This study aimed to determine the effect of fingerroot powder supplementation on the performance of laying hens, egg quality, and malondialdehyde in the egg yolk. A total of 192 Hy-Line brown laying hens aged 43 weeks were divided into four treatment groups with 48 hens in each group (8 replicates; 6 birds each). The treatments were the control (basal diet), basal diet supplemented with vitamin E at 250 mg/kg, basal diet plus 20 g fingerroot powder/kg, and basal diet plus 40 g fingerroot powder/kg. Despite its name, Canine distemper virus (CDV) infection is not restricted to canids but extends to several species of the order Carnivora. The extent of CDV exposure in captive carnivores in Thailand has never been estimated before. Therefore, the objective of this study is to estimate CDV exposure in captive carnivores in Thai zoos using a pseudotype-based neutralization assay (SNT). Flavobacterium oreochromis is an opportunistic bacteria and causative agent of columnaris disease that affects freshwater fish worldwide. To initiate infection, the bacteria are required to attach and form biofilm on the fish's skin. This study aimed to describe the AMR characteristics and transferable R plasmids in Escherichia coli isolated from meat ducks reared in an open house system. One hundred seventy-seven (n=177) commensal E. coli were previously isolated from duck cloacal swabs. In this study, all were biochemically confirmed and examined for their susceptibilities to 15 antimicrobial agents by broth microdilution method. Transfer of R plasmids was tested by biparental mating method followed by plasmid replicon typing (PBRT) and plasmid multi-locus sequence typing (pMLST). It is becoming clear that exposure to different types of microplastic can impair the nonspecific immune system of crustaceans. Therefore, this study investigated the impact of high-density polyethylene microplastic particles (HDPE-MP) in feed on nonspecific immune system gene expression of Pacific white shrimp (Litopenaeus vannamei). PEDV and PDCoV are recognized as important pathogens causing enteric disease in swine herds worldwide. During 2014-2015, several outbreaks of PEDV occurred in many provinces in Thailand, and the clinical signs were more severe than those of classical PEDV in Thailand during 2008-2012. Programmed cell death-ligand 1 (PD-L1), an immune check points molecule, is expressed differently in various tissues and plays an important role in common cancers. The objectives of this study are to investigate the expression of canine PD-L1 mRNA and protein in normal tissues and common cancers and to determine the correlation of PD-L1 expression with histologic grading of the malignant mammary gland tumor (MGT). In the normal tissue, the highest PD-L1 mRNA expression was observed in lymph node (P<0.01). In the cancer tissues, the highest PD-L1 mRNA expression was observed in lymphoma, but not significant (P=0.224). The PD-L1 mRNA expression was higher in benign tumors compared to malignant MGT (P=0.093) but not significant. Immunohistochemistry (IHC) was performed in formalin fixed paraffin embedded tissue of normal and MGT tissue using newly developed anti-canine PD-L1 monoclonal antibody (mAb) (clone 5D2). Immunostaining was detected on epithelial cells, transitional cell, parenchymal cells, alveolar macrophages and immune cells. The immunostaining score of malignant MGTs was higher than benign MGT (P<0.05). The immunostaining score was not significantly different among histological sub-types and histological grades. The lack of correlation between PD-L1 mRNA and protein expression in canine malignant MGTs was observed. In conclusion, this anti-canine PD-L1 mAb could be used as predictive biomarker for canine MGT. This study has fulfilled and expanded a knowledge of the PD-L1 checkpoint molecule in dogs and highlights a potential of PD-L1 antibodies using for immunotherapy in canine MGTs, similar to their application in human tumors.

Effects Of Supplemented Intra- And Extra-Cellular Trehalose On Cryopreservation Of Feline Oocytes, Saengtawan Arayatham

Study 1 aimed at examining the effects of different stages of oocyte maturation and trehalose concentrations on freezing ability and developmental capability of feline oocytes. The immature and matured feline oocytes were cryopreserved and thawed. They were assessed for developmental competence. The effects of different concentrations of trehalose (0, 0.125, 0.25, 0.5, 1 M) on the freezing ability of feline oocytes were tested. The cryopreservation generally induced poor embryo development, while immature oocytes were more susceptible to cryodamage compared with the matured oocytes (P<0.05). Incubation of oocytes with high concentrations of trehalose (0.5, 1 M) significantly reduced the developmental competence. Optimal concentrations of trehalose during cryopreservation of immature and matured oocytes were different (0.125 M and 0.25 M, respectively). It is concluded that stages of meiotic maturation affected the freezing ability of feline oocytes. Optimal concentration of trehalose was differently required for immature and matured feline oocytes. Study 2 aimed at examining the effect of membrane-permeable trehalose on the freezing ability of feline oocytes matured in vitro. intracellular trehalose (trehalose hexaacetate; Tre-(OAc)6) was synthesized from trehalose precursor and subjected to spectroscopic characterization. The membrane permeability of the Tre-(OAc)6 was investigated by incubating oocytes with different concentrations of Tre-(OAc)6 (3, 15, and 30 mM) and optimum concentration and the toxicity of Tre-(OAc)6 were subsequently assessed. The effects of Tre-(OAc)6 on freezing ability in terms of apoptotic gene expression and developmental competence of in-vitro matured oocytes were examined, respectively. The Tre-(OAc)6 permeated into the ooplasm of cat oocytes in a dose- and time-dependent manner. The highest concentration of intracellular trehalose was detected when the oocytes were incubated for 24 h with 30 mM Tre-(OAc)6. For the toxicity test, incubation of oocytes with 3 mM Tre-(OAc)6 for 24 h did not affect maturation rate and embryo development. However, high doses of Tre-(OAc)6 (15 and 30 mM) significantly reduced maturation and fertilization rates (P < 0.05). In addition, frozen-thawed oocytes treated with 3 mM Tre-(OAc)6 significantly upregulated anti-apoptotic (BCL-2) gene expression compared with the control (0 mM) and other Tre-(OAc)6 concentrations (15 and 30 mM). Oocyte maturation in the presence of 3 mM Tre-(OAc)6 prior to cryopreservation significantly improved oocyte developmental competence in terms of cleavage and blastocyst rates when compared with the control group (P < 0.05). Our results lead us to infer that increasing the levels of intracellular trehalose by Tre-(OAc)6 during oocyte maturation improves the freezing ability of feline oocytes, albeit at specific concentrations. Study 3 aimed at examining the effects of combination of intra-cellular (Tre-(OAc)6) and extra-cellular (α,α-trehalose) trehalose on freezing ability of feline oocytes. Optimal concentrations of Tre-(OAc)6 (0, 1.5, and 3 mM) and trehalose (0, 0.125, 0.25, and 0.5 M) were investigated for cryopreservation of matured feline oocytes. The combination of the two optimal concentrations were subsequently examined. The results showed that 3 mM of Tre-(OAc)6 improved post-thawed developmental competence, while extra-cellular trehalose at 0.25 M significant enhanced freezing ability of feline oocytes (P<0.05). The combination of intra- and extra-cellular trehalose significantly improved developmental capability of frozen-thawed matured feline oocytes compared to non-trehalose control.

Improper use of antibiotics in swine could reduce commensal bacteria and possibly increase pathogen infections via the gut resistome. This study aimed to compare the metaproteomic profiles of gut resistome and related metabolism in the cecal microbiota of fattening pigs raised under antibiotic-free (ABF) conditions with those of ordinary industrial pigs (controls, CTRL). The top three relative abundant microbials in both groups were Escherichia coli, Ruminococcus and Lactobacillus, followed by Bacteroides and Bifidobacterium. The primary goal of this research was to assess the situation analysis and knowledge, attitude, and practices (KAP) of poultry practitioner veterinarians (PPV) and broiler poultry farmers (BPF) regarding antimicrobial use (AMU) and antimicrobial resistance (AMR) in Nepal. The study area covered approximately 88.1% of Nepal's poultry population. A total of 327 PPV from 56 districts and 500 BPF from 40 districts of seven provinces participated. AMU situation and demographic information were collected and analyzed using descriptive statistics and logistic regression analysis to determine associations between variables. The gastrointestinal infectious diseases caused by bacteria have been a major problem in the Thai swine industry. Porcine proliferative enteropathy (PPE), also known as porcine ileitis, is an enteric infectious disease caused by Lawsonia intracellularis. Porcine ileitis has been an economic threat to the global pork production system due to various types of diarrheas. Serological diagnosis can be applied for determining pathogen-antibodies seroprevalence and the timing of L. intracellularis infection when introduced to the herd. Streptococcus agalactiae is a highly contagious pathogen with a significant economic impact on bovine mastitis. The research aims to achieve three objectives: 1) identifying and characterizing S. agalactiae strains responsible for bovine mastitis based on their phenotypic and genotypic characteristics, 2) determining susceptibility profiles to antimicrobial agents, and 3) applying molecular epidemiological analysis to explore intramammary infections in Thai dairy herds. We analyzed a collection of one hundred S. agalactiae isolates from clinical and subclinical mastitis cases in thirteen dairy herds in central region of Thailand. Ungulate malaria parasites, including Plasmodium caprae that infects the domestic goats, have received little attention. There are only few studies about the prevalence and mitochondrial genome analyses. This study, therefore, aimed to investigate the natural infection dynamics of P. caprae and its mosquito vectors. Besides, genome analyses based on nuclear genes of P. caprae and other related haemosporidian parasites were also carried out to obtain an in depth understanding about their evolutionary history and origin. Pancreatic beta-cell replacement is recognized for feasible type 1 diabetes (T1D) treatment. However, in post-transplantation, the autoimmune destruction incidentally attacks the activity and survival of beta-cells are reported in animal and human. To address these concerns, the generation of immortalized, biocompatible beta-cells, and the engraftment platform are insightfully investigated. The stepwise chemical process was used for in vitro Insulin-producing cells (IPCs) production from mouse gingival fibroblast-induced pluripotent stem cells (mGF-iPSCs). Cannabinoids extracted from the hemp plant Cannabis sativa have been suggested as a cancer therapy due to their anti-tumor effects on several human tumors. Morphological and molecular data analyses have been performed on Clinostomum to assess its diversity. However, the molecular occurrence of Clinostomum spp. in aquaculture in Thailand has yet to be reported. The present study was carried out to (i) investigate the association between boar semen microbiota and semen qualities and (ii) determine the relationship between boar bacteria contamination and longevity of boar semen preserved for 3 days in BTS with and without gentamicin. A total of 20 Duroc ejaculates were collected from standard quality boars (n=10) and high-quality ones (n= 10). The aims of the present study were to investigate the effect of altrenogest treatment from 109 to 112 days of gestation in combination with double intramuscularly of PGF2alpha administrations at 113 days of gestation on the variation of farrowing in sows and its side effects on the sow colostrum yield, colostrum IgG, newborn piglet characteristics and piglet survival rate until seven days of postnatal life. Feline arterial thromboembolism (ATE) is an acute and distressing clinical presentation with high mortality and poor prognosis. ATE may be related to cardiogenic causes such as the underlying myocardial disease or non-cardiogenic causes. Echocardiography is the routine diagnostic tool to evaluate cardiac structural changes and myocardial function in cats. This study aims to evaluate the risk factor and an association between Bartonella spp. infection and immune status by using retroviral-infected cats as an immunocompromised model and to develop the recombinant B. henselae specific antigen protein (17kDa and GroEL)-based ELISA test for antibody detection against Bartonella spp. infection in cats.
