Photodynamic therapy (PDT) is a minimally invasive treatment that uses light-activated photosensitizers to selectively destroy diseased tissue through the generation of reactive oxygen species. However, conventional in vitro models often fail to accurately reproduce the pharmacokinetics of photosensitizer accumulation and clearance observed in vivo. This study investigated protoporphyrin IX (PpIX) accumulation following 5-aminolevulinic acid (ALA) exposure under physiologically relevant conditions using two-dimensional (2D) monolayers and three-dimensional (3D) spheroids. By comparing continuous ALA exposure with transient exposure followed by ALA washout, this work provides a more biologically relevant framework for studying PpIX kinetics and optimizing photodynamic therapy.

This thesis investigates the relationship between macroscopic rheological signals and underlying microscopic dynamics in complex fluids, specifically focusing on ”living polymers” within the cetyltrimethylammonium bromide (CTAB) and sodium salicylate (NaSal) surfactant system. The research addresses the inverse parameterization problem, demonstrating how bulk measurements like zero-shear viscosity can mask fundamentally different physical topologies, such as purely cylindrical, reptating micelles versus highly branched networks. Through the successful synthesis of viscoelastically ”degenerate” samples, the study utilizes an array of characterization techniques including frequency sweeps, Large Amplitude Oscillatory Shear (LAOS) to resolve these unique underlying states. This thesis develops a documented, reproducible procedure for building diffraction-limited microscope objectives entirely from catalog singlets, for imaging ultracold 6Li atoms in a ring-trap experiment. The objectives must resolve micron-scale features through a 5 mm fused-silica vacuum window, operate at multiple wavelengths, fit inside a 48 mm magnet bore, and contain no conductive or magnetic material, because the surrounding coils switch 0.1 T fields on microsecond timescales. Commercial long-working-distance objectives are universally housed in metal, which the eddy-current constraint rules out, and custom fabrication of a suitable non-conductive matched pair is estimated at nearly $200k.

Quantum sensors achieve exceptional measurement sensitivity through coherent, well-isolated quantum systems, but practical deployment is hindered by destructive readouts that require repeated state preparation and create long system dead times. Optical cavities enable continuous, nondestructive measurements with minimal back-action, yet integrating high-finesse cavities into scalable quantum devices remains limited by existing fabrication methods. To overcome these constraints, an adaptive CO₂ laser-milling platform was developed. Guided by glass thermodynamics and melt dynamics, this closed-loop system uses in-situ phase-shifting interferometry to sculpt complex fused-silica surfaces with sub-nanometer root-mean-square roughness. Weak value amplification (WVA) has emerged as a powerful technique that enhances measurement precision. However, traditional table-top WVA setups and on-chip demonstrations have not achieved an amplification over 20 dB. The practical limit of the amplification is the fidelity of photon post-selection. To address this limit, we design a weak value device with an over 30 dB interferometer extinction ratio, where the stray light in the dark port is minimized with thermally tunable phase shifters. Background: Breast cancer progression and metastasis are closely linked to alterations in the mechanical properties of tumor cells and their microenvironment. Softer, more deformable cells are often associated with higher metastatic potential. While atomic force microscopy (AFM) is the current gold standard for mechanical characterization, it is limited to surface measurements and can damage 3D cultures. This dissertation presents two experimental investigations at the intersection of quantum sensing and precision optical instrumentation. The primary project demonstrates optically detected nuclear magnetic resonance (NMR) of 13C nuclear spins in diamond, using state-selective Landau-Zener transitions under microwave frequency sweeping to bidirectionally transfer spin polarization between nitrogen-vacancy (NV) electron spins and remote 13C nuclear spins. This enables optical polarization and readout of large ensembles of polarized nuclear spins at low magnetic fields and room temperature, with spin dephasing times limited by longitudinal relaxation of nearby NV electron spins. Fourier-transform spectrometers are widely used for spectral measurements. Changes in the field of view during measurement introduce oscillations into the measured spectra known as scene-change artifacts. Field-of-view changes also introduce uncertainty about which target the measured spectrum represents. Though scene-change artifacts are often present in dynamic data, their significance is disputed in the current literature. This work presents a theoretical framework and experimental validation for scene-change artifacts. Field-of-view changes introduce variable interferogram offsets, which standard processing techniques assume are constant. Mid-wave infrared (MWIR) optical systems require compact, broadband, manufacturable components whose performance can be predicted and measured reliably. Silicon metalenses are attractive because silicon combines high refractive index, MWIR transparency, and semiconductor-compatible fabrication. This dissertation develops a scale-bridging design–fabrication-modeling–metrology framework for MWIR silicon metalenses.

A shared full-aperture framework connects meta-atom libraries, physical layout generation, angular-spectrum propagation, point-spread-function calculation, and focal-plane energy metrics. Building on this foundation, the Dispersive Sweatt Model (DSM) incorporates meta-atom dispersion into conventional ray-tracing software, enabling broadband co-optimization of metasurfaces and refractive elements. This study aimed to explore the potential of modern spectroscopy in the authentication of organic and non-organic chicken eggs using near-infrared spectroscopy (NIRS) and laser-induced breakdown spectroscopy (LIBS) spectra. A total of 175 eggs were analyzed, which were grouped into seven categories based on the source of feed given: 100% organic, 100% non-organic, 75% organic, 75% non-organic, 50% organic, free-range chickens, and eggs obtained from the local traditional market. While many methods of time resolution are already established using classical resources, they fail to combine high precision outcomes with large-scale implementations. The overarching motivation of this work is the development of seedless, non-intrusive, laser-based diagnostics for supersonic airflow in wind tunnels. To advance this goal, this dissertation fo-cuses on three-photon excitation in a research-grade argon beam within a tabletop vacuum system, which offers a controlled environment for testing and refining the approach. An experiment was performed to demonstrate the pump-probe spectroscopy of Rubidium (Rb) vapor, revealing the nonlinear atom-laser interactions in the 5S1/2 ↔ 5D5/2 transition in Rb that can be used to generate quantum entanglement between two laser beams. The setup included a tunable infrared diode laser, optical mirrors, photodiode-detectors, a beam-splitter, and a temperature- regulated rubidium gas chamber, all of which are controlled by the TeachSpin laser controller and monitor. Advancing biomedical imaging requires platforms that combine high-resolution, speed, and stability across biological scales. This dissertation presents three innovations designed to address limitations of conventional light-sheet and oblique plane imaging. First, we introduce Reflected Inline Detection in Epi-Oblique Plane Microscopy (RIDE-OPM), a compact and drift-resistant system that eliminates a tertiary detection path while maintaining alignment flexibility, enabling robust long-term live imaging. Second, we present a high-speed Axially Swept Light-Sheet Microscopy (ASLM) platform for cleared tissues, using dual foci and synchronized sweeping to achieve isotropic sub-micron resolution at up to 40 frames-per-second, a fourfold improvement over standard ASLM. In this dissertation a sensing method applying to any physical quantity that modifies optical phase is developed. Two pulses are produced inside a synchronously pumped Optical Parametric Oscillator, generating two identical, undistinguishable frequency combs. The physical quantity to be measured applies a small phase shift/round trip to one of the pulses, resulting in a frequency shift of the corresponding comb. The latter frequency is measured as a beat by interfering the two combs on a detector. A world record resolution, close to the quantum limit, of 0.033 nanoradian (corresponding to 0.006 fm in displacement) is achieved. A detailed analysis of …

Two methods for improving the sensitivity of nitrogen vacancy quantum sensors in diamond are explored. First, by passively concentrating the magnetic flux, three orthogonally oriented ferrite truncated cone pairs amplify the field isotropically by 19 times, allowing measurement of Earth's field without a bias field and increasing sensitivity. Through thorough analysis, modeling and tuning a novel 3-dimensional flux concentrator system, we achieve a fractional standard deviation of less than 1% anisotropy and quantify minimal deadzones and ambient temperature-limited variations below 40 nT/hour. Considerable progress has been made in recent decades in the understanding of gravity wave (GW) dynamics. However, there remain important aspects and effects that are poorly understood. In particular, the coupling and deposition of their energy and momentum into the mesosphere lower thermosphere (MLT) region and the interaction and instability dynamics that are directly associated with GW breaking. Using an Advanced Mesosphere Temperature Mapper (AMTM) several strong wave breaking events were observed in the summer of 2015 at USU’s BLO research station. We present an analysis of how the group delay dispersion in silica glass affects the envelope of non-Gaussian laser pulses. In particular, we examine femtosecond pulses in the ultraviolet regime with cosine squared and symmetric exponential envelope shapes. For both envelope shapes, our results show a linear or semi-linear dependence of both the maximum electric field strength and duration on the group delay dispersion when the pulse passes through the silica glass medium with a thickness between 1 μm and 10 μm. This computational analysis is done using a fast Fourier transform method.

In this thesis we propose an interferometric scheme to retrieve the dynamics of an electron wave packet emitted from two distinct residual photo-ion channels in the ionization of an atom by an ultra-short UV pulse. EPAS (Entangled-Photoelectron Attosecond Spectroscopy) works by having a non-overlapping UV and few-cycle IR pulse, where the IR is tuned near the transition frequency between two electronic bound states of the photo-ion. This dissertation explores the interactions and applications of optical angular momentum within magneto-optical materials. Beginning with a theoretical and experimental analysis of multiple reflection and refraction phenomena within magneto-optical material. We derive and verify the dependence of refractive indices on optical spin angular momentum and magneto-optical magnetization, resulting in nonreciprocal elliptical and linear polarization beam splitting effects and wavevector discretization. We fabricate a magnetless slab waveguide isolator with minimal optical loss. Extending our study to optical orbital angular momentum, we introduce a perturbation to the electronic transition model in bismuth-substituted iron garnets. We utilize a method using frequency combs to construct waves that feature superoscillations—local regions of the wave that exhibit a change in phase that the bandlimits of the wave should not otherwise allow. This method has been shown to create superoscillating regions that mimic any analytic function—even ones well outside the bandlimits—to an arbitrary degree of accuracy. We experimentally demonstrate that these waves are extremely robust against noise, allowing for accurate reconstruction of a superoscillating target function thoroughly buried in noise. Surface analytical techniques are essential for engineering nano-detectors allowing characterization of their chemical composition, electromechanical properties, and physical structure. In the first chapter, the positron impact-induced secondary electron (PIISE) energy spectra and yield from single-layer graphene (SLG) and multi-layer graphene (MLG) grown on a polycrystalline Cu substrate have been presented. In the second chapter, an extensive catalog of Positron Induced Auger Spectra is presented to aid in the analysis of elemental composition of various materials. A Simple And Low-Cost Method For Generating Short Laser Pulses, Daniel Fisher

A simple, portable method is described and demonstrated for generating short optical pulses (~ 1 μsec) from a continuous wave (CW) source using a rotating pinhole. A CW 633nm HeNe beam was focused onto a 225 μm diameter stainless steel pinhole, which was rotated up to 1000 Hz using an optical chopper wheel. The pulsed light was then focused onto a silicon photodetector and read by an oscilloscope. This novel setup aims to bridge the gap between pulsed laser performance and price for wide application in spectroscopy methods, industrial laser peening, and medical treatments. In recent years, the negatively charged nitrogen- vacancy (NV) center has emerged as a promising solid-state color center capable of measuring magnetic fields with high sensi- tivity and spatial resolution under ambient conditions. In this thesis I will discuss how we perform super resolution magnetic microscopy and acquire magnetic field images of nanoparticle samples at 100 nm resolution. I will explain how 3D flux concentrators increase magnetic field amplitude and allows us to measure vector component magnetic fields as low as 50 µT with a diamond magnetometer, without the use of an additional bias magnetic field. The effectiveness of free-space laser communications is limited due to wavefront deformations caused by atmospheric optical turbulence. To determine these deformations, we propose a wavefront sensor that utilizes the angular selectivity of an optical transmission filter to measure the first derivative of the wavefront. The transmission filter converts the gradients of the incident wavefront into an intensity distribution. For each direction (x and y) this distribution is captured twice, with different angles between filter and optical axis for each measurement.
