There has always been a connection between biology and architecture. Michael Pawlyn, in Biomimicry in Architecture, coined “biomechanical designs” to describe structural techniques derived from creatures. This study investigates chitin-based creatures. Crustaceans, insects, and microscopic organisms all have exoskeletons in nature. With a focus on crabs, other crustaceans and bugs, this study examines and evaluates the formal characteristics of chitin-based organisms. The architectural creations of OXMAN, NOX, which was founded by Lars Spuybroek; and Lebbeus Woods are among those that are analyzed and studied due to their inspiration from living things. Much scholarly research analyzes extreme misogynistic language in “manosphere” forums, due to their connections to radicalization and violence. However, less attention has been given to the prevalence of such content in mainstream media, such as podcasts. The Joe Rogan Experience (JRE) podcast, a prominent and influential platform, has been praised for its wide-ranging discussions but critiqued for fostering and amplifying harmful ideologies, including misogyny. Despite its controversy, limited research has explored how pre-existing misogynistic rhetoric in online communities might shape or influence the content of JRE episodes. This study focuses on the characterization of a SABP2-interacting protein (SIP)-432. SIP432 is a putative premnaspirodiene oxygenase-like P450 enzyme. Premnaspirodiene is a 15-carbon compound that can be converted into antimicrobial compounds by a hydroxylation reaction. The interaction of SIP432 with SABP2, a critical component of salicylic acid-mediated plant immunity, implies a role for SIP432 in plant stress signaling. This study uses the T-DNA knockout mutant of Arabidopsis thaliana SIP432 (homolog of NtSIP432) to help understand its role in abiotic and biotic stress responses. The purpose of this study was to determine the prevalence rates of the tetracycline resistance genes tetA, tetB, and tetM in environmental Escherichia coli isolates. Samples were from both an urban and an agricultural waterway were filtered and incubated with Coliscan™ medium to differentiate for fecal coliforms. The resulting colonies were streaked onto EMB and then TSA plates to ensure isolation and presence of E. coli. All media used were supplemented with 10 µg/mL tetracycline in order to maintain selective pressure on the isolates. In eukaryotes, the structure and function of the Sm core and its associated small nuclear ribonucleoproteins (snRNPs) play a significant role in pre-mRNA splicing. These snRNPs exhibit a high degree of conservation. While plants also utilize alternative splicing, the conservation of plant snRNAs and other non-coding RNA such as snoRNA and scaRNA, which assemble the Sm core, remains largely unexplored. In this study, plant sequences were compared with human sequences using the multiple sequence alignment program Jalview to determine the conservation of snRNPs in other non-coding RNA. The complexity of the proteome requires strategies that efficiently characterize protein functions. Activity-Based Protein Profiling (ABPP) is a method that identifies enzyme activity by targeting key catalytic residues. This study focused on histidine, a residue involved in enzymatic catalysis, metal coordination, and protein-protein interactions. Despite its importance, histidine-specific chemical probes are limited due to its variable reactivity. This work developed and characterized a histidine-specific probe using a pyrocarbonate-based mechanism. Singleresidue assays confirmed selective reactivity of the probe with histidine over other nucleophilic amino acids. Chloroviruses, of the family Phycodnaviridae, are large, dsDNA viruses with a genome of 290 to 410 kb. These virus genomes are so large that they are classified as “giant viruses”. Within this genome, up to 16 tRNAs and 330 to 450 proteins are encoded. Among the massive amounts of genes and proteins are unique carbohydrate synthesis/metabolism genes, which are responsible for the glycosylation of chloroviruses major capsid proteins. In most viruses, these proteins are glycosylated by the host enzymes located in the endoplasmic reticulum. Instead, Chloroviruses encode their own glycosylation machinery; essentially decorating themselves. Furthermore, these chlorovirus glycans are unlike …

Senescent cells (SCs) have been documented to accumulate with age in the organism. Recent studies have shown that reducing the number of SCs in an organism result in tissue rejuvenation and extension of life expectancy. Intermittent fasting is reported to have multiple health benefits and is speculated to reduce the burden of SCs. Nothobranchius furzeri are a new model organism for studying aging and for which there are short- and long-lived strains available. We hypothesize that long- and short-lived strains of N. furzeri will accumulate SCs at different rates; and that fasting will reduce the burden of SCs.

Tris(pyrazolyl)methane (Tpm) ligands have a central carbon with 3 pyrazole rings attached. These were discovered in 1970 and have since been synthesized with many substituents on the rings. Based on the parent structure, my goal was to synthesize a fluorinated Tpm derivative. By adapting existing procedures, I have synthesized a Tpm derivative with trifluorinated methyl groups in the 3 and 5 positions of the pyrazole ring. The fluorinated ligand is expected to be electron withdrawing and exhibit interesting chemistry when bound to a metal center. Future directions of the project will be discussed.

This project aims to discover a way to improve the safety of and reduce adverse side effects of disulfide-linked antibody-drug conjugates. Disulfide-cleavable linkers for antibody-drug conjugates function by exploiting the differential concentrations of dominant thiol species in the blood plasma and in the cytosol of malignant cells. This project aimed to identify a relationship between the stability of disulfide cleavable linkers in the blood plasma and disulfide linker structure, to prevent premature release of the cytotoxic drug. Comprehensive data analysis on crystal growth in microgravity is a requisite to inform understanding of prior work. Our research constructed and analyzed a database containing data gathered from research focusing on semiconductors grown in microgravity. Data curation spanned multiple decades, countries, and semiconductor materials. All relevant reported information was recorded in the database, notably data regarding the metrics used to determine the impact space had on crystal growth: size, structure, uniformity, and performance. A new transition metal borate hydrate, Co6B24O39(OH)6(H2O)6·2.21H2O, was synthesized via a mild hydrothermal method using boric acid as the melt. The compound crystallizes in the trigonal space group P 3‾ with lattice parameters a = b = 13.17840(10) Å, c = 12.3287(2) Å, and a unit cell volume of 1854.27(4) Å3 (Z = 2). This borate hydrate crystallizes in a new structure type comprised of layers containing [BO3] trigonal planar units and [BO4] tetrahedra that are linked by CoO6 and Co(OH)2(H2O)4 octahedra via corner-sharing. G protein-coupled receptors (GPCR) can bind multiple ligands, leading to differential signaling events termed biased signaling. C-C chemokine receptor 7 (CCR7) is a GPCR known to have biased signaling between the two ligands it binds to, CCL19 and CCL21. Binding of C-C Chemokine Ligand 19 (CCL19) promotes a high level of CCR7 phosphorylation, while CCL21 leads to a much lower level of CCR7 phosphorylation; findings observed in HEK and dendritic cells. GPCR kinases (GRKs), which can control the phosphorylation patterns of a GPCR, are hypothesized to regulate biased signaling of a receptor. GRK expression differs between cell lines and cell …

Sometimes conveying research can be difficult through two-dimension models. In my project, I aimed to highlight the significance of three-dimensional visualization in science, thereby enhancing the understanding of complex biomedical concepts. Through my work in Dr. Steven Gregory’s lab, I have been studying the ribosome – the macromolecular machine that utilizes messenger RNA and transfer RNA to link together amino acids thereby synthesizing proteins. Proteins are critical for most processes that occur within every cell. The innovations scholar program spans roughly five months and included two trips to Mayo Clinic Rochester. The team of four undergraduate students and an MBA student worked with Mayo Clinic ventures to assess the viability of a few patents. The project included a 40-page comprehensive paper as well as a 30-minute presentation. The specific details are protected under an NDA agreement however our key takeaways are shared on the poster.

Alcohol use disorder (AUD) and obesity are major public health concerns, contributing to more than 28 million deaths annually. Despite extensive research, the mechanisms underlying excessive alcohol and food consumption remain incompletely understood. Emerging evidence suggests that neuro-immune crosstalk plays a pivotal role in feeding and alcohol-induced behaviors, with inflammation as a key pathological hallmark. Toll-like receptors (TLRs), traditionally recognized for their role in innate immunity, have recently been implicated in modulating neural responses to ethanol. Biofilms are structured communities of bacterial cells encased in a self-produced extracellular matrix, composed of polysaccharides, extracellular DNA, and proteins. Bacteria in a biofilm pose significant challenges in healthcare due to their resistance to disinfectants and tolerance to antibiotics. In some species, histone-like proteins were found to be important structural components of the biofilm. Nε-Lysine acetylation is a widespread post-translational modification (PTM) in bacteria, which can alter a protein’s function. Multiple myeloma (MM), a cancer of plasma B cells, is a hematological malignancy in which patients inevitably relapse and develop drug resistance. Mcl-1, a member of the anti-apoptotic subgroup of Bcl-2 family proteins, plays a critical role in the progression of multiple myeloma and contributes significantly to drug resistance. Elevated Mcl-1 expression is observed in approximately 52% of MM patients at diagnosis, increasing to 81% at relapse. Breast cancer is the most common cancer diagnosed in female adults, with 2.3 million new cases worldwide in 2020. Plants have been used in traditional medicine and are a potential source of pharmaceuticals. One example is the large Lamiaceae (mint) family. The Salvia genus, commonly known as the sages, is the largest genus in Lamiaceae with almost 900 known species. Glucagon-like peptide-1 receptor agonists (GLP-1RAs) are synthetic analogs of glucagon-like peptide-1 (GLP-1) used to treat obesity and diabetes by reducing blood glucose levels and appetite. While early rodent studies suggested a link between GLP-1RAs and thyroid cancer pathogenesis, evidence in humans remains inconclusive, with randomized controlled trials not supporting that link. Due to the rapid, widespread use of these drugs, concerns have expanded to other obesity-associated cancers, with conflicting findings on their role in cancer progression. My thesis discusses the work that I have conducted on two myosins in Toxoplasma gondii, which is an obligate intracellular parasite that has chronically infected around 10% of the American population. It causes toxoplasmosis and severe health consequences for individuals with compromised immune systems or when infections occur in utero. However, immunocompetent individuals are asymptomatic or present with subclinical symptoms. In the Heaslip lab, I investigated two myosins in the parasite: Myosin L and Myosin F. The current world's life-threatening illnesses have amplified multidrug resistance infections, bringing about immune system mayhem, thus, the quest for novel antimicrobial compounds with a broad spectrum of action. The epidermal growth factor (EGF) receptor cascade plays a crucial role in the survival and proliferation of tumor cells. Tyrosine kinase inhibitors (TKIs) are a class of drugs that inhibit epidermal growth factor receptors (EGFRs), thereby preventing the downstream signal transduction. Despite their importance, models that link spatial receptor dynamics to tumor growth remain scarce. Further, TKIs act through selective mechanisms, inhibiting active, inactive, or all receptor states, which poses a challenge to traditional modeling approaches.

Cerium oxide nanoparticles (CNPs) are recognized for their ability to scavenge reactive oxygen species (ROS), making them promising candidates for mitigating oxidative damage in biomedical applications. However, many conventional synthesis methods limit the formation of Ce³⁺ states, which are critical for this antioxidant function. Building upon previous findings that glucose can promote Ce³⁺ formation, this study investigates the broader impact of both reducing (glucose, fructose, galactose) and non-reducing (dextran, sucrose, cyclodextrin) natural sugars on CNP synthesis. CNPs were synthesized in the presence of these sugars, and the surface densities of Ce³⁺ were measured. Results showed that reducing sugars significantly increased …

Members of the periplasmic binding protein superfamily are involved in the selective passage of ligands through bacterial cell membranes. The hyperthermophilic eubacterium Thermotoga maritima encodes a putative periplasmic branched chain amino acid binding protein (TM1135). The gene was cloned into pET21a(+), overexpressed in Escherichia coli, purified using Ni-NTA chromatography, and confirmed with SDS-PAGE and MALDI mass spectrometry. Biophysical characterization of the protein was performed with fluorescence spectroscopy, absorbance spectroscopy, and circular dichroism experiments to investigate structural stability and ligand binding ability. Ischemic injury contributes to a range of global pathologies. Ischemia/Reperfusion Injury (IRI) is a paradoxical phenomenon that involves an initial restriction of blood flow followed by a sudden restoration of perfusion. This type of injury takes place in conditions such as myocardial infarction, acute kidney disease, and ischemic stroke and often leads to cellular death. Additionally, IRI exacerbates post-surgical outcomes and plays a role in graft dysfunction and transplant rejection. Despite efforts to develop therapies targeting known IRI pathways, clinical trials have largely been unsuccessful, underscoring the need for alternative mechanisms and biomarkers associated with IRI-induced apoptosis. The preservation of complex biological materials in the frozen state has been well established and plays a critical role in the advancement of clinical applications and the pharmaceutical industry. Cryopreservation of complex biological materials has several clinical applications, including vaccine preservation, bone marrow transplants, cell therapeutics, and increasing the shelf life of red blood cells (RBCs) for transfusion. However, many challenges are present, including the use of toxic protective agents and the financial burden of maintaining extremely low temperatures between 193.15 and – 77.6 K (Parihar, Kumar et al. 2023). Lyophilization, or freeze-drying, presents an opportunity to mitigate such challenges. …
