Climate change during the last 40 years has had a serious impact on agriculture and threatens global food and nutritional security. From over half a million plant species, cereals and legumes are the most important for food and nutritional security. Although systematic plant breeding has a relatively short history, con- ventional breeding coupled with advances in technology and crop management strategies has increased crop yields by 56 % globally between 1965− 85, referred to as the Green Revolution. Nevertheless, increased demand for food, feed, fiber, and fuel necessitates the need to break existing yield barriers in many crop plants. In …

Carinata (Brassica carinata A. Braun), a non-food oilseed crop and an alternative bio-jet fuel feedstock, has received attention for its potential as a low-input option for production in the semi-arid region of the Northern Great Plains of the United States. Research addressing the impacts of nitrogen (N) and sulfur (S) fertilizers on soils and greenhouse gas (GHG; CO2, N2O, and CH4) emissions from carinata production are limited. Thus, objective of this study was to evaluate the impact of different rates of N and S fertilizers applied to carinata on soil properties and GHG emissions. Field experiments were conducted in 2017 …

Large datasets of greenhouse gas and energy surface-atmosphere fluxes measured with the eddy-covariance technique (e.g., FLUXNET2015, AmeriFlux BASE) are widely used to benchmark models and remote-sensing products. This study addresses one of the major challenges facing model-data integration: To what spatial extent do flux measurements taken at individual eddy-covariance sites reflect model- or satellite-based grid cells? We evaluate flux footprints—the temporally dynamic source areas that contribute to measured fluxes—and the representativeness of these footprints for target areas (e.g., within 250–3000 m radii around flux towers) that are often used in flux-data synthesis and modeling studies. We examine the land-cover composition …

In 2008, we published the first set of guidelines for standardizing research in autophagy. Since then, this topic has received increasing attention, and many scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Thus, it is important to formulate on a regular basis updated guidelines for monitoring autophagy in different organisms. Despite numerous reviews, there continues to be confusion regarding acceptable methods to evaluate autophagy, especially in multicellular eukaryotes. Here, we present a set of guidelines for investigators to select and interpret methods to examine autophagy and related processes, and for reviewers to …

The identity of an ecological regime is central to modern resilience theory and our understanding of how systems collapse and reorganize following disturbance. However, resilience-based models used in ecosystem management have been criticized for their failure to integrate disturbance outcomes into regime identity. Assessments are needed to understand how well these classifications represent ecosystem responses that occur over management relevant time scales. We tracked post-wildfire forest and grassland dynamics 27 years after wildfire in eastern ponderosa pine savanna. We tested for differences between the assigned identity of a site (forest or grassland) versus classifications based on the site's disturbance history …

This research used the existing whole-exome sequences of eight Thai rice varieties, including the standard checks for tolerance and susceptibility, and the previously reported salt-tolerant genes to understand the mechanisms of salt tolerance in Thai rice, as well as the bulk-segregant analysis (BSA) to identify salt-tolerant QTLs in ‘Jao Khao’, the salt tolerant Thai rice. Salt responsive characteristics were used to classify tolerant and susceptible genotypes at the seedling stage. Phylogenetic analysis of SNPs from exome sequences showed that the Thai rice varieties were monophyletic and distantly related to 'Pokkali' and …

Neuroprotective Effect Of Passiflora Edulissims Against Amyloid Beta-Induced Neurotoxicity, Shuqin Cao Jan 2021

Neuroprotective Effect Of Passiflora Edulissims Against Amyloid Beta-Induced Neurotoxicity, Shuqin Cao

Chulalongkorn University Theses and Dissertations (Chula ETD)

Alzheimer’s disease (AD) is a common and devastating disease characterized by pathological aggregations of beta-amyloid (Aβ) plaques extracellularly, and Tau tangles intracellularly. While our understandings of the aetiologies of AD have greatly expanded over the decades, there is no drug available to stop disease progression. This study involves two parts. First, we demonstrate the potential of P. edulis pericarp (PP) extract in protecting against Aβ-mediated neurotoxicity in mammalian cells and Caenorhabditis elegans models of AD. We show PP protects against memory deficit, neuronal loss, and promotes longevity in the Aβ model of AD via stimulation of mitophagy, the selective cellular …

Effects Of Sacubitril/Valsartan On Cardiac Function, Mitochondrial Quality And Autophagy In A Rat Model With Chronic Mitral Regurgitation, Lalida Tantisuwat

Chulalongkorn University Theses and Dissertations (Chula ETD)

Sacubitril/Valsartan (S/V), an angiotensin receptor blocker-neprilysin inhibitor (ARNi), has demonstrated beneficial outcomes in heart failure (HF) patients. However, the effects of the drug in mitral regurgitation (MR) condition are not thoroughly elucidated. The objective of this study was to investigate the effects of S/V compared with valsartan (Val) on cardiac function, mitochondrial quality and autophagy in a rat model with chronic MR. MR was induced in male, Sprague-Dawley, rats (n=24), while the other 6 rats underwent sham surgery. Four weeks post-surgery, the MR rats were randomly divided into 3 groups: MR (n=8), Val (n=7), and S/V (n=8); one rat was …

Extending Animal Welfare Science To Include Wild Animals, Walter Veit, Heather Browning Jan 2021

Extending Animal Welfare Science To Include Wild Animals, Walter Veit, Heather Browning

Animal Sentience

Ng’s (2016) target article built on his earlier work advocating a science of welfare biology (Ng 1995). Although there were problems with the models proposed in Ng’s original paper regarding the balance of pleasure and suffering for wild animals, his call for a science of wild animal welfare was a sound one. This does not require a new discipline but just an extension of the existing frameworks and methods of animal welfare science to include wild animals.

Analysis Of Protein-Protein Interaction Network From Leukocyte Transcriptomic Profiles In Severe Covid -19 Patients, Pakorn Sagulkoo Jan 2021

Analysis Of Protein-Protein Interaction Network From Leukocyte Transcriptomic Profiles In Severe Covid -19 Patients, Pakorn Sagulkoo

Chulalongkorn University Theses and Dissertations (Chula ETD)

Coronavirus disease 2019 (COVID-19) still provides global public health issues although several vaccines and antiviral agents have been developed. Some patients experience severe conditions needed medical intensive care, and some are dead due to the failure of treatments. Therefore, identifying the key genes and underlying molecular mechanisms is necessary to discover precisely targeted drugs. Analysis of protein-protein interaction (PPI) networks provides invaluable information to find disease mechanisms and effective alternative drugs. Hence, PPI network analysis based on leukocyte transcriptomic profiles of severe COVID-19 collected from Gene Expression Omnibus (GEO) DataSets was proposed for this study. A network diffusion method called …

Structural Characterization And In Vitro Antiproliferative Activity Of Non-Specific Lipid Transfer Protein 1 (Nsltp1) From Fennel (Foeniculum Vulgare) Seeds, Mekdes Megeressa

Pharmaceutical Sciences (PhD) Dissertations

Non-specific lipid transfer proteins (nsLTPs) are cationic proteins involved in intracellular lipid shuttling, in growth and reproduction, as well as in defense against pathogenic microbes. Even though the primary and spatial structures of some nsLTPs from different plants indicate their similar features, they exhibit distinct lipid-binding specificities signifying their various biological roles that dictate further structural study. The present study determined the complete amino acid sequence, in silico 3D structure modeling, and in vitro antiproliferative activity of nsLTP1 from fennel (Foeniculum vulgare) seeds.

Fennel is a member of the family Umbelliferae (Apiaceae) native to southern Europe and the …

Amphiphilic Cell-Penetrating Peptides Containing Natural And Unnatural Amino Acids As Drug Delivery Tools And Antimicrobial Agents, David Salehi

Pharmaceutical Sciences (MS) Theses

Cell-penetrating peptides containing arginine as positively charged residues and tryptophan or diphenylalanine as hydrophobic residues were synthesized. The synthesis was accomplished through the Fmoc solid-phase peptide synthesis in the presence of HBTU and DIPEA. The side-chain protected linear peptides were cleaved from the resin and cyclized in the presence of DIC and HOAt in the solution phase overnight. MALDI-TOF mass spectrometry was used to characterize the peptides.

The cytotoxicity of the synthesized peptides was determined in CCRF-CEM (human, lymphoblast peripheral blood), and HEK-293 (human, embryonic epithelial kidney healthy) cells using the MTS assay. A concentration of 10 µM was found …
