An Emulsion Dye Laser, Kenneth Lee Matheson, James M. Thorne

A laser dye which is insoluble in water has been dissolved in hexane and emulsified in a water matrix. When pumped with a nitrogen laser, this mixture was observed to lase. The emulsion is superior to a simple hexane solution because the excellent thermo-optical properties of the water matrix help prevent refractive-index gradients from degrading laser performance. This is a useful characteristic for flash-pumped dye lasers, laser-pumped dye lasers, and liquid filters. Another type of solvent system, a critical solution, is also discussed. For certain dyes, a critical solution has even better thermo-optical properties because of its ability to absorb …

The purpose of this research was to investigate the effect of selenium upon methyl mercury poisoning in Medaka fish (Oryzias latipes) embryos. Selenium has been shown to provide a protective mechanism against mercury poisoning in some adult animals. Three experiments were done. In the first, selenium and methyl mercury were put directly in the growing medium of the eggs. In the second experiment, selenium was included in the diet of the parent fish. In the third part, methyl mercury exposure was delayed three days. The results of the first experiment were quite dramatic and showed a significant difference when selenium …

A Unified Radiometric Assay System For The Gaba-Glutamate Regulating Enzymes, Robert C. Dinwoodie

The purpose of this paper was to develop a single assay system for the enzymes which regulate GABA and glutamate concentrations in brain and nerve tissue. Since all the enzymes produce L-glutamate, their activities were measured by coupling them to L-glutamate decarboxylase. Enzymatic activity was determined by measuring the release of co2 from radioactive substrates. The glutamate decarboxylase was obtained from a commercial acetone powder by simplifying existing procedures. The glutamate decarboxylase produced was of sufficient purity to be used in the coupled assays, which were checked with commercial preparations of each enzyme, where available, and with crude brain homogenates. …

Many new restriction-modification (R-M) system which are closely linked to serB and allelic to the hsdSB system of S. typimurium occur in different Salmonella serotypes. In PI transduction studies with these systems, recombinants were isolated which fell into one of three classes with reference to their specific restriction phenotype. Recombinants that restricted like the donor, recombinants that restricted like the recipient and recombinants that had lost the ability to restrict. This latter group having lost the recipient SB restriction without gaining the specific donor restriction and here called "zero recombinants,” were of special interest and were investigated.

4-Fluoroisoleucine was produced by ammonolysis of 2- bromo-4-fluoro-3-methylpentanoic acid, which resulted from the bromofluorination of 4-methyl-2-pentenoic acid. It did not inhibit Plasmodium berghei in mice at 640 mg/kg and was not toxic to the animals. The fluoroamino acid inhibited Aspergillus niger, Trichoderma viride, Myrothecium verrucaria, Trichophyton mentagrophytes, and Mucor mucedo in Czapek solution agar at a concentration between 104 and 103 μg/ml. Growth of Escherichia coli was inhibited 25% at 900 μg/ml in a defined medium

The study of metabolic compartmentation in brain has been under investigation for some twenty years. Most interpretations of the data have focused on attempts to assess the relative contributions of glia and neurons to the metabolic pictures since anatomically and functionally they are distinct. It is very probable that these cell types contain other metabolic compartments based on anatomic and subcellular subdivisions. An Immunochemical Study Of The Combining Sites Of The Second Lectin Isolated From Bandeiraea Simplicifolia (Bs Ii), Charles Wood, E. A. Kabat, S. Ebisu, Irwin J. Goldstein

Two lectins with different binding specificities have been isolated from extracts of seeds of Bandeiraea simplicifolia. The first, Bandeiraea lectin I [11] was specific for terminal non-reducing αDGalactosyl residues. It reacted with B substances from human ovarian cysts and with several galactomannans to form precipitin lines in agar gels. Polysaccharides with terminal αDGalactosyl residues, such as larch galactan, did not react. The lectin agglutinated B erythrocytes strongly but also reacted to a lower titre with A1 and very weakly with A2 erythrocytes [15, 28] indicating that terminal non-reducing αDGalNAc [24] can …

Lycopodium flabelliforme (Fern.) Blanch. sporophytes were collected from a colony west of Flat Rock in Crawford County, Illinois. The material was either ground to a coarse grade using a Wiley Mill (grindate), left whole and untreated (fresh) or water extracted with a Waring blender (extract). The effect of the sporophyte material was tested on the germination of seeds and on the growth of seedlings of Raphnus sativa L. Sporophytes subjected to the grindate treatment showed the most inhibitory effects. It was shown that the alledged inhibitory substance was heat stable and able to be concentrated. The zero‐frequency shear wave dip appearing in the depolarized Rayleigh spectrum in benzonitrile has been studied as a function of concentration and temperature. The solution study was carried out at constant viscosity equal to the viscosity of liquid benzonitrile at each temperature. The result indicates that the presence of shear wave fine structure does not depend on the collective orientational fluctuations. The orientational and vibrational relaxation times of benzonitrile were measured at various concentrations and temperatures. The orientational relaxation times show no concentration dependence at any temperature, suggesting that the pair correlation is negligible at all concentrations. The orientational relaxation …

Human placental alkaline phosphatase was purified by butanol extraction, methanol precipitation and chromatography on DEAE-cellulose. The pH optimum was found to be 10.9. The enzyme was competitively inhibited by di-ethyl-p-nitrobenzyl-phosphonate and copper sulfate. An increase in enzymatic activity occurred in the presence of the phosphate ion (HPO4) and ATP.

The enzyme was crystallized from a 40% saturated solution of ammonium sulfate and shown to be homogeneous by sedimentation analysis. Homogeneity of the enzyme was also demonstrated using poly-acrylamide-gel-electrophoresis. At a pH of 7.5% the electrophoretic pattern yielded a single band and showed no significant contamination.

Lake Mead is a deep, subtropical, moderately productive, desert impoundment with a negative heterograde oxygen profile occurring during; the summer stratification. investigations of the Boulder Basin of Lake Mead by the University of Nevada were initiated in November 1971. The primary objective of the study was to determine what effects industrial and sewage effluent from the Las Vegas metropolitan area, discharged into Las Vegas Bay, have had on the water quality and limnological conditions of Boulder Basin. The Relationship Of Reduced Levels Of Atp To Inhibited Chondrogenesis In The Production Of Limb Deformity Induced By 6-Aminonicotinamide, Val C. Sheffield

6-aminonicotinamide (6-AN), an analogue and antagonist of nicotinamide, impairs cartilage formation and results in shortening of the limb when administered to chick embryos. 6-AN forms an abnormal NAD analogue (6-ANAD), which inhibits the activity of NAD-dependent enzymes associated with the production of ATP. The mechanism of action of 6-AN was studied by measuring the biosynthesis of protein and DNA, and the sulfation and glycosylation of chondroitin sulfate in control and 6-AN treated cartilage from 8-day chick embryos. The rotational and vibrational relaxation times of acetonitrile–carbon tetrachloride solutions were investigated as a function of concentration, viscosity, and temperature using depolarized Rayleigh and Raman scattering. Using a Fabry‐Perot interferometer and single frequency laser source, we have shown that reliable results for the single particle orientational correlation times (τs) for CH3CN can be obtained by carrying out a concentration dependent depolarized Rayleigh scattering study. Raman scattering was shown to yield inconsistent results for τs in CH3CN. At constant viscosity, it was found that the Rayleigh scatteringrelaxation time (τRay) of CH3 …

Protoplasts were isolated from tobacco and barley leaves in sucrose or mannitol using commercially available cellulases and macerozymes. Barley growth and protoplast isolation and purification conditions were optimized so that protoplasts were obtained in high yields free of unwanted debris and organelles. A technique for processing barley and tobacco protoplasts for examination by scanning electron microscopy was developed in which protoplasts seem to have maintained their structural integrity. Barley and tobacco protoplasts took up 3H-B. subtilis DNA, 125I-B. subtilis DNA or 125I-M. luteus DNA as a linear function of time (0-6 hr) and DNA concentration (0-200 μg/ml). Up to 16 …

Intermolecular Potential Surfaces From Electron Gas Methods. Ii. Angle And Distance Dependence Of The A’ And A” Ar–No(X^2ii) Interactions, Glen C. Nielson, Gregory A. Parker, Russell T. Pack

Angle dependent intermolecular potential energy surfaces for the two states (2A’ and 2A”) that arise from the interaction of ground (X^2II) state NO with Ar are calculated using the electron gas model to obtain the short range interactions. The average and difference of the two interaction energies are fit to analytic forms convenient for use in scattering calculations and joined smoothly onto the long range van der Waals potential previously determined. The results, which appear to be of useful accuracy, and the applicability of the electron gas model to such open shell–closed shell interactions are discussed.

Protein subunit structure in the nuclear envelope has not been previously described. Covalent crosslinking of polypeptides provides a method for studying the subunit structure of protein-rich systems. In this study, several polypeptide crosslinking methods have been used to determine associations among polypeptides in isolated chicken erythrocyte nuclear envelope. Interchain associations that exist in the isolated membrane were fixed by crosslinking the polypeptides in the isolated nuclear envelope. Crosslinked and uncrosslinked polypeptides were resolved by electrophoresis in a dissociating detergent system. Each agent caused distinctive alterations in the gel pattern. Various methods were employed to detect changes in the peripheral degradation of thyroxine (T4) in adult Uinta ground squirrels (Citellus armatus) collected at two different altitudes (5400 and 9000 feet.) Plasma half-life of injected T4-125I (tl/2) was significantly decreased with altitude, as was T4-125I distribution space (TDS) and T4 degradation and secretion rate (TSR). In vitro deiodination of T4-125I by heart homogenates was not affected, while urinary excretion of thyroxine derived iodide was increased at high altitude 44-70 hours post-injection. Both total and free serum T4 concentrations were reduced at high altitude, although not significantly. The cause of the reduced …

1. The waters of Las Vegas Bay, a heavily utilized recreational resource, receive discharges from a variety of municipal and industrial waste sources. The U.S. Environmental Protection Agency has determined, on the basis of numerous studies, that the present water quality violates state and federal standards and constitutes a public nuisance. Consultants have advised the Sewage and Wastewater Advisory Committee that rapid abatement of the alleged pollution conditions can be achieved by an advanced wastewater treatment (AWT) plant.
