A Bisphosphonate With A Low Hydroxyapatite Binding Affinity Prevents Bone Loss In Mice After Ovariectomy And Reverses Rapidly With Treatment Cessation, Abigail A. Coffman, Jelena Basta-Pljakic, Rosa M. Guerra, Frank H. Ebetino, Mark W. Lundy, Robert J. Majeska, Mitchell B. Schaffler

Publications and Research

Bisphosphonates (BPs) are a mainstay of osteoporosis treatment; however, concerns about bone health based on oversuppression of remodeling remain. Long-term bone remodeling suppression adversely affects bone material properties with microdamage accumulation and reduced fracture toughness in animals and increases in matrix mineralization and atypical femur fractures in patients. Although a “drug holiday” from BPs to restore remodeling and improve bone quality seems reasonable, clinical BPs have long functional half-lives because of their high hydroxyapatite (HAP) binding affinities. This places a practical limit on the reversibility and effectiveness of a drug holiday. BPs with low HAP affinity and strong osteoclast inhibition …

Age And Sex Differences In Load-Induced Tibial Cortical Bone Surface Strain Maps, Alessandra Carriero, Behzad Javaheri, Neda Bassir Kazeruni, Andrew A. Pitsillides, Sandra J. Shefelbine

Publications and Research

Bone adapts its architecture to the applied load; however, it is still unclear how bone mechano-adaptation is coordinated and why potential for adaptation adjusts during the life course. Previous animal models have suggested strain as the mechanical stimulus for bone adaptation, but yet it is unknown how mouse cortical bone load-related strains vary with age and sex. In this study, full-field strain maps (at 1 N increments up to 12 N) on the bone surface were measured in young, adult, and old (aged 10, 22 weeks, and 20 months, respectively), male and female C57BL/6J mice with load applied using a …

Lumbopelvic Stability And Trunk Muscle Contractility Of Individuals With Chronic Ankle Instability, Ryan S. Mccann, Kelly Johnson, Ashley M.B. Suttmiller

Rehabilitation Sciences Faculty Publications

BACKGROUND: Chronic ankle instability (CAI) results in hip neuromuscular impairments that can perpetuate dysfunction through reduced lumbopelvic stability and subsequent malpositioning of the lower body during functional movement. Lumbopelvic stability might be further impaired through changes in trunk muscular contractility. However, lumbopelvic stability and trunk muscle morphology have not been compared between individuals with and without CAI.

PURPOSE: To compare lumbopelvic stability and trunk muscle contractility between individuals with and without chronic ankle instability (CAI) and determine if lumbopelvic stability and trunk muscle contractility are associated with self-reported function.

STUDY DESIGN: Case-control study.

METHODS: Ten individuals with CAI, 10 …

Simulation Of A Rat Muscle-Tendon Unit With Hill-Type Model Dynamics And The Study Of Viscoelasticity In A Collagen Molecule Via Molecular Dynamics, Veronica Siko Jan 2021

Simulation Of A Rat Muscle-Tendon Unit With Hill-Type Model Dynamics And The Study Of Viscoelasticity In A Collagen Molecule Via Molecular Dynamics, Veronica Siko

West Chester University Master’s Theses

The field of biological science has established that tendons transfer muscular forces to adjacent bones, but there is a dearth of information about the underlying physical principles of these interactions and how the property of viscoelasticity (displayed in tendons as a difference in mechanical response to stress with differing stretching rates) is encompassed in the collagen of tendons. This thesis details the results of concentric and eccentric contractions of the rat muscle-tendon unit (MTU) with and without viscoelasticity (concentric contraction requires active shortening of the muscle, while eccentric contraction requires active lengthening of the muscle) (Lovering & Brooks, 2014). Once …

Collagen microfiber-based constructs have garnered considerable attention for ligament, tendon, and other soft tissue repairs, yet with limited clinical translation due to strength, biocompatibility, scalable manufacturing, and other challenges. Crosslinking collagen fibers improves mechanical properties; however, questions remain regarding optimal crosslinking chemistries, biocompatibility, biodegradation, long-term stability, and potential for biotextile assemble at scale, limiting their clinical usefulness. Here, we assessed over 50 different crosslinking chemistries on microfluidic wet-extruded collagen microfibers made with clinically relevant collagen to optimize collagen fibers as a biotextile yarn for suture or other medical device manufacture. The endogenous collagen crosslinker, glyoxal, provides extraordinary fiber ultimate tensile …

Acoustic Detection Of Developmental Dysplasia Of The Hip In Models Representing Neonates, Pinak Raodeo Jan 2021

Acoustic Detection Of Developmental Dysplasia Of The Hip In Models Representing Neonates, Pinak Raodeo

Honors Undergraduate Theses

Developmental dysplasia of the hip (DDH) is a condition that involves the dislocation of the head of the femur in the acetabulum of the pelvic bone. Although it may not interfere with a child's range of motion during infancy, DDH can cause various effects over time such as joint pain, abnormal gait, and even paralysis. It is crucial to catch this phenomenon early on so that permanent disability is not introduced to the patient. In this study, an excitation device was used to send a broadband frequency signal through a hip joint simulated by a 3D printed bone apparatus consisting …

Purpose:
Knee sleeves have become widely popular in the exercise realm, especially for knee support during back squats. Knee sleeves are successful in providing frontal plane knee support during functional tasks, but have not been investigated in back squats. Knee wraps, a somewhat similar elastic material, provide elastic energy that increases weight lifted during back squats. Thus, it is possible the thick neoprene knee sleeves could prove advantageous for back squats. The purpose of this study was to examine the effects of knee sleeves on weight lifted, knee biomechanics, and muscle activations during back squats.

Methods:
Fifteen resistance trained men …

Predicting Total Length Of Fish In Minnesota Lakes Using Bony Structure Dimensions, Jay Holland Jan 2021

Predicting Total Length Of Fish In Minnesota Lakes Using Bony Structure Dimensions, Jay Holland

Journal of Earth and Life Science

This study seeks to establish a relationship between a fish’s total length to the size of various bony structures of prey fishes. Three important prey fish species in Minnesota were selected for examination in this study, including Black Crappie Pomoxis nigromaculatus, Bluegill Lepomis macrochirus, and Yellow Perch Perca flavescens. Length and weight dimensions of each fish’s otoliths, cleithra, and vertebrae were associated to total lengths of the fish they came from. In many dietary studies, the need for these relationships is strongly related to the disfigurement or degradation of prey fish in stomach samples. With accurate enough regression equations, researchers …

Cellular Bioenergetics Regulates Cell Proliferation During Mammalian Regeneration, Sandeep Saxena Jan 2021

Cellular Bioenergetics Regulates Cell Proliferation During Mammalian Regeneration, Sandeep Saxena

Theses and Dissertations--Biology

Mammalian system consists of stress-sensing molecules that regulates their cellular response against damage, injury and oncogenic stress. During vertebrate regeneration, cells responding to injury re-enter the cell cycle and proliferate to form new tissue. Cell cycle re-entry or arrest is at least partly regulated by cellular senescence which negatively impacts the proliferative pool of cells during regeneration. What remains unclear is whether cells in regenerating systems possess an increased propensity to proliferate and are refractory to signals that induce senescence. My thesis work has focused on how fibroblasts from the ear pinna differentially regulate healing in highly regenerative mammals (e.g., …

The Role Of Hamstring Contraction During Running: An Analysis, Theodore Hu Jan 2021

The Role Of Hamstring Contraction During Running: An Analysis, Theodore Hu

Senior Honors Theses

The hamstring muscle group, spanning the posterior aspect of the proximal lower limb, generates large amounts of contractile force during running. Researchers have examined this power generation within different phases of the stride cycle. Forces generated by the hamstrings hold implications for both running efficiency and the role of strengthening for injury prevention. The goal of this review is to examine the hamstring’s unique physiology as a bi-articular muscle, its role in running stride, various training philosophies, and the specific impact on running economy contributing to overall running performance. The summation of research presented shows that the hamstring muscle group …

The Impact Of Combined Flexion And Compression On The Mechanical Integrity Of The Annulus Fibrosus, Keaton Briar Jan 2021

The Impact Of Combined Flexion And Compression On The Mechanical Integrity Of The Annulus Fibrosus, Keaton Briar

Theses and Dissertations (Comprehensive)

Introduction: Intervertebral disc (IVD) herniation is characterized by an expulsion of nucleus pulposus (NP) material through the annulus fibrosus (AF). The AF contains two major adhesive structures, the intralamellar matrix and the interlamellar matrix, which act to maintain the strength of the AF and prevent NP material migration. As a herniation occurs, clefts form within the intralamellar matrix, pushing the NP between adjacent collagen fibres; meanwhile, delamination of the interlamellar matrix causes the NP to pool between layers of the AF. Further, herniation more readily occurs in a combined loading scenario of both compression and flexion. Flexion, and in particular …

Estimating Infant Hip Joint Moments Using A Novel Musculoskeletal Model, Yeram Lim, Victor Huayamave, Christine Walck, Erin Mannen, Safeer Siddicky

Publications

Musculoskeletal models offer a convenient method to non-invasively study human development, function, and pathologies. While adult musculoskeletal models have been extensively studied [1], infant populations have been widely underserved. However, understanding infant joint biomechanics during the first year of life is vital considering abnormal loads may be detrimental to growth and development. Joint moments from an infant musculoskeletal model could help assess pathologies such as developmental dysplasia of the hip (DDH). Previous computational studies involving infant biomechanics [2, 3] lacked the modeling of active muscle response which may affect musculoskeletal development [4]. As such, the purpose of this study is …
