Myocarditis/dilated cardiomyopathy (DCM) patients can develop autoantibodies to various cardiac antigens and one major antigen is β1-adrenergic receptor (β1AR). Previous reports indicate that animals immunized with a β1AR fragment encompassing, 197–222 amino acids for a prolonged period can develop DCM by producing autoantibodies, but existence of T cell epitopes, if any, were unknown. Using A/J mice that are highly susceptible to lymphocytic myocarditis, we have identified β1AR 171–190, β1AR 181–200, and β1AR 211–230 as the major T cell epitopes that bind major histocompatibility complex class II/IAk or IEk …

Fifteen New Zealand rabbits were randomly assigned to one of 3 dietary treatment groups of 5 animals each and fed pelleted, extruded, or muesli diets in a completely randomized design experiment. Rabbits were placed in individual cages with ad libitum access to water and food for 45 days acclimation followed by 30 days experimental period. Feed intake of rabbits fed pelleted and extruded diets was greater (p < 0.05) than rabbits fed the muesli diet (125.6 and 130.4 vs. 91.9 g/d), but weight change and feed efficiency were not affected by treatment. Diet digestibility among the treatments was inconsistent when comparing results obtained from total fecal collection and AIA (please define) as an internal marker. Rabbits fed extruded and pelleted diets had lower (p < 0.05) cecal pH (6.42 and 6.38 vs. 7.02, respectively), and higher (p < 0.05) production of SCFA (18.5 and 19.0 vs. 11.7 mM, respectively) than those fed muesli. The fermentation products from rabbits fed pelleted and extruded diets had a greater proportion of butyrate and less propionate than rabbits fed muesli. The results of this study indicate that the basal dietary composition had a greater impact on diet utilization and cecal fermentation than food form.

The Ebola virus (EBOV) trimeric envelope glycoprotein (GP) precursors are cleaved into the receptor-binding GP1 and the fusion-mediating GP2 subunits and incorporated into virions to initiate infection. GP1 and GP2 form heterodimers that have 15 or two N-glycosylation sites (NGSs), respectively. Here we investigated the mechanism of how N-glycosylation contributes to GP expression, maturation, and function. As reported before, we found that, although GP1 NGSs are not critical, the two GP2 NGSs, Asn563 and Asn618, are essential for GP function. Further analysis uncovered that Asn563 and Asn618 …

Silibinin, extracted from milk thistle (Silybum marianum L.), has exhibited considerable preclinical activity against prostate carcinoma. Its antitumor and chemopreventive activities have been associated with diverse effects on cell cycle, apoptosis, and receptor-dependent mitogenic signaling pathways. Here we hypothesized that silibinin’s pleiotropic effects may reflect its interference with epigenetic mechanisms in human prostate cancer cells. More specifically, we have demonstrated that silibinin reduces gene expression levels of the Polycomb Repressive Complex 2 (PRC2) members Enhancer of Zeste Homolog 2 (EZH2), Suppressor of Zeste Homolog 12 (SUZ12), and Embryonic Ectoderm Development (EED) in DU145 and PC3 human prostate cancer cells, …

Mycobacterium avium subsp. paratuberculosis (MAP) is the etiologic agent of Johne’s disease in ruminants causing chronic diarrhea, malnutrition, and muscular wasting. Neonates and young animals are infected primarily by the fecal–oral route. MAP attaches to, translocates via the intestinal mucosa, and is phagocytosed by macrophages. The ensuing host cellular immune response leads to granulomatous enteritis characterized by a thick and corrugated intestinal wall. We review various tissue culture systems, ileal loops, and mice, goats, and cattle used to study MAP pathogenesis. Many bacteria regulate the expression of virulence factors via carbon catabolite responsive elements. In Gram-positive bacteria, the predominant mediator of carbon catabolite repression is the catabolite control protein A (CcpA). Hyperglycemia is a widespread disorder that predisposes individuals to an array of symptoms and an increased risk of infections. In hyperglycemic individuals, the bacterium Staphylococcus aureus causes serious, life-threatening infections. The importance of CcpA in regulating carbon catabolite repression in S. aureus suggests it may be important for infections in hyperglycemic individuals. Appropriate embryonic and fetal development significantly impact pregnancy success and, therefore, the efficiency of swine production. The pre-implantation period of porcine pregnancy is characterized by several developmental hallmarks, which are initiated by the dramatic morphological change that occurs as pig blastocysts elongate from spherical to filamentous blastocysts. Deficiencies in blastocyst elongation contribute to approximately 20% of embryonic loss, and have a direct influence on within-litter birth weight variation. This Peer Review of Teaching Project portfolio focuses on the Animal Physiology II course which is required for first year veterinary medicine students. Weekly quizzes assess baseline knowledge and had been administered individually and in groups. I hypothesized the discontinuation of group quizzes would increase student effort when preparing for quizzes. Unit exams involve scenario-based questions and require students to apply information. I hypothesized the implementation of group exams would help under-performing students improve their ability to apply information they had learned. Pestivirus infections have a huge economic impact on livestock production. In 2014, an aborted fetus from a sheep flock suffering from abortus and diarrhea was submitted for virological diagnosis. Due to the positive result of the sample for pestivirus and continuing clinical symptoms in the flock, all of the animals were sampled individually. Blood samples for serum and peripheral blood mononuclear cells were collected from 93 animals (5 rams, 26 lambs, and 62 sheep). The present study was performed to design sucuk production with microencapsulated and free cells of Lactobacillus rhamnosus and to investigate the effect of this production method on probiotic viability and quality characteristics on the final product. L. rhamnosus was microencapsulated with an optimal coating material combination by using response surface methodology and subsequently used in sucuk production as a probiotic culture. Microencapsulation protected L. rhamnosus against gastric and other stress conditions in sucuk composition. The aim of this study was to examine the effect of the prepartal administration of the monensin release capsule on dry matter intake (DMI), milk yield, and some reproductive parameters of dairy cows. This study investigated how environmental structuring in the growing units of goat kids influences their ability to cope with social isolation and weaning-induced stress. Two experimental groups were arranged in an enriched (E) or barren (B) environment and each of the two groups had 10 female and 10 male kids. During the social isolation, the goat kids in group E started bleating earlier (P = 0.1612) and more frequently (P = 0.0125). The present study was carried out to evaluate outer membrane protein (OMP)-based relatively simple dot-ELISA assays for the diagnosis of acute canine leptospirosis and to identify the differential expression of leptospiral OMPs for identifying the markers of acute infection, if any. Among 91 canine serum samples screened, 63 samples were found to be positive by MAT. All the samples were tested by OMP-based IgM and IgG dot ELISAs. The sensitivities of IgM and IgG dot ELISA in comparison to MAT (titer: ≥1:400) were found to be 100%, whereas the specificities were found to be 89.71% and 45.31%, respectively. IgM and …

The most prospective and actual trend in farm animal breeding is studying gene polymorphisms affecting the productive traits. The objective of this investigation was to study the effect of polymorphisms in intron 1 of the swine POU1F1 gene on number of days to 100 kg (days to 100 kg), length of body (LB), backfat thickness (BF), and number of piglets born alive (NBA). A study was conducted to compare the clinical presentation and pathology of some organs associated with experimental infection of Nigeria field isolates of Trypanosoma vivax and Trypanosoma congolense in the Yankasa breed of sheep. The present study aimed to identify whether vitamin E and C and their combination in basal freezing medium improved the viability of postthaw transfected spermatogonial stem cells (SSCs). For this purpose, SSCs were harvested by enzymatic digestion twice. After enrichment and culture on Sertoli cell feeder layer, SSCs were characterized by using alkaline phosphatase staining and expression of oct-4 and c-kit genes as specific stem cell markers. These functions may facilitate survival in harsh conditions, but the role of GR from the liver of the Arabian camel (Camelus dromedarius) is unknown. Sixty multiparous Holstein-Friesian dairy cows made up the study sample. Group M (n = 20) received 300 mg/day monensin (Kexxtone, Elanco) for 21 days prior to the expected calving date. The distribution of infection by the species Brucella ovis in rams and abortion cases in sheep in Croatia was investigated in the period from 2011 to 2015. The investigation relied on serological testing in rams and sheep after abortion conducted using a complement fixation test and immunosorbent assay conducted with bacteriological testing of the testes in seropositive rams and aborted material. Seropositive reactions were confirmed in 1100 (6%) of rams samples and in 12 (2.3%) of ewe samples. Isolated strains were also confirmed on the molecular level. Perineal hernia (PH) in dogs normally requires surgical reconstruction of the pelvic diaphragm. The use of fascia lata grafts has been reported and described in previous studies for the correction of PH after failure of the conventional herniorrhaphy. n order to reduce estrus synchronization cost, utilization of residual progesterone (P4) in the used controlled internal drug release (CIDR) device upon sanitization can be possible. Two datasets were compiled from 8 and 7 research articles involving 3434 (2653 heifers + 771 lactating cows + 10 ovariectomized cows) and 7301 cattle (3879 heifers + 3422 lactating cows) to evaluate blood P4 concentration and conception rate, respectively, in response to repeatedly used CIDR devices after sanitization via autoclaving and disinfection. As the number of CIDR usages increased, blood P4 concentration decreased linearly (y = -0.484X + 3.135, R2 = 0.99, P < 0.001). Blood P4 concentration (1.52 vs. 2.20 ng/mL; P < 0.002) and conception rate (40.76 vs. 32.96%, P < 0.05) for lactating cows were lower than those for heifers. The sanitization method did not affect blood P4 concentration, but reused CIDR device subjected to disinfection was associated with reduced conception rate. In summary, depending upon the initial P4 load (1.38-1.90 g), the CIDR device could be used twice in lactating cows and four times in heifers after autoclaving to achieve target blood P4 concentrations.

The objective of this work was to present the first case of Brucella suis biovar 2 isolation in outdoor reared pigs in Serbia. B. suis biovar 2 has not previously been detected in Serbia, from either wild boar or outdoor reared pigs. In our case, brucellosis was discovered in the region of Srem, which geographically constitutes a part of northwestern Serbia. Outdoor reared pigs in the Srem region are part of an extensive breeding system in the field and woods.
