Purification And Analysis Of A Pteroic Acid Conjugate, Naveen Kumar Ganji

All Capstone Projects

(FR) is overexpressed on the cell membrane in cancer tissues and activated macrophages associated with rheumatoid conditions. However, FR is restricted in normal healthy tissues. Folic acid has shown high affinity to folate receptors and thus can be used in targeting the tumor cells. Folic acid residue, pteroic acid is also used in several studies that are involved in targeting cancer cells. Several examples include technetium -99m radiotracer labelled pteroyl-lys conjugates, pteroic acid-conjugated nitroheterocyclic phosphoramidates..

In this project, we were successfully able to conjugate pteroic acid with polyethylene Glycol-Amine to create an elegant building block(pte-PEG-amine) for synthesis of future imaging …

Synthesis Of Copper-Selenium Complex, Sakvarit Nitrathorn Jan 2017

Synthesis Of Copper-Selenium Complex, Sakvarit Nitrathorn

Chulalongkorn University Theses and Dissertations (Chula ETD)

This research proposed to synthesize metal-organic frameworks (MOFs) from selenium-containing ligands and to study crystal structures of synthesized MOFs. MOFs can be designed in many ways, depended on types of metal center and linkage ligands. Selenium-containing organic molecules were rarely found, but they were a good choice for medical field. In this work, we tried to synthesize selenium-containing ligands from Schiff base reaction and Friedel-Craft acylation. However, the reactions were unsuccessful. In the other hand, we synthesized a coordination compound from commercial ligands, selenite ion from sodium selenite and seleno-L-cystine, which is amino acid. The experiment results showed that dark …

Impact Of Timber Harvest On Mercury Mobilization In A Mixed Pine And Hardwood Forest And Adjacent Lake In Mississippi, Mary Kristen Dickerson Jan 2017

Impact Of Timber Harvest On Mercury Mobilization In A Mixed Pine And Hardwood Forest And Adjacent Lake In Mississippi, Mary Kristen Dickerson

Electronic Theses and Dissertations

With 65% of land area covered in forest, Mississippi is a leader in the timber industry. Whereas forestry provides numerous benefits for humans and wildlife, forest harvest has been hypothesized to increase mercury (Hg) mobilization to aquatic systems. In a forest, Hg mostly accumulates in the upper layer of soil, where associates with organic matter. This top soil is exposed after clear-cutting and thus more susceptible to runoff. Harvest may also change the net balance between Hg methylation and demethylation by microorganisms through changes in redox conditions and addition of carbon sources from decaying logging residues. To better understand the …

Quantitative Analysis Of Tobacco-Specific Nitrosamines And Their Precursor Alkaloids In Tobacco Extracts, Celeste T. Wilkinson Jan 2017

Quantitative Analysis Of Tobacco-Specific Nitrosamines And Their Precursor Alkaloids In Tobacco Extracts, Celeste T. Wilkinson

Theses and Dissertations

Tobacco-specific nitrosamines (TSNA) are carcinogenic constituents derived from alkaloids in tobacco. Researchers are actively exploring several avenues to reduce TSNA levels in tobacco products like moist snuff tobacco. The focus of the research presented within is the quantitative analysis of TSNA in tobacco, specifically N’-nitrosonornicotine (NNN), 4-(methylnitrosamino)-1(3-pyridyl)-1-butanone (NNK), N’-nitrosoanatabine (NAT), and N’-nitrosoanabasine (NAB).

Tobacco alkaloids and nitrosamines in tobacco are currently analyzed by different instrumentation due to orders of magnitude difference in their concentrations, chromatographic separation challenges due to structural similarities, and similar mass fragmentation patterns. An analytical column using silica and 1,2-bis(siloxy)ethane hybrid particles of 1.7 µm size is …

Elucidation Of The Cation And The Anion Doping Mechanism Of Nanoparticulate Manganese Arsenide: Effect Of Doping On The Magnetostructural Properties, Surangi Hasitha Roshini Pimmachcharige Jan 2017

Elucidation Of The Cation And The Anion Doping Mechanism Of Nanoparticulate Manganese Arsenide: Effect Of Doping On The Magnetostructural Properties, Surangi Hasitha Roshini Pimmachcharige

Wayne State University Dissertations

The mechanism of solution-phase synthesis of P-doped type-B MnAs nanoparticles has been quantitatively assessed and the knowledge obtained is extended to independently control nanoparticle size and P incorporation in order to understand the roles of size and dopant concentration on the magnetostructural properties. During the solution-phase synthesis, the dimensions of the nanoparticles change as the monomer concentration in the bulk solution is depleted and the particles become uniform in size and shape when the temperature stabilizes. The temperature at which the nanoparticles were isolated controls the particle size and polydispersity. High temperature isolation of nanoparticles is required to achieve narrow …

Differential Activation Of Dead Box Rna Helicases Rhlb And Rhle By Hfq/Srnas And Their Target Mrnas, Amit Kumar Jan 2017

Differential Activation Of Dead Box Rna Helicases Rhlb And Rhle By Hfq/Srnas And Their Target Mrnas, Amit Kumar

Wayne State University Theses

Number of small RNA (sRNA) gene regulators have mounted in E. coli over the years whereas the number of validated protein partners has not changed considerably. Hfq has remained the only well studied global regulatory partner of sRNAs in E. coli. However, direct or indirect involvement of other protein partners has always been speculated. Study from Blasi lab has shown that CsdA, one of the five DEAD-box RNA helicases of E. coli, is required for the DsrA mediated upregulation of rpoS under cold stress condition. Previous study from our lab has identified two other DEAD-box RNA helicases, RhlB and RhlE, …

Tightly regulated cysteine CA proteases play a major role in maintaining the homeostasis within cells. Subsequently, when these proteases are dysregulated and mislocalized they disrupt healthy cell dynamics and contribute to many life-threatening pathologies such arteriosclerosis, osteoporosis and cancer. As such many pharmaceutical companies and research teams are highly interested in these proteases as targets. One emergent strategy is the spatiotemporal control of biological processes. In relation to this, a series of spatiotemporally controlled …

The Decline And Recovery Of Thermal Oxidative Stability Of Ultra-Low Sulfur Diesel Blends, Abigail Schoor Cohen Jan 2017

The Decline And Recovery Of Thermal Oxidative Stability Of Ultra-Low Sulfur Diesel Blends, Abigail Schoor Cohen

Electronic Theses and Dissertations

Dissertation supervised by Professor Bruce D. Beaver

The oxidative stability of ultra-low sulfur diesel (ULSD) has generally been expected to continuously decline over time. However, recent studies have suggested that it may fluctuate.1-2 In this study, the oxidative stability of stored commercially purchased ULSD was monitored with a methanol extraction method.3 In this methodology, proposed by Hardy and Wechter, fuel blends are extracted with methanol before and after thermal stressing.4 The methanol-soluble layer contains the oxygenated and oxidizable components of the fuel (SMORS) and the change in this mass upon stressing thus represents the oxidative stability of …

Target Based Design And Synthesis Of Heterocycles In The Potential Treatment Of Cancer And Opportunistic Infection, Shruti Choudhary Jan 2017

Target Based Design And Synthesis Of Heterocycles In The Potential Treatment Of Cancer And Opportunistic Infection, Shruti Choudhary

Electronic Theses and Dissertations

Dose limiting toxicity and development of multidrug resistance by the tumors are the major limitations of current cancer chemotherapy. Microtubule targeting agents (MTAs) are a structurally diverse set of compounds that disrupt microtubule dynamics and exert their anticancer effect. Among the various classes of such agents, the colchicine site binding agents are particularly important as they circumvent the Pgp and β-III tubulin mediated clinical resistance. These resistance mechanisms, when manifested, are a major reason for the failure of clinically used agents such as taxanes and vinca alkaloids. A series of monocyclic pyrimidine analogs were designed and synthesized as colchicine site …

Nitric Oxide Release From Poly(Lactic-Co-Glycolic Acid) Nanoparticles And Titanium Alloy, Nina A. Reger Jan 2017

Nitric Oxide Release From Poly(Lactic-Co-Glycolic Acid) Nanoparticles And Titanium Alloy, Nina A. Reger

Electronic Theses and Dissertations

Current methods for the treatment of bacterial infection involve the use of systemic antibiotics, which are high concentrations of antibiotics delivered over a long period time. Unfortunately, the use of systemic antibiotics can cause harmful side effects to the patient and increases the possibility for antibiotic resistance. The delivery of antibiotics or alternative antimicrobial compounds, such as nitric oxide, directly to the site of infection would decrease the amount of antibiotic necessary to treat a bacterial infection.

Poly (lactic-co-glycolic acid)/polyvinyl alcohol nanoparticles and a titanium- aluminum-vanadium metal oxide alloy implant were surface functionalized to deliver nitric oxide. Polymer nanoparticles can …

Rationalizing The Band Gap Tunability Of Semiconductors Via Electronic Structure Calculations, Matthew N. Srnec Jan 2017

Rationalizing The Band Gap Tunability Of Semiconductors Via Electronic Structure Calculations, Matthew N. Srnec

Electronic Theses and Dissertations

The polymorphs of titanium dioxide and various diamond-like semiconductor materials are promising candidates in photovoltaic solar cell applications. Several of these polymorphs have been studied with experimental and computational methods, which often aim at tuning the electronic structure, particularly the band gap value of the crystalline solid. Prior studies report that the addition of a substituent into the structure of titanium dioxide decreases its band gap value, but the reasons for this are unknown. Possible explanations for the change in band gap involve the substituent atom's crystal radius, electronegativity, and ionization energy. Understanding the cause of these changes will provide …

Photodynamic Cancer Therapy A Study Of The Photochemical Properties Of Meso-Tetra (2,3,4-Trifluorophenyl) Porphyrin And Meso-Tetra (2,3,5,6-Tetrafluorophenyl) Porphyrin, Delanie L. Newberry Jan 2017

Photodynamic Cancer Therapy A Study Of The Photochemical Properties Of Meso-Tetra (2,3,4-Trifluorophenyl) Porphyrin And Meso-Tetra (2,3,5,6-Tetrafluorophenyl) Porphyrin, Delanie L. Newberry

Honors College Theses

Photodynamic therapy(PDT) is a type of cancer treatment that utilizes photosensitive molecules that absorb photons and use that energy to create cytotoxic molecules. Porphyrins are photosensitive compounds that can be used selectively at the site of cancerous tumors. Their selectivity is due to the fact the porphyrins are generally not toxic in absence of light; in the presence of light, the drug will be activated to produce the cytotoxic molecule, singlet oxygen(1O2 ). 9,10 diphenyl anthracene(DPA) was used to simulate the cancer cells and monitor how effective the porphyrin was in creating 1O2, because DPA reacts with 1O2 form diphenyl …

Preventing Gene Expression Through The Interruption Of Dna Binding, Olivia Perdue Jan 2017

Preventing Gene Expression Through The Interruption Of Dna Binding, Olivia Perdue

Honors College Theses

The interruption of DNA binding is a gateway to the inhibition of unregulated expression of genes which can possibly lead to illnesses such as cancer and epilepsy. The goal of this research is to produce high affinity DNA binding molecules which could act to displace the binding of natural transcription factor thus reducing the over- or under-expression of various proteins. Literature shows that the interruption of DNA binding can be completed using both organic and inorganic means, such as by means of natural transcription factors like the NF-κB protein or with metal-ligand complexes in order to bind DNA. NF-κB, a …

Iodine-Initiated Hydration Of Internal Alkynes, Nicholas J. Shuber, Karelle Aiken Jan 2017

Iodine-Initiated Hydration Of Internal Alkynes, Nicholas J. Shuber, Karelle Aiken

Honors College Theses

The iodine-initiated hydration of internal alkynes is a novel, green, and inexpensive synthetic pathway in comparison with the commonly used transition metal catalyzed reactions. This can be a useful alternative in many fields, academic or industrial. This experimental design has been used successfully on terminal alkynes and is now being extended to internal alkynes. Through systematic probing of the reaction conditions, we have gleaned sufficient information to determine a strong mechanistic hypothesis based on neighboring group participation. Here in, we will report our result with a series of internal, keto alkynes reacted under ambient conditions with iodine in “wet” solvent. …

Structure-Reactivity Relationship Of Bio-Inspired Copper Complexes Containing Polypyridylamine Derivatives, Pattira Suktanarak

Chulalongkorn University Theses and Dissertations (Chula ETD)

In this dissertation, bio-inspired copper complexes were designed to study the influence from ligands, nuclearity, solvents and secondary coordination on reactivity toward small-molecule activation. This structure-reactivity relationship was examined in oxygen reduction reactions (ORR), and applied in detection of ascorbic acid (AsH2). First, copper(II) complexes containing polypyridyl-derivative ligands (i.e., Cu(dpa), Cu(adpa) and Cu2(addpa)) were synthesized and fully characterized by X-ray crystallography. The Cu(I) complexes which are active species for ORR were generated using AsH2 as a reducing agent. Redox states of the metal were examined by UV-Vis, NMR and EPR. It was found that copper complexes with various ligand topologies …
