Small GTPases of the Ras superfamily are membrane-bound molecular switches that regulate nearly all major cellular processes, and their dysregulation drives cancers, neurological disorders, and metabolic diseases. While anchored to cellular membranes, the catalytic domains of small GTPases undergo orientation dynamics, adopting distinct configurations that can occlude or expose effector-binding surfaces and modulate signaling output. These dynamics have been characterized in the Ras oncoproteins, but whether they extend across the superfamily and influence allosteric druggability has remained unknown.

Warming, sea-level rise, and changing herbivory patterns are affecting ecosystem processes in northern coastal soils, including carbon and nitrogen cycling, although their combined effects are poorly understood. We studied the impact of these changes in a full-factorial microcosm experiment using four distinct soil types from the Yukon-Kuskokwim Delta of western Alaska. Soil was collected from three coastal wetlands, which differ in flooding frequency and use by migratory geese, and one rarely-flooded upland tundra used by geese in late summer. Due to the overuse of antibiotics in agriculture, the overprescription of antibiotics, and the misuse of antibiotics by patients, antibiotic resistance is spreading at an alarming rate. As antibiotic resistance spreads, the antibiotics we depend on to treat common infections become ineffective. Therefore, it is imperative to develop novel treatments for infections that do not increase the prevalence of antibiotic resistance. Honey is a naturally occurring substance that has a complex composition including compounds such as flavonoids, that contribute to potential health benefits. Quantitative analysis of flavonoids in honey can be challenging due to their similar structural characteristics, matrix effects, and chromatographic peak separation issues. This study aimed to establish calibration curves for five flavonoids–kaempferol, luteolin, chrysin, myricetin, and quercetin– to estimate their concentrations in five honey samples from varying geographic origins. The purpose of this thesis is to explain the current state of alternative oxygen carriers and propose a new hemoglobin-based oxygen carrier (HBOC) as a blood substitute/ blood therapeutic. Blood donation has drawbacks: risk of spread of infection, short storage time, and blood typing, and the current shortage. There are several possible modifications for hemoglobin to create a carrier (HBOC) which address specific issues related to allowing free hemoglobin to exist extra cellularly. Due to the diverse environments that mobile phones are exposed to via human handling they become reservoirs of microorganisms. The objective of the current study was to determine the level and character of bacterial contamination on mobile phones by identifying bacterial strains, and the degree of antimicrobial susceptibility. Precise indoor positioning system remains a significant challenge within Wireless Sensor Networks (WSNs) due to the instability and noise sensitivity of Received Signal Strength Indicator (RSSI) data. Recent agricultural applications of engineered metallic nanoparticles (ENPs) have raised concerns about their uptake and fate in plants. To understand this critical issue, this study investigated the fate and uptake of silver (Ag) and zinc oxide (ZnO) ENPs in maize (Zea mays L.) after single or binary ENP exposure in soil. Results indicated that both Zn and Ag elemental concentrations in roots increased with increasing ENP dosage. Ag NPs in the tissues were detected, but not Zn NPs. The current study aims to find an alternate oral contrast media for use in magnetic resonance cholangiopancreatography (MRCP) that satisfies the following criteria, the greatest imaging quality safety, no or few side effects, and low cost. The present study created an oral contrast agent sample (solution) by dissolving a one tablet of manganese sulfate supplement (taken daily dose) in 200 ml of distilled water. Pakistan's freshwater bodies host significant fish species, but molecular identification is still in its infancy, necessitating the use of morphological and molecular DNA barcoding techniques. The study confirmed the species of the order Siluriformes found in the River Ravi, Pakistan, using DNA extracted from muscle tissue, sequenced for the COI gene, and obtained accession and Barcode Index Numbers from GenBank and BOLD databases. In the BLAST search, identities ranged from 99.23–100% and 99.52–100% in the GenBank and BOLD databases, respectively. The study revealed that the Kimura 2-Parameter genetic distance increased from lower to higher taxonomic levels: within species (0.00%) < within genus (15.17%) < within family (19.77%). The study estimated average nucleotide differences (104.964) and nucleotide diversity (0.16099) but found Tajima's neutrality test to be statistically insignificant. The neighbor-joining tree displayed closely linked species under a node, whereas divergent species were grouped under distinct nodes. The COI gene-based DNA barcoding aids in Pakistan's fish resource inventory, monitoring, and management, providing valuable input for traditional methods.

Type 2 diabetes is a chronic metabolic disorder characterized by hyperglycemia and chronic inflammation. This study investigated the anti-inflammatory and insulin-modulating effects of Tithonia-Curcuma-Moringa (TCM) formulation in type 2 diabetes model. Male BALB/c mice were divided into six groups: normal control, type 2 diabetes control, metformin treatment, and three TCM formulation treatment groups with three different dose combinations. Type 2 diabetes was induced using a high-fat diet combined with streptozotocin injection, supported by sucrose administration. This study aims to analyze research trends in reading literacy within science learning from 2015 to 2025 through a Systematic Literature Review (SLR) following the PRISMA (Preferred Reporting Items for Systematic Reviews and Meta-Analyses) framework. The initial screening of 150 articles from the WoS Core Collection and SINTA databases yielded 29 articles that met the inclusion criteria. The analysis focused on publication dynamics, contributing countries, participant characteristics, research variables, and the science subjects investigated. The Mitotic Exit Network (MEN) is a signaling pathway that allows a dividing cell to exit cytokinesis. Dbf2 is a MEN protein kinase in yeast, which provides a simplified model for studying this pathway. By altering Dbf2’s activity through phosphorylation mutants, its effect on cytokinesis can be studied. Transformed cells with a mutant and degron Dbf2 allele are prompted to destroy the degron Dbf2 protein, leaving only mutated Dbf2 behind. DNA nanostructures have garnered increasing interest in their application as drug delivery vehicles. However, DNA is susceptible to degradation under physiological conditions. To remedy this, we have devised a novel method to coat DNA nanostructures with the polysaccharide chitosan (a natural derivative of chitin). Chitosan binds to DNA electrostatically and inhibits nuclease activity that would normally degrade DNA in a physiological environment. Iqg1 plays crucial roles in cytokinesis, including the assembly, contraction, and disassembly of the actomyosin ring, as well as the coordination of contraction and septation. Given that cytokinesis must occur post-mitosis, Iqg1 is regulated by the cell cycle. This study explores hypotheses regarding Iqg1 regulation: it’s binding to action is negatively influenced by the cyclin-dependent kinase (CDK) Cdc28 and positively influenced by dephosphorylation via Cdc14. Newly developed yeast degron tagged strains allow for rapid depletion of specific proteins. Iqg1 mutations that alter serine to threonine, which allows phosphorylation but not dephosphorylation, in the degron strain allow testing of mutant phenotypes. …

Asphalt is a relatively inexpensive material commonly used for road pavements; however, asphalt ages from exposure to oxygen and UV radiation, leading to loss of elasticity, which leads to cracks and potholes. To slow or reverse asphalt aging, pavement preservation treatments are in use. It’s unclear whether these treatments truly rejuvenate the pavement structure or merely offer a short-term benefit. Nuclear Magnetic Resonance (NMR) relaxometry is an analytical technique that uses the recovery of excited nuclear magnetization as a parameter to gain insights into molecular environments. Cataract is a leading cause of vision impairment worldwide and are primarily caused by oxidative stress that damages and aggregates lens proteins, leading to lens opacification. However, the eye's anatomical barriers limit the penetration and bioavailability of antioxidant therapies. To address this challenge, a dendrimer-based nanogel with a built-in reactive oxygen species (ROS)-scavenging capability developed by us was employed to deliver the antioxidant N-acetylcysteine (NAC) to the lens. NAC was loaded into a generation-5 PEGylated poly(amidoamine) dendrimer (G5-PEG-TK, termed the GPT) nanogel. This project aims to develop a folic acid-attached carbon dot nanoparticle system (FA-CDs-GEM) for the targeted delivery of gemcitabine to cancer cells. While gemcitabine is widely used, its effectiveness is limited because a short in vivo half life and lack of selectivity, which lead to severe side effects. Carbon dots offer a simple and modifiable platform as the drug delivery and bioimaging agent. Conjugation of folic acid to the carbon dots enables selective targeting of cancer cells that overexpress folate receptors. Programming DNA-binding proteins is an effective strategy for developing therapeutics that regulate gene expression. These proteins can directly bind to DNA to activate or repress transcription of specific genes. Zinc finger proteins are a class of DNA-binding proteins commonly found in eukaryotic organisms. Their DNA-binding function is derived from unique amino acid sequences within each domain that recognizes specific nucleotides, allowing each finger to selectively recognize three DNA base pairs. Adjoining of multiple domains in tandem allows recognition of a specific DNA sequence. The Lafayette River, a shallow, eutrophic tributary of the Chesapeake Bay, experiences seasonal harmful algal blooms (HABs). While management efforts focus on reducing external nutrient inputs, internal loading from benthic sediments remains poorly constrained. This study quantifies spatial and seasonal variability in nutrient and radium isotope fluxes across the sediment–water interface at three sites where HABs are known to initiate and represent contrasting sediment textures and hydrodynamic conditions. Sediment cores and porewater samples were collected in summer 2023 and winter 2024 to measure grain size, porosity, dissolved nutrients (NH₄⁺, NOₓ⁻, PO₄³⁻), and radionuclide activities (²²⁴Ra and ²²⁸Th). This study examines the factors influencing Senior High School students’ preferences for Bachelor of Secondary Education (BSEd) majors in Bagac and Morong, Bataan, with particular focus on the low interest in Mathematics and Science specializations despite increasing demand for STEM educators. Using a descriptive quantitative design, data were collected from 369 students through a structured survey and analyzed using frequency and percentage distributions. The results reveal a strong preference for non-STEM majors, with Social Studies (42.12%) and English (35.04%) dominating, while Science (15.43%) and Mathematics (7.395%) received significantly lower interest.
