Human whipworm (Trichuris trichiura) infects approximately 1 in 15 people worldwide, representing the leading infectious cause of colitis and subsequent, inflammatory bowel disease (IBD). Current control measures focused on mass deworming have had limited success due to low drug efficacies. Vaccination would be an ideal, cost-effective strategy to induce protective immunity, leading to control of infection and transmission. Here we report the identification of whey acidic protein, a whipworm secretory protein, as a strong immunogen for inducing protective efficacy in a surrogate mouse T. muris infection model. We describe noncovalent, reversible asparagine ethylenediamine (AsnEDA) inhibitors of the Plasmodium falciparum proteasome (Pf20S) β5 subunit that spare all active subunits of human constitutive and immuno-proteasomes. The compounds are active against erythrocytic, sexual, and liver-stage parasites, against parasites resistant to current antimalarials, and against P. falciparum strains from patients in Africa. The β5 inhibitors synergize with a β2 inhibitor in vitro and in mice and with artemisinin. P. falciparum selected for resistance to an AsnEDA β5 inhibitor surprisingly harbored a point mutation in the noncatalytic β6 subunit. Movement disorders are uncommon manifestations of neurocysticercosis. When present, most are secondary to parenchymal lesions in the basal ganglia. Rarely, movement disorders can occur in racemose/extraparenchymal neurocysticercosis, an aggressive variant frequently associated with cerebrospinal fluid outflow obstruction and hydrocephalus. Appropriate treatment can reverse neurological manifestations.

Schistosomiasis, caused by trematodes in the genus Schistosoma, is a widespread neglected tropical disease with the species S. mansoni infecting over 100 million people. We aimed to better understand the snail host and parasite responses during intramolluscan stages of infection by performing dual RNA-Seq on field-collected snails with natural infections from western Kenya. We collected uninfected Biomphalaria pfeifferi, B. pfeifferi with a patent cercariae-producing S. mansoni infection, and B. pfeifferi exposed to field-collected S. mansoni at 1 and 3d (days post infection).

BACKGROUND: Blood flukes of the genus Schistosoma cause schistosomiasis-a neglected tropical disease (NTD) that affects more than 200 million people worldwide. Studies of schistosome genomes have improved our understanding of the molecular biology of flatworms, but most of them have focused largely on protein-coding genes. Small non-coding RNAs (sncRNAs) have been explored in selected schistosome species and are suggested to play essential roles in the post-transcriptional regulation of genes, and in modulating flatworm-host interactions. However, genome-wide small RNA data are currently lacking for key schistosomes including Schistosoma haematobium-the causative agent of urogenital schistosomiasis of humans.

Tick-borne diseases spread through enzootic transmission cycles that often involve ticks parasitizing bird hosts. Some avian species are competent reservoirs, amplifying the pathogens that cause tick-borne illnesses in humans. Prescribed burns in forests have the potential to reduce tick-borne disease risk if they limit interactions between ticks and infectious wildlife hosts. Although prescribed burns are increasingly being used for a variety of habitat management purposes, little is known about how they affect tick-host interactions. We hypothesize that if prescribed fires reduce tick abundance, then birds in burned forest plots will host fewer ticks than birds in unburned forest plots. Experimental …

Background: Primaquine (PQ) is the only FDA-approved drug for radical cure of Plasmodium vivax (P.v) malaria, but treatment can result in life-threatening hemolysis if given to a glucose-6- phosphate dehydrogenase deficient (G6PDd) patient. Therefore, the G6PD status of the patient with P.v must be known prior to prescribing PQ. However, patient G6PD status in rural malaria endemic settings is generally unknown, illuminating the need for reliable point of care G6PD diagnostic tests as a prerequisite to safely administer PQ. Schistosomiasis is a major neglected tropical disease. Control of schistosomiasis currently relies on a single drug, praziquantel, and despite its efficacy against the all schistosome species that parasitize humans, it displays some problematic drawbacks and alone is ineffective in counteracting adverse pathologies associated with infection. Moreover, due to the development of the potential emergence of PZQ-resistant strains, the search for additional or alternative antischistosomal drugs have become a public health priority. Parasitic helminths infect over 1 billion people worldwide, while current treatments rely on a limited arsenal of drugs. To expedite drug discovery, we screened a small-molecule library of compounds with histories of use in human clinical trials for anthelmintic activity against the soil nematode Caenorhabditis elegans. From this screen, we found that the neuromodulatory drugs sertraline, paroxetine, and chlorpromazine kill C. elegans at multiple life stages including embryos, developing larvae and gravid adults. These drugs act rapidly to inhibit C. elegans feeding within minutes of exposure. The liver fluke Opisthorchis viverrini is a food-borne, zoonotic pathogen endemic to Thailand and adjacent countries in Southeast Asia. The adult developmental stage of the O. viverrini parasite excretes and secretes numerous proteins within the biliary tract including the gall bladder. Lesions caused by the feeding activities of the liver fluke represent wounds that undergo protracted cycles of healing and re-injury during chronic infection, which can last for decades. Controlled human hookworm infection (CHHI) is a central component of a proposed hookworm vaccination-challenge model (HVCM) to test the efficacy of candidate vaccines. Critical to CHHI is the manufacture of Necator americanus infective larvae (NaL3) according to current Good Manufacturing Practice (cGMP) and the determination of an inoculum of NaL3 that is safe and reliably induces patent infection. Schistosomiasis is an important parasitic disease, touching roughly 200 million people worldwide. The causative agents are different Schistosoma species. Schistosomes have a complex life cycle, with a freshwater snail as intermediate host. After infection, sporocysts develop inside the snail host and give rise to human dwelling larvae. We present here a detailed step-by-step video instruction in English, French, Spanish and Portuguese that shows how these sporocysts can be manipulated and transferred from one snail to another. This procedure provides a technical basis for different types of ex vivo modifications, such as those used in functional genomics studies.

เหาโค (Cattle lice) เป็นแมลงดูดเลือดชนิดหนึ่งซึ่งเป็นสาเหตุหลักของปัญหาสุขภาพสัตว์ทั่วโลก หลายงานวิจัยได้เปิดเผยข้อมูลการตรวจพบเชื้อแบคทีเรียก่อโรคในเหาโคและเหาโคอาจมีศักยภาพที่สามารถทำหน้าที่เป็นพาหะนำโรคต่าง ๆ ที่สามารถติดต่อโดยเหา และการศึกษาชนิดของเหาโคโดยใช้เทคนิคทางอณูชีววิทยาและการตรวจหาเชื้อแบคทีเรียก่อโรคในประเทศไทยยังไม่มีการรายงาน งานวิจัยนี้มีวัตถุประสงค์เพื่อตรวจหาเชื้อแบคทีเรียก่อโรคในเหาโคและจำแนกชนิดของเหาโคที่เก็บจากพื้นที่ 3 จังหวัดของประเทศไทย โดยตัวอย่างเหาโคทั้งหมด 109 ตัวอย่าง จะนำมาสกัดดีเอ็นเอและใช้เทคนิค PCR ศึกษาบนตำแหน่งยีน 18S rRNA ที่ได้ทำการพัฒนาออกแบบขึ้นเพื่อระบุชนิดของเหาโค และตรวจหาเชื้อ Bartonella spp., Acinetobacter spp. และ Rickettsia spp. จากนั้นนำมาวิเคราะห์หาลำดับนิวคลีโอไทด์และสร้างแผนภูมิวิวัฒนาการ จากผลลำดับนิวคลีโอไทด์ของยีน 18S rRNA แสดงให้เห็นว่าชนิดของเหาโคในงานวิจัยนี้แบ่งออกเป็น 2 กลุ่ม คือ Haematopinus quadripertusus และ Haematopinus spp. ที่มีความสัมพันธ์ใกล้ชิดกับ H. tuberculatus และสามารถตรวจพบเชื้อ Bartonella spp. ทั้งยีน gltA และ rpoB ทั้งหมด 25 ตัวอย่างจาก 109 ตัวอย่าง (22.93%) พบทั้งในระยะไข่และตัวเต็มวัย เมื่อทำการวิเคราะห์ลำดับนิวคลีโอไทด์และศึกษาแผนภูมิวิวัฒนาการของยีน gltA และ rpoB พบว่าเชื้อ Bartonella spp. ในงานวิจัยนี้มีความใกล้ชิดกับสายพันธุ์ B. bovis งานวิจัยนี้เป็นการรายงานครั้งแรกของการตรวจพบเชื้อ Bartonella spp. ในเหาโคที่เก็บจากพื้นที่ 3 จังหวัดของประเทศไทย และทำให้ทราบถึงข้อมูลชนิดของเหาโคในประเทศไทยโดยใช้เทคนิคทางอณูชีววิทยา ซึ่งข้อมูลเหล่านี้อาจนำมาใช้เพื่อระบุศักยภาพของเหาโคที่จะทำหน้าที่เป็นพาหะนำโรคและอาจถ่ายทอดเชื้อแบคทีเรียก่อโรคเหล่านี้จากสัตว์มาสู่มนุษย์

Infecting upwards of a billion people worldwide, hookworm is one of the most prevalent parasitic infections affecting the world today. Those infected experience hookworm disease, characterized by severe anemia, which can lead to malnutrition, low birth weights, and physical and mental impairments. Currently, the benzimidazoles albendazole and mebendazole are used as the primary treatment; however, resistance to these drugs is emerging. Due to this resistance, finding new anthelmintic compounds active against hookworm is a priority. Leishmaniasis is a parasitic disease caused by protozoans of the genus Leishmania. This vector-born disease, transmitted by biting phlebotomine sandflies, typically manifests in one of three ways. The cutaneous form of the disease is characterized by localized lesions of the skin and is by far the most common manifestation.
