Enzymatic And Nonenzymatic Mechanisms For Ferric Leghemoglobin Reduction In Legume Root Nodules, Manuel Becana, Robert V. Klucas

Evidence is presented for the operation in nodules of at least four systems for restoring functional ferrous leghemoglobin (Lb2+) from its inactive, ferric form. (i) Reduction of ferric leghemoglobin (Lb3+) by a reductase. The enzyme is a flavoprotein of 100 kDa with two equally sized subunits and exhibits a Km of 9 µM for soybean Lb3+ component a and a K. of 51 µM for NADH. NADPH is only 30% (initial velocities) as effective as NADH. Lb3+ reductase converts 215 nmol of Lb3+to Lb2+ •CO (or Lb2+ •O2 …

Morphological features of mermithid nematodes (Mermithidae) were studied with scanning electron microscopy, using hexamethyldisilazane-air drying in comparison with critical point drying via liquid carbon dioxide. Although general morphologic preservation of both HMDS-dried and CP-dried specimens was similar, structural features of the complex cuticle and internal organization were more easily resolved at higher magnifications in the HMDS-dried nematodes. These features include the superficial cuticular annulations, the fibrillar inner cuticle and peg-like microtrabeculae. The previously undescribed microtrabeculae are of special interest since they may facilitate an interaction of the mermithid (and perhaps nameatodes in general) musculature with its body wall that, at …

We partially characterized a cell extract (CE) from Actinobacillus pleuropneumoniae serotype 1 and used the CE to test the efficacy of secreted proteins against disease. Secreted products from 4-h culture supernatants were precipitated with 20% polyethylene glycol. Analysis of the CE indicated the presence of protein, endotoxin, and carbohydrate. Hemolytic activity to bovine erythrocytes and cytotoxic activity to porcine mononuclear leukocytes was also demonstrated. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis of the CE from a 4-h culture showed a major band at 110 kilodaltons (kDa), while a CE of a 26-h culture indicated the presence of a number of additional …

Mus musculus , the house mouse (laboratory strain), Peromyscus gossypinus. the cotton mouse, Eumeces laticeps. the broad-headed skink, and Cavia porcellus, the guinea pig, were successively infested five times with larvae of Ixodes scapularis. the blacklegged tick. Tick feeding success, engorgement weight and molting success were measured after each infestation. In general, a greater percentage of ticks (PM. musculus and E. laticeps than on P. gossypinus or Q. porcellus. P. gossypinus expressed partial resistance, measured in percentage of ticks feeding, during the third infestation, but showed increased tolerance during the fourth and …

High performance liquid chromatography (HPLC)methodologies for the determination of tamoxifen and its metabolites 4-hydroxytamoxifen and n-desmethyltamoxifen, and using cis-tamoxifen as an internal standard have been developed.
Clinical analyses of tamoxifen and its metabolites in human serum and semen, were performed as follows: The analytes were extracted from serum and semen with hexane, the hexane evaporated, the dried extract then reconstituted with methanol, followed by reverse phase high performance liquid chromatography.
Between-run coefficients of variation (CV'S) for tamoxifen and n-desmethyltamoxifen for serum sample analyses, (n = 15) were, 4% and 13% at mean (SD) concentration of 140.2 (6.2) and 71.8 (9.8) …

The purpose of this study was to treat normal human fibroblasts with known tumor promoters and observe queuine modification of tRNA. The queuine insertion enzyme, tRNAguanine ribosyltransferase, was studied in vivo and in vitro. Tumor promoter-treated human fibroblast cultures exhibited variable queuine-insertion rates with a transient inhibition that correlated with variable levels of queuine modified tRNA over time in culture from passages 3 — 8. In contrast the in vitro studies showed that phorbol cetera and saccharin actually increased insertion with strong evidence indicating phosphorylation as a positive modulating force of the enzyme activity. Fermentation of dilute-acid-pretreated mixed hardwood and Avicel by Clostridium thermocellum was comparedinbatchandcontinuouscultures.Maximumspecificgrowthratesper hourobtainedon cellulosic substrateswere 0.1inbatchcultureand>0.13incontinuousculture.Cellyields(gramsofcellsper gram of substrate)inbatchculturewere 0.17forpretreatedwoodand0.15forAvicel.Ethanolandacetatewere the mainproductsobservedunderalconditions.Ethanol:acetateratios(ingrams)were approximately1.8:1in batchcultureand generallyslightlylessthan 1:1incontinuousculture.Utilizationofcellulosicsubstrateswas essentially complete in batch culture. A prolonged lag phase was initialy observed in batch culture on pretreated wood; the length of the lag phase could be shortened by addition of cell-free spent medium. In continuousculturewith-5g ofglucoseequivalentper literinthefeed,substrateconversionrelativeto theoreticalrangedfrom0.86ata dilutionrate(D)of0.05/hto0.48ata D of0.167/hforAvicelandfrom0.75 ata D of0.05/hto0.43ata D of0.11/hforpretreatedwood.Atfeedconcentrationsof<4.5g ofglucose equivalentperliter,conversionofpretreatedwoodwas80to90%atD= 0.083/h.Lowerconversionwas obtainedathigherfeedsubstrateconcentrations,consistentwitha limitingfactorotherthancellulose.Free Avicelaseactivitiesof12to84mU/mlwere observed,withactivityincreasinginthisorder:batchceliobiose, batchpretreatedwood< batchAvicel,continuouspretreatedwood< continuousAvicel.Freecellulaseactivity was higheratincreasingextentsofsubstrateutilizationforbothpretreatedwoodandAvicelunderal conditions tested. Flexible-shelled eggs of squamate reptiles are strongly influenced by their surrounding microclimate, particularly the hydric conditions of the nest. The eggshell water vapor conductance of an egg can be an indication of its requirement for water from the nest. Ackerman et al. (1985. Physiol. Zool. 58:129-137) have described the scaling relationship for eggshell water vapor conductance (GH2O) of flexible-shelled reptilian eggs (GH2O = 981.8M0.52). Inbred strain 2 guinea pigs were vaccinated with Mycobacterium bovis BCG or were left unvaccinated. They were maintained for 6 weeks on defined, isocaloric diets containing either 30% (control animals) or 10% (animals receiving low protein) ovalbumin as the sole protein source. Animals were challenged by the respiratory route with a low dose of virulent M. tuberculosis H37Rv and killed 4 weeks later. Protein-malnourished animals were not protected by previous vaccination with BCG. Lymphocytes isolated from various tissues were tested in vitro for proliferative responses to mitogen (concanavalin A) and antigen (purified protein derivative [PPD]), production of interleukin-2 (IL-2), and …

A comparison of the relationship of fermentation to chemical composition was made for forages which were wilted and ensiled at 35 to 45 percent dry matter. Trial I consisted of three forages ensiled in October, 1981: interseeded soybeans and grain sorghum. whole plant corn with added anhydrous ammonia, and whole plant corn with shelled corn added at a rate of 150 kg/t of fresh forage. Trial II consisted of two forages ensiled in 1982: interseeded soybeans and pearlmillet, and wheat. Temperatures of fermentation were collected, and chemical composition during the first 25 days of fermentation analyzed.

Basic research into reproductive physiology has begun to identify possible new approaches to controlling fertility. Gossypol, a natural polyphenol compound isolated from cottonseed oil, has been shown to be an effective male antifertility agent. The decomposition products of gossypol which are thought to possess more potent antifertility effects than the parent compound gossypol have been studied extensively in more recent research. The purpose of this research is to develop techniques and methods which could be used to identify decomposition products of gossypol. A rapid, reliable, and sensitive method for simultaneous determination of 3-methoxy-4-hydroxyphenylglycol (MHPG), Homovanillic acid (HVA), and 5-hydroxyindoleacetic acid (5- HIAA) in plasma is described. The method is based on high performance liquid chromatography with electrochemical detection. Samples were filtered with Amicon Centrifree micropartition systems and extracted with organic solvent before chromatography. Chromatographic separation of sample components was achieved isocratically in a C18 reverse-phase column. Quantitation was performed by measuring the peak height ratios of the sample components to an internal standard. Analytical recoveries ranged from 50.78 (SD 4.2%) to 67.94 (SD 3.28). A microwell based competitive enzyme immunoassay for determination of total thyroxine and triiodothyronine in human serum was developed with use of horseradish peroxidase conjugate. The assay, which can be performed in only 1.5 hours at room temperature, involves simultaneous incubation of T3- or T4- HRPO conjugate and serum sample in anti-T3 or anti-T4 gamma globulin fraction coated polystyrene microwells.

Several cell surface proteins (Mr = 120,000, 90,000, 63,000 and 47,000) apparently integral to embryonic fibroblast plasma membranes were extracted with detergent and isolated by collagen affinity chromatography. Certain of these proteins (Mr = 120,000, 90,000, and 47,000) were specifically eluted from collagen affinity columns by synthetic peptides containing the amino acid sequence arginyl-glycyl-aspartic acid (RGD). These data show that a number of collagen binding proteins exist on the embryonic fibroblast cell surface. Some of the proteins may be collagen receptors binding to RGD sequences in the collagen molecule while at least one of the proteins (Mr = 63,000) recognizes …

Fetal rat lung (FRL) cells incubated in the presence of retinoic acid or dexamethasone exhibit altered 125I-epidermal growth factor (EOF) binding capacity. Retinoic acid enhances 125I-EGF binding 3-fold following 24 hours of 37° incubation. In contrast, FRL cells incubated in the presence of dexamethasone display a 2-fold reduction in 125I-EGF binding capacity. The change in binding capacity, induced by either hormone, was due to alterations in the average number of receptors per cell as determined by Scatchard analysis of 125I-EGF binding. In the absence of retinoic acid or dexamethasone, FRL cells were calculated to have approximately …

The intent of this dissertation is to demonstrate that ferritin iron may be involved in alloxan, diquat, and radiation-dependent damage and that ceruloplasmin may afford protection by limiting the availability of ferritin iron. Initial studies demonstrated that the alloxan radical formed by glutathione-dependent reduction of alloxan reductively mobilized ferritin iron and the released iron catalyzed lipid peroxidation. SOD and catalase were without effect, whereas ceruloplasmin effectively inhibited both iron release and lipid peroxidation. Using 137Cs gamma-radiation it was demonstrated that radiolytically generated superoxide released iron from ferritin with an efficiency of 67%. N-Nitrosamines and N-nitrosamides have been reported to be mutagenic. N-Nitrosamides are direct mutagens that need no activation to be mutagenic, whereas N-nitrosamines need to be enzymatically activated to exert their mutagenicity. Oxidative demethylation of nitrosamines is a commonly accepted activation mechanism. Another pathway of nitroso compounds, denitrosation, has recently been proposed. The mechanism of denitrosation, however, is still unknown.
The purpose of this study was to use a photo -reaction model to explore the possible denitrosation mechanism of N-nitroso compounds. A novel 0.6 kb LINE family in human DNA, designated L2Hs, has been described (Musich and Dykes 1986). Studies employing clone N6.4, containing three 0.6 kb segments of this family, indicate that these sequences are interspersed and moderately repetitive. Two additional variant sequences of the L2Hs family, N6.1 and N6.3, have been identified. Restriction mapping of each cloned segment indicates similarities among N6.4, N6.3 and N6.1. When the cloned DNAs were cleaved with restriction enzymes and subjected to cross-hybridization, each cloned insert produced a pattern indicating that the sequences contained in N6.1 and N6.3 are represented in at least one …

BTV-2, 10, 11, 13 and 17 were purified by three discontinuous sucrose gradients. Viral proteins were separated from the genomic double-stranded RNA using a newly developed KCI-SDS (sodium dodecyl sulfate) precipitation method. Cyanogen bromide (CNBr) peptide mapping of VP5 and VP7 from bluetongue virus (BTV) serotypes 2, 10, 11, 13 and 17 was then done using silver-stained SDS-polyacrylamide gels. The antigenic epitopes of VP5 and VP7 digested with CNBr were also investigated with polyclonal antibodies produced against KCl-SCS precipitated BTV-13 viral proteins. Human cultured leukemia cells appear to have a decreased amount of inosine in their tRNA. When cells with inosine deficient tRNA are placed in a hypoxanthine fortified media, they incorporate hypoxanthine into their tRNA by the action of the enzyme tRNA-hypoxanthine ribosyl transferase. This generates the nucleoside inosine in the tRNA. The cultured human leukemia cell lines, CCRF-CEM, HL-60, and HGPRT(-) HL- 60, incorporate hypoxanthine into their tRNA, as determined by tRNA isolation, hydrolysis, and HPLC analysis. The purpose was to determine whether dogs and hamsters contain the same sperm-specific protamine and testis specific proteins (TP) as other eutherian mammals previously studied. The proteins (protamine and TPs) were isolated from spermatids and mature spermatozoa, and were subjected to electrophoresis in an acid-urea, polyacrylamide gel system according to the procedure of Panyim and Chalkley (1962). The results indicated that, regardless of the anatomical location of sperm acquisition, acid precipitation technique, or denaturing agents used, epididymal and testicular samples from both the dog and hamster contained protamine in elongated spermatids and mature spermatozoa. The most significant problem in maintaining platelet viability during storage is the decrease in pH resulting from accumulation of lactic acid formed by platelet anaerobic glycolysis. This study investigates nutrient alternatives to glucose for maintenance of morphology and functional integrity during platelet storage in a well-defined synthetic medium and correlates this data with purine nucleotide levels determined by high-performance liquid chromatography (HPLC) and other in vitro measures of platelet quality. Platelets prepared from pooled ABO- and Rh- identical platelet-rich plasma were stored as concentrates in the synthetic medium without added nutrients (control) and with added substrates.

Limnological monitoring was conducted in Las Vegas Bay and Boulder Basin from April to December of 1988. The purpose of the monitoring was to (i) document possible changes in water quality resulting from decreased phosphorus loading and increased ammonia in Las Vegas Wash, and (ii) establish a data base for evaluating the adequacy of water quality standards.

Current advances in fertility treatment emphasize extracorporeal procedures, such as in vitro fertilization (IVF)/embryo transfer. IVF is now used to treat several types of infertility, including male factor and idiopathic infertility. However, IVF is time consuming, costly, unpredictable, and often yields a low pregnancy rate. Thus, it would be valuable to devise laboratory procedures for better prediction of IVF outcome.
The male infertility rate can be as high as 50%. Since sperm count and percent of motility alone do not appear to be strong predictors of IVF outcome especially when these parameters are not severely abnormal, a multivariable analysis of …

Platelet factor 4 (PF4), which is released by platelets during coagulation, binds very tightly to negatively charged oligosaccharides such as heparin. To date, six other proteins are known that are homologous in sequence with PF4 but have quite different functions. The structure of a tetramer of bovine PF4 complexed with one Ni(CN)42− molecule has been determined at 3.0 Å resolution and refined to an R factor of 0.28. The current model contains residues 24–85, no solvent, and one overall temperature factor. Antifungal studies were made of mixtures of minimal inhibitory concentrations (MICs) of 8-quinolinol and its bischelates with copper(II), zinc(II), and manganese(II) and with mixed ligand chelates composed of 8-quinolinol, copper(II) and a second ligand including salicylic acid, 3-hydroxy-2-naphthoic acid, 3,5-diiodosalicylic acid, and 4-bromo-3-hydroxy-2-naphthoic acid. Mixtures of the MICs of the bischelates of 8-quinolinol with copper(II) and zinc(II) and copper(II) and manganese(II), as well as 7-iodo-8-quinolinol and its bischelate with copper(II), and 8-quinolinol and 5-iodo-8-quinolinol were also studied against six fungi: Aspergillus niger, Aspergillus oryzae, Trichoderma viride, Myrothecium verrucaria, Mucor cirinelloides, and Trichophyton mentagrophytes. The glp-1 gene functions in two inductive cellular interactions and in development of the embryonic hypodermis of C. elegans. We have isolated six mutations as recessive suppressors of temperature-sensitive (ts) mutations of glp-1. By mapping and complementation tests, we found that these suppressors are mutations of known dumpy (dpy) genes; dpy genes are required for development of normal body shape. Based on this result, we asked whether mutations previously isolated in screens for mutants defective in body shape could also suppress glp-1(ts). From these tests, we learned that unselected mutations of eight genes required for normal C. elegans morphogenesis, including …

Estrogen receptor (ER) is a ligand-dependent transcriptional regulator. ER protein has several functionally distinct regions, including a DNA-binding domain in its midportion, a ligand binding domain in the carboxy terminal region, and at least one domain, in the amino terminal region, that influences the magnitude of transcriptional enhancement (1,2). We now report the sequence of a frequently occurring ER genetic variant that carries two point mutations in the region of the gene encoding the amino terminal domain of the protein.
