Background: Equine besnoitiosis, caused by Besnoitia bennetti, and equine protozoal myeloencephalitis (EPM), caused by Sarcocystis neurona and Neospora hughesi are relevant equine diseases in the Americas that have been scarcely studied in Europe. Thus, a serosurvey of these cystogenic coccidia was carried out in Southern Spain. A cross-sectional study was performed and serum samples from horses (n = 553), donkeys (n = 85) and mules (n = 83) were included. An in-house enzyme-linked immunosorbent assay (ELISA) was employed to identify a Besnoitia spp. infection and positive results were confirmed by an a posteriori western blot. For Neospora …

Disclosed is an in vivo equine disease model and a method of preparing the disease model for equine herpesvirus-1 neurological disease comprising a horse having a low preexposure level of herpesvirus-specific CTL precursors wherein the horse is experimentally infected with a neuropathogenic strain of equine herpesvirus or a mutant thereof. Also disclosed is a method of quantifying the risk factors and predicting the development of clinical neurologic signs of equine herpesvirus-1 neurological disease in a horse. It is well known that arginine vasotocin (AVT) in birds is involved in physiological homeostasis such as cardiovascular, osmotic regulation as well as reproductive functions. Pertinent to these physiological functions, AVT immunoreactive (-ir) neurons in the hypothalamus have been found associated with hemorrhage, dehydration, oviposition and other physiological regulation. Evidence, however, suggests that AVT also plays significant roles in modulating behavior, memory, stress, and food intake. This dissertation research addresses the latter two neuroendocrine functions of AVT in detail within the chicken brain. Research has recently demonstrated that larval zebrafish show similar molecular responses to nociception to those of adults. Our study explored whether unprotected larval zebrafish exhibited altered behaviour after exposure to noxious chemicals and screened a range of analgesic drugs to determine their efficacy to reduce these responses. This approach aimed to validate larval zebrafish as a reliable replacement for adults as well as providing a high-throughput means of analysing behavioural responses. Background: ELISAs are known to have a higher diagnostic sensitivity than the agar gel immunodiffusion (AGID) when employed for serological diagnosis of equine infectious anaemia (EIA). For this purpose, an “in-house” and five commercial ELISAs available in Italy were assessed by the National Reference Centre for EIA for their analytic specificity (Sp); precocity, defined as capability of detecting first antibodies produced during a new infection; precision based on repeatability and reproducibility, estimated from the coefficient of variation (CV); accuracy, estimated from multiple K and relative Sp and sensitivity (Se). Seeds of Carica papaya L. (Caricaceae) are a promising source of investigation for a novel anthelmintic to treat equine strongyle infestation. Strongyles are small redworms that attach to the intestinal wall in the equine. They can cause colic, poor hair coat, poor body condition, and, in severe cases, death. MicroRNAs (miRNAs) are small, non-coding RNAs which are produced throughout the body. Individual tissues tend to have a specific expression profile and excrete many of these miRNAs into circulation. These circulating miRNAs may be diagnostically valuable biomarkers for assessing the presence of disease while minimizing invasive testing. In women, numerous circulating miRNAs have been identified which change significantly during pregnancy-related complications (e.g. chorioamnionitis, eclampsia, recurrent pregnancy loss); however, no prior work has been done in this area in the horse. Cereal grains are often included in equine diets. When starch intake exceeds foregut digestion starch will reach the hindgut, impacting microbial ecology. Probiotics (e.g., lactobacilli) are reported to mitigate GI dysbioses in other species. This study was conducted to determine the effect of exogenous lactobacilli on pH and the growth of amylolytic and lactate-utilizing bacteria. Feces were collected from 3 mature geldings fed grass hay with access to pasture. Stable flies are among the most important blood feeding ectoparasites of cattle worldwide. The livestock industry is losing more than $2 billion dollar annually in the United States alone. Moist decaying vegetative material with bacteria supports larval development. Temperature and diet quality are important factors for stable fly development, affecting development rates, adult survival, and size, but how temperature affects adult size and interacts with nutrition is unknown. Mastitis is a devastating disease of dairy animals in Pakistan that occurs in clinical and subclinical forms. Various bacterial pathogens are associated with mastitis and the susceptibility of these pathogens remains inconsistent to various antibacterial drugs. This study was aimed to assess the prevalence of mastitis in and around the city of Peshawar, Pakistan. A total of 2,791 milk samples (Bovine, Ovine and Caprine) were subjected to White side test and Surf field mastitis test. The objective of this study was to evaluate type 2 porcine reproductive and respiratory syndrome virus (PRRSV)-based modified live vaccine in a herd with concurrent circulation of type 1 and type 2 PRRSV. Type 2 PRRSV-based modified live vaccine improved average daily gain by 48.24 grams/pig/day (631.17 grams/pig/day in the vaccinated group vs 582.93 grams/pig/day in the unvaccinated group; P < 0.05). Pathological examination indicated that the vaccination effectively reduced microscopic lung lesions compared with the control animals. The protection of this vaccine against type 1 and type 2 PRRSV provides clinical control of co-infection with both genotypes under field conditions.

An irregular, subcutaneous mass (2x3x2 cm) located between the first and second digits of the left hindlimb of a nine-year-old, male, captive capybara (Hydrochoerus hydrochaeris) was biopsied. Microscopically, the mass scattered throughout the dermis. The neoplastic cells were arranged in solid-sheet patterns and comprised of pleomorphic histiocytic cells with pleomorphic nuclei and abundant eosinophillic cytoplasm. Mitotic figures were 2-3 cells/HPF. Numerous, multinucleated, giant cells, up to 10 nuclei/cells, were also prominently observed and found scattered throughout the mass. Immunohistochemically, the neoplastic cells were strongly positive for vimentin and ionized calcium-binding adaptor molecule 1 (Iba-1), while immunoreactivity for cytokeratin was negative. …

To date, the interferon gamma (IFN-γ) assay has become an important diagnostic tool which is applied alternatively to define infection of intracellular pathogens. In this study, the molecular characteristics of IFN-γ in Rusa deer (Rusa timorensis), Hog deer (Axis porcinus), Sambar deer (Rusa unicolor) and Eld’s deer (Rucervus eldii) were determined. Among the cervid family, the analysis of IFN-γ cDNA sequences indicated 5 polymorphism nucleotide positions. Deduced IFN-γ amino acid compositions among the cervid group were negligibly different (98-100% homology). Preliminary quantification of IFN-γ mRNA expression in the cervidae by reverse transcription quantitative real-time polymerase chain reaction (RT-qPCR) was demonstrated. …

The objective of this study was to isolate antimicrobial resistant bacteria from animals, humans and environment, and to characterize them based on phenotypic attributes of resistance. A total of 521 samples were collected from animals (284), humans (69) and environment (168) which resulted in isolation of 615 non-duplicate resistant isolates (to either amoxycillin-clavulanate, ceftazidime, cefixime, meropenem, or imipenem). Antimicrobial susceptibility test (AST) revealed the highest rate of resistance to cefixime (49.3%) followed by ceftazidime (45%), meropenem (38.1%), amoxicillin-clavulanate (35.3%), and imipenem (13%). This study aimed to determine the treatment response of canine nasal tumor treated with only linear accelerated radiation therapy (LINAC) using volumetric measurement criteria at Kasetsart University Veterinary Teaching Hospital between May 2013 and August 2015. Medical records of the patients including age, breed, sex, histopathological diagnosis, clinical stage, types of radiation therapy and complications of treatment were recorded. All 20 dogs were irradiated with total dose ranging from 36 to 54 Gray (Gy). Nonalcoholic steatohepatitis (NASH) patients have elevated plasma saturated free fatty acid levels. These toxic fatty acids can induce liver cell death and our recent results demonstrated that the biliary epithelium may be susceptible to lipotoxicity. Here, we explored the molecular mechanisms of cholangiocyte lipoapoptosis in cell culture and in an animal model of NASH. Treatment of cholangiocytes with palmitate (PA) showed increased caspase 3/7 activity and increased levels of cleaved poly (ADP-ribose) polymerase and cleaved caspase 3, demonstrating cholangiocyte lipoapoptosis. Interestingly, treatment with PA significantly increased the levels of microRNA miR-34a, a pro-apoptotic microRNA known to be elevated in NASH. …

Porcine reproductive and respiratory syndrome (PRRS) is characterized by reproductive failure of sows and respiratory problems of nursery and growing pigs. Current management strategies mainly focus on preventing PRRS virus (PRRSV) infection through vaccination. However, the currently available modified live vaccines are not sufficient to eradicate the virus and do not provide complete immunity from infection. Periodontal diseases are the most common infectious diseases of dogs. Up to now, the knowledge about periodontopathic bacteria in dogs remains sparse. The objectives of this study were to apply the Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis (PCR-DGGE) technique, cloning and 16S rRNA gene sequencing to the investigation into subgingival plaque bacteria from healthy and periodontitis dogs; and compare DGGE patterns from the subgingival plaque of dogs and humans. Although Asian elephant is listed among the endangered species, the number of populations is over the carrying capacity in some areas, resulting in human-elephant conflict, as well as African elephants. High aggression associated with musth and female reproductive pathology are observed in captive elephants. This study was done to investigate the viability and effectiveness of cytochrome c oxidase I (COI) gene in DNA barcoding for identification of Chinese native chickens. Two sequences of the COI gene of 64 chickens of four Chinese native breeds (16 of each breed) were analyzed by amplification and sequencing. Results showed that the bar1 sequence of COI had lower polymorphism compared to bar2. Bar1 had 1 polymorphic site and 2 haplotypes, while bar2 had 4 polymorphic sites and 8 haplotypes.
