Various microbial communities in marine biofilms cause biofouling on submerged surfaces, posing challenges to marine industries. Despite their ecological and economic importance, biofilms' biomechanical and biochemical responses to hydrodynamic shear stress are insufficiently understood, particularly in dynamic flow conditions. To fill this gap, our study uses an innovative fiber-optic hyperspectral imaging (HSI) system and a supercontinuum laser source to examine marine biofilms' structure, composition, and detachment behavior at different shear stress levels.

High-power fiber lasers are steadily increasing in maximum power and rapidly approaching theoretical maximum power output from a single fiber aperture. The low thermal conductivity of silica fibers is an inherent limitation to the dissipation of heat from the fiber core, and the buildup of heat within the laser gain media is the principal driver of transverse mode instability within fiber systems. Real-world materials, particularly biological structures such as feathers exhibit complex appearances that vary spatially across their surfaces. The field of computer graphics provides a means of understanding such surfaces through material modeling which uses both analytical models and data acquired from light-surface interactions. There are many efforts within the past decade in measuring materials for graphics, but common limitations in these works include not accounting for spatially varying properties and reliance on neural networks and synthetic datasets.

Bovine in-vitro production(IVP) of embryos is a growing field for producing offspring with desirable genetics in the cattle industries. While IVP is effective, oocyte cytoplasmic maturation is compromised, and bovine oocytes matured in-vitro have reduced metabolic activity than those matured in-vivo. Mitochondria are the central unit of oocyte metabolism, producing ATP through OXPHOS. Photobiomodulation is a light treatment reported to improve metabolic activity. In five experiments, we measured the effects of photobiomodulation treatment with red LED at 16-(L-16) and 20 h(L-20) of bovine oocyte in-vitro maturation on subsequent embryonic development, mitochondrial activity and nuclear progression. This thesis presents the comprehensive design, fabrication, and demonstration of advanced high-power, high-efficiency single-mode semiconductor lasers operating at a wavelength of 9xxnm. We begin with the design of the laser epitaxial structure, serving as the cornerstone for achieving high-power high-efficiency lasers. Our methodology integrates a semi-analytical calculation model, which accounts for Longitudinal Spatial Hole Burning (LSHB) and Two-Photon Absorption (TPA) effects, facilitating a thorough exploration of how design parameters influence output power and conversion efficiency. This approach offers an effective and time-efficient epitaxial structure optimization strategy compared to conventional full 3D simulation models.

Optically stimulated luminescence (OSL) dosimetry was utilized to calibrate Al2O3:C powder dosimeters, available commercially as the nanoDot® from Landauer Inc., and compare the dosimeter response to radium-226 (226Ra) and cesium-137 (137Cs). The signal from the OSL was quantified using a microSTARii® OSL reader also produced by Landauer Inc. Dose-response curves were developed for 226Ra and 137Cs experiments (5 dosimeters each) at thirteen absorbed doses. Individual dosimeter response was tracked by serial number. Laser shock peening (LSP) is an important material surface strengthening technique that uses laserinduced shock waves to cause severe plastic deformation near material surfaces. It can significantly improve material properties like hardness, strength, fatigue life, and corrosion resistance and is widely used in industrial sectors such as automotive, aerospace, nuclear, and medical areas. In confocal single-molecule FRET experiments, the joint distribution of FRET efficiency and donor lifetime distribution can reveal underlying molecular conformational dynamics via deviation from their theoretical Forster relationship. This thesis presents the theoretical development of orbital angular momentum (OAM) based wavelets for the analysis of localized OAM information in space. An optical probing system for generating and detecting these wavelets is demonstrated; individual wavelets can scan the environment in 10µs or less. The probing system was applied to a three-dimensional atmospheric turbulence distribution to obtain a continuous wavelet transform of the angular information of the turbulent propagation path about a fixed radius. An entire continuous wavelet transform was measured in 3.8ms; the measurements are much faster than the turbulence and give insight into the short time scale of …

This dissertation uses porous silicon as a material platform to explore novel optical effects in three domains: (i) It studies dispersion engineering in integrated waveguides to achieve high performance group index sensing. With proper design parameters, the sensor waveguides can theoretically achieve 6 times larger group index shift compared to the actual bulk effective refractive index shift. We demonstrate the guided mode confinement factor to be a key parameter in design and implementation of these waveguides. In this thesis, light transmission through a single sub-wavelength slit in a silver thin film with periodic gratings at the exit side of the slit has been studied in both the visible and mid-infrared wavelength ranges.
In the visible wavelength range, we first investigate the enhancement of plasmonic light beaming efficiency by near field resonance in a subwavelength metallic slit-groove beaming structure. We show that by varying the film thickness and the separation distance of the beaming grating, the intensity of the near field can be greatly enhanced by resonance, which leads to the increase of the beaming efficiency.
