Reproduction In West Virginia Populations Of The Southern Two-Lined Salamander (Eurycea Cirrigera), Timothy R. Brophy, Thomas K. Pauley

We describe reproduction in West Virginia populations of the southern two-lined salamander (Eurycea cirrigera) and provide insights into the behavioral ecology of this wide-ranging urodele. The first signs of reproduction are evident well before the arrival of Spring. Sexually mature adults inhabit the cold, rocky streams of southwestern West Virginia in early February. Females arc known by the presence of large oocytes visible through the body wall and males by their extremely swollen heads. Breeding occurs in the stream during late March and is marked by the presence of gravid females with sperm caps in their posterior cloacae. Eggs are …

Immune responses against cardiac myosin and group A streptococcal M protein have been implicated in the pathogenesis of rheumatic heart disease. Although cardiac myosin is known to produce myocarditis in susceptible animals, it has never been investigated for its role in production of valvular heart disease, the most serious sequelae of group A streptococcal infection in acute rheumatic fever. In our study, cardiac myosin induced valvulitis in the Lewis rat, and epitopes responsible for production of valvulitis were located in the rod region. The stable isotope compositions of organic carbon (OC) and nitrogen, contents of OC and nitrogen, and C/N ratios for four short cores, which were recovered from Msansani Bay (WO 57), off Msimbazi River (WO 76) and Dar es Salaam harbour (WO 62 and WO 74), are used to document temporal variation in the sources and diagenetic alteration of organic matter (OM). The δ15N and δ13C values at sites WO 57, WO 62 and WO 74 show a trend of down-core decrease to the base of the core, and such a down-core trend could be related to diagenetic loss of compounds …

Circular permutation analysis has detected fairly strong sites of intrinsic DNA bending on the promoter regions of the yeast GAL1–10 and GAL80 genes. These bends lie in functionally suggestive locations. On the promoter of the GAL1–10 structural genes, strong bends bracket nucleosome B, which lies between the UASG and the GAL1 TATA. These intrinsic bends could help position nucleosome B. Nucleosome B plus two other promoter nucleosomes protect the TATA and start site elements in the inactive state of expression but are completely disrupted (removed) when GAL1–10 expression is induced. Recent advances in the technique of capillary electrophoresis have demonstrated fast, highly efficient separation of mixtures of intact microbes. This paper describes the application of this technique for the separation of microbial aggregates of Micrococcus luteus, Saccharomyces cerevisiae, or Alcaligenes faecalis. The aggregates of these microbes were resolved into several highly efficient peaks with analysis times under 10 min and efficiencies approaching 1 000 000 plates m-1 in some cases. A reproducible relationship was found between the electrophoretic mobility and the aggregation number or size of the cluster under a given set of experimental conditions. Previously, we reported the expression of chimeric lipopolysaccharides (LPS) in Escherichia coli strain JM109 (a K-12 strain) transformed with plasmids containing Haemophilus influenzae lipooligosaccharide synthesis genes (lsg) (Abu Kwaik, Y., McLaughlin, R. E., Apicella, M. A., and Spinola, S. M. (1991) Mol. Microbiol. 5, 2475–2480). In this current study, we have analyzed the O-deacylated LPS and free oligosaccharides from three transformants (designated pGEMLOS-4, pGEMLOS- 5, and pGEMLOS-7) by matrix-assisted laser desorption ionization, electrospray ionization, and tandem mass spectrometry techniques, along with composition and linkage analyses. Anti-streptococcal antibodies cross-reactive with N-acetyl-bD-glucosamine (GlcNAc) and myosin are present in the sera of patients with rheumatic fever (RF). However, their role in tissue injury is not clear. In this study, we show that anti-GlcNAc/anti-myosin mAb 3.B6 from a rheumatic carditis patient was cytotoxic for human endothelial cell lines and reacted with human valvular endothelium and underlying basement membrane. Reactivity of mAb 3.B6 with the valve was inhibited by human cardiac myosin > laminin > GlcNAc. After the theory of spontaneous generation was discredited, only religious explanations were offered to explain the origin of life. Alexander Oparin (1894-1980), an atheist, suggested that natural chemical reactions produced biological molecules that came together to form the first living thing. Later, Stanley Miller tested this hypothesis and produced chemical "building blocks" but not life itself. In spite of much progress, there is still no clear consensus on how life originated on Earth. Some scientists are even looking to outer space for the origin of life.

Reproduction and health were studied in a beaver (Castor canadensis) population from Prince William County, Virginia during 1998. Copulating beavers were observed in Quantico Creek at Prince William Forest Park on 22 January. Seven females from Quantico Marine Base were trapped between January and May, sacrificed, and dissected. Those reproductively active weighed over 39 pounds (17.7 kg) and were trapped before 1 March. Mean litter size based on counts of corpora lutea was 4.80 young (3-7); however, litter size based on the number of embryos present was only 2.75 (1-3), comparable to most others reported in the literature. The class I epitope of streptococcal M protein is an epidemiological marker for acute rheumatic fever (ARF)-associated serotypes of group A streptococci and is recognized by anti-M protein monoclonal antibody (MAb) 10B6. Using MAb 10B6, we determined the relationship between the class I epitope of M protein and the a-helical coiled-coil protein myosin. MAb 10B6 reacted by enzyme-linked immunosorbent assay and Western blotting with human cardiac myosin and rabbit skeletal myosin and its heavy meromyosin (HMM) subfragment. An Nmr Investigation Of The Effect Of Hydrogen Bonding On The Rates Of Rotation About The C-N Bonds In Urea And Thiourea, Karl A. Haushalter, Janice Lau, John D. Roberts

The interaction between urea and tetrabutylammonium acetate was investigated in dimethylformamide/ dimethyl sulfoxide solutions using ¹H and 15^N NMR. The chemical-shift behavior of the urea protons is consistent with a urea-acetate hydrogen-bonded complex involving both carboxylate oxygens and the urea hydrogens trans to the carbonyl oxygen with K_assoc = 120 ± 10. Line shape analysis of the temperature-dependent ¹H NMR spectra show that ∆G^‡ for rotation about the C-N bond of urea changes only slightly from 11.0 ± 0.1 to 11.2 ± 0.1 kcal/mol on 1:1 molar addition of tetrabutylammonium acetate to a dilute solution of urea. Polygonum perfoliatum L. (Polygonaceae) or mile-a-minute weed, from eastern Asia, has been spreading through wild and disturbed areas of the mid-Atlantic United States. It has a wide ecological amplitude and is found on stream banks, moist thickets, roadsides, nurseries, wood-piles, clearings, and ditches. Polygonum perfoliatum presents serious problems for reforestation because it thrives where forests are clear-cut. A southward direction of spread indicates that the species will probably proliferate in at least some southern states. Mechanical control is not likely to be completely effective because seeds are often left behind. No appropriate biocontrols are known. The plant poses a threat …

Giant salvinia (Salviniaceae) is a potentially serious aquatic weed that is native to Brazil. It has been reported in more than 20 countries, but is not established in the U.S. at this time. Mitchell and Tur (1975) reported that three years after the formation of the Kariba Reservoir in Africa, giant salvinia blanketed 21.5% or 1003 km2 of the reservoir surface area. Transmission electron microscope images were obtained of fractions of poly(1-tetradecene sulfone) and poly(cyclohexene sulfone) cast from very dilute solutions (0.007%, wt/vol) and rapidly freeze-dried on a mica surface. The samples were then vertically platinum-carbon (Pt-C) replicated with 9 +/- 0.3-A Pt-C and held together with 128 A of electron-transparent evaporated carbon. The Pt-C coating enlarges the molecular chain diameters by approximately 5 A, so that a single polysulfone chain has an apparent diameter of 9-12 A in the transmission electron microscope. Poly(1-tetradecene sulfone) forms short helical regions that show irregular helical turns of pitch 7-18 A, two to eight turns …

Phospholipids are the major structural components of the lipid portion of biological membranes. The study of the thermal properties of these phospholipid provides a systematic understanding of the relationships between the chemical structures and functional properties of phospholipid molecules in artificial and biological membranes. This study examined the thermal behavior of phospholipids in the solid state using DSC and TG coupled with FITR. The phospholipids were found to have phase transitions below the melting point. Egg-yolk lecithin had four transitions below its melting point; sphingomyelin had two phase transitions below its melting point. Phosphatidylethanolamine and phosphatidylserine had complex phase transitions. …

The first preliminary structure of a surface lipooligosaccharide from Haemophilus ducreyi has been determined. The major oligosaccharide was released by mild acid hydrolysis and analyzed by liquid secondary ion and tandem mass spectrometry. The mass spectral data combined with composition and methylation analysis yielded the most probable structure; Gal1----4GlcNAc1----3Gal1----4Hep1----6Glc1----( Hep1----2Hep1----)3,4Hep1---- KDO, where the reducing terminal 3-deoxy-D-manno-octulosonic acid (or KDO) exists in an anhydro form. This anhydro species results from the elimination of a phosphate from C-4 of KDO during mild acid hydrolysis. The core heptose trisaccharide consists of L-glycero-D-manno-heptose, but analysis of the peracetylated sugars indicated that the 1,4-linked heptose …

Consumption Rates, Evacuation Rates And Diets Of Pygmy Killifish, Leptolucania Ommata, And Mosquitofish, Gambusia Affinis In The Okefenokee Swamp, J. Douglas Oliver

I studied feeding dynamics of Leptolucania ommata and Gambusia affinis in the Okefenokee Swamp. Both fishes mainly ate insect larvae (such as Chironomidae) and Cladocera. Evacuation rates ranged from 0.143 (L. ommala in winter) to 0.279/hour (L. ommala in summer). Daily food consumption (dry weight) ranged from 24.2 (L. ommata in winter) to 148.3 mg/g/day (G. affinis in summer). Maximum consumption by both species was estimated at 26.31 mg/t m2 / day, in summer. These values are consistent with other observations supporting a hypothesis that invertebrate prey production is substantial in these blackwater wetlands.
