีความสัมพันธ์กับโรคออทิซึมสเปกตรัมหรือไม่ ทางผู้วิจัยได้ทำการวิเคราะห์แบบ meta-analysis โดยใช้ข้อมูลการแสดงออกของยีนที่ผิดปกติจากการได้รับบิสฟีนอลเอที่ได้ตีพิมพ์ไปแล้วและใช้การวิเคราะห์แบบ hypergeometric distribution หาความสัมพันธ์ระหว่างกลุ่มยีนดังกล่าวกับรายชื่อยีนจากฐานข้อมูลโรคออทิซึมสเปกตรัมต่าง ๆ สำหรับการศึกษาถึงผลของการได้รับบิสฟีนอลเอขณะตั้งท้องต่อการแสดงออกของยีนที่มีความเกี่ยวข้องกับโรคออทิซึมสเปกตรัมนั้น คณะผู้วิจัยได้นำสมองส่วนฮิปโปแคมปัสที่แยกได้จากลูกหนูที่แม่ได้รับบิสฟีนอลขณะตั้งท้องมาวิเคราะห์ทางทรานสคริปโตมิกส์ด้วยเทคนิค RNA-seq และใช้โปรแกรม Ingenuity Pathway Analysis วิเคราะห์อินเตอร์แอกโตมและบทบาทหน้าที่ทางชีวภาพของกลุ่มยีนที่มีการแสดงออกผิดปกติ ยีนที่มีความสัมพันธ์กับโรคออทิซึมสเปกตรัมถูกคัดเลือกเพื่อยืนยันด้วยเทคนิค quantitative RT-PCR นอกจากนี้เซลล์ระบบประสาทที่แยกได้จากลูกหนูถูกนำมาเพาะเลี้ยงเพื่อศึกษาความยาวของสายใยประสาท การศึกษาพบว่า จากการวิเคราะห์ทาง meta-analysis ยีนที่มีการแสดงออกผิดปกติจากการได้รับบิสฟีนอลเอมีความสัมพันธ์อย่างมีนัยสำคัญกับหน้าที่และโรคออทิซึมสเปกตรัม การวิเคราะห์ RNA-seq ในสมองส่วนฮิปโปแคมปัสที่แยกได้จากลูกหนูที่แม่ได้รับบิสฟีนอลเอขณะตั้งท้องพบว่ายีนที่มีการแสดงออกผิดปกติมีความสัมพันธ์กับโรคออทิซึมสเปกตรัม เป็นที่น่าสนใจอย่างยิ่งว่ากลุ่มยีนที่เคยมีการรายงานว่ามีความผิดปกติในโรคออทิซึมสเปกตรัม ได้แก่ Auts2 Foxp2 และ Smarcc2 แสดงออกผิดปกติอย่างจำเพาะต่อเพศ การวิเคราะห์เชิงอินเตอร์แอกโตมพบว่ายีนที่มีการแสดงออกผิดปกติจากการได้รับบิสฟีนอลเอมีความเกี่ยวข้องอย่างมีนัยสำคัญกับบทบาทหน้าที่ทางระบบประสาทที่เคยมีรายงานในผู้ป่วยออทิซึมสเปกตรัม การวิเคราะห์ความยาวของสายใยประสาทพบว่าการได้รับบิสฟีนอลเอส่งเสริมความยาวของสายใยประสาทและการแตกแขนงของสายใยประสาทแต่ลดขนาดของตัวเซลล์อย่างมีนัยสำคัญทางสถิติ จากการศึกษาครั้งนี้พบว่าการได้รับบิสฟีนอลเอขณะตั้งท้องส่งผลต่อการแสดงออกที่ผิดปกติของยีนในสมองส่วนฮิปโปแคมปัสและกลุ่มยีนดังกล่าวมีความเกี่ยวข้องกับโรคออทิซึมสเปกตรัมและแสดงออกจำเพาะในแต่ละเพศประกอบกับการได้รับบิสฟีนอลเอขณะตั้งท้องส่งเสริมความยาวของสายใยประสาท การศึกษานี้แสดงให้เห็นว่าการได้รับบิสฟีนอลเอขณะตั้งครรภ์อาจเป็นสาเหตุหรือเพิ่มความเสี่ยงต่อโรคออทิซึมสเปกตรัมและอาจจะเป็นหลักฐานที่อธิบายถึงความชุกของโรคนี้ในเพศชายที่มากกว่าเพศหญิงซึ่งควรศึกษาเพิ่มเติมในอนาคต

Human cytomegalovirus (HCMV) is a ubiquitous viral pathogen. In individuals with fully functioning and mature immune systems, HCMV is associated with mild symptoms prior to establishing latency. In individuals with naïve or compromised immune systems, HCMV is capable of causing severe organ damage. HCMV is the leading infectious cause of congenital birth defects and a major non-genetic cause of hearing loss. Unfortunately, antiviral treatment options lack diversity due to limited knowledge of virion replication. If HCMV replication were better understood, new antiviral treatments could be developed.

Wastewater application to land can be a useful tool for mitigating impacts of nutrient enrichment on aquatic systems. A land application treatment system at VCU’s Rice Rivers Center in Charles City County, VA provided an opportunity to study the impact of wastewater addition on the biogeochemistry of forests representative of the Mid-Atlantic Coastal Plain. Nutrient concentrations in throughfall and leachate were measured at Treatment and Control sites to assess differences in nutrient deposition and retention. These nucleic acid binding proteins in prokaryotes can play a vital role in many cellular processes, including replication, transcription, gene expression, recombination, and repair, to name a few. SH3 and multiple ankyrin repeat domains 3 (SHANK3) is a multidomain scaffold protein that is highly augmented in the postsynaptic density (PSD) of excitatory glutamatergic synapses within the central and peripheral nervous systems. SHANK3 links neurotransmitter receptors, ion channels, and other critical membrane proteins to intracellular cytoskeleton and signal transduction pathways. Mutations in SHANK3 are linked with a number neuropsychiatric disorders including autism spectrum disorders (ASDs). Intellectual disability, impaired memory and learning, and epilepsy are some of the deficits commonly associated with ASDs that result from mutations in SHANK3. Niemann Pick Type C (NPC) disease is a fatal childhood neurological disease caused by mutations in the NPC-1 protein, resulting in cholesterol buildup in the late endosomes. StarD4 and StarD5 are cholesterol binding proteins that play a role in the intracellular cholesterol transport. In this study we overexpress StarD4 and StarD5 in in vitro and in vivo models, and find evidence of amelioration of the NPC phenotype. This study demonstrates that the overexpression of these proteins has the potential to be a therapeutic treatment for NPC disease.

DNA methylation is necessary for learning, memory consolidation and has been implicated in a number of neuropsychiatric disorders. Obtaining high quality and comprehensive data for the three common forms of methylation in brain is challenging for methylome-wide association studies (MWAS). To address this we optimized a panel of enrichment methods for screening the brain methylome. Results show that these enrichment techniques approach the coverage and fidelity of the current gold standard bisulfite based techniques. Inflammation is a homeostatic response to tissue injury or infection, which is normally short- lived and quickly resolves to limit tissue damage. In contrast, chronic inflammation has been linked to a variety of human diseases, including cancers such as glioblastoma multiforme (GBM). GBMs are very aggressive tumors with very low patient survival rates, which have not improved in several decades. GBM tumors are characterized by necrosis and profound inflammation; with cytokines secreted by both GBM cells and the tumor microenvironment. The objective of this project was to identify important sources of bacterial loading in Plaster Creek (Kent County). The following goals were set to reach this objective: 1) the relative contributions of bacterial loading for 10 major tributaries will be assessed and the five most egregious contributors will be prioritized; 2) molecular markers will be used to try to identify the primary taxonomic sources, and 3) to pinpoint bacterial loading locations using molecular markers and scent-trained canines in sub-watersheds.

A key idea in de novo modeling of a medium-resolution density image obtained from cryo-electron microscopy is to compute the optimal mapping between the secondary structure traces observed in the density image and those predicted on the protein sequence. When secondary structures are not determined precisely, either from the image or from the amino acid sequence of the protein, the computational problem becomes more complex. We present an efficient method that addresses the secondary structure placement problem in presence of multiple secondary structure predictions and computes the optimal mapping. Three-dimensional density maps of biological specimens from cryo-electron microscopy (cryo-EM) can be interpreted in the form of atomic models that are modeled into the density, or they can be compared to known atomic structures. When the central axis of a helix is detectable in a cryo-EM density map, it is possible to quantify the agreement between this central axis and a central axis calculated from the atomic model or structure. Skeletal muscle wasting occurs as a corollary of numerous physiological conditions, including denervation, immobilization, and aging. The E3 ubiquitin ligases MuRF1 and MAFbx are induced under nearly all atrophy conditions and are believed to play a key role in protein degradation. Data in this thesis provides interesting new evidence that MuRF1 may also act as a transcriptional modulator of atrophy-induced genes or atrogenes. The transcriptional regulation of MuRF1 and MAFbx were characterized using a reporter gene system and exhibited repressed activity in C2C12 cells overexpressing MuRF1. The conserved DnaA-oriC system is used to initiate replication of primary chromosomes throughout the bacterial kingdom; however, bacteria with multipartite genomes evolved distinct systems to initiate replication of secondary chromosomes. In the cholera pathogen, Vibrio cholerae, and in related species, secondary chromosome replication requires the RctB initiator protein. Here, we show that RctB consists of four domains. The structure of its central two domains resembles that of several plasmid replication initiators. RctB contains at least three DNA binding winged-helix-turn-helix motifs, and mutations within any of these severely compromise biological activity. Myo-inositol phosphates (phytates) are important biological molecules produced largely by plants to store phosphorus. Phytate is very abundant in many different soils making up a large portion of all soil phosphorus. This review assesses current phytase science from the perspective of its substrate, phytate, by examining the intricate relationship between the phytate-hydrolyzing enzymes and phytate as their substrate. Specifically, we examine available data on phytate’s structural features, distribution in nature and functional roles. Adult neurogenesis, the process of generating mature neurons from neuronal progenitor cells, makes critical contributions to neural circuitry and brain function in both healthy and disease states. Neurogenesis is a highly regulated process in which diverse environmental and physiological stimuli are relayed to resident neural stem cell populations to control the transcription of genes involved in self-renewal and differentiation. Understanding the molecular mechanisms governing neurogenesis is necessary for the development of translational strategies to harness this process for neuronal repair. Politics and science do not always go hand-in-hand. Nowhere was this more clear than in the Flint Water Crisis. Negligence towards growing levels of lead poisoning in drinking water led to incredibly deleterious effects on Flint's citizens. The chemistry of equilibrium and the shortcomings of local leaders led to Flint's water becoming a crisis.

SMYD3 plays a key role in cancer cell viability, adhesion, migration and invasion. SMYD3 promotes formation of inducible regulatory T cells and is involved in reducing autoimmunity. However, the nearly “closed” substrate-binding site and poor in vitro H3K4 methyltransferase activity have obscured further understanding of this oncogenically related protein. Here we reveal that SMYD3 can adopt an “open” conformation using molecular dynamics simulation and small-angle X-ray scattering. This ligand-binding-capable open state is related to the crystal structure-like closed state by a striking clamshell-like inter-lobe dynamics.
