We investigated the effects of drop volume on the development of mouse presumptive zygotes to the completely hatched blastocyst stage. For this aim, we established 3 experimental groups. In the first group, we cultured hybrid presumptive zygotes to the hatched blastocyst stage in 10 presumptive zygotes per 5, 10, 20, 40 and 50 µl drops. In the second group, we used microinjected hybrid zygotes for culture. In the last group, we cultured CD-1 mouse presumptive zygotes to the hatched blastocyst stage. Milk progesterone level is a good marker to evaluate the functional status of the corpus luteum in all farm animals. The present study was conducted to monitor changes in milk progesterone concentrations at the time of oestrus and post-mating for diagnosing early pregnancy in Anatolian water buffaloes. Twenty-one buffaloes reared at the Kocatepe Agricultural Research Institute, Afyon, Turkey, were used. The types and rates of acrosome and other (head, mid-piece and tail) abnormal spermatozoon types were determined by the Spermac® staining technique. Semen from 5 stray tom cats under the same management conditions was used. Semen collection was performed under general anesthesia by electro-ejaculator once a week for 5 weeks. After ejaculation the semen was diluted by 100 µl of 0.9% NaCl solution and stained with Spermac® stain for morphological evaluation. Occurrence Of Aflatoxin M_1 In Some Cheese Types Sold In Erzurum, Turkey, Mustafa Gürses, Ahmet Erdoğan, Bülent Çeti̇n

Turkish Journal of Veterinary & Animal Sciences

Sixty-three samples of cheese consisting of 23 White cheeses, 14 Kaşar cheeses, 11 Tulum cheeses, 9 Civil cheeses and 6 Lor cheeses (whey-curd), were analyzed for the occurrence of aflatoxin M_1 (AFM_1) using enzymatic immunoassay. In 28 of 63 samples (44.44%), the presence of AFM_1 was detected in concentrations between 7 ng/kg and 202 ng/kg. The mean levels of AFM_1 were 28.08 ng/kg in White cheeses, 22.80 ng/kg in Kaşar cheeses, 74.05 ng/kg in Tulum cheeses, 12.32 ng/kg in Civil cheeses and 15.95 ng/kg in Lor cheeses.

Milk yields and milk compositions of 2 different goat breeds with high milk yields, the Damascus (Shami) goat and the German Fawn x Hair goat crossbred (B_1), were monitored during lactation in order to evaluate their performance under Eastern Mediterranean conditions as an alternative to the Hair goat which has a low milk yield and shorter lactation period. Body fat composition and growth performance of sea bass juveniles fed with different kinds of vegetable oils instead of fish oil were studied. Feeding trials were carried out with 600 sea bass (Dicentrarchus labrax L.) juveniles with on initial live weight of 7.58 ± 0.13 g. Experimental feeds contained fish oil, soybean oil, sunflower oil, corn oil and olive oil. A Scanning Electron Microscope Examination Of Heligmosomum Costellatum, Kader Yildiz, Külti̇gi̇n Çavuşoğlu

The morphology of Heligmosomum costellatum, a nematode of field mice (Microtus epiraticus), was described by scanning electron microscope. The scanning electron microscopic view of this nematode revealed that the anterior end was surrounded by 2 cephalic vesicles. The 2 copulatory spicules of the male were enveloped in membrane and the male bursa was large. The female posterior end was characterized by a caudal spine. The body of this parasite had transverse ridges.

An Investigation Of Quality Changes In Anchovy (Engraulis Encrasicolus, L. 1758) Stored At Different Temperatures, Sevi̇m Köse, Mehmet Emi̇n Erdem

Turkish Journal of Veterinary & Animal Sciences

Quality changes in anchovy caught at different times during storage at ambient and refrigerated temperatures were investigated in 3 different batches. Sampling was carried out each day during storage. The results showed no significant differences among the anchovy batches except for bacteriological counts and trimethylamine (TMA) values at refrigerated temperatures. Storage time and temperature had a significant effect (P < 0.001, P < 0.05) on sensory, bacteriological and chemical values. Bacteriological counts and chemical values increased significantly (P < 0.05, P < 0.001) depending on time, while sensory results decreased (P < 0.05). According to sensory results, samples were acceptable for 1 day at ambient temperatures and for 2 days refrigerated. This study showed that refrigeration did not extend the shelf-life of fresh anchovies by more than a day compare to ambient temperature.

Phylogenetic relationships in some Turkish dog breeds were investigated to determine protein polymorphisms by electrophoretic analysis. Blood samples were collected from 276 dogs including the Kangal, Akbash and German Shepherd as well as Doberman Pinscher, Setter, Pointer and Labrador breeds. Enzymes and proteins were separated electrophoretically using isoelectrofocusing gel, starch gel and polyacrylamide gel. Polymorphism on albumin (Alb), postalbumin-1 (Poa-1), postalbumin-3 (Poa-3), transferrin (Tf), and esterase (ArE) loci was detected, while no polymorphism was observed on the hemoglobin (Hb) locus. To study the effect of acute fluoride poisoning on nitric oxide and methemoglobin formation, 250 mg/kg bw sodium fluoride was applied alone and verapamil was applied together with fluoride. Blood nitric oxide (Griess reaction) and calcium levels; hemoglobin, methemoglobin and hematocrit values; and erythrocyte counts were determined and compared with those of the controls. After the fluoride application it was found that there was a relative relationship between the increase in nitric oxide and methemoglobin levels and the decrease in calcium, hemoglobin and hematocrit levels and erythrocyte count. The reproductive activities of Silurus glanis were investigated over 12 months in Menzelet Reservoir, in the East Mediterranean region of Turkey. Total length of the examined 245 S. glanis varied from 33.4 cm (220 g) to 195.0 cm (48,000 g), and age distribution ranged between 1 and 14. The aim of this study was to isolate Edwardsiella ictaluri from diseased rainbow trout obtained from a commercial farm in the Ankara region, Turkey. Growth of the microorganism after 36 h of incubation was higher and purer at 28 ºC than at 37 ºC on all the organs (liver, spleen, kidney) of the 10 fish examined. In this paper, a simple and rapid method for the purification of 6-phosphogluconate dehydrogenase from rat erythrocytes together with an analysis of the kinetic behavior and some properties of the enzyme are considered. The purification steps comprised high-speed centrifugation, 20-50% ammonium sulfate precipitation and 2', 5'-ADP Sepharose 4B affinity gel chromatography. The yield was 78.4% and the specific enzyme activity was 5.15 EU/mg proteins. The present study was designed to evaluate serum biochemical and haematological parameters and serum nitrate concentrations in cattle suffering from foot- and -mouth disease (FMD). For this purpose, 30 Swiss Brown cattle aged between 3 and 5 years were used. This study was designed to determine the health status of dairy cattle, and the rates of and reasons for culling on dairy farms in Kars. A 2-stage stratified sampling strategy was used to select localities and farms. The study involved an interview with the farmers and regular visits to the farms. The farm prevalence levels of abortion, foot- and -mouth disease (FMD), respiratory problems, leptospirosis, blackleg, hardware (TRP), bloat and anthrax were 46.7%, 57.8%, 4.4%, 13.3%, 4.4%, 11.1%, 8.8% and 0% in 2001 and 53.3%, 62.2%, 17.8%, 17.8%, 4.4%, 11.1%, 11.1% and 8.8% in 2002, respectively. The structural diversity of oligosaccharides found in glycoconjugates is enormous. This is due to the number of different ways in which sugar monomers may be linked to each other regarding linkage position, anomeric configuration, pyranosidic or furanosidic ring form and chain branching. It has been proposed that these factors contribute to the exquisite potential of oligosaccharides to establish a code system of biological information. The information contained in these structures is decoded by complementary sites present on carbohydrate binding proteins (lectins).

The Determination Of Microbial Flora, Water Activity And Chemical Analyses In Smoked, Canned Mussels (Mytilus Galloprovincialis, L.), Gülgün F. Ünal Şengör, Hani̇fe Kalafatoğlu, Hüseyi̇n Gün

This study reports the water activity, moisture, sodium chloride, pH, acidity, vinegar, total plate count, fecal coliform, coliform and Clostridium botulinum of canned mussels processed by 2 different smoking techniques. The results indicated that the average measured water activity was 0.971 ± 0.001 for liquid smoked canned mussel with curry sauce and 0.982 ± 0.001 for traditional smoked mussel. The moisture of samples varied between 69.440% for traditional smoked mussel and 76.35% for salting before traditional smoking. The moisture, salt, pH, acidity and vinegar were significantly different in some of the groups (P < 0.05). Mussels smoked using liquid and oak wood sawdust, in tomato puree sauce, curry sauce and sunflower oil sauce, were sterilized at 120 ± 1 ºC and 5.2 ± 0.1 F_0. In none of the products was swelling or C. botulinum detected at 37 ºC or 55 ºC. Prior to canning, the mussels were investigated for fecal coliform, coliform and total plate count and found to carry no risk in terms of these parameters.

Many chemical anesthetics and drugs have been used in fish stripping, surgery and transportation, but their undesirable side effects on the body metabolism during treatment are not usually taken into consideration. In this study, we investigated the inhibition effect of some chemical anesthetics, such as Tranquil and tricaine methanesulfonate (MS-222), and hypothermia on glucose 6-phosphate dehydrogenase (G6PDH) activity. The effects of different extenders and cryoprotectants on the viability and fertilizing ability of frozen spermatozoa from the mirror carp, Cyprinus carpio L. (1758), were investigated. Semen was collected from anesthesized males by the abdominal massage method. Having determined the main spermatological properties (volume, motility, duration of motility, total spermatozoa number, concentration and pH), the pooled ejaculates were diluted with 3 extenders containing different cryoprotectants (15% DMSO, 15% DMA, 15% Glycerol) individually. Because of the interaction between nitric oxide (NO) and the superoxide anion radical, the effect of ischemia-reperfusion on intestinal contractility regulated by the nitrergic system was investigated in the present study.
