Rolling Circle Mutagenesis Of Gst-Mcherry To Understand Mutation, Gene Expression, And Regulation, Jessica Cole, Amanda Ferguson, Verónica A. Segarra, Susan Walsh
Biology Faculty Publications and Presentations
Undergraduates are often familiar with textbook examples of human mutations that affect coding regions and the subsequent disorders, but they may struggle with understanding the implications of mutations in the regulatory regions of genes. We have designed a laboratory sequence that will allow students to explore the effect random mutagenesis can have on protein function, expression, and ultimately phenotype. Students design and perform a safe and time-efficient random mutagenesis experiment using error-prone rolling circular amplification of a plasmid expressing the inducible fusion protein glutathione S-transferase (GST)-mCherry. Mutagenized and wild-type control plasmid DNA, respectively, are then purified and transformed into bacteria …
Middle And High-School Teacher Perceptions Of The Mathematics Georgia Standards Of Excellence, Marnie Dutcher
Middle And High-School Teacher Perceptions Of The Mathematics Georgia Standards Of Excellence, Marnie Dutcher
Theses and Dissertations
Public education in the United States had frequent curriculum changes throughout history. The curriculum initiatives of the past had a focus on improving education in schools based on one of four topics: (a) common standards, (b) preparing students for post-secondary education, (c) improving the quality of education, and (d) increasing the rigor of the content (Wallender, 2014). In February 2015, the Georgia State Board of Education approved revisions to the Common Core Mathematics Curriculum, which was the third set of mathematics curriculum standards implemented in Georgia since 2012. The standards were named The Georgia Standards of Excellence Mathematics Curriculum (GSE). …

Despite an increased recognition of the role that ‘informal’ learning spaces (e.g. museums, aquariums, other cultural institutions) have in children’s science education (NRC, 2015), there remains a gap between the goals and values of ‘informal’ and ‘formal’ (i.e. school-based) learning sectors. Moreover, the potential for informal spaces and institutions to also play a role in initial teacher preparation is only beginning to be realized. Here, we present our Science Teacher Learning Ecosystem model and explain how it frames the design of our elementary science teacher education coursework. We then use this framework to describe learning experiences that are collaboratively planned …

Although bioinformatics, the use of computational science to study biology, has become imperative in many areas of the biological sciences and related career paths, introductory biochemistry courses may disregard practical knowledge on bioinformatics. For this reason, we merged a hands-on activity module into an undergraduate biochemistry course in two ways. First, we incorporated bioinformatics modules for building phylogenetic trees by aligning the active sites of 10 chosen related α-amylase enzymes using freely available data. Secondly, we chose three of those 10 α-amylase enzymes to compare the 3D structure of their active sites. This module gives the students an opportunity to …
