Investigating The Role Of Tryptophan In The Native Photo-Crosslinking Of A Cys-Containing Transmembrane Helix To A Membrane Lipid Double Bond, Michael Jay Telehany

Our research group has previously discovered that a Cys residue within a transmembrane (TM) helix can directly photo-crosslink to a membrane lipid via an oxythiolation addition reaction to the double bond within the lipid, with a reaction yield of 10-20%. The pH-low Insertion Peptide (pHLIP) serves as our model TM helix, while the POPC lipid bilayer is our model membrane. We had long believed that the Trp residues within the sequence of pHLIP served as the key chromophore for this reaction, and thus they were essential for the formation of the TM helix-lipid adduct. The medial prefrontal cortex (mPFC) is one of several brain structures that is contained in the mesolimbocortical pathway and controls many processes involved in alcohol use disorder (AUD). There is vast glutamatergic output from the mPFC to other mesolimbocortical regions; neuroplastic changes to this system are believed to be closely tied to the development and maintenance of AUD. Astrocytes are key regulators of glutamatergic transmission through their uptake of excess glutamate from the synapse and recycling back into neurons. Intimate partner violence (IPV) is physical, psychological, or sexual abuse by a current or former romantic partner. The long- and short-term consequences can lead to severe repercussions. To prevent these consequences of IPV, we need to screen for it and assess it. Across cultures, IPV is present and should be assessed regardless of linguistic boundaries. Spanish is the most widely used language in the U.S. other than English; therefore, assessment instruments that can reliably capture IPV in Spanish are necessary. Opioid misuse has been identified to be related to problematic release of dopamine in the brain that is otherwise satiated by other alternative rewards. As mediated by patterns of positive and negative reinforcement, prolonged opioid use may transition into misuse and development of opioid use disorder. In human populations, men and women differ in their acquisition of drug use and escalation to drug misuse. Women quickly surpass their male counterparts in their transition from use to misuse in what is coined the telescoping effect. Alcohol use disorder (AUD) is a serious chronic brain disease; in 2021 there were ~29.5 million people in the U.S. diagnosed with AUD. Individuals with AUD often show cognitive impairment such as risky decision-making, difficulties with impulse control, and working memory deficits. This impairment is associated with structural and functional changes in the prefrontal cortex (PFC), an extensively interconnected region of the frontal lobe involved in executive control of goal-directed behaviors. Lung cancer is one of the most common and fatal cancers today, infecting 1 in 16 people, and killing well over a million patients per year. Currently, lung cancer is detected usually in stages three and four by quite costly and invasive methods, which shows only a survival rate of 10%. Breath sensors provide a low-cost and non-invasive technique for early screening of lung cancer. TLR7 agonists are small molecules that are useful within cancer immunotherapy due to their ability to stimulate the TLR7 pathway resulting in NFκB activation, and cytokine release.1 Due to the risk of toxicity when delivered systemically, our team has employed a particular type of drug-delivery technology, Antibody Drug Conjugates (ADCs) to deliver these payloads directly to tumor tissue. In this study, I investigated the individual and additive effects of both acculturation and enculturation on experiencing psychological intimate partner violence (IPV) victimization within low-income Latinx males. I hypothesized that there would be a significant positive association between acculturation and psychological IPV victimization, a significant negative association between enculturation and psychological IPV victimization, and a significant negative association between the additive effects of acculturation and enculturation on psychological IPV victimization. In this work I investigate changes in historical metal consumption practices over time at Biry/Ahr House, a frontier settlement in Castroville, Texas. Alsatian immigrants settled this site in Medina County as part of the empresario system of the 1840s. Biry/Ahr House was home to two families, the Birys (1844-1921) and the Groff/Tschirharts (1921-1972). Using vibrant materialism, consumerism, and collections-based research, I address the ways in which metal usage changed over time at the site as well as the factors behind these changes. Dominance And Radical Submission In Seventeenth-Century Drama: Chastity, Fairness, And Silence In Portrayals Of Mar(R)Iam(Ne), Sophonisba, And Cleopatra, Laura S. Deluca

This thesis conducts an extensive reading of early modern English playwrights’ interpretation of ancient royalty. I survey a series of seventeenth-century plays concerning Mariamne I, the Carthaginian noblewoman Sophonisba, and Cleopatra VII. I argue that the English stage produced two models of ancient royalty. Mar(r)iam(ne) and Sophonisba personify one model, functioning as white, seemingly obedient figureheads. I document playwrights portraying their men as reducing them to their chastity and fairness, or lack thereof. Despite the inactivity of these objectified women, the qualities that these men obsess over catalyze masculine irrationality. The other model, which Cleopatra embodies, encompasses blackness and defiance. …

Microtubules and the desire to visualize them has been of utmost importance in understanding the behavior of the cytoskeleton. Even further, scientists are using this knowledge to elucidate cancerous activities as they occur within cells. In order to acquire this knowledge, the following methods have been established: the expression of tubulin after it has been fused with a fluorescent protein, the insertion of tubulin that has been exogenously labeled, and the use of fluorescent-probe conjugated microtubule ligands, such as paclitaxel. Protein carbonylation is one manifestation of oxidative stress, which is characteristic of many diseases. Our lab has developed several coumarin-hydrazine based fluorescent probes capable of detecting carbonylation in live cells via the formation of hydrazone conjugates. As an effort to expand and improve on these achievements, this work presents new data regarding the synthesis and characterization of three such probes. Two of the probes—julolidine coumarin hydrazide (JCH) and trifluoromethyl coumarin hydrazine (TFCH)—were previously synthesized by our lab. Advancements in the field of analytical chemistry have greatly expanded the development of biosensors for the detection of a wide array of diseases. This study aims to optimize an affordable paper-based nanocomposite biosensor that utilizes surface-enhanced Raman spectroscopy (SERS). Specifically, it investigates the preparation parameters for a paper-based SERS substrate, including nanoparticle administration and drying procedures. A particular focus of this work is to assess how the wax-defined paper channels can effectively enhance SERS intensity. Antibody-drug conjugates (ADCs) are an increasingly popular modality for targeted drug delivery in many oncological and immunological applications. The paratope of a monoclonal antibody (mAb) directs the delivery of a conjugated therapeutic payload to antigen expressing cells, resulting in a controlled transport of payload to a desired cell type. Internalization of the ADC followed by lysosomal degradation results in the release of a payload to perform its biochemical function. An effective way to promote student engagement in the laboratory is to substitute traditional reagents for recognizable, real-world products. In this work, two laboratory experiments are outlined – one geared toward undergraduate general chemistry students and one for high school introductory level chemistry students – which replace solutions of metal salts with tattoo inks in the classic flame test experiment. I will be examining temporality in British texts about Egypt across time. In order to achieve this, I analyze the play Antony and Cleopatra (1606) by William Shakespeare, and put it in conversation with Pharos, the Egyptian (1899) by Guy Newell Boothby. I will also be discussing Alexandria (2009) by Lindsey Davis, as a demonstration that the pattern in my findings is enduring. Identity In The Late Woodland Northeast: Interpreting Communities Of Practice From Paste Composition At The Thomas/Luckey And The Losey 3 Sites, Douglas S. Riethmuller

Thomas/Luckey’s 13th -15th and Losey 3’s 14th-17th century occupations in the Late Woodland Northeast contain assemblages with incongruous regional pottery types; Kelso Corded and an assumed non-local Shenks Ferry. I argue the presence of Shenks Ferry vessels at these two sites indicates the movement of people who reproduced their natal designs upon arrival, rather than trade. The question of whether identity and communities of practice can be discerned from pottery decorations and paste was answered by analyzing sherds with pXRF. While pottery types are based on visual attributes, pXRF looks at elemental composition. Decoration is mimicable, but paste is not; …

The SagS protein is a two-component regulatory system in Pseudomonas aeruginosa that works to independently regulate biofilm formation and antibiotic tolerance. Previous work found that these two pathways are controlled by two distinct sets of amino acids within the sensory domain of SagS that are thought to be potential ligand binding sites. Alanine Serine Cysteine Transporter 2 (ASCT2) is a member of the solute carrier 1A (SLC1A) family of transport proteins. It is a Na+ dependent, obligatory neutral amino acid exchanger with the capability to transport glutamine and it is a primary glutamine transport protein in cancer cells. This glutamine transport capability confers an important physiological role for ASCT2 in the maintenance of intracellular amino acid pools for various metabolic and cell signaling pathways.
