The effects of free radicals in the liver tissue of 20 mice were studied. The mice, separated into acute and chronic groups, were injected with phorbol myristate acetate (PMA) to trigger the release of free radicals. Seroprevalence of Babesia ovis in Awassi sheep was studied in Urfa and environs between November 1997 and October 1998. A total of 607 serum samples were collected from sheep in 12 different localities, and tested for the presence of antibodies to B. ovis by using an indirect ELISA. Blood smears were also prepared from 110 sheep. The overall prevalence of B. ovis infection was 41.02%. B. ovis was only detected in two blood smears examined (1.82%). The purpose of the present study was to examine the effects of vitamin A and zinc interaction in laying hens fed on diets supplemented with two different levels of vitamin A (0, 10.000 IU/kg) in combination with five different levels of zinc (0, 25, 50, 100, 200 mg/kg) on egg yield, plasma and yolk concentration of zinc, and the levels of plasma triglyceride and phosphorus for 12 weeks. Expression Of Bifunctional Genes Encoding Xylanase And \Beta(1,3-1,4)-Glucanase In Gram-Positive Bacteria, Mehmet Sai̇t Eki̇nci̇, Harry J. Flint

The xylanase and \beta(1,3-1,4)-Glucanase activities encoding gene from the cellulolytic rumen anaerobe bacterium was cloned into Escherichia/Streptococcus shuttle vector for introduction into Gram-positive bacteria, including the rumen facultative anaerobe bacterium Streptococcus bovis. Activities due to the cloned gene decreased in the stationary phase in batch cultures of S. bovis, reflecting the sensitivity of the cloned enzymes to inactivation in the presence of accumulated lactic acid. Cloned gene activity was detected in the culture supernatant, indicating recognition of the cloned gene signal peptide by Gram-positive bacteria.

In this study, the influences of cumulus cells during in vitro fertilization (IVF) and their subsequent effects on mouse embryo development in vitro were studied in inbred and hybrid strain mice. Embryos at the 2-cell stage obtained after in vitro fertilization were transferred to the oviduct of foster mothers to assess their implantation rate. Mouse oocytes, which were intact at the time of in vitro fertilization, demonstrated less polyspermic fertilization and more blastocyst development than those inseminated without any cumulus cells. Identification Of A Lactose Utilization And Copper Resistance Plasmid In Lactococcus Lactis Subsp. Lactis Mcl64, Mustafa Akçeli̇k

Turkish Journal of Veterinary & Animal Sciences

Lactose negative and copper sensitive (Lac-/Cus) mutants of Lactococcus lactis subsp. lactis MCL64 were isolated after extended incubation periods at 32ºC and 37ºC. A 30.0 kb plasmid was absent in all Lac-/Cus mutants. These results showed that lactose metabolism and copper resistance were linked to the 30.0 kb plasmid in strain MCL64. Using MCL64 as a donor, the 30.0 kb plasmid transferred into Lac-/Cus recipient strains P81 and MCL64-90 with the frequencies of 4.1x10-5- 8.2x10-3 per donor cell, respectively. All transconjugants were found to be of a Lac+/ Cur phenotype.

The effects of outdoor raising on the survival and growth of Simmental calves in individual hutches were studied at Kazova State Farm, the location having a mixture of Black Sea and semiarid continental climate conditions. A total of 63 female calves born in each of the four seasons between March 1996 and February 1997 were used. The animals were kept in individual hutches for 3 months (preweaning period) and then in groups of 10 until 6 months of age (postweaning period). The purposes of this study were to determine the effects of oviposition time on egg quality and to examine the possibility of estimating egg shell weight using a formula. The interactions in some egg quality measurements were also studied. During the experiment, 80 white and 64 brown layers were housed in cages and fed with a standard layer ration. In this study, the growth, age composition, sex ratio, condition factor and spawning period of whiting (Merlangius merlangus euxinus Nordmann, 1840) on the eastern part of the Trabzon-Yomra coast were investigated. During the study, a total of 1730 fish (1122 females and 608 males) ranging in age from group I to IX were examined. In this study, the effects of experimentally induced hyperthyroidism and hypothyroidism on the erythrocyte, leucocyte and haematocrit values, the amount of haemoglobin and plasma glucose, cholesterol, triglyceride, creatinine and lactate dehydrogenase (LDH) levels were investigated. A total of 45 4-week-old female Japanese quail (Coturnix coturnix Japonica) were used. In the survey study, a total of 19 canine mast cell tumors were examined with regard to sex, age, breed of dogs, tumor location, and pathological findings in the period of 1960-1999. The majority of tumors occurred in males (78%) and between the ages of 6 and 11 years (67%). The pointer was predisposed to development of tumors (35%). The hindquarters and hind legs were most commonly involved, followed by the extragenital inguinal area and trunk. The tumors clearly had a site preference for the posterior region of the body (79%). The study was carried out to investigate the effects of sunflower meal with an enzyme mixture containing \alpha-amylase, ß-glucanase, cellulase, lipase and protease substituted for soybean meal in broiler diets on performance, intestinal weight, viscosity and pH, as well as the frequency of sticky droppings and abdominal fat. In the trial, a total of 960 one-day-old Avian broiler chicks were used. The trial lasted 6 weeks. A total of 12 experimental diets were supplied ad libitum. Survey Of European Foulbrood Disease In Bee Farms Located In Elazığ And The Surrounding Provinces, Hali̇l Şi̇mşek, Canan Özcan

Turkish Journal of Veterinary & Animal Sciences

A total of 250 specimens from 110 bee farms located in Elazığ and the surrounding provinces were taken in 1999 and 2000. The specimens were examined for European Foulbrood Disease. Under macroscopic examination clinical symptoms were noted in 12 specimens, and Paenibacillus alvei was isolated and identified from 8 of these specimens. As a result of the antibiotic sensitivity test, the isolate was found sensitive to enrofloxacin, streptomycin and erythromycin 75%, 75%, and 62,5% of the time respectively. In this study, the presence of European Foulbrood Disease was determined in 3.2% of cases in Elazığ and the surrounding provinces.

This research was carried out with the aim of improving the conservation and feed value characteristics of citrus pulps. In order to ensure and improve citrus pulp conservation even for short periods (1 week), several methods intended to inhibit yeast activity were examined. Orange and grapefruit pulps were used as feed materials and three heads of merino male yearlings fitted with rumen cannula were used as animal materials. One hundred and twenty thoroughbred breeder mares with endometritis or infertility were investigated for the detection of antibodies to Taylorella equigenitalis by the passive haemagglutination (PHA) test and ELISA between 1997 and 1999 in the Marmara region and the two tests were compared. Fifty-eight (48.33 %) of 120 sera were found to be positive by the PHA test and 52 (43.33%) by ELISA. 22 of the sera were found to be positive by the PHA test reacted at 1/64 titer, 27 at 1/128, 7 at 1/256 and 2 at 1/512 titer. Two studies were conducted to evaluate if Fertirelin acetate (an analogue of GnRH) and Gentamycin sulphate would improve pregnancy rate in healthy cows or in nonpregnant cows although they were inseminated three times or more. In this study 250 raw milk and 254 herby cheese samples collected from Van city center and neighboring villages were investigated in terms of Listeria species. For Listeria isolation,the method recommended by the FDA was used.Of the raw milk samples,6 (2.40 %)were found to be positive with regard to Listeria; 3 (1.20 %)had L.monocytogenes, 1 (0.40 %)L.innocua and 1 (0.40 %)L.welshimeri. In this study,antibodies against Brucella melitensis in aborted sheep were investigated with enzyme-linked immunosorbent assay (ELISA), standard tube agglutination test (SAT), rose bengal plate test (RBPT), complement fixation test (CFT), 2-mercaptoethanol microagglutination test (2-ME-MAT)and dithiothreitol microagglutination test (DTT-MAT). For this purpose,the genital organs of 6100 ewes from different breeds and ages were examined.There were pathomorphologic changes in 274 cases (4.49%)in the uterus,70 cases (1.14%)in the cervix,and 61 cases (1.00%)in the vagina.Inflammatory changes were primarily lesions (2.68%)observed in the uterus.These changes were catarrhal endometritis (0.52%), purulent metritis (0.32%),necrotic endometritis (0.11%), chronic nonpurulent endometritis (0.95%),granulomatous endometritis (0.13%),pyometra (0.11 %)and parametritis (0.52%). This study was carried out to determine the incidence of C.burnetii infection using the Indirect Fluorescent Antibody (IFA)Test in ewes in Elazığ and neighboring provinces.Seropositivity was detected in 38.59 %(71/184)of aborted ewes and in 11.01 %(25/227)of unaborted ewes.The seropositivity rates were 20 %in Elazığ,20% in Malatya, 27.95 %in Bingöl and 27.27% in Muş. Antibody titers ranged from 1/80 to 1/1280. Q fever should be considered an important cause of abortions in ewes in Elazığ and neighboring provinces.

In this study, 14 adult albino guinea pigs, weighing 300-350 g, were divided into two equal groups. The small intestinal segments (distal ileum)taken from the animals were kept in vitro in the isolated organ bath, and the isotonic contractions of the tissues produced by amitraz and acetylcholine were recorded on a polygraph.
