Chronic low back pain is a global socioeconomic crisis compounded by an absence of reliable, curative treatments. The predominant pathology associated with chronic low back pain is degeneration of intervertebral discs in the lumbar spine. During degeneration, nerves can sprout into the intervertebral disc tissue and be chronically subjected to inflammatory and mechanical stimuli, resulting in pain. Pain arising from the intervertebral disc, or disc-associated pain, is a complex, multi-faceted disorder which necessitates valid animal models to screen therapeutics and study pathomechanisms of pain.

Open-source laboratory equipment has become an increasingly popular alternative to commercially available options due to its simple implementation, capability of user-customized modifications, and cost-effectiveness. Presented here is the development of a programmable and open-source fluidic system comprised of three custom-built syringe pumps for cell culture applications. The fluidic system was directly integrated with a multiwell plate on an environmentally controlled microscope platform for live-cell imaging and time-lapse analyses of cellular responses as culture conditions are adjusted using the programmable fluidic system. Objective: Left ventricular assist devices (LVADs) are utilized as a bridge to transplant or as destination therapy for patients with end-stage heart failure. Although cardiac offloading from these devices rarely leads to complete remodeling and functional recovery, the use of mesenchymal cells to modulate heart failure has been explored in recent years due to its intrinsic regenerative properties. Eagle Medical Incorporated is a contract medical device packaging and sterilization company. The company purchases thermoformed medical packaging trays, which maintain the sterility of medical devices, from various manufacturers. To ensure packaging quality and to prevent cleanroom contamination, Eagle Medical inspects and sterilizes each blister tray that they order. This process is an essential non-value-added activity that creates a bottleneck. Cleanroom employees must stop packaging medical devices and attend to the processing of blister trays and packaging solutions. The blister trays arrive at Eagle’s facility in nested stacks. Serious gaming is the practice of using video games, either commercial or specifically designed, for physical rehabilitation. Serious games have emerged in the last decade as a way to increase patient involvement in rehabilitation, increase the likelihood of patients continuing treatment after progress stalls, and generally increase mental health during treatment. Because of its extreme rarity, the genetic disease arthrogryposis multiplex congenita (AMC) and the needs of individuals with the diagnosis are often overlooked. AMC refers to the development of nonprogressive contractures in disparate areas of the body and is characterized by decreased flexibility in joints, muscle atrophy, and developmental delays. Positron Emission Tomography (PET) scans can utilize a radioactive tracer, in this case 2-deoxy2-[fluorine-18] fluoro-D-glucose (18F-FDG), to visualize malignant tumors in cancer patients. The uptake was compared to glucose to understand the difference in thermal properties, which contribute to the ability to image the cancerous cells. The uptake of 18F-FDG by cancer cells and the imaging process of positron emission tomography were reviewed from a thermodynamic perspective. Gastrointestinal and neurological imaging techniques were reviewed to understand the role of PET imaging in different areas of the human body.

Hydrogen sulfide (H2S) is a gasotransmitter critical in maintaining microcirculation homeostasis. Impaired microcirculation occurs in multiple disease states such as peripheral vascular disease, diabetes mellitus (DM), and hypertension. Early detection and identification of patients with DM who are at risk for heart attack, stroke and amputation due to microvascular disease is crucial. The goal of this research is to characterize the electromagnetic properties of biological and non-biological materials at terahertz (THz), millimeter-wave, and microwave frequency bands. The biological specimens are measured using the THz imaging and spectroscopy system, whereas the non-biological materials are measured using the microwave and millimeter-wave free-space system. These facilities are located in the Engineering Research Center at the University of Arkansas. The THz imaging system (TPS 3000) uses a Ti-Sapphire laser directed on the photoconductive antennas to generate a THz time domain pulse. FLASH ultra-high dose rate radiotherapy (RT) is one of the most rapidly growing subfields of radiation oncology today due to its potential to increase the limits of the therapeutic ratio. Athletes and medical personnel need reliable methods to monitor vital signs and detect potentially life-threatening issues before they occur. Electrocardiograph (ECG) monitors are currently the best method of detecting heart-related health issues. Current ECG monitors worn by athletes are difficult to use and lack the ability to track ECGs in real time. Through research and interviews with various clinicians, further problems have been identified that include poor electrode adhesion, and a lack of compatibility with the equipment worn by athletes. Localized chemical delivery plays an essential role in the fundamental information transfers within biological systems. Thus, the ability to mimic the natural chemical signal modulation would provide significant contributions to understand the functional signaling pathway of biological cells and develop new prosthetic devices for neurological disorders. In this paper, we demonstrate a light-controlled hydrogel platform that can be used for localized chemical delivery in a high spatial resolution. Combining vibrating mesh nebulizers with additional new technologies leads to substantial improvements in pharmaceutical aerosol delivery to the lungs across therapeutic administration methods. In this dissertation, streamlined components, aerosol administration synchronization, and/or Excipient Enhanced Growth (EEG) technologies were utilized to develop and test several novel devices and aerosol delivery systems. The first focus of this work was to improve the poor delivery efficiency, e.g., 3.6% of nominal dose (Dugernier et al. 2017), of aerosolized medication administration to adult human subjects concurrent with high flow nasal cannula (HFNC) therapy, a form of continuous-flow non-invasive ventilation (NIV). This paper discusses fetal development throughout pregnancy from conception to full-term status as well as fetal lung development specifically from conception until full-term status. EPICS is a service-learning design program run through Purdue University. It strives to teach students design skills through providing for individuals, communities, and organizations in the surrounding area while mirroring engineering industry standards. BME (Biomedical Engineering) is a team within EPICS that strives to serve community partners through biomedical applications. William Sevick is an elementary school student with a congenital arm amputation. William and his family have been working with the BME team for the past three years designing assistive devices with the purpose of improving his actions in daily life such as eating, playing games, and riding his bike.

The study aims were to develop fracture prediction models by using machine learning approaches and genomic data, as well as to identify the best modeling approach for fracture prediction. The genomic data of Osteoporotic Fractures in Men, cohort Study (n = 5130), were analyzed. After a comprehensive genotype imputation, genetic risk score (GRS) was calculated from 1103 associated Single Nucleotide Polymorphisms for each participant. Data were normalized and split into a training set (80%) and a validation set (20%) for analysis. Random forest, gradient boosting, neural network, and logistic regression were used to develop prediction models for major osteoporotic fractures …

Although Support Vector Machines (SVM) are widely used for classifying human motion patterns, their application in the automatic recognition of dynamic and static activities of daily life in the healthy older adults is limited. Using a body mounted wireless inertial measurement unit (IMU), this paper explores the use of an SVM approach for classifying dynamic (walking) and static (sitting, standing and lying) activities of the older adults. Specifically, data formatting and feature extraction methods associated with IMU signals are discussed. The scope of this project includes designing a non-invasive bubble detector for peripheral venous and arterial lines. For minimally invasive cardiac procedures, arterial and venous access is necessary for these procedures as is constant flow through the tubing to prevent clotting and to administer drugs. Air bubbles are common in these lines and can be consequential if enough air is introduced into the human system. Air embolisms are the most common and relevant issues during these procedures. Chronic venous insufficiency (CVI) is a form of cardiovascular disease that is caused when valves in the leg become incompetent. Current treatment methods serve to manage symptoms, but there are currently no ways to treat the underlying cause of CVI. A venous valve prosthetic made from a xenograft of a bovine jugular vein is one possible treatment method currently in the research phase. Prosthetic valves must be tested with an accelerated wear tester prior to approval for clinical testing. Although such testers exist for heart valves, physiological differences between heart and venous valves restrict the use those testers on venous …

Currently, 18.2 million adults aged 20 and older are diagnosed with Coronary Artery Disease (CAD) (Benjamin et al., 2019). Stenosis is the most common intervention. However, when a patient has a bifurcated artery, treatment becomes more difficult and is often unsuccessful. This project created a new stent and balloon complex that was tested in vitro using a gel phantom artery model. Two separate prototypes have been created and tested so far, with improvements made upon each. Testing is still underway with Prototype 2.
