Metastatic prostate cancer (PCa) in the bone induces bone-forming lesions that contribute to progression and therapy resistance. PCa-induced bone originates from endothelial cells (EC) that have undergone endothelial-to-osteoblast (EC-to-OSB) transition in response to tumor-secreted BMP4. Current strategies targeting PCa-induced bone formation are lacking. Here, we show that activation of retinoic acid receptor (RAR) inhibits EC-to-OSB transition and reduces PCa-induced bone formation. Treatment with palovarotene, an RARγ agonist being tested for heterotopic ossification in fibrodysplasia ossificans progressiva, inhibited EC-to-OSB transition and osteoblast mineralization in vitro and decreased tumor-induced bone formation and tumor growth in several osteogenic PCa models, and similar effects …

Broadly neutralizing antibodies (bNAbs) that target the membrane-proximal external region (MPER) of HIV gp41 envelope, such as 4E10, VRC42.01 and PGZL1, can neutralize > 80 % of viruses. These three MPER-directed monoclonal antibodies share germline antibody genes (IGHV1 - 69 and IGKV3 - 20) and form a bNAb epitope class. Furthermore, convergent evolution within these two lineages towards a 111.2GW111.3 motif in the CDRH3 is known to enhance neutralization potency. We have previously isolated an MPER neutralizing antibody, CAP206 - CH12, that uses these same germline heavy and light chain genes but lacks breadth (neutralizing only 6 % of …

Medical Autopsy For Sudden Unexplained Death In Saudi Arabia: A Call To Action, Wael Alqarawi, Abdulelah Alsaeed, Malak Alghamdi, Ahmed Hersi, Tarek Kashour, Khaldoon Aljerian
Journal of the Saudi Heart Association
Sudden unexplained death (SUD) is a sudden, unexpected, and unexplained death in an individual older than 1 year. It is one of the most devastating and tragic events to families and the community at large, particularly when it happens at a young age. Finding the cause of SUD is extremely important in order to prevent its recurrence in the family, and to help understand the epidemiology of SUD in the community. It has been well-established that the most effective way of finding the cause of SUD is by performing a medical autopsy. In many countries, medical autopsy is mandated in …

Objective: To investigate and compare the impact of pre-heating of an ormocer-based and a methacrylate-based restorative materials at different temperature on intra-pulpal temperature with that at room temperature. Materials and Methods: In the current study, two bulk-fill composite systems; one ormocer-based, and one methacrylate-based were used according to their manufacturer instructions. A total of 30 sound maxillary human molars was selected, and prepared by removal of occlusal enamel and part of dentin till remaining dentin thickness becomes 2 mm above highest pulp horn. The specimens were assigned equally into two halves, according to type restorative material used (n=15 for each). …

Transformative, Noetic, And Transpersonal Experiences During Personal Development Workshops, Helané Wahbeh, Cassandra Vieten, Garret Young, Agnes Cartry-Jacobsen, Dean Radin, Arnaud Delorme
International Journal of Transpersonal Studies Advance Publication Archive
The global personal development market was valued at $38.28 billion in 2019 and is expected to grow an additional 5% from 2020 to 2027. Many of these workshops promise to be transformational. This secondary analysis study examined transformative, transpersonal, and noetic aspects of personal development workshops. We found that 74% of post-survey records endorsed that participants experienced a moment of clarity or profound insight during their workshop. In addition, 66% endorsed that participants had experienced at least one noetic experience, and 84% endorsed at least one transpersonal experience. These analyses provide preliminary evidence for the transformational potential of personal development …

Preparation, Characterization And Pharmacokinetics Of Tolfenamic Acid-Loaded Solid Lipid Nanoparticles, Wei Xu, Zhaoyou Deng, Yifei Xiang, Dujuan Zhu
Faculty and Student Publications
The clinical use of nonsteroidal anti-inflammatory drugs is limited by their poor water solubility, unstable absorption, and low bioavailability. Solid lipid nanoparticles (SLNs) exhibit high biocompatibility and the ability to improve the bioavailability of drugs with low water solubility. Therefore, in this study, a tolfenamic acid solid lipid nanoparticle (TA-SLN) suspension was prepared by a hot melt–emulsification ultrasonication method to improve the sustained release and bioavailability of TA. The encapsulation efficiency (EE), loading capacity (LC), particle size, polydispersity index (PDI), and zeta potential of the TA-SLN suspension were 82.50 ± 0.63%, 25.13 ± 0.28%, 492 ± 6.51 nm, 0.309 ± …

Gunshot Wound To The Chest With Retained Epicardial Bullet., Toba Bolaji, Abuoma C Ekpendu, Frederick Giberson
Transitional Year Resident Research
Gunshot wounds remain the most common cause of penetrating injuries in children and adolescents and the second leading cause of death among youth in the United States. Penetrating cardiac injuries carry a significantly increased mortality rate. The extent of damage caused depends on the type of firearm, the bullet used, the velocityand the trajectory. Therefore, rapid diagnosis and treatment is of the utmost importance. We report a case of a 19-year-old boy who presented to ouremergency department (ED) after sustaining a gunshot wound (GSW) to the right chest. In the ED, the patient was stabilized and a large hematoma was …

Background: Mental health and sleep problems are prevalent in the workforce, corresponding to costly impairment in productivity and increased health care use. Digital mindfulness interventions are efficacious in improving sleep and mental health in the workplace; however, evidence supporting their pragmatic utility, potential for improving productivity, and ability to reduce employer costs is limited.
Objective: This pragmatic, cluster randomized controlled trial aimed to evaluate the experimental effects of implementing a commercially available mindfulness app—Calm—in employees of a large, multisite employer in the United States. Outcomes included mental health (depression, anxiety, and stress), sleep (insomnia and daytime sleepiness), resilience, productivity impairment …

Therapeutic Jurisprudence In Practice: Applying Goffman’S Dramaturgical Approach To Mental Health Courts, Karen A. Snedker
Sociology Faculty Books and Book Chapters
"This chapter draws on qualitative data from two MHCs [mental health courts] to show how team members translate the tenets of therapeutic jurisprudence into the performance of their social roles in both formal and informal court settings. I argue that the successful operation of teamwork lies at the heart of MHC effectiveness. Judges, acting as both audience and team member, can undermine the work of the team. MHCs clearly rely upon a well-functioning collaborative team, working partially in backstage settings, with a judge willing to alter frontstage performances for the sake of collaboration and therapeutic effects for clients."

Background
Acute myocardial dysfunction in patients with sepsis is attributed to oxidative stress, inflammation, and cardiomyocyte loss; however, specific drugs for its prevention are still lacking. Tetrahydrocurcumin (THC) has been proven to contribute to the prevention of various cardiovascular diseases by decreasing oxidative stress and inflammation. This study was performed to investigate the functions and mechanism of action of THC in septic cardiomyopathy.
Methods
After the oral administration of THC (120 mg/kg) for 5 consecutive days, a mouse model of sepsis was established via intraperitoneal lipopolysaccharide (LPS, 10 mg/kg) injection. Following this, cardiac function was assessed, pathological section staining was …
