Management and control of the low back pain as an important health problem in the industrial societies necessitates to investigate how the risk of this disease is affected by aging. Many studies have demonstrated clinical applications for the use of dental pulp stem cells (DPSC) for the treatment of various conditions. This has driven medical and scientific interest in the collection, isolation and banking of DPSC tissues for research into these potential therapies. Few studies to date have evaluated the viability of DPSC following long-term cryopreservation. The purpose of this study is to evaluate the effects of cryopreservation on dental pulp-derived stem cells (DPSC) viability over a period of three years. Hydrocephalus is a neurological disorder that typically requires a long-term implantation of a shunt system to manage its symptoms. These shunt systems are notorious for their extremely high failure rate. More than 40% of all implanted shunt systems fail within the first year of implantation. On average, 85% of all hydrocephalus patients with shunt systems undergo at least two shunt-revision surgeries within 10 years of implantation. A large portion of this high failure rate can be attributed to biofouling-related obstructions and infections. Statistical data suggests that increased use of evidence-based medical therapies has largely contributed to the decrease in American death rate caused by heart disease. And my studies are about two applications of magnetic resonance imaging (MRI) as a non-invasive approach in evidence-based health care research. In my first study, the achievement of a pulmonary valve replacement surgery was assessed on a patient with tetralogy of Fallot (TOF). Human footwear is not yet designed to optimally relieve pressure on the heel of the foot. Proper foot pressure assessment requires personal training and measurements by specialized machinery. This research aims to investigate and hypothesize about Preferred Transition Speed (PTS) and to classify the gait phase of explicit variances in walking patterns between different subjects. An in-shoe wearable pressure system using Android application was developed to investigate walking patterns and collect data on Activities of Daily Living (ADL). Many studies aim to understand the fundamentals of kicking commonly displayed by soccer players [4,6,10,16,17,18,24,25,28,29,30,34,36,38,40]. Of those studies, most are limited to a two-dimensional (2D) analysis using high-speed cameras for position tracking or utilizing electromyography to observe the activity of select muscles [4,6,18,25,29,36]. The few studies that investigate kicking using a three-dimensional (3D) model are limited in their position tracking capabilities and focus mainly on joint flexion potentials and foot speed.

Several surgically corrective procedures are considered to treat Adult Acquired Flatfoot Deformity (AAFD) patients, relieve pain, and restore function. Procedure selection is based on best practices and surgeon preference. Recent research created patient specific models of Adult Acquired Flatfoot Deformity (AAFD) to explore their predictive capabilities and examine effectiveness of the surgical procedure used to treat the deformity. The models’ behavior was governed solely by patient bodyweight, soft tissue constraints, and joint contact without the assumption of idealized joints. Mesenchymal stem cells are derived from a variety of human tissues and are being bioengineered and studied for possible uses in the advancement of medicine. Recent efforts are being focused on Dental Pulp Stem Cells (DPSC's) due to the accessibility of this tissue. Many factors influence DPSC quality and quantity, including the specific methods used to isolate, collect, concentrate, and store these isolates once they are removed. Ancillary factors, such as the choice of media, the selection of early versus late passage cells, and cryopreservation techniques may also influence the differentiation potential and proliferative capacity of DPSC isolates.

A computational framework was implemented and validated to better understand the hypertensive artery remodeling in both geometric dimensions and material properties. Integrating the stress-modulated remodeling equations into commercial finite element codes allows a better control and visualization of local mechanical parameters. Both arterial thickening and stiffening effects were captured and visualized. An adaptive material remodeling strategy combined with the element birth and death techniques for the geometrical growth were implemented. The purpose of this project was to create a strider, a type of standing rehabilitation device, for children in developing countries who have trouble walking and supporting their full body weight. The project was initially brought to us by Cal Poly professor Brian Self, who had visited a clinic in San Marcos and determined that there were children there who had difficulties with walking and were a need for a rehabilitation device. Patient outcomes regarding fixation quality can be affected by the healing environment and human factors such as bone quality and surgeons’ perception. Lung transplantation is the last recourse for patients with severe respiratory failure. However, transplantable lungs do not keep up with the demand, and 15.4% of patients die awaiting transplant. An alternative therapy is extracorporeal life support (ECLS), which uses a pump and an oxygenator to serve as a bridge to transplant. However, this technology limits ambulation and requires ICU care. This novel study implements pumpless arteriovenous (AV) ECLS technology to remove CO2 via the subclavian vessels, providing respiratory support and hypercapnia (excessive CO2 retention) sympton relief for patients while allowing ambulation.

Quantitative evaluation of shear stress in the vessel wall due to the presence of asymptomatic gas emboli is lacking. The goal of this work was to assess the impact of chronic asymptomatic gas emboli on the risk of atherosclerosis through a custom-built cardiovascular flow simulator. Gas bubbles were created by forced air from a syringe pump. The influences of embolism injection rate, pulse rate, and time-averaged flow rate on the wall mean shear stress were investigated at resting and elevated heart rate conditions. It is the aim of this study to investigate a novel mouthguard design that incorporates the use of a nickel-titanium (Nitinol) layer and thin foam layer in addition to EVA layers. It is thought that the Nitinol layer can distribute the force of an impact and that the thin foam layer may absorb this distributed force better than a solid EVA mouthguard of the same thickness. IPMCs are one of the most widely used types of electro-active polymer actuator, due to their low electric driving potential and large deformation range. Altered pressure in the developing left ventricle (LV) results in altered morphology and tissue material properties. Mechanical stress and strain may play a role in the regulating process. This study showed that confocal microscopy, three-dimensional reconstruction, and finite element analysis can provide a detailed model of stress and strain in the trabeculated embryonic heart. The method was used to test the hypothesis that end-diastolic strains are normalized after altered loading of the LV during the stages of trabecular compaction and chamber formation. Pectus excavatum (PE) also known as sunken or funnel chest is the most common chest wall deformity and affects 1 in every 1,000 children. In PE patients, the middle lower portion of the sternum is depressed producing concave appearance of the anterior part of the chest wall. This problem can be corrected with minimally invasive surgery developed by Dr. Donald Nuss in 1986. A specially made steel bar is implanted to lift the sternum to its normal position. The Piernas de Vida team partnered with the Vida Nueva Clinic of Prostheses and Orthoses in Choluteca, Honduras to design, plan, and implement a prosthetic foot that could be manufactured entirely by the technicians at the clinic in order to reduce their costs and ultimately allow them to serve more patients each year. Currently, the clinic uses a prefabricated prosthetic lower limb provided by the International Committee of the Red Cross (ICRC) which can be adapted to both transDfemoral and transDtibial patients. During voiced speech, the larynx provides quasi-periodic acoustic excitation of the vocal tract. Following a laryngectomy, some people speak using an electrolarynx which replaces the excitatory function of the absent larynx. Drawbacks of conventional electrolarynx designs include the buzzing monotonic sound emitted, the need for a free hand to operate the device, and difficulty experienced by many laryngectomees in adapting to its use. Despite these shortcomings, it remains the preferred method of speech rehabilitation for a substantial minority of laryngectomees. The humerus of a racing horse Thoroughbred is highly susceptible to stress fractures at a characteristic location as a result of cyclic loading. The propensity of a Thoroughbred to exhibit humeral fracture has made equines useful models in the epidemiology of stress fractures. In this study, a racing Thoroughbred humerus was simulated during training using a 3D finite element model in conjunction with a bone remodeling algorithm. Most patients escape diagnosis of subtalar joint instability for years before treatment. Therefore, a clinically relevant technique to diagnose subtalar joint instability will improve its management. Towards this goal, a six degree of freedom positioning and loading device was developed to apply physiologic loads to the hindfoot. Because the ideal loading configuration to detect subtalar joint instability is unknown, the device was designed to apply rotational and translational forces simultaneously. This thesis presents an improved oscillotonometric system for the measurement of human blood pressure. The study included:
1. The design of a hydraulic occlusive cuff,
2. The investigation of the wave forms taken from the blood pressure measurements, and
3. The design of a mechanism for the simulation of human blood pressure pulse.
