This project will analyze the effects of chain length and saturation of fatty acids on saponification processes, as well as the effects of the base used in the reaction. Stearic acid, lauric acid, and oleic acid will be used for the fatty acid comparisons, and sodium hydroxide and potassium hydroxide will be used for the base comparisons. Stearic acid is considered a long chain fatty acid, while lauric acid is considered a short chain fatty acid. Oleic acid is a monounsaturated fatty acid. Chalcones are a class of naturally occurring compounds found in plants, which have a general structure of Ar–CO–CH=CH–Ar’. They have shown significant potential as tunable organic fluorophores due to their extended π-conjugation and structural simplicity. In this study, a 2-hydroxy-substituted chalcone featuring a terminal dimethylamino group was synthesized via a pyrrolidine-catalyzed aldol condensation and characterized for its photophysical properties. The electron-donating and -accepting ends of the molecule establish an intramolecular charge transfer (ICT) pathway, while the hydroxyl group enables excited state intramolecular proton transfer (ESIPT), leading to complex emission behavior. The focus of this project was the plant Dalea candida and its phytochemistry. The plant compounds were isolated and purified by an array of chromatographic techniques. The structures of the pure compounds were determined by one-dimensional and two-dimensional 1H and 13C NMR spectroscopy, supported by infrared and ultraviolet spectroscopy, experimental and calculated circular dichroism spectroscopy, and mass spectrometry. Plant parts were worked on separately since they typically differ in content. Purified compounds will be tested for biological activities. Facilitated Glucose transporters (GLUTs) are membrane proteins responsible for the uptake of simple sugars and other structurally similar metabolites in all mammalian cells. One of the hallmarks of cancer is dysfunctional sugar metabolism, known as the Warburg effect, and one of the manifestations of that effect is abnormal GLUT expression. There are 14 known GLUT proteins with different expression patterns in different tissues, and tumors tend to express certain GLUTs that are not present in surrounding healthy tissue. Nicotinamide adenine dinucleotide (NAD+/NADH) plays a critical role in cellular metabolism, with abnormally elevated NADH levels linked to disrupted metabolic processes and cancer. In this study, we developed near-infrared fluorescent probes to selectively measure NADH concentrations. We present five cyanine-based probes, which were optimized by modifying the connection bridges to fine tune electronics. Two NAD(P)H-biosensing probes consisting of 1,3,3-trimethyl-3H-indolium and 3-quinolinium acceptors, linked by thiophene, A, and 3,4-ethylenedioxythiophene, B, bridges are detailed. They exhibit near-infrared fluorescence at 742.1 nm and 727.2 nm for probes A and B, respectively, and generate absorbance signals at 690.6 nm and 685.9 nm. Probe A …

Every heartbeat depends on cyclical contraction-relaxation produced by the interactions between myosin-containing thick and actin-based thin filaments (TFs) arranged into a crystalline-like lattice in the cardiac sarcomere. Therefore, the maintenance of thin filament length is crucial for myocardium function. The thin filament is comprised of an actin backbone, the regulatory troponin complex and tropomyosin that controls interactions between thick and thin filaments. Thin filament length is controlled by the tropomodulin family of proteins; tropomodulin caps pointed ends of thin filaments, and leiomodin (Lmod) promotes elongation of thin filaments by a “leaky-cap” mechanism. The development of structurally defined carbon nanostructures has gained significant attention due to their unique electronic, optical, and mechanical properties. Cycloparaphenylenes (CPPs), also known as carbon nanohoops, and carbon nanobelts (CNBs) represent key molecular segments of carbon nanotubes (CNTs), offering a bottom-up approach to precise nanotube construction. This dissertation focuses on the synthesis, structural characterization, and functionalization of CPPs, as well as the development of novel synthetic pathways for CNBs. Within the field of biochemistry, there has been a significant development of ratiometric fluorescent probes designed for mitochondrial pH detection; however, many of these probes have emissions within the visible spectrum and require long synthesis times with low-yield reactions. However, in recent years, microwave-assisted syntheses of organic molecules have led to significant increases in reaction yields and quicker reaction times. In this work, two common reactions in probe synthesis, Heck coupling, and Knoevenagel condensations are investigated using microwave-assisted synthesis. The ERα transcription factor that induces tumor growth is a potential target for breast cancer treatment. Each monomer of the ERα DNA-binding domain (ERαDBD) homodimer has two conserved (Cys)4-type zinc fingers, ZF1 (N-terminal) and ZF2 (C-terminal). Electrophilic agents release Zn2+ by oxidizing the coordinating Cys of the more labile ZF2 to inhibit dimerization and DNA binding. Microsecond-length molecular dynamics (MD) simulations show that greater flexibility of ZF2 in the ERαDBD monomer leaves its Cys more solvent accessible and less shielded from electrophilic attack by sulfur-centered hydrogen bonds than ZF1 which is buried in the protein. Enteroviruses initiate genomic replication via a highly conserved mechanism that is controlled by an RNA platform, also known as the 5' cloverleaf (5'CL). Here, we present a biophysical analysis of the 5'CL conformation of three enterovirus serotypes under various ionic conditions, utilizing CD spectroscopy, size-exclusion chromatography, and small-angle X-ray scattering. In general, a tendency toward a smaller monomeric hydrodynamic radius in the presence of salts was observed, but the exact structural signature of each 5'CL varied depending upon the serotype. Exposure to organophosphate-based nerve agents and pesticides poses significant health and security threats to civilians, soldiers, and first responders. Despite extensive efforts to develop chemical detoxification agents for use in topical applications on exposed skin surfaces and for intravenous injections, there remains an unmet need for effective, non-hazardous decontaminating agents. The current state-of-the-art decontaminating agent, Dekon-139 (2,3-butanedione oxime, potassium salt), exhibits adverse effects when applied to the skin.

"Organofluorine chemistry plays a critical role in synthetic chemistry due to the unique properties of fluorine. However, the S-trifluoromethylation of thiols remains underexplored, with existing methods often relying on toxic and expensive organometallic catalysts. Given the advantages of S-trifluoromethylated compounds, including enhanced pharmacological activity and improved metabolic stability, we developed a green and cost-effective method for the synthesis of S-CF3 compounds using diacetyl as the organic catalyst and Langloi's reagent as the trifluoromethylating reagent. This reaction, which proceeds under mild conditions using visible light photoredox catalysis, works across various substrate scopes, including aromatic, heteroaromatic, and aliphatic thiols.

Azobenzene is a photoswitchable molecule known for its high conversion yield and repeatability. The inclusion of an azobenzene moiety with a known inhibitor of your target of interest allows for greater study of function as well as potential therapies. Three derivatives were designed from azobenzene and known inhibitor of EAATs, TBOA, to synthesize photoswitchable inhibitors of EAATs. Further investigation into the function of EAATs and development of a specific inhibitor for EAAT3, an important glutamate transporter in the brain required for proper function, can then be conducted. Preptin, a 34-amino-acid peptide derived from proinsulin-like growth factor II, has emerged as a promising therapeutic candidate for both Type 2 diabetes mellitus and osteoporosis. Its dual functionality—stimulating insulin secretion without inducing hypoglycemia and promoting osteoblast proliferation—positions it as a unique alternative to traditional sulfonylureas and osteogenic agents. One important cell signaling pathway is the PI3K pathway because it influences many key cellular functions such as proliferation and apoptosis. Among the enzymes that are involved in this pathway, the SH2-containing inositol-5’-phosphatase (SHIP) plays an important role by maintaining the balance of PI(3,4,5)P3 and PI(3,4)P2. Questions regarding the sustainability of petroleum-based surfboard waxes gave rise to the development and marketing of a sustainable alternative called “Eco” surf wax. The chemistry behind surf waxes is generally unknown and there is no published research on the area of surf waxes. In this analysis, the chemical properties of both types of waxes were investigated, to determine the difference between non-eco and eco waxes and between different water temperature rating waxes. 15 surf wax samples (10 non-eco, 5 eco) were extracted into organic and inorganic fractions and analysed by FT-IR, GC-MS, and HTGC-FID/MS. Tyre particles are a dominant source of microplastic pollution in our environment. However, their chemical and physical properties are poorly understood and no standardised methods for identifying and quantifying them exist. This study looked to develop a method to identify tyre particles using two tyre samples of differing particle sizes, fine (62 - 125 µm) and coarse (250 – 500 µm). Both samples underwent ultrasound-assisted extraction before being analysed via gas chromatography-mass spectrometry (GC-MS) in both full scan and SIM mode, using benzothiazole as a marker. Acetylcholinesterase (AChE) inhibitors are currently the only effective treatment for people with Alzheimer’s disease (AD), a fatal neurodegenerative disease that affects 50 million people worldwide at present but is expected to rise dramatically over the next 30 years. The aim of this research was to synthesise an AChE inhibitor derivative of tacrine, 4-chloro-2(((1,2,3,4-tetrahydroacridin-9yl)imino)methyl)phenol (chlorophenol tacrine). This was achieved by first synthesising tacrine, evidencing successful synthesis and assaying purity of the product, then, synthesising a simple derivative, acetyl tacrine, before chlorophenol tacrine. The use of exocyclic enol ethers as nucleophiles in carbon-carbon bond forming reactions has been underutilized in organic synthesis. This work describes the use of 6-iodomethyl-2-methylenetetrahydropyran as a bis-exocyclic enol ether equivalent for the synthesis of pyran derivatives. Use of this linchpin allows for bidirectional elaboration of the pyran core by two directional carbonyl ene. The development of a convergent, modular synthesis of 2,6-disubstituted-4H-pyrans in a three-step sequence starting from 6-iodomethyl-2-methylenetetrahydropyran is described. Initial carbonyl ene reaction of 6-iodomethyl-2-methylenetetrahydropyran with aldehydes or activated ketones was followed by elimination of iodide to give cross oriented bis-enol ethers that contain both endocyclic …

Oxadecalins are privileged scaffolds found in various natural products that show biological activity, such as platelet aggregating factor antagonism and activation of adenyl cyclase. As such, various synthetic methods such as intramolecular substitution reactions and [4+2] cycloadditions have been developed for the construction of these important intermediates. Studies towards the synthesis of a simplified core of the labdane diterpene, forskolin, are described. Forskolin is the only known supplement to naturally raise cAMP levels, thereby displaying important pharmacological properties. Multi-drug resistance poses a serious threat to future generations and historical antibiotic pipelines; consequently, the medical and economic burdens associated with treating multidrug-resistant bacteria are substantiated. In this context, P. aeruginosa (PA) emerges as one of the most significant healthcare challenges, being a leading cause of resistance-associated mortality worldwide. Despite modern efforts to combat these poor outcomes, drug-resistant cases continue to rise, and the need for novel antibiotic treatment is apparent.
