In 2016, guidelines for diagnostic Next Generation Sequencing (NGS) have been published by EuroGentest in order to assist laboratories in the implementation and accreditation of NGS in a diagnostic setting. These guidelines mainly focused on Whole Exome Sequencing (WES) and targeted (gene panels) sequencing detecting small germline variants (Single Nucleotide Variants (SNVs) and insertions/deletions (indels)). Since then, Whole Genome Sequencing (WGS) has been increasingly introduced in the diagnosis of rare diseases as WGS allows the simultaneous detection of SNVs, Structural Variants (SVs) and other types of variants such as repeat expansions. Background: Obsessive-compulsive disorder (OCD) is an often disabling and chronic condition that is normally assessed using diagnostic interviews or lengthy self-report questionnaires. This makes routine screening in general health settings impractical, and as a result OCD is often under-(or mis-)recognized. The present study reports on the development of an ultra-brief version of the Obsessive-Compulsive Inventory-Child Version (OCI-CV) which may be administered routinely as a screener for pediatric OCD.
Method: A total of 489 youth diagnosed with OCD, 259 non-clinical controls, and 299 youth with other disorders completed the OCI-CV and other indices of psychopathology. Using item analyses, we extracted five …

Meiosis has a principal role in sexual reproduction to generate haploid gametes in both sexes. During meiosis, the cell nucleus hosts a dynamic environment where some genes are transcriptionally activated, and some are inactivated at the same time. This becomes possible through subnuclear compartmentalization. The sex body, sequestering X and Y chromosomes during male meiosis and creating an environment for the meiotic sex chromosome inactivation (MSCI) is one of the best known and studied subnuclear compartments. Vascular ring is a rare congenital anomaly in which the abnormal origin of the aorta or its branches and pulmonary arteries leads to encircling and compression of the trachea and esophagus. A right aortic arch (RAA) with an aberrant left subclavian artery is one of the most common forms of vascular ring. Here, we report a case of a prenatally diagnosed vascular ring resulting from an RAA with an aberrant left subclavian artery. Pregnancy in women with hypertrophic cardiomyopathy is not well described. In this retrospective study, we analyzed data on pregnant women with hypertrophic cardiomyopathy who were under follow-up care in the cardiology department of a tertiary care hospital. We reviewed data on all women registered in the hypertrophic cardiomyopathy cohort and those who attended the cardio-obstetric clinic and delivered between January 2010 and June 2019. From these 2 groups, we identified 7 pregnant women with hypertrophic cardiomyopathy who delivered during this period. Background
The aim of this study was to evaluate clinical outcomes in the published literature on medical and surgical management of diabetic foot osteomyelitis (DFO).
Methods
A PubMed and Google Scholar search of articles relating to DFO was performed over the dates of January 1931 to January 2020. Articles that involved Charcot arthropathy, case reports, small case series, review articles, commentaries, nonhuman studies, and non-English articles were excluded. The Quality Assessment of Diagnostic Accuracy Studies (QUADAS-2) tool was used to rate the bias of each study. A meta-analysis was performed using random-effects and inverse variance methods. PURPOSE: To examine the relations between patient-reported outcomes (PROs) within a conceptual model for adults with sickle cell disease (SCD) ages 18 - 45 years enrolled in the multi-site Sickle Cell Disease Implementation Consortium (SCDIC) registry. We hypothesized that patient and SCD-related factors, particularly pain, and barriers to care would independently contribute to functioning as measured using PRO domains.
METHODS: Participants (N = 2054) completed a 48-item survey including socio-demographics and PRO measures, e.g., social functioning, pain impact, emotional distress, and cognitive functioning. Participants reported on lifetime SCD complications, pain episode frequency and severity, and barriers to healthcare.

Acupuncture and moxibustion have been accepted as add-on options for primary dysmenorrhea (PD); however, the clinical evidence is still inadequate. We searched AMED, CENTRAL, EMBASE, PubMed, Web of Science, CBM, CNKI, VIP, Wangfang database, ANZCTR, ClinicalTrials.gov, and the WHO ICTRP, from their inception to February 2021. The pooled analysis of 13 RCTs with 675 participants for VAS showed that acupuncture and moxibustion were more effective in managing PD than the control group with the MD of −1.93 (95% CI [−2.80, −1.06] and −2.67 (95% CI [−4.96, −0.38]). This study utilised feedback from older adults during balance-challenging, elastic band resistance exercises to design a physical activity (PA) intervention. Methods: Twenty-three active participants, aged 51 – 81 years, volunteered to perform a mini balance evaluation test and falls efficacy scale, and completed a daily living questionnaire. Following a 10 min warm-up, participants performed eight pre-selected exercises (1 × set, 8 – 12 repetitions) using elastic bands placed over the hip or chest regions in a randomised, counterbalanced order with 15 min seated rests between interventions. To investigate if Clostridium (Clostridioides) difficile infection (CDI), traditionally thought of as hospital-acquired, can be genomically linked to hospital or community environmental sources, and to define possible importation routes from the community to the hospital.
In 2019, C. difficile was isolated from 89/300 (29.7%) floor and 96/300 (32.0%) shoe sole samples at a tertiary hospital in Western Australia. Non-toxigenic C. difficile ribotype (RT) 010 predominated among floor (96.6%) and shoe sole (73.2%) isolates, while toxigenic RT 014/020 was most prevalent among contemporaneous clinical cases (33.0%) at the hospital. Introduction: Biomarkers that predict the risk of immune-mediated adverse events (irAEs) among patients with non-small cell lung cancer (NSCLC) may reduce morbidity and mortality associated with these treatments. Methods: We carried out high resolution human leucocyte antigen (HLA)-I typing on 179 patients with NSCLC treated with anti-program death (PD)-1/program death ligand (PDL)-1. Toxicity data were collected and graded as per common terminology criteria for adverse event (CTCAE) v5.0. We used 14.8-week for landmark analysis to address lead-time bias to investigate the correlation between HLA-I/II zygosity, supertypes and alleles with irAE. Sequencing batch reactors are efficient wastewater treatment systems, however, they are not adapted to treat aquaria. We aimed to compare the efficacies of a modified single-tank sequencing batch reactor (SSBR) and a regular overflow filtration system. Zebrafish were maintained in an acrylic aquarium connected to an SSBR or in a tank connected to a regular overflow filtration system for 27 weeks. Water change or aeration using air stone was not performed. The water quality in both groups was within the normal range until 26 weeks, after which the nitrite and nitrate levels increased. The microbial quality of 45 EBN consisting of raw unclean and raw clean Edible-birdnest (EBN) from two different premises were analyzed for 8 weeks. Using 3M™ Petrifilm, Total Aerobic Count (TAC) of raw clean EBN and Total Coliform Count (TCC) for raw unclean, 3 days, 2 weeks and 4 weeks EBN in both premises exceeded the limit stated in by the Malaysian Standard (MS 2334:2011; SIRIM/DVS 3:2015). A total of 264 bacteria and 208 fungi species were isolated. Bacteria isolated was identified using bioMérieux’s API® kit and a series of biochemical tests. Comparison Of Biomechanical Properties Associated With The Diameter And Insertion Depth Of Bone Anchors In The Femoral Condyle Of Toy Breed Cadaver Dogs, Yun-Hee An, Mu-Young Kim, Hun-Young Yoon

The objective of the present study was to compare the biomechanical properties of implanted bone anchors based on diameter and insertion depth to yield selection criteria for bone anchor size in toy breed dogs. Twenty toy breed dog cadavers weighing < 5 kg underwent placement of five types of veterinary bone anchors (short diameter, short length [SDSL]; short diameter, medium length [SDML]; medium diameter, medium length [MDML]; medium diameter, long length [MDLL]; and long diameter, medium length [LDML]) at a predetermined femoral attachment site. Anchor screw depth/femoral condyle width (FCW) and anchor screw diameter/femoral condyle length (FCL) were measured using radiography. The yield load, Young’s modulus and failure load were measured and the causes of failure for each construct were recorded. The anchor screw depth/FCW was < 50%, 50%–75%, ~50%, and 75%–100% in the SDSL, SDML, MDML, and MDLL groups, respectively. The yield load, Young’s modulus and failure load were higher in the SDML and MDLL groups than in the SDSL and MDML groups. The anchor screw diameter/FCL was 12%–15% and 24%–30% in the SDML and LDML groups, respectively. No differences in biomechanical parameters were found between the SDML and LDML groups. The cause of failure in all constructs was pull-out of the bone anchor, except for distal femur fracture in five LDML constructs. In conclusion, when implanting bone anchors in toy breed dogs, the insertion depth should be > 50% of the FCW, regardless of diameter. Moreover, distal femur fracture can occur if the bone anchor diameter/FCL ratio exceeds 24%.

Environmental enrichment methods are meant to encourage animals to display their complete ethogram while under human care in zoos and aquariums. In this study, we sought to characterize the effectiveness of environmental enrichment (involving the inclusion of toys in the living environment) on captive beluga whales in the National Museum of Marine Biology and Aquarium. Our results indicate that belugas with toys spend less time floating and swimming repetitively than do belugas without toys. The presence of toys was also shown to enhance social interactions among captive belugas. Overall, our findings indicate that the provision of toys can enrich the …

To evaluate the effect of P. mirifica supplementation on ovarian activity in crossbred Thai native does, a study was carried out for three consecutive estrus cycle periods: 1) D0-D21(pre-treatment), 2) D21-D42 (treatment) and 3) D42-D63 (post-treatment); D0, D21 and D42 were the days of expected estrus. The aim of this study was to clone, express and identify the truncated S1 gene of nephrotropic infectious bronchitis virus (IBV) and granulocyte-monocyte colony stimulating factor (GM-CSF) of chicken. Firstly, two genes were amplified by polymerase chain reaction (PCR) and cloned into pMD18-T vector. BACKGROUND: Antibiotic use drives antibiotic resistance.
OBJECTIVES: To systematically review the literature and estimate associations between prior exposure to antibiotics across World Health Organization's (WHO) AWaRe categories (Access, Watch, Reserve) and isolation of critical and high-priority multidrug resistant organisms (MDROs) on the WHO priority pathogen list.
DATA SOURCES: Embase, Ovid Medline, Scopus, Cochrane Database of Systematic Reviews, Cochrane Central Register of Controlled Trials, and ClinicalTrials.gov (from inception to 20/08/2020).

Axicabtagene ciloleucel (axi-cel) is a standard-of-care for patients with relapsed or refractory (r/r) large B cell lymphoma who have received 2 or more lines of prior therapy. Excessive substance use and substance use disorders (SUDs) are common, serious and relapsing medical conditions. They frequently co-occur with other diseases that are leading contributors to disability worldwide. While heavy substance use may potentiate the course of some of these illnesses, there is accumulating evidence suggesting common genetic architectures. In this narrative review, we focus on four heritable medical conditions - cardiometabolic disease, chronic pain, depression and COVID-19, which are commonly overlapping with, but not necessarily a direct consequence of, SUDs. We find persuasive evidence of underlying genetic liability that predisposes to both SUDs and chronic pain, depression, and COVID-19. …

OBJECTIVE: Pulmonary hypertension is a lethal disease characterized by pulmonary vascular remodeling and is mediated by abnormal proliferation and migration of pulmonary arterial smooth muscle cells (PASMCs). Platelet-derived growth factor BB (PDGF-BB) is the most potent mitogen for PASMCs and is involved in vascular remodeling in pulmonary hypertension development. Therefore, the objective of our study is to identify novel mechanisms underlying vascular remodeling in pulmonary hypertension.
METHODS: We explored the effects and mechanisms of PTPRD downregulation in PASMCs and PTPRD knockdown rats in pulmonary hypertension induced by hypoxia.
