Diabetes mellitus is an ever increasing pandemic. Type 1 diabetes mellitus (T1DM), although not the most prevalent form of diabetes mellitus, has had an “average annual increase of 3.09%, meaning the incidence of T1DM would increase 40% from 1998 to 2010”. T1DM leads to many complications: adverse side effects such as retinopathy, neuropathy, and nephropathy. These comorbidities are costing the United States billions of healthcare dollars per year. Delaying complications result in cost savings and improved quality of life, which can be obtained by tightly controlling blood sugars. However, strict control is difficult to attain. Trans-Atlantic Correlation Of Upper Cretaceous Marine Sediments, Marcus R. Ross, David E. Fastovsky

Upper Cretaceous marine deposits from the Mid-Atlantic region of North America (Delaware, Maryland, and New Jersey) and the Maastricht area (southern Netherlands, and nearby Belgium and Gennany) are correlated across the Atlantic using a variety of macro invertebrates, nannofossils, and sequence stratigraphy. Four late Cretaceous Mid-Atlantic sequences, the Marshalltown, Englishtown, Merchantville, and Navesink, span the upper Santonian to lowennost Danian, and have direct correlatives in the Maastricht area. Correlations between the Mid-Atlantic and the Maastricht regions (respectively) are as follows: the upper Santonian to lower Campanian Merchantville and Matawan formations with the Achen and lower Vaals fonnations; the middle Campanian …

Experimental autoimmune myocarditis (EAM) can be induced in the Lewis rat by cardiac myosin or its cryptic S2-16 peptide epitope (amino acids1052 to 1076). To investigate cellular mechanisms and the role of antigen-presenting cells in regulation of myocarditis, we induced protection against EAM in Lewis rats by administration of S2-16 peptide in incomplete Freund’s adjuvant (IFA). Protection to EAM was associated with activation of S2-16-reactive splenocytes secreting high levels of interleukin (IL)-10 and reduced levels of interferon- and IL-2. Adoptive transfer of S2-16:IFA-induced splenocytes producing IL-10 suppressed myocarditis induction in syngeneic recipients, suggesting their regulatory cell nature. However, exposure of …

Two Purified Domains Of Telomerase Reverse Transcriptase Reconstitute Sequence-Specific Interactions With Rna, C. M. O'Connor, Cary K. Lai, Kathleen Collins

Telomerase reverse transcriptase (TERT) and telomerase RNA (TER) function together to create a uniquely specialized polymerase. Here we have described for the first time domains of bacterially expressed Tetrahymena TERT that interacted directly with TER in the absence of assembly chaperones. We used quantitative binding assays to define TER sequence requirements for recognition by the high affinity RNA binding domain and an independent N-terminal RNA interaction domain. The TERT RNA binding domain and N-terminal RNA interaction domain had distinct, nonoverlapping requirements for TER sequence and structure that together accounted for all of the sites of TER contact inferred for full-length …

The release of the 1000th complete microbial genome will occur in the next two to three years. In anticipation of this milestone, the Fellowship for Interpretation of Genomes (FIG) launched the Project to Annotate 1000 Genomes. The project is built around the principle that the key to improved accuracy in high through put annotation technology is to have experts annotate single subsystems over the complete collection of genomes, rather than having an annotation expert attempt to annotate all of the genes in a single genome. Synthesis Of The Novel Crown Ether 1 ,9-Dioxa-4,6-Dithiacycloundecane, Cassie Booth
Capstone Research Projects
Attempts to produce novel crowns, incorporating a thioacetal fragment, were made via two synthetic pathways. Despite the well-established reactions found in path A (see figure 3), this method failed to produce the desired crown. Path B, a one-pot reaction originally designed to close rings via the addition of a thioacetal fragment, yielded an oily product that was purified with a silica gel column (48.3% yield). Initial analysis by 1H NMR showed a structure consistent with that of the desired crown (figure 2). Mass spectroscopy was used to verify product identity, confirming the creation of our target molecule 1 ,9-dioxa-4,6-dithiacycloundecane.

Inmunofluorescence staining of murine NIH3T3 fibroblasts grown at high density shows that conventional nucleoside diphosphate (NDP) kinases A and B localize to a sensory organelle, the primary cilium. Similar results are obtained with Xenopus A6 kidney epithelial cells, suggesting that NDP kinases are a universal component of the primary cilium. The translocation of NDP kinase into primary cilia depends on size, taking place only when cilia reach a critical length of 5-6 μm. In mature cilia, NDP kinases are distributed along the ciliary shaft in a punctate pattern that is distinct from the continuous staining observed with acetylated α-tubulin, a …

We used immunofluorescence techniques to determine the localization of nucleoside diphosphate (NDP) kinase in NIH-3T3 fibroblasts. We found that cytoplasmic NDP kinase can be separated into two populations according to subcellular localization and response to extracellular stimuli. Specifically, within minutes of stimulation of resting fibroblasts with serum, growth factors or bombesin, a portion of NDP kinase becomes associated with membrane ruffles and lamellipodia. Another pool of NDP kinase accumulates independently of stimulation around intracellular vesicles. Transfection of cells with activated Rac mimics, whereas expression of dominant negative Rac inhibits, the effects of extracellular stimulation on the translocation of NDP kinase …

Hypothalamic proopiomelanocortin (POMC) neurons are critical for controlling homeostatic functions in the mammal. We used a transgenic mouse model in which the POMC neurons were labeled with enhanced green fluorescent protein to perform visualized, whole-cell patch recordings from prepubertal female hypothalamic slices. The mouse POMC-enhanced green fluorescent protein neurons expressed the same endogenous conductances (a transient outward K current and a hyperpolarization-activated, cation current) that have been described for guinea pig POMC neurons. In addition, the selective -opioid receptor agonist DAMGO induced an outward current (maximum of 12.8 1.2 pA), which reversed at K equilibrium potential (EK), in the majority …

C75, a recently derived compound that potently suppresses feeding and induces weight loss, has been proposed to act mainly by inhibiting fatty acid synthase (FAS) in central neurons that control feeding. For example, normal, fasting- associated, hypothalamic increases in neuropeptide Y (NPY)/Agouti-related protein (AGRP) expression and decreases in proopiomelanocortin (POMC)/cocaine and amphetamine regulated transcript (CART) expression were reported to be blocked by C75. Using loose-patch extracellular recording in acute slices, we tested the effect of C75 on anorexigenic POMC neurons and orexigenic NPY neurons of the hypothalamic arcuate nucleus, which were identified by promoter-driven GFP expression, as well as on …

Effects Of A Yec Apologetics Class On Student Worldview, David A. Dewitt, Steve Deckard, Sharon E. Cargo

Creationism is a significant component of an overall Christian worldview. A Creation Worldview Test (CWT) was administered to students both before and after completion of a required Apologetics course dealing with the creation/evolution controversy. Pre-test analysis revealed that incoming students tend to have a creationist worldview, however their CWT scores reflected a degree of uncertainty or neutrality on scientific creation and the age of the earth. Post-test analysis showed a significant shift toward a young earth creationist (YEC) view. Implications for teaching apologetics and development of a Creationist worldview based on a Six-day Young Earth perspective were explored and identified.
