Last year, approximately 45 million acres of American farmland were planted with crops that had been genetically engineered to either produce their own pesticides, or withstand herbicides. The long and short-term effects of this biotechnology on humans and the environment, are being studied, but remain unknown. Leading the way in the field of bioengineered crops is the Missouri-based Monsanto Company. Monsanto believes that current agricultural practices are inconsistent with sustainable development. The NewLeaf potato is one of three products Monsanto has created to feed the growing world population. The genetically engineered crop produces, in every cell of the plant, a …

Drip irrigation is one of the most efficient systems in delivering water to the plant root zone. Research has shown that the saturated, or nearly saturated, surface beneath the emitter may increase evaporation thereby reducing the irrigation efficiency. To increase the efficiency of surface applied drip irrigation on permanent tree crops a sand tube irrigation (STI) method was developed and tested. The sand tube method consists of removing a soil core beneath the emitter and filling the void with coarse sand. Members of a panel of stress-responsive biosensors have been used to study the effect of megahertz frequency ultrasound onEscherichia coli. Insonification causes acoustic cavitation, the collapse of oscillating microbubbles in solution, which can damage bacterial cells. A focused 1-MHz ultrasound transducer, capable of generating a spatial peak pulse average intensity of 500 W/cm2, was used to treat liquid bacterial cultures. Stress-responsive promoters fused to luxCDABE allowed the continuous measurement of light produced as a result of protein damage, DNA damage, oxidative stress, and membrane perturbation. A promoter responsive to ammonia limitation was not transcriptionally activated under test conditions. In contrast …

Solid-state fermentation has the potential to produce inexpensive enzymes for use in high-volume industrial applications. Process parameters such as substrate moisture content and length of fermentation can have a significant effect on the amount and timing of enzyme production. This study was conducted in two stages, a screening stage and an optimization stage, to determine the effects of moisture content of the substrate, surfactant addition upon inoculation, depth of the substrate, and duration of fermentation on xylanase activity produced by Trichoderma longibrachiatum. Pasture runoff can contribute to elevated concentrations of nutrients, solids, and bacteria in downstream waters. The objective of this study was to determine the effects of vegetative filter strip (VFS) length on concentrations and transport of nitrogen, phosphorus, solids and fecal coliform in runoff from plots treated with cattle manure. Three plots with dimensions of 2.4 × 30.5 m were used. The upper 12.2 m of each plot was treated with cattle manure, while the lower 18.3 m acted as a VFS. Runoff produced by rainfall simulators was sampled at VFS lengths of 0, 6.1, 12.2, and 18.3 m and …

Pesticides used in urban environments can be transported in runoff to downstream waters and cause adverse environmental consequences. This experiment assessed the effects of post-application irrigation depth (0, 6.4, and 12.7 mm) and formulation (liquid and granular) on concentration and transport of diazinon (a pesticide commonly used for lawn insect control) in runoff from “tall” fescue (Festuca arundinacea Schreb.) plots. The post-application irrigation was applied using rainfall simulators immediately following diazinon application. The rainfall simulators were again used approximately 2 h after diazinon application to apply the equivalent of a heavy rainfall (64 mm/h for approximately 1.5 h) to generate …

As a part of a National Pork Producers Council educational program, our research and extension team at the University of Kentucky was linked with an independent commercial swine producer to test the NCPIG model against observed commercial on-farm data. This experience provided improved information for model development as well as increased producer insight into the data input needs and potential benefits of modeling. Detailed production information comparisons between the NCPIG model and producer data are presented for summer time conditions to assess the validity of the NCPIG model for simulation of grow-finish swine performance. Land application of horse stall bedding and municipal sludge can increase runoff concentrations of nutrients, organic matter, and bacteria as well as steroidal hormones such as estrogen. Concentrations of materials in runoff from sites treated with animal manure can be reduced by aluminum sulfate, or alum [Al2(SO4)3•14H2O] treatment. A sensor for measuring diffuse reflectance of milk during the typical 6-h culture of cottage cheese was installed in a local manufacturing facility. Diffuse reflectance was found to increase slowly during the first three hours of the culture and increase rapidly toward the end of fermentation. The correlation between parameters generated from the diffuse reflectance profile and cutting time was sufficient to develop an algorithm for cutting time prediction. An algorithm incorporating tmax (time from adding culture to the maximum rate of change in reflectance) and slope of the reflectance curve at tmax predicted the operator selected cutting …

Three spatial data sets consisting of high spatial resolution (1 m) remote sensing images acquired in 12 spectral bands, an on-the-go yield map, and a Digital Elevation Model were co-registered and evaluated for spatial variability studies in a Geographic Information Systems environment. Separate on-the-go yield maps were developed for 3, 5, and 12 statistically significant mean yield classes. For each yield class, the corresponding mean spectral and elevation data were extracted. The relationship between mean spectral and yield data was strongly linear (r = 0.99). Also, a strong linear relationship between mean yield and elevation data (r = 0.92) was …

Vegetative filter strips (VFS) reduce losses of nutrients, solids, and other materials from land area treated with fertilizers and manures. A number of models are available that simulate nutrient and sediment transport in VFS. While VFS effectiveness is considered to depend on lengths of pollutant source area and VFS areas, few published studies have tried to validate these models using variable pollutant source area and VFS area. The objective of this study was to validate an event-based nutrient transport model (Chaubey et al., 1995) that simulates soluble nutrient transport in VFS. The Korean character is composed of several alphabets in two-dimensional formation and the total number of Korean characters exceeds eleven thousand. Therefore, the previous approaches to Korean cursive characters pay most of their attention to segmenting a character into alphabets accurately. However, it is difficult because the boundaries of alphabets are not apparent in most cases. We propose an alphabet-based method without assuming accurate alphabet segmentation. In the proposed method, a cursive character is segmented into substrokes by a set of segmenting conditions. The toxicity of common compounds of lead, cadmium and zinc was evaluated in waters similar to that found in the world's largest lead producing area in Missouri. Static, acute toxicity tests were performed using fathead minnows (Pimephales promelas) and water fleas (Daphnia magna) respectively. Test organisms were subjected to varying amounts of sulfide, carbonate, chloride and sulfate salts of lead, zinc and cadmium mixed in hard, alkaline waters typical to this region. Median lethal concentrations were calculated using nominal versus measured metal concentrations. We have fabricated a novel low off-state leakage current thin-film transistor (TFT) using a chlorine incorporated amorphous silicon [a-Si:H(:Cl)] and amorphous silicon (a-Si:H) stacked active layer, in which conduction channel is formed in a-Si:H and a-Si:H(:Cl) is photo-insensitive material. The off-state photo-leakage current of the a-Si:H(:Cl)/a-Si:H TFT is much lower than that a conventional a-Si:H TFT.

Transesterification of beef tallow was investigated. The solubility of ethanol in beef tallow was much higher than that of methanol. At 100 °C the solubility of methanol was 19% (w/w). The solubility of ethanol in beef tallow reached 100% (w/w) at about 68 °C. For the distribution of methanol between beef tallow methyl esters (BTME) and glycerol, the percentage of total methanol in the glycerol phase was higher than that in the fatty acid methyl ester (FAME) phase in a simulated system at room temperature. At 65-80 °C, however, the percentage of total methanol in FAME (60% (w/w)) was higher …

A dissolved oxygen probe and an ion specific electrode were used to study the lead-induced metabolic inhibition in a toluene-enriched microbial population. Predicted toxicity values were compared to the actual toxicity responses using a free metal ion toxicity model (TM) which linked metabolic inhibition with lead activity. Experimentally derived values for the model parameters (lead activity and a lead distribution coefficient) were used in the TM. It was postulated that cellular metabolism is disrupted by the conformational changes to the cell's plasma membrane produced by lead ion adsorption. The predicted toxicity values were higher than the actual toxicity response. This …

The effectiveness of management practices in improving quality of runoff from agricultural land areas has been reported based primarily on results from plot- and field-scale studies. There is limited information available on watershed scales, particularly when the dominant agricultural land use is pasture. Experimental autoimmune myasthenia gravis (EAMG) is an animal model of human myasthenia gravis (MG). In mice, EAMG is induced by immunization with Torpedo californica acetylcholine receptor (AChR) in complete Freund's adjuvant (CFA). However, the role of cytokines in the pathogenesis of EAMG is not clear. Because EAMG is an antibody-mediated disease, it is of the prevailing notion that Th2 but not Th1 cytokines play a role in the pathogenesis of this disease. To test the hypothesis that the Th1 cytokine, interferon (IFN)-gamma, plays a role in the development of EAMG, we immunized IFN-gamma knockout (IFN-gko) (-/-) mice and wild-type (WT) …
