Application of chemotherapeutic agents in current cancer treatment has been limited by adverse effects as poor selectivity results in systemic toxicity; most chemotherapy approaches also experience inherited or acquired drug resistance which lead to reduced treatment outcome. Research efforts have focused on the discovery of novel chemotherapies that overcome the limitations mentioned above. Proteins inherently fluctuate between conformations to perform functions in the cell. For example, they sample product-binding, transition-state-stabilizing and product-release states during catalysis, and they integrate signals from remote regions of the structure for allosteric regulation. However, there is a lack of understanding of how these dynamic processes occur at the basic atomic level. This gap can be at least partially addressed by combining variable-temperature (instead of traditional cryogenic temperature) X-ray crystallography with algorithms for modeling alternative conformations based on electron-density maps, in an approach called multitemperature multiconformer X-ray crystallography (MMX). This study explores the potential of Raman spectroscopy, coupled with multivariate regression techniques and a protein separation technique (ion exchange chromatography), to quantitatively monitor diagnostically relevant changes in high molecular weight proteins in liquid plasma. Measurement protocols to detect the imbalances in plasma proteins as an indicator of various diseases using Raman spectroscopy are optimised, such that strategic clinical applications for early stage disease diagnostics can be evaluated. The MLR family of COMPASS-like complexes are histone methyltransferase complexes that are associated with the activation of gene enhancers. In D. melanogaster, Cara mitad (Cmi, also known as Lpt) and Trithorax related (Trr) are central subunits of a complex orthologous to mammalian Lysine methyltransferase 2 C and D (KMT2C and KMT2D, also known as MLL3 and MLL2/4) that catalyze H3K4 monomethylation. Recent strategies for anticancer drug design have been focused on utilizing antibody as a drug or targeted moiety for targeted drug delivery. Antibody−drug conjugates (ADCs) have become a promising new class of targeted therapeutic agents for treatment of cancer. ADCs are designed to preferentially direct a cytotoxic drug to a cell-surface antigen recognized by an antibody. However, there are some challenges in developing ADCs, such as limited solid tumor penetration, high manufacturing costs and antibody-drug stoichiometry. Over the past two decades, protein kinases have been intensively investigated as targets to treat neoplastic diseases. Many protein kinase inhibitors not only have therapeutic potential but are becoming invaluable reagents for the study of cell signaling.

We aspired to use our Cyclin-Dependent Kinase 8 inhibitor, Q-12, as a probe for biomarker discovery for CDK8 inhibitor sensitive tumor types. Q-12 shows potent inhibition of cell viability and induction of apoptosis process in some triple-negative breast cancer and colorectal cancer cell lines in vitro. The traditional anticancer drugs are distributed in vivo through systemic blood circulation with a very small portion reaching the tumor site. Targeted drug delivery systems are developed in efforts to concentrate the drug molecules in the tissue of interest while reducing the drug distribution to healthy tissues to reduce the side effects. Liposomes are colloidal systems composed of amphiphilic molecules that assemble into vesicle structures in aqueous media. They are common carriers for targeted drug delivery with the advantages of low toxicity, low immunogenicity and the ability of encapsulating both lipophilic and hydrophilic drugs.

The sarco/endoplasmic reticulum calcium ATPase (SERCA) is the major regulator of Ca2+ levels in the cell. Deficient calcium handling in the heart has been linked to heart failure, a leading cause of death in developed countries. As of today, targeting SERCA to enhance cardiac function has not been successful due to lack of details about SERCA structural dynamics during Ca2+ transport.

The title compound, [NiBr(C25H29N2O)], contains an NiII atom with a slightly distorted square-planar coordination environment defined by one O and two N atoms from the 2-{[(8-aminonaphthalen-1-yl)imino]methyl}-4,6-di-tert-butylphenolate ligand and a bromide anion. The Ni—O and Ni—N bond lengths are slightly longer than those observed in the phenyl backbone counterpart, which can be attributed to the larger steric hindrance of the naphthyl group in the structure of the title compound. The molecule as a whole is substantially distorted, with both the planar naphthalene-1,8-diamine and imino–methyl– phenolate substitutents rotated against the NiN2OBr plane by 38.92 (7) and 37.22 (8), respectively, giving the molecule …

Nucleic acids are complex macromolecules that are susceptible to both endogenous and exogenous damage. This study explored damage resulting from interactions with environmental nucleophilic toxins, such as a variety of diols and amines found in industry. These nucleophiles can react with electrophilic groups, such as 5-formylcytosine. 5-formylcytosine is an oxidation product of the epigenetic base 5-methylcytosine. It is typically removed by thymine DNA glycosylase (TDG) but is known to accumulate in the genome, making the formyl group susceptible to attack. Many signal transduction pathways are regulated by guanine nucleotide-binding (G?) proteins, which function as molecular switches fluctuating between active and inactive conformations. Proper function depends on three flexible switch regions that are involved in the relatively slow hydrolysis of GTP. Deep sequencing studies have found mutations in the GNAS and GNAI1 genes involved in tumorigenesis, among which include a mutation corresponding to a highly conserved arginine residue in the switch II region. Angiogenesis in NSCLC has been identified as important therapeutic target in combination with EGFR TKIs. However, only small incremental advancements have been made for the use of angiogenesis inhibitors in NSCLC and it remains elusive why the inhibition of VEGF-mediated neovascularization is not therapeutically efficacious. I present experimental evidence that a subpopulation of NSCLC cells with EGFR TKI-induced EMT contributes toward the attenuation of the response to EGFR TKI therapy. Durian (Durio zibethinus M.), a major economic fruit, actively produces infamous sulfur-containing compounds during ripening. However, sulfur metabolism which is important in this fruit is rarely investigated. In this study, the integration of metabolomics using CE-TOF/MS and transcriptomics using de novo RNA-seq assembly was performed to reveal metabolic changes involved in sulfur metabolism and chemical composition related to fruit quality. During ripening, the expression level of genes involved in sulfur metabolism is concurred with the reduced sulfate ion content in placenta and the up-accumulation of sulfur metabolism-related metabolites in pulps. A paper-based analytical devices (PADs) provides a great benefit in medical diagnostics, especially in the developing world. The first part of this dissertation describes the effect of sample viscosity on PADs for the measurement of pH and nitrite concentration in human saliva. Preliminary study found that viscosity had a significant effect on the calibration curve of nitrite and a consistent curve could be generated after added washing buffer. Nitrite measurement in artificial saliva with viscosity 1.87-5.10 mPa-s was tested on the proposed device and also showed a good correlation with the standard spectrophotometry method (p=0.8484, paired sample t-test, n=20). Mango (Mangifera indicia L.) is a popular tropical fruit which has a great value in both domestic and international markets. Mango is a climacteric fruit which ethylene production increases dramatically, thus leading to a quick ripening process and a short postharvest life. Polyamines (PAs) have been reported to be involved in fruit ripening. Changes in endogenous free polyamines accumulation were measured in two mango cultivars; ‘Nam Dok Mai No.4’ and ‘Nam Dok Mai Si Tong’. Mangoes were allowed to ripen at 25±1 ºC after harvest. Acute hepatopancreatic necrosis disease (AHPND) caused by the bacterium Vibrio parahaemolyticus carrying toxin produced plasmid (VPAHPND), has led to severe mortalities in farmed penaeid shrimp throughout Asia. Our studies demonstrated that a chronic non-lethal heat shock (chronic-NLHS) could enhance resistance of the Pacific white shrimp Penaeus vannamei to VPAHPND infection. Shrimp exposed to chronic-NLHS (28 °C to 38 °C, 5 min for 7 days) had higher survival rate (>50%) than that of the control non-heated shrimp (20%) when they were challenged with VPAHPND. It is widely used as a nutritional supplement and a precursor for the synthesis of food additives. In the RNA interference (RNAi) process, small RNAs called microRNAs (miRNAs) modulate gene expression post-transcriptionally by acting as guides to direct Argonaute (Ago)-containing RNA-induced silencing complexes (RISCs) to targeted mRNAs. The Penaeus monodon miR-750 was previously reported as the highly upregulated miRNA during white spot syndrome virus (WSSV) infection. In this study, the computational program and 2D gel electrophoresis technique were used to identify miR-750 target genes. Produced by levansucrase that uses sucrose as a substrate, levan and levan oligosaccharides (GFn) have potential applications in food and pharmaceutical industries such as prebiotics, anti-obesity and anti-tumor agents. Previous study reported that N251A and N251Y mutants of levansucrase from Bacillus licheniformis RN-01 could effectively produce short-chain oligosaccharides upto GF3, instead of long-chain levan synthesis. This study hypothesized that these mutations probably reduced GF3 binding affinity in the active site of levansucrase that contains fructosyl-Asp93 intermediate and caused GF3 to be in an unfavorable orientation for transfructosylation; therefore, levansucrase could not effectively extend GF3 to GF4. The previous study of OsCam1-1 showed that the transgenic KDML105 rice over-expressing OsCam1-1 was more tolerant to salt stress than the wild type, so to investigate the role of calmodulin encoded by OsCam1-1 in the salt-responsive mechanism, the transcriptome approach using RNA-seq technique was applied. Transcriptome profile of the transgenic KDML105 rice over-expressing OsCam1-1 under 4 hr of salt stresss (150 mM NaCl) revealed that several cell processes were affected; signaling, hormone-mediated regulation, transcription, secondary metabolism, stress responses, lipid metabolism, glycolysis, TCA cycle, glyoxylate cycle, photosynthesis, and carbohydrate metabolism. The IKK-NF-κB signaling cascade is one of the crucial responsive mechanisms in inflammatory and immune responses. The key kinase proteins called inhibitor of kappa B kinases (IKKs) serve as the core elements involved in cascade activation. Here, the complete open reading frames of IKK homologs including PmIKKβ, PmIKKε1 and PmIKKε2, from the black tiger shrimp Penaeus monodon were identified and characterized for their functions in shrimp antiviral responses. The PmIKK transcripts were widely expressed in various examined tissues and the PmIKKε protein was detected in all three types of shrimp hemocytes. Human immunodeficiency virus type-1 (HIV-1) can cause acquired immunodeficiency syndrome (AIDS), a world serious public health problem. HIV-1 protease (HIV-1 PR) and reverse transcriptase (HIV-1 RT) are necessary target enzymes which have been used for antiretroviral drugs development. Herein, Lignosus rhinocerus (LR), Auricularia polytricha (AP) and Astraeus spp. (AH) crude extracts were screened for inhibitory activities on both enzymes. Crude hexane extract of AP (APH) exhibited significant inhibition on both HIV-1 PR and RT activities not only in an in vitro non-cell based but also cell based assays. วิทยานิพนธ์นี้เป็นการนำเสนองานวิจัย การพัฒนาวิธีการใหม่เพื่อควบคุมและเฝ้าระวังโรคติดเชื้อ โดยประกอบด้วยเนื้อหาสำคัญสองส่วน ได้แก่ (i) การพัฒนาแบบจำลองเซลล์ลำไส้ของมนุษย์ที่เพาะเลี้ยงบนกระดาษที่มีการดัดแปรพื้นผิว เพื่อใช้ประโยชน์ในการศึกษากลไกการบุกรุกของเชื้อจุลชีพก่อโรคในระบบทางเดินอาหาร และ (ii) การพัฒนาต้นแบบไบโอเซนเซอร์แบบไร้สายเพื่อการตรวจติดตามการเจริญของเชื้อจุลชีพก่อโรค โดยในส่วนแรก เซลล์ลำไส้ของมนุษย์ HT-29 จำนวน 200,000 เซลล์ ถูกเพาะเลี้ยงในบริเวณชอบน้ำของกระดาษกรอง Whatman No.1 โดยกระดาษถูกดัดแปรพื้นผิวด้วยสารแมทริกซ์นอกเซลล์ (Extracellular matrix: ECM) ชนิดต่าง ๆ ได้แก่ Matrigel, Collagen-1 และ Laminin พื้นที่เพาะเลี้ยงเซลล์มีขนาด 12.56 ตารางมิลลิเมตร ภายใต้สภาวะที่เหมาะสม เซลล์ HT-29 ที่ถูกเพาะเลี้ยงบนกระดาษ มีชีวิตรอดได้ถึง 28 วัน และแสดงลักษณะของเซลล์ที่เจริญเต็มที่ โดยมีการแสดงออกของวิลไล (villi) ที่ผิวเซลล์ด้านบน (apical) และมีการแสดงออกของโปรตีน ZO-1 ซึ่งแสดงถึงเซลล์มีการสร้างไทต์จังก์ชันเกิดขึ้น จากการใช้สารเชื่อมพันธะสำหรับเชื่อมพันธะ ECM แต่ละชนิด พบว่าเมื่อใช้สารเจนิพินในการเชื่อมพันธะ เซลล์มีชีวิตรอดทั้งหมด (100 -112.8%) ในขณะที่มีเพียง 1.1 - 67.3 % ของเซลล์รอดชีวิต หากใช้สารกลูตารัลดีไฮด์ในการเชื่อมพันธะ แต่การใช้สารเชื่อมพันธะทั้งสองชนิด ทำให้เซลล์ไม่สามารถแบ่งตัวมีรูปร่างแบบสามมิติได้ เมื่อสังเกตด้วย SEM เซลล์มีลักษณะแบน การดัดแปรพื้นผิวกระดาษด้วยสาร ECM ชนิด Matrigel ส่งผลให้เซลล์มีชีวิตรอดมากที่สุดและสาร ECM ชนิด Laminin ทำให้เซลล์เกิดการแปรสภาพ (cell differentiation) ได้ดีที่สุด งานวิจัยส่วนที่ 2 เป็นการพัฒนาไบโอเซนเซอร์ไร้สายสำหรับติดตามการเจริญของเชื้อจุลชีพก่อโรค ได้แก่ เชื้อ Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Streptococcus gordonii และ Candida albicans พบว่าเชื้อดังกล่าวสามารถสร้างไบโอฟิล์มบนผิวของขั้วไฟฟ้าคาร์บอนอสัณฐาน (glassy carbon electrode) และสามารถให้ค่าศักย์ไฟฟ้าที่เป็นลบได้ …

Gamma-Hydroxybutyrate (GHB) is an endogenous short-chain fatty acid formed from Gamma-aminobutyric acid (GABA). Clinically, GHB is marketed in the United States as Xyrem to treat narcolepsy with cataplexy and in Europe for the treatment of alcohol withdrawal and narcolepsy. However, the illicit use and abuse of GHB occurs due to its sedative/hypnotic and euphoric effects. Monocarboxylate transporters (MCTs and SMCTs) are integral membrane proteins that control the bidirectional transport of endogenous substrates including lactate, acetate and pyruvate.
