4-(2-Methylbutyl)aminodipicolinic acid (H2MEBADPA) has been synthesized and fully characterized in terms of aqueous phase protonation constants (pKa's) and photophysical measurements. The pKa's were determined by spectrophotometric titrations, utilizing a fully sealed titration system. Photophysical measurements consisted of room temperature fluorescence and frozen solution phosphorescence as well as quantum yield determinations at various pH, which showed that only fully deprotonated MEBADPA2– is appreciably emissive. The fluorescence of MEBADPA2– has been determined to be quenched by hydroxide and methoxide anions, most likely through base-catalyzed excited-state tautomerism or proton transfer. This quenching phenomenon has been quantitatively explored through steady-state and time-resolved fluorescence measurements. …

A Bulk Water-Dependent Desolvation Energy Model For Analyzing The Effects Of Secondary Solutes On Biological Equilibria, Daryl K. Eggers

A new phenomenological model for interpreting the effects of solutes on biological equilibria is presented. The model attributes changes in equilibria to differences in the desolvation energy of the reacting species that, in turn, reflect changes in the free energy of the bulk water upon addition of secondary solutes. The desolvation approach differs notably from that of other solute models by treating the free energy of bulk water as a variable and by not ascribing the observed shifts in reaction equilibria to accumulation or depletion of solutes next to the surfaces of the reacting species. This study examines the properties of a 4 × 2 matrix of aqueous cations and anions at concentrations up to 8.0 M. The apparent molar water volume, as calculated by subtracting the mass and volume of the ions from the corresponding solution density, was found to exceed the molar volume of ice in many concentrated electrolyte solutions, underscoring the nonideal behavior of these systems. The work is related with the finding out of magnetic phases in strongly anisotropic high-temperature superconductor Bi1,7Pb0,3Sr2Ca2Cu3O10-δ in the temperature region where the superconductor is in the normal state. It was studied the temperature dependence of the paramagnetic probe EPR line width. In the normal state at T>Tc near 175 K it was revealed a pick in the temperature dependence of line width. In this region it was observed the time increase of the line width with the characteristic time ~ 17 min. This shows the possibility of magnetic phase formation in this material.

The investigation of temperature dependence of damping and period of vibrations of HTSC superconductive cylinder of Bi-Pb-Sr-Ca-Cu-O system suspended by a thin elastic thread and performing axial-torsional vibrations in a magnetic field at temperatures above the critical one for the main phase Tc=107 K were carried out. It was observed some "chaos" temperature region in the temperature interval 107-138 K, where it is seen separate ripples of dissipation and oscillation frequency. It is assumed that the "chaos" region could point to a possibility of existence of other magnetic and more high-temperature phases as single islands in a normal materials matrix.

Organically-modified siloxanes were used as host materials to examine the influence of surface chemistry on protein conformation in a crowded environment. The sol–gel materials were prepared from tetramethoxysilane and a series of monosubstituted alkoxysilanes, RSi(OR′)3, featuring alkyl groups of increasing chain length in the R-position. Using circular dichroism spectroscopy in the far-UV region, apomyoglobin was found to transit from an unfolded state to a native-like helical state as the content of the hydrophobic precursor increased from 0 to 15%. At a fixed molar content of 5% RSi(OR′)3, the helical structure of apomyoglobin increased with the chain length of the R-group, …

The secondary structures of two proteins were examined by circular dichroism spectroscopy after adsorption onto a series of organically modified silica glasses. The glasses were prepared by the sol-gel technique and were varied in hydrophobicity by incorporation of 5% methyl, propyl, trifluoropropyl, or n-hexyl silane. Both cytochrome c and apomyoglobin were found to lose secondary structure after adsorption onto the modified glasses. In the case of apomyoglobin, the α-helical content of the adsorbed protein ranged from 21% to 28%, well below the 62% helix found in solution. In contrast, these same glasses led to a striking increase in apomyoglobin structure …

A judicious change in the selected transition used for circular polarization excitation will overcome the low oscillator strength limitation of the currently allowed magnetic-dipole 5D1 ← 7F2 (Eu(III)) transition chosen for circularly polarized luminescence (CPL) measurement. The proposed allowed magnetic-dipole 5D1 ← 7F0 (Eu(III)) transition will facilitate the detection of CPL from the Eu(III) systems of interest. CPL on the acetonitrile solution of the chiral tris complex of Eu(III) with (R,R)-N,N′-bis(1-phenylethyl)-2,6-pyridinedicarboxamide ([Eu((R,R)-1)3]3+), recently suggested as an effective and reliable CPL calibrating agent, confirms the feasibility of the proposed experimental procedure. Three mammalian nuclear lamin proteins, lamin B1, lamin B2 and the lamin A precursor, prelamin A, undergo canonical farnesylation and processing at CAAX motifs. In the case of prelamin A, there is an additional farnesylation-dependent endoproteolysis, which is defective in two congenital diseases: Hutchinson-Gilford progeria (HGPS) and restrictive dermopathy (RD). These two diseases arise respectively from defects in the prelamin A substrate and the enzyme (ZmpSte24) that processes it. Recent work has shed light on the roles of the lamin proteins and the enzymes involved in their farnesylation-dependent maturation. The genetic diseases Hutchinson-Gilford progeria syndrome (HGPS) and restrictive dermopathy (RD) arise from accumulation of farnesylated prelamin A because of defects in the lamin A maturation pathway. Both of these diseases exhibit symptoms that can be viewed as accelerated aging. The mechanism by which accumulation of farnesylated prelamin A leads to these accelerated aging phenotypes is not understood. Here we present evidence that in HGPS and RD fibroblasts, DNA damage checkpoints are persistently activated because of the compromise in genomic integrity. Inactivation of checkpoint kinases Ataxia-telangiectasia-mutated (ATM) and ATR (ATM- and Rad3-related) in these patient cells can partially overcome their …

A new phase is detected within 100micrometers of 24 DEG ab grain boundary (GB) in YBa2Cu3O7-d 50 nm films on SrTiO3 by enhanced (001) anomalous scattering. Site identification and temperature dependence is interpreted using crystallographic weights to distinguish enhanced scattering from total electron yield and fluorescence spectra. The c-axis, c0 indicates that only ortho-I phase is present far from GB, both ortho-I and II phases are present near GB. The phase c0 is constant versus temperature across the transition to superconductivity.

Enhanced (001) anomalous scattering by YBa2Cu3O7-δ 50 nm films on SrTiO3 substrates with and without a grain boundary versus temperature is interpreted using crystallographic weights to distinguish it from total electron yield and fluorescence spectra. The power of diffraction enhancement is to ascertain the film oxygen composition from the changes in the c-axis, c0 as the film surface is scanned across the grain boundary, and to determine that c0 is constantversus temperature across the superconducting phase transition.

By means of contactless mechanical method of the measurement of energy losses in superconductors, the anisotropy of dissipation processes has been studied in single crystal high-temperature layered superconductors of Yba2Cu3O7-d system, being in mixed state. The observed anisotropy of energy losses indicates the possibility of the existence of the symmetry of order parameter of dx2-y2 type in these single crystals.

Comparison of Is/I0 with TEY/I0, F/I0 and A, at the Cu L2,3 and Ba M4,5 edges indicates that YBa2Cu3O7 (YBCO) 50 nm films scatters the circular components of linearly polarized light in a different way. The YBCO, Ba L2,3 XAS shows indication of Faraday rotation near Tc. Hilbert analysis of data was done.

The onset of electro magneto-optic effects, observed at the Ba L2,3 edges synchrotron X-ray absorption by a YBa2Cu3O7 single crystal, 10 K above the transition temperature to superconductivity, Tc ~ 92 K is used to identify the role played by the Ba donor layer in the transition to superconductivity in the CuO2 layers. Negative permeability leads to Faraday rotation of the transmitted beam below T = 112 to 56 K for the 22 μm thick single crystal (c-axis orientation of 8π/18 relative to εX-rays) and sharp changes in the density of empty final states lead to zero transmitted radiation in …

Absorption, emission, and excitation spectra for solid-state and solution of Tb(III), Dy(III), and Gd(III) complexes with the polypyridine ligand 6,6‘-bis[bis(2-pyridylmethyl)-aminomethyl]-2,2‘-bipyridine (C36H34N8) are presented. Measurements of excited-state lifetimes and quantum yields in various solvents at room temperature and 77 K are also reported and used to characterize the excited-state energetics of this system. Special attention is given to the characterization of metal-to-ligand energy transfer efficiency and mechanisms. The measurement of circularly polarized luminescence (CPL) from the solution of the Dy(III) complex following circularly polarized excitation confirms the chiral structure of the complexes under study. Objective— The majority of apoptotic cells in atherosclerotic lesions are macrophages. However, the pathogenic role of macrophage apoptosis in the development of atherosclerosis remains unclear. Elevated expression of Bax, one of the pivotal proapoptotic proteins of the Bcl-2 family, has been found in human atherosclerotic plaques. Activation of Bax also occurs in free cholesterol-loaded and oxysterol-treated mouse macrophages. In this study, we examined the effect of Bax deficiency in bone marrow-derived leukocytes on the development of atherosclerosis in low-density lipoprotein receptor-null (LDLR−/−) mice. We present a comparative study in terms of structural properties deduced from X - ray diffraction diagrams between YBa2Cu3O7 (YBCO) thin films fabricated on SrTiO3 (STO) single crystal substrates and bicrystallines substrates with a symmetrical tilt angle of 24 degrees. Periodic Lattice Distortions (PLD) have been observed around different Bragg peaks in YBCO thin films deposited on STO bicrystals while only diffraction peaks have been measured in the diagrams corresponding to the YBCO thin films deposited on STO single crystal substrates. Only in regions situated a 3.5 mm at both sides of the grain boundary the PDL have been investigated. …

The onset of electro magneto-optic effects, observed at the Ba L2,3 edges synchrotron X-ray absorption by a YBa2Cu3O7 single crystal, 10 K above the transition temperature to superconductivity, Tc ~ 92 K is used to identify the role played by the Ba donor layer in the transition to superconductivity in the CuO2 layers. Negative permeability leads to Faraday rotation of the transmitted beam below T = 112 to 56 K for the 22 μm thick single crystal (c-axis orientation of 8π/18 relative to εX-rays) and sharp changes in the density of empty final states lead to zero transmitted radiation in …

Type II restriction enzymes are homodimeric systems that bind four to eight base pair palindromic recognition sequences of DNA and catalyze metal ion-dependent phosphodiester cleavage. While Mg(II) is required for cleavage in these enzymes, in some systems Ca(II) promotes avid substrate binding and sequence discrimination. These properties make them useful model systems for understanding the roles of alkaline earth metal ions in nucleic acid processing. We have previously shown that two Ca(II) ions stimulate DNA binding by PvuII endonuclease and that the trivalent lanthanide ions Tb(III) and Eu(III) support subnanomolar DNA binding in this system. The increased transmission, observed in the EXAFS region of their X-ray absorption spectra, as cuprate materials go through the superconducting transition temperature Tc is correlated with a critical increase in Abrikosov Vortex expulsion in zero magnetic field as the temperature T →Tc.
