Flavodoxin is a small, highly stable protein that contain a single FMN cofactor used to transfer single electrons at low potentials. The organism Rhodopseudomonas palustris contains a long-chain flavodoxin. Long-chain flavodoxins are characterized by a 20 amino acid loop that is proposed to allow interactions with partner proteins. We plan to utilize this protein as a model to build our repertoire with protein fluorination and fluorine NMR in flavoproteins. This tool kit will then be applied to the study of a partner protein of interest that is capable of performing electron bifurcation. We have incorporated m-fluoro tyrosine into flavodoxin and …

The receptor binding protein of parainfluenza virus, hemagglutinin- neuraminidase (HN), is responsible for actively triggering the viral fusion protein (F) to undergo a conformational change leading to insertion into the target cell and fusion of the virus with the target cell membrane. For proper viral entry to occur, this process must occur when HN is engaged with host cell receptors at the cell surface. It is possible to interfere with this process through premature activation of the F protein, distant from the target cell receptor. Polymer brush patterns have a central role in established and emerging research disciplines, from microarrays and smart surfaces to tissue engineering. The properties of these patterned surfaces are dependent on monomer composition, polymer height, and brush distribution across the surface. No current lithographic method, however, is capable of adjusting each of these variables independently and with micrometer-scale resolution. Here we report a technique termed Polymer Brush Hypersurface Photolithography, which produces polymeric pixels by combining a digital micromirror device (DMD), an air-free reaction chamber, and microfluidics to independently control monomer composition and polymer height of each pixel. Herein we report an in-depth study on the cytotoxic mechanism of action of four developmental cytotoxic copper(II) complexes: [Cu(phen)2]2+ (Cu-Phen); [Cu(DPQ)(Phen)]2+ (Cu-DPQ-Phen); [Cu(DPPZ)(Phen)]2+; and [Cu(DPPN)(Phen)]2+ (where Phen = 1,10-phenanthroline, DPQ = dipyrido[3,2-f:20,30-h]quinoxaline, DPPZ = dipyrido[3,2-a:20,30-c]phenazine, and DPPN = benzo[i]dipyrido[3,2-a:20,30-c]phenazine). This complex class is known for its DNA intercalative properties and recent evidence—derived from an in vivo proteomic study—supports the potential targeting of mitochondrial function. Therefore, we focused on mitochondrial-mediated apoptosis related to cytotoxic activity and the potential impact these agents have on mitochondrial function. ”There is a growing demand for non-surgical means of cataract treatment. This dissertation presents three bodies of work that reflect the early-stage development of eye drop formulations aimed at delaying cataract progression. These formulations consist of the antioxidant 2-mercaptopropionylglycine (MPG) loaded onto nanodiamond particles.
Cataractogenesis is linked to oxidative damage to lens proteins. To investigate the potential of MPG for protection against oxidative damage, A549 cells were incubated in 0.6 mM tert-butylhydroperoxide (tBHP). Cells exposed to tBHP without MPG exhibited elevated levels of reactive oxygen species, which led to the depletion of the vital antioxidant glutathione and, ultimately, apoptosis. Co-administration …

Oxidative stress-induced neuroinflammation is the prominent feature of neurodegenerative disorders, and is characterized by a gradual decline of structure and function of neurons. Many biochemical events emerge thanks to the result of this neurodegeneration, and ultimately provoke neuroinflammation, activation of microglia, and oxidative stress, leading to neuronal death. งานวิจัยนี้มุ่งเน้นศึกษาปัจจัยที่มีผลต่อการเกิดตำหนิและหาแนวทางปรับปรุงกระบวนการผลิตเพื่อลดปริมาณตำหนิชนิดฟองอากาศและรีม ด้วยการนำตัวแปรจากกระบวนการผลิตได้แก่ การชั่งน้ำหนักส่วนผสม อุณหภูมิในการหลอม อุณหภูมิในการขึ้นรูปและองค์ประกอบทางเคมีของกระจก มาหาความสัมพันธ์กับปริมาณตำหนิที่เกิดขึ้นด้วยการวิเคราะห์การถดถอย จากผลการวิเคราะห์ตำหนิฟองอากาศด้วยเทคนิคแมสสเปกโทรเมตรีและเทคนิครามานสเปคโทรสโคปีพบว่าฟองอากาศส่วนใหญ่พบแก๊ส CO2 เป็นองค์ประกอบอยู่ที่ 30 ถึง 70 เปอร์เซ็นต์ ซึ่งเกิดจากการสลายตัวทางความร้อนของวัตถุดิบและการไล่ฟองอากาศที่ไม่สมบูรณ์ และจากความสัมพันธ์ของปริมาณฟองอากาศและตัวแปรจากกระบวนการผลิตพบว่า อุณหภูมิการหลอมที่สูงขึ้น ปริมาณ fining agent ที่น้อยลง และกำลังการผลิตที่เพิ่มขึ้นส่งผลให้ตำหนิชนิดฟองอากาศเพิ่มขึ้น จากการศึกษาตำหนิชนิดรีมโดยใช้กล้องจุลทรรศน์อิเล็กตรอนแบบส่องกราดร่วมกับการวัดการกระจายพลังงานของรังสีเอกซ์ในการวิเคราะห์องค์ประกอบธาตุของตำหนิชนิดรีมเทียบกับบริเวณโดยรอบโดยพบว่ารีมมีปริมาณธาตุ Si และ Na สูงกว่าบริเวณโดยรอบ ซึ่งเกิดจากการชั่งวัตถุดิบที่คลาดเคลื่อนไปจากสูตรการผลิต โดยเกิดจากวัตถุดิบติดค้างบริเวณกรวยรับวัตถุดิบ ส่งผลให้องค์ประกอบทางเคมีของส่วนผสมไม่สม่ำเสมอ ยิ่งความแตกต่างของน้ำหนักส่วนผสมจากสูตรการผลิตยิ่งมาก จะทำให้ปริมาณรีมเพิ่มสูงขึ้น ซึ่งสามารถความผิดพลาดจากการชั่งนี้สามารถแก้ไขได้โดยติดตั้งค้อนลมเพื่อเคาะวัตถุดิบให้ลงไปในโม่ผสมวัตถุดิบครบถ้วนตามสูตรการผลิต

Metabolomics data are complex with a high degree of multicollinearity. As such, multivariate linear projection methods, such as partial least squares discriminant analysis (PLS-DA) have become standard. Non-linear projections methods, typified by Artificial Neural Networks (ANNs) may be more appropriate to model potential nonlinear latent covariance; however, they are not widely used due to difficulty in deriving statistical inference, and thus biological interpretation. Herein, we illustrate the utility of ANNs for clinical metabolomics using publicly available data sets and develop an open framework for deriving and visualising statistical inference from ANNs equivalent to standard PLS-DA methods.

Mitochondria are essential organelles as the site of respiration in eukaryotic cells and are involved in many crucial functions in cell life. Dysfunction of mitochondrial metabolism and irregular morphology have been frequently found in human cancers. The capability of imaging mitochondria as well as regulating their microenvironment is important both scientifically and clinically. Mitochondria penetrating peptides (MPPs), certain peptides that are composed of cationic and hydrophobic amino acids, are good candidates for mitochondria targeting. This paper reports on syntheses of 3-hydroxybenzaldehyde (1) and tetradecanal (2) from the same renewable starting material (cashew nut shell liquid or CNSL), thus demonstrating the potential of the agro-waste cashew nut shells (CNS) as a source of valuable fine and industrial chemicals. The strategy employed in accomplishing the synthesis started with extraction of CNSL followed by isolation of anacardic acid (4) and decarboxylation to give cardanol (5). Compound 5 was hydrogenated followed by protection of phenolic group to give 3-pentadecylphenyl acetate (7). Accumulation and allocation of biochemicals in plants are affected by various factors. The influence of seasons and vertical zonation on biochemical contents of seagrasses Cymodocea rotundata and Thalassia hemprichii from Mjimwema seagrass meadow along the coast of Dar es Salaam were assessed. Samples were collected in 2016 during wet/rainy (April–May) and dry (June–July) seasons. Concentrations of crude protein, soluble carbohydrates, phosphorus, lipid, and proline were determined using standard methods. Significant higher levels of phosphorus, lipid, protein and proline were observed during the dry season than in wet season (p < 0.05). Likewise, seagrass population occurring at the shore zone expressed higher levels of biochemical contents than those found at the subtidal zones. The highest concentration of protein was recorded in Thalassia hemprichii (5.8 mg/g dw)), while those of lipid and carbohydrates (27.1 and 53.1 mg/g dw, respectively) were recorded in Cymodocea rotundata. The two species studied did not vary significantly in their biochemical concentrations in both seasons. Generally it was observed that the concentrations of studied biochemicals were up-regulated during the dry season and varied depending on the locations where the seagrass grow. Thus, both season and zonation played roles in biochemical accumulation in the seagrasses understudy, however not interactively.

The peroxisome proliferator-activated receptor gamma (PPARγ) is a ligand-dependent transcription factor of the nuclear receptor superfamily that controls the expression of a variety of genes involved in fatty acid metabolism, adipogenesis, and insulin sensitivity. PPARγ is a target for insulin-sensitizing drugs, and it plays a significant function in prostate cancer. PPARγ antagonists have anti-proliferative effects in a broad range of hematopoietic and epithelial cell lines. Chamber oxidation experiments conducted at the Fire Sciences Laboratory in 2016 are evaluated to identify important chemical processes contributing to the hydroxy radical (OH) chemistry of biomass burning non-methane organic gases (NMOGs). Based on the decay of primary carbon measured by proton transfer reaction time-of-flight mass spectrometry (PTR-ToF-MS), it is confirmed that furans and oxygenated aromatics are among the NMOGs emitted from western United States fuel types with the highest reactivities towards OH. In this study, Anacyclus pyrethrm Link (roots) is a medicinal plant from the Asteraceae family, endemic to Morocco and is widely used in traditional medicine; was investigated for their total phenolics, flavonoids contents and antioxidant activities. The quantitative estimation of total flavonoids and phenols by the colorimetric method showed that the extracts were prepared by fractionation of the crude hydromethanolic extract (ethyl acetate and n-butanol). The mannose-6-phosphate/insulin-like growth factor II (M6P/IGF2) receptor is a transmembrane protein known to sequester growth factors from the extracellular matrix. This behavior suggests a mechanism of tumor suppression. Structurally, the receptor’s extracellular region is segmented into 15 homologous repeats, which are divided further into 5 triplet domains, labelled 1-3, 4-6, 7-9, 10-12, and 13-15. What is notable about the triplets is their propensity to form dimers with triplets on a second M6P/IGF2 receptor. In chapter 1, we describes asymmetric synthesis towards C29-C34 moiety of fragment A of the Antascomicin B. The non-immunosuppressant Rapamycin, Ascomycin, and Tacrolimus (FK506), strongly binds with FKBP12, the ligand FKBP12 complexes responsible for immunosuppressive activity. Antascomicin B structurally related to Rapamycin, Ascomycin, and Tacrolimus (FK506), binds strongly to FKBP12, yet does not shown immunosuppressive activity. The effects of the dihydrouracil lesion in DNA were studied using two dimensional NMR spectroscopy. The sequence used was based off of the Drew-Dickerson Dodecamer, with the cytosine in the three position replaced by a dihydrouracil. All of the nonexchangeable proton chemical shifts, with the exception of the H2, H5’, and H5’’, of the lesioned DNA were identified using NOESY spectra and then compared to the chemical shift values of the Drew Dickerson Dodecamer. Most research has focused on the interaction of the glycon of substrates and their inhibitors within the active site of glycoside hydrolases. The inhibitors employed to probe these interactions generally had small aglycons (i.e. a hydrogen atom, amidines, small aliphatic groups, or benzyl groups). Designed model transmembrane peptides and oriented 2H and 15N solid-state nuclear magnetic resonance (NMR) spectroscopy were used to analyze how simple sequence modifications can influence peptide structure, behavior and dynamics as well as for determining the pKa of glutamic acid at the membrane interface. The GW5,19ALP23 (acetyl-GGALW(LA)6LWLAGA-amide) peptide framework adopts a well-defined tilted orientation in lipid bilayers (DLPC, DMPC and DOPC) and undergoes low amounts of dynamic motion. The sequence was initially modified by moving the Trp residues outwards to positions 4 and 20. Veratrum californicum, commonly referred to as corn lily or Californian false hellebore, grows in high mountain meadows and produces bioactive steroidal alkaloids, including cyclopamine. Cyclopamine is a potent inhibitor of the Hedgehog (Hh) signaling pathway. Our lab has optimized methods to extract cyclopamine and related steroidal alkaloids from V. californicum, and implemented in-house Hh signaling bioactivity evaluation of these steroidal alkaloids using a Shh-Light II cell assay. New techniques have recently been developed and applied to capture reactive nitrogen species, including nitrogen oxides (NOx D NOCNO2), nitrous acid (HONO), nitric acid (HNO3), and particulate nitrate (pNO3 ), for accurate measurement of their isotopic composition. Here, we report – for the first time – the isotopic composition of HONO from biomass burning (BB) emissions collected during the Fire Influence on Regional to Global Environments Experiment (FIREX, later evolved into FIREX-AQ) at the Missoula Fire Science Laboratory in the fall of 2016.
